Objective To explore the characteristics of prospective memory ( PM ) deficit in acute stroke patients with cognitive impairment.MethodsSixty patients suffering from acute stroke who met the inclusion criteria in screening with the neurobehavioral cognitive status examination (NCSE) were enrolled into the experimental group. Sixty healthy participants who matched the experimental group in gender distribution and average age and education level were assigned as a control group. All the participants completed several neuropsychological evaluations, including the Chinese version of the Cambridge prospective memory test ( C-CAMPROMT), the Chinese version of the Rivermead behavior memory test (C-RBMT), a digit-span test (DS), the Chinese version of Stroop's word-color test (C-SWCT) and a color trail test (CTT).ResultsTime-based PM (TBPM) scores in the control group were significantly higher than in the experimental group. At the intention initiation stage TBPM scores in the control group were also significantly better than in the experimental group. The TBPM performance of the experimental group was significantly worse than that of the controls when PM performance was compared with other cognitive functions controlled for.ConclusionsThe acute stroke patients with cognitive impairment showed greater TBPM performance deficits than the controls. This may have resulted from impairment at the intention initiation stage. TBPM deficits may exist independently. If so, they could serve as an assessment of cognitive impairment after stroke.

Objective To investigate the effect of photochemical gene transfection on colon cancer cells mediated by polycation. Methods Based on SW480 and HT29 colon carcinoma cell culture, the intracellular localization of meso-tetraphenylporphine disulfonate(TPPS_(2a)) were observed by laser confocal scanning microscopy(LCSM).Photochemically induced cytotoxicity of TPPS_(2a) on SW480 and HT29 cell lines was evaluated by MTT assay.By flow cytometry, the light-induced transfection of EGFP plasmids mediated by PEI was studied. Results TPPS_(2a) exhibited typical vesicular localization in two colon carcinoma cell lines. The photochemical cytotoxicity based on TPPS_(2a) increased with light dose. The light dose of IC_(50) was 22 mW/cm~(2) for 6 min for SW480 cells and for 12 min for HT29 cells(wavelength: 405 nm).The light treatment based on TPPS_(2a) induced an increase in transfection efficiency to(29.61?1.15) for SW480 cells and(23.47?0.62) for HT29 cells(P

Objective To study the synergistic effects of Mitomycin C in photodynamic therapy for colonic carcinoma. Methods The colon carcinoma cell line SW800 was assigned to be treated with 002 ?g/ml Mitomycin C, or 2 ?g/ml PSD-007 (photosensitizer), or Mitomycin C and PSD-007 (PSD+MC group). The cells without any special treatment served as control. Fluorescence spectrophotometer and MTT assay were used to detect the content of intracellular photosensitizer and survival rate of SW700 cells under photodynamic therapy. The obtained data was analyzed with SPLM statistical software. Results After PSD was added for 4, 8, 16 and 24 h, intracellular PSD level of PSD+MC group increased significantly as compared with that of PSD group. The intracellular PSD contents of PSD group, PSD+MC group had a strong and negative correlation with cell survival rate respectively after photodynamic therapy, but the cell survival rate curve shifted to the left obviously. The best killing effects on SW700 cells of photodynamic therapy could be obtained when the SW700 cells had been exposed to photosensitizer for 8-16 h. Conclusion There is insignificant influence of lowconcentration of Mitomycin C alone on cell survival rate. The better synergistic effect of Mitomycin C on killing the colon carcimoma cells can be achieved by enhancing the intracellular content of photosensitizer for 6-8 h before photodynamic therapy.

Objective To make better use of the antitumor effects of TNF related apopotosis inducing ligand (TRAIL) ,we con-struct a new fusion protein including TRAIL ,for the sake of improving its purity and targeting to hepatocytes .Methods Fusion protein concluding His label protein and human recombinant TRAIL which linked by constructed specifically u-PA cleavage site . TRAIL protein was obtained by disintigrating from the fusion protein which had been purified .Results The purified His-rh-sTRAIL was demonstrated to be cleavable by u-PA and possess enhanced antitumor effect ,it′s toxicity to hepatocytes was reduced . Conclusion The fusion protein His-rhsTRAIL has antitumor effect on colon cancer cell with high efficiency ,highly targeting and low-toxicity .

Objective To evaluate the reliability and criteria validity of the Chinese Cambridge Prospective Memory Test (C-CAMPROMPT) for testing Chinese patients with acquired brain injury. Methods The CAMPROMPT was translated into Chinese. The translated Version was then used with 30 'normal persons' and 39 with acquired brain injury and memory problems. Correlation analysis and Cronbach's a were used to evaluate reliability and criteria validity of the C-CAMPROMPT. Results Correlation and Cronbach's a were in the range 0.697 to0.951.The total and event-based scores were positively corrlated with all three field Chinese Version of Rivermead Behavior Memory Test scores. Time-based C-CAMPROMPT scores, however, were negatively correlated with Chinese Version of the Stroop Word-Color Test results. Conclusion The reliability and validity of the C-CAMPROMPT were high enough to be used to test the prospective memory of acquired brain injury patients in the clinic.

Objective To examine the inter- rater reliablity of Myoton-3 Myometer in assessment of muscle tone in healthy subjects. Methods 20 healthy volunteers were assessed their muscle tone of biceps brachii and flexor carpi radialis muscles in relaxed state with Myoton-3 by 2 testers within 24 h. The frequency of damping oscillations (F value) measured by Myoton-3 was as the characteristic of the muscletone. The intraclass correlation coefficient (ICC) and Bland-Altman analysis were performed. Results The ICC was 0.72~0.88 and 0.79~0.89 in triple scan and ten-time scan pattern, respectively. The Bland-Altman analysis revealed no systematic errors between testers. ConclusionThe Myoton-3 Myometer is reliable between testers for measuring the muscle tone in healthy adults.

Objective To evaluate the swallowing function of patients with dysphagia after brainstem injury using manofluorography (MFG),analyzing the abnormal biomechanical and kinematic parameters as well as any correlation between changes in the pharynx and the upper esophageal sphincter (UES) measured manometrically and changes in the kinematics of the hyoid bone. Methods Thirteen patients with dysphagia after brainstem injury (the patient group) and 13 healthy participants (the control group) underwent manofluorography.Kinematics and biomechanical changes during swallowing were compared between the two groups and the correlations between the observations were analyzed. Results The patient group showed significantly lower maximum pressure and rate of pressure change at the base of the tongue and in the hypopharynx,as well as less hyoid anterior displacement, smaller and briefer UES opening, but significantly higher minimum pressure at UES relaxation. The duration of tongue root elevation and hypopharynx pressure was also shorter than in the control group,on average. There was a negative correlation between hyoid anterior displacement and the minimum pressure on UES relaxation in the control group, and a positive correlation between hyoid anterior displacement and the maximum pressure at the base of the tongue and in the hypopharynx in the patient group. Conclusions The concurrent use of manometry and video-fluorography for evaluating dysphagia can be an objective and effective diagnostic tool for the comprehensive evaluation of swallowing function.
