1.Discussion on pathology experimental teaching reform of medical undergraduate students

The traditional experimental teaching of pathology has defect in training the students' operating ability and mobilizing the initiative of students to observe.Pathology Department of North Sichuan Medical College has developed pathology specimens-making competition for undergraduate students.The students observed,chose and made specimens by themselves after sufficient review of the relevant pathology knowledge.The competition has mobilized the enthusiasm of students effectively and developed their operating ability.This form of teaching should become beneficial supplement to the experimental teaching of pathology.
2.Scanning electron microscopy of Schistosoma nanjingi

Objective To examine the tegmental structure of Schistosoma nanjingi using scanning electron microscopy. Methods Adult schistosomes were obtained from infected rabbits with cercariae shedding from Oncomelania snails, which were infected with miracidia of Schistosoma nanjingi. The adult schistosomes were fixed with 4% glutaradehyde, and then, the samples were prepared with the conventional procedures and the schistosomes were examined with a scanning electron microscope (SX-40). Results There were two types of male and female adult worms. For the big male worm, there were big spines and deep cavity on the surface of middle back and some small spines on the surface of middle abdomen; for the small male worm, there were many small spines on the surface of whole back and abdomen. As the big female worm, there were some small spines on the whole tegumental surface. As the small female worm, there were some small spines on the surface of back and abdomen, but the shape of spines was different between the spines of back and abdomen. On the tail surface of the small female worm, there were two types of spines. The spoke-like acetabulum was found. The sensory organelle papillae with or without cilia were found on the tegmental surface of both male and female woems. Conclusion The tegmental structure of Schistosoma nanjingi is much different from that of Schistosoma japonicum.
3.Clinicopathologic analysis of 3299 patients with thyroid tumors in Nanchong

Objective:To determine the incident tendency and constituent ratio of thyroid tumors, patient age, and histology, as well as present scientific data for the prevention and treatment of thyroid tumors in Nanchong. Methods:Data were collected in the Affili-ated Hospital of North Sichuan Medical College from 2001 to 2015. Microsoft Excel and SPSS 17.0 were used to analyze the data statis-tically. Results:A total of 3299 cases of thyroid tumors were observed for a span of 15 years. The cases of benign tumors were 2503, while those of malignant tumors were 796. Most cases of benign (33.0%) and malignant (27.3%) tumors occurred in the 40-49 age group. At 35 years old and below, the proportion of patients with thyroid malignant tumors was 23.5%. The number of male patients was 566, while that of female patients was 2733, resulting in a male-to-female ratio of 1:4.8. Conclusion:As the incidence of thyroid tumors increase every year, so does the constituent ratios of thyroid malignant tumors. The onset of tumors in men and women were significantly different. These findings should attract more clinicians' and preventive researchers' attention. Targeted detection, preven-tion, and control need to be carried out immediately.
4.Effect of inhibitor HNP1 transfection on tumor growth of human nasopharyngeal carcinoma cell line HNE-1.

OBJECTIVE:
The aim of this study is to investigate the effects of inhibitor HNP1 transfection on proliferation and tumor growth of human nasopharyngeal carcinoma cell line HNE1.
METHOD:
HNE1 cells were transfected with HNP1 by liposome, and the cytotoxic effect was detected by MTT, In vivo tumor growth test was performed in nude mice inoculated with transfected HNE1 cells. The therapeutic effect of HNP1 was evaluated in the inoculated tumors, alpha-defensin 1 expression was detected in implanted tumor tissues by immunohistochemical stain.
RESULT:
HNP1 transfection significantly inhibited the proliferation of HNE1 cells. MTT assay confirmed cytotoxic effect. In vivo study showed tumor volume was significantly smaller in HNP1 transfection group than that in control group (P < 0.01). Immunohistochemical stain showed alpha-defensin expression was increased in HNP1 transfection group.
CONCLUSION
HNP1 transfection can inhibit the proliferation of HNE1 cells, as well as tumor growth.

5.Effects of acupuncture on serotonin, histamine, substance P, and tryptase levels at sensitized points in model rats with knee osteoarthritis

Objective:
To elucidate the differences in manual acupuncture effectiveness at sensitized points by investigating the mechanisms of local skin action at different sensitization points in rats with knee osteoarthritis (KOA).
Methods:
Forty Sprague–Dawley rats were equally divided into control, model (1 mg of monoiodoacetate into the right knee joint cavity), sham operation, manual acupuncture at right Tianjing acupoint (MAR-SJ 10), and left SJ 10 groups. Safranine-O and fast green staining were used to assess the modeling. The morphological and functional changes in mast cells (MCs) were assessed during acupoint sensitization using toluidine blue and immunofluorescence staining. The levels of serotonin, histamine, substance P (SP), and tryptase at skin acupoints and serum levels of IL-β, IL-6, and TNF-α were detected using ELISA.
Results:
After 14 days of treatment, the number of MCs and their degranulation rates were statistically higher in the model group than in the control group (both P < .001). After applying acupuncture, the levels of 5-HT, HA, and SP at skin acupoints were lower than those in the model group (all P < .05), and tryptase level was higher (both P < .05). Tryptase level was higher on the skin at the MAL-SJ 10 acupoint than that on the MAR-SJ 10 acupoint (P = .004). Compared with the model group, the serum levels of IL-1β, IL-6, and TNF-α in the MAR-SJ 10 and MAL-SJ 10 groups were lower (all P < .05).
Conclusion
Acupuncture at KOA-sensitized acupoints mitigates joint injury in KOA rats and may bidirectionally regulate local MCs of these acupoints. This finding not only enhances the reference value of sensitizing points in clinical diagnosis and treatment, but also contributes to the understanding of the biological mechanisms underlying acupuncture intervention at sensitizing points.
