Objective To investigate the relationship between rules of local metastasis and the operation for rectal cancer. Methods Literatures about rules of local metastasis and operation for rectal cancer were reviewed and analysed. Results The study of rules of local metastasis results in the change and refinement of the operation for rectal cancer. Conclusion With the development of basic research for rectal cancer, the operation for rectal cancer is changing and refining, and the result of the treatment is more favourable.

Objective To investigate the role of T cell factor-4 (TCF-4) in the carcinogenesis of colorectal cancer. Methods Relevant references about TCF-4 and the carcinogenesis of colorectal cancer, which were published recently domestic and abroad, were collected and reviewed. Results For TCF-4 gene, multiple isoforms are generated by way of alternative splicing, which encode different proteins. TCF-4 protein is sequence-specific DNA binding protein and is incapable of activating or repressing transcription independently, but it can interact with distinct partners to lead to different effects through multiple domains. Conclusion TCF-4 might be viewed as nuclear vehicles targeting other auxiliary proteins to a specific set of promoters and functions as molecular switch during the carcinogenesis of colorectal cancer.

Objective To study the current research status of minichromosome maintenance protein 2 (MCM2), the cell cycle proliferation marker, in the diagnosis of colorectal carcinoma. Methods Literatures about the application of MCM2 in the study of colorectal carcinoma were collected and reviewed. Results MCM2, as a marker of cell dysplasia and malignancy, was usually used in the study of carcinoma. The study on expression of MCM2 in the cell of colorectum in different proliferational stage might help to screen colorectal carcinoma as early as possible. Conclusion As a relatively specific and sensitive marker of cell proliferation, MCM2 might become a promising mark for diagnosing colorectal carcinoma in the early stage.

Bone marrow-derived mesenchymal stem cell (BMSC) transplantation is one ot the most popular therapeutic measures in severe acute pancreatitis (SAP). However, technical challenges and ethical concern have hindered its clinical application. Paracrine factor, as a new safe and easy handing therapeutic measure, can work comparably effective as BMSC transplantation in SAP therapy, but bio-safe risks could be greatly reduced. In this paper, we reviewed the therapeutic effect and potential mechanism of paracrine factors in the treatment of SAP. The injection of paracrine factors yielded from cultured cell suspension will be a new cell therapeutic measure for SAP.

Journal of Clinical Pediatrics 2015;(4):326-329
ObjectiveTo explore the roles of procalcitonin in differentiating bacterial infection from other inlfammatory processes and identifying bacterial species.MethodsThe blood culture results of 253 hospitalized children diagnosed with sepsis from January 2010 to June 2014 were analyzed retrospectively and its relationship with PCT was also analyzed. ResultsIn 253 children with sepsis, the blood culture results of 124 children showed positive (49.01%). There were 71 (28.06%) cases of Gram-positive infection and 53 (20.95%) cases of Gram-negative infection. The median level of PCT in 253 patients was 1.6 (0.4-4.5) ng/ml. According to the PCT levels, all patients were divided into low level group (0.05-0.5 ng/ml,n=86), middle level group (0.5-2.0 ng/ml,n=55), high level group (2.0-10 ng/ml,n=75) and extremely high level group (10-300 ng/ml,n=37). The positive rates of bacterial culture were signiifcantly different among different levels of PCT (χ2=69.14,P<0.01). In low lev-el group the positive rate of bacterial culture was the lowest and in other three groups the positive rate was all above 60%. The detection rate of Gram-positive and Gram-negative bacteria was signiifcantly different among different levels of PCT (P<0.01). In high level group the detection rate of Gram-positive bacteria was the highest while in extremely high level group the rate of Gram-negative bacteria was the highest.ConclusionsThe level of PCT is helpful for the differentiation of bacterial infection from other inlfammatory processes in children with sepsis and abnormally elevated PCT level may indicates Gram-negative infection.

Objective To establish a mouse model of acute necrotizing pancreatitis. Methods Thirty six male ICR mice were randomly divided into control group ( n =6) and experimental group ( n =30). Each of the animals in the experimental group received 7 intraperitoneal injections of caerulein (50 ?g/kg body weight) in 0.9% NaCl at hourly intervals over 6 hours. The animals in the experimental group were killed at 9,18,24,48 and 72 hours respectively after the first caerulein injection. The control animals received the same volume of 0.9% NaCl without caerulein. The animals in the control group were killed at the 18th hour after the first intraperitoneal injection. The severity of acute necrotizing pancreatitis was evaluated in terms of amylase level, pancreatic weight/body weight and the histological changes. Variance analysis was employed in the processing of these data. Results Both amylase level and pancreatic weight elevated 9 hours after the first caerulein injection, and correlated with the course of pancreatitis. The maximums of both alterations were observed at the same time point (18 hours after the first injection of caerulein). Prominent interstitial inflammation and acinar cell necrosis occurred at the 18th hour, and the histological score for pancreatitis reached a maximum ( P

Objective To study the prognostic value of circumferential resection margin(CRM)status. Methods Specimens of 62 patients with rectal cancer, who underwent total mesorectal excision from December,2001 to June,2002,were examined using whole-mount sections. All patients were followed up for 4~60 months. Results Circumferential resection margin involvement(CRMI)was identified in 12 specimens.while only 4 of them were cancer positive in the resection margin.Analyzed by logistic regression test, the occurrence of CRMI was related to tumor differentiation [ P=0.015,Exp(B)=0.131]and lymph node metastases[P=0.013,Exp(B)=7.488 ],but not tumor distance to anal verge(P=0.246).The difference in local recurrence rate, overall recurrence rate,disease-free survival rate and overall survival rate between CRMI and that of CRM negative, were significant(P=0.002,0.000,0.000 and 0.003,respectively). Conclusions CRMI is an accurate prognostic factor of recurrence and postoperative survival, and its occurrence is mainly determined by biologic characteristics of the tumor.TME could decrease the incidence of CRMI.

Objective To investigate the effects of endothelin-1 on the proliferation of HCT-116 cells. Methods The expression of endothelin-1 and endothelin receptors A and B in the HCT-116 cells was detected by RT-PCR and immunohistochemistry method. The effects of endothelin-1 at different concentrations and action time points on HCT-116 cells were measured by MTT assay and were analyzed by variance analysis. Results The expression of endothelin-1 and endothelin receptors A and B at the gene and protein levels in HCT-116 cells was detected.Endothelin-1 had significant effects on the proliferation of HCT-116 cells. The proliferation peak of HCT-116 cells was reached when the concentration of endothelin-1 was 1×10-7mol/L and the action time was 48 hours. Conclusions Endothelin-1 promotes proliferation of HCT-116 cells in a dose-and time-dependent manner, and the process is mediated by endothelin receptors.
