1.Preparation of transferrin and RGD co-modified PLGA nanoparticles and its targeting to melanoma

Objective To prepare transferrin and Arg-Gly-Asp polypeptide co-modified nanoparticles(TF/RGD-NPs)and evaluate its targeting efficiency to melanoma.Methods The co-modified nanoparticles were prepared by emulsion method and its appearance,particle size and Zeta potential were evaluated.The cellular uptake experiment and melanoma tumor spheroids penetration test were used to evaluate the affinity and ability to penetrate tumor tissues of TF/RGD-NPs to melanoma B16 cells. Results The particle diameter of co-modified nanoparticles was(113.4 ±12.5)nm and the Zeta potential was(4.53 ±2.15)mV.In vitro uptake test demonstrated that the efficacy of cellular uptaken TF/RGD-NPs by B16 cells were 2.7 times and 2.9 times to TF-NPs and RGD-NPs,respectively,the differences were all significant(P<0.05 ).Tumor spheroid penetration test results showed that TF/RGD-NPs has good affinity to melanoma cells.Conclusion TF/RGD-NPs can target to melanoma B16 cell efficiency in vitro,it may be serve as a potential drug delivery system for targeting melanoma.
2.Effect of embelin and doxorubicin on the proliferation of MCF-7 cell and growth of MCF-7 bearing nude mice

Objective To explore the effect of X-linked inhibitor of apoptosis protein(XIAP)combined with doxorubicin on the proliferation of MCF-7 cell and growth of MCF-7 bearing nude mice.Methods The anti-proliferation efficiency of doxorubicin and embelin were determined by MTT assay.MCF-7 cell apoptosis induced by embelin and doxorubicin were detected by Flow cytometry.Anti-tumor ability of embelin and doxorubicin were evalued with tumor spheroids test.MCF-7 cell were xenografted to mice to establish the animal model,which was used to evaluate the effect of anti-tumor. Results Compared with saline group,embelin group and doxorubicin group,the combination(doxorubicin +embelin)group inhibited the growth of MCF-7 cells effectively(P<0.01).The combination group induced the apoptosis of MCF-7 cells more effectively than doxorubicin alone(P<0.01), and significantly inhibit the growth of tumor in vitro and in vivo than other groups(P<0.01).Conclusion The combination of doxorubicin and embelin may be used as a potentially effective treatment method for breast cancer.
3.Studies on analgesic effect and toxicity of Weifuqitong Capsule

AIM: To study the analgesic effect and toxicity of Weifu qitong Capsule (Rhizoma Corydalis, cortex Magnoliae Officinalis, Radix Aucklandiae, etc.) to provide experimental data for its clinical application. METHODS: The analgesic effect of Weifu qitong Capsule was assessed by hot-plate test and writhing test, its acute and subacute toxicity studies were also conducted. RESULTS: Taking high dose of Weifu qitong Capsule, after 4.5 h (6.249 g/kg) WFQC could increase pain threshold in hot-plate test. 4.Significance of dynamic detection of free DNA in hypertensive cerebral hemorrhage

Objective To study the relationship between the dynamic change of free DNA in cerebrospinal fluid (CSF)and the tissue injury in the patients with hypertensive intracerebral hemorrhage.Methods 54 cases of hypertensive intracerebral hemor-rhage were divided into 3 groups according to the glasgow coma scale(including 17 mild cases,21 moderate cases and 16 severe ca-ses).2 mL of CSF was collected for extracting free DNA on 1,3,7,15 d after stroke onset.The free DNA level was measured by the fluorescent real-time PCR.Results The free DNA level in the severe group was significantly higher than that in the mild and mod-erate groups.The free DNA level in the abnormal intracranial pressure group was higher than that in the normal intracranial pres-sure group;the intracranial infection group was higher than the non-infection group.Conclusion The free DNA level has certain value for diagnosing the craniocerebral injury in hypertensive intracerebral hemorrhage and is conducive to monitor the occurrence of postoperative complications.
5.Analysis and Diagnosis on Backhand Twist Technique of Leading Men’s Player of National Table Tennis Team

Objective To make analysis and diagnosis on backhand twist technique used by player A, who is the leading men’s player of national table tennis team, so as to provide references for improving his backhand twist technique. Methods The three-dimensional kinematics test and analysis were used. The backhand twist techniques of player A and those of player B who has good backhand twist techniques were compared by quantitative data and picture analysis. Results At the stage of swinging racket backward, the racket swing amplitude, shoulder angle and wrist angle of player A were significantly smaller than those of player B. The roll angle of trunk of player A was significantly larger than that of player B. At the stage of swinging and hitting the ball, the shoulder angle, elbow angle of player A were significantly bigger than those of player B, while the increasing amplitude of shoulder angle and elbow angle, as well as the changing amplitude in roll angle of trunk of Lin Guoyuan were significantly smaller than those of player B. At the stage of swinging racket forward, the shoulder angle and elbow angle of player A were significantly bigger than those of player B. The increasing amplitude of shoulder angle and elbow angle, as well as the roll angle of trunk and its changing amplitude of player A were significantly smaller than those of player B. Conclusions The racket swing amplitude of player A was smaller, and the distance between the racket and the ball of player A was close at the end of swinging racket backward stage. During swinging and hitting the ball stage, the hitting point was far from the body, the shoulder joint was not stable enough to support, so that the wrist was used more. The center of gravity was not enough to force forward, and the outburst power was not concentrated. At swinging racket forward stage, player A’s braking was not active enough, which affected the stability of hitting the ball. At hitting the ball stage, the torsion of the trunk was smaller, and the waist power was not concentrated. On the basis of unaffecting the forehand outburst power, player A should slightly adjust his backhand twisting technique, or appropriately increase the the racket swing amplitude and torsion of the body. In the process of hitting the ball, the sequence of outburst power was the waist, the forearm and the wrist.
