1.THE DETERMINATION AND CONTENTS OF LUTEIN,ZEAXANTHIN AND ?-CAROTENE IN VEGETABLES IN BEIJING REGION

Objective To determine the contents of lutein,zeaxanthin and ?-carotene in 31 vegetables.Method High performance liquid chromatography method with Develosil C30 column and ultraviolet detector was used to detect lutein,zeaxanthin and ?-carotene at wavelength of 480nm.A gradient elution method with elution A (acetonitrile:methol=75:25) and B (methyl-tert-butyl-ether) was used to separate the carotenoids in both raw and cooked vegetables.Results The retention time of lutein,zeaxanthin and ?-carotene was 10.355,10.732 and 20.789 min respectively.The contents of lutein,zeaxanthin and ?-carotene were linearly related with the peak area.Analysis showed that dark leafy vegetable such as amaranth,Chinese celery leaf,parsley,spinach and pakchoi petiole contained the highest amount of lutein.Zeaxanthin was detected only in dehydrated daylily.There was little or no loss of lutein,zeaxanthin and ?-carotene but an average increase in lutein and ?-carotene in vegetables after cooking.Conclusion Vegetables,especially dark leafy vegetables are major sources of dietary lutein,and ?-carotene.Consuming cooked vegetables tends to increase the intakes of lutein,zeaxanthin and their bioavailability.
2.Clinical features and CYBB gene mutation analysis of chronic granulomatous disease in 6 neonate infants

Objective To determine the clinical features and gene expression of neonatal chronic granulomatous disease(CGD) in newborns,in order to provide the evidence for early diagnosis,treatment and infection prophylaxis and so as to improve prognosis.Methods The clinical data of 6 neonates with CGD who hospitalized in Beijing Children's Hospital,Capital University of Medical Sciences,from Jan.to Dec.2013 were retrospectively analyzed,to summarize their clinical features and gene expression.Results All patients were male,mean age of onset in 6 patients was 19 days old.Six patients were onset with cough and fever,3 cases with skin abscesses.All patients had special lung images and their respiratory burst test were all positive.Six patients were all X-linked recessive heredity with mutation in CYBB gene,4 cases carried missense mutation,1 case carried frameshit mutation,1 case had shear point disappear.All patients were improved during hospitalization with anti-infective therapy including antifungal and antibiotic drugs.After their discharge,the patients were followed up for 1 year,among them 5 patients taking antibiotics according to doctor's advise hadn't got any severe infections or other complications,1 patient did not receive antibiotics according to doctor's advise,caught respiratory infection was occured once per 1-2 months and died of multiple organ failure in 5 months due to severe infection.Conclusions For newborn with respiratory symptoms such as cough and fever,with apparently nodular and massive radiologic infiltrates,CGD cases should be highly suspected.Long term prognosis is good in children with prophylactic medication and they may have a longer infection interval.
3.The Study of Hypertension and its Related Factors among Mongolian Residents in Hulunbeier Region

Objective To investigate the prevalence of hypertension and its related factors in Hulunbeier Mongolian region. Method A stratified multistage sampling was used to select residents aged 18 and over in a town of Hulunbeier region. 453 persons served as subjects for this questionnaire survey. Their blood pressure, height, weight, waist circumference were measured and body mass index (BMI) were calculated. Then an analysis of the prevalence of hypertension and its related risk factors were performed. Results The hypertension prevalence rate was 34.5%, the standardized rate was 45.4%, and the prevalence in men (48.9%) was significantly higher than that in women (25.4%) (P50 (OR=11.312, 95%CI 4.309~22.132), BMI (2.255, 1.180~4.311), gender (OR=2.788, 1.461~5.323) and drinking (2.306, 1.287~4.134) were significantly related to hypertension. Conclusion The standardized prevalence of hypertension in Hulunbeier Mongolian region is higher than the average prevalence in China, 2002. Old age, abdominal obesity, overweight or obesity, male and alcohol drinking were the main risk factors for hypertension in this region. The government and the health sector should strengthen the health education of healthy lifestyle and health promotion and the control.
4.Effect of heating in a calorstat to the oxygen-carrying function of red blood cell

Objective To study the change of the oxygen-carried function of stored red blood cells,which was warmed in room temperature and calorstat.Methods Randomly removed from 4 ℃storage refrigerator,red blood cell suspension were put in room temperature 30 min (group N)and 37℃ calorstat 30 min (group H),and samples taken to examine electrolyte,blood gas,free Hb,ATP content,2,3-DPG content,oxygen affinity(P50).Results In two groups,blood pH value and blood Na+ did not change obviously due to heating.The content of blood K+ and free Hb increased,glucose fell,but there was no statistically significant difference.The content of ATP and 2,3-DPG in eryth-rocyte increased and P50 increased after heating (P <0.05),which increased more significantly in group H compared with group N (P<0.05).Conclusion Heating stored blood up to 37 ℃ can in-crease the oxygen-carried function of red blood cells,which contribute to improve the effectiveness of transfusion.
5.Role of MMP2/TIMP1 in the vascular remodeling after angioplasty in rabbits

AIM: To discuss the effects of matric metalloproteinasez and tissue inhibitor 1 (MMP2/TIMP1) on the vascular remodeling after angioplasty and the regulatory effect of tongxinluo on it in rabbits. METHODS: 65 male white rabbits (2.5-3.5 kg; 6-8 months) were divided into 4 groups: control group (n=5, normal diet ), injury group (n=20, normal diet plus intimal injury), high-lipid group (n=20, high cholesterol diet plus intimal injury) and tongxinluo group (n=20, tongxinluo 0.5 mg?kg~-1?d~-1, 4 weeks, also high cholesterol diet plus intimal injury). Under X-ray the narrow parts of the vessel underwent angioplasty. 6.Effect of microRNA-101 on apoptosis of rabbit condylar cartilage cells by inhibiting target gene SOX9

Objective:To explore the effect of microRNA-101 on apoptosis of condylar cartilage cells and the specific mechanism of molecular biology. Methods: IL-1 was used to stimulate and establish the model of apoptosis of condylar cartilage cells. The expression change of miR-101 in control group was compared with that in IL-1 stimulation group by qRT-PCR. Overexpression and down-regulation models of miR-101 were established by transfecting Mimics and Inhibitor and verified by qRT-PCR. Flow cytometry was used to detect the effect of miR-101 overexpression and down-regulation on apoptosis. Target gene of miR-101 was analyzed and calculated through bioinformatics. Western blot and Luciferase report assay were used to detect whether Sox9 could become the target gene of miR-101. Results:qRT-PCR results showed that IL-1 stimulation could cause the increase of miR-101 expression. After the transfection of rabbit condylar cartilage cells by Mimics and Inhibitor, qRT-PCR results confirmed the significant effect of miR-101 overexpression and down-regulation. It was confirmed by flow cytometry that overexpression of miR-101 could promote the apoptosis of condylar cartilage cells, and down-regulation of miR-101 could reduce the apoptosis. It was confirmed by Western blot and Luciferase report assay that Sox9 was the target gene of miR-101, and miR-101 inhibited SOX9 expression through complementary pairing with 3’UTR of Sox9 mRNA. Conclusions:miR-101 can promote the apoptosis of condylar cartilage cells through inhibiting the protein level of target gene SOX9.
8.Prevalence of hypertension among urban residents in Mongolian region of Hulunbeier

Objective To investigate the prevalence of hypertension among adult residents in Mongolian region of Hulunbeier. Methods A total of 1536 adult residents were selected by using cluster sampling method, and 1496 completed the questionnaire survey. Blood pressure was measured for each participant. Results The prevalence of hypertension was 31.1%. Higher prevalence of hypertension was found in male ( 42. 0% vs 24. 5%, P < 0. 05 ) and the elderly. No statistically significant difference of prevalence of hypertension was investigated in different nationalities (P > O. 05 ). Conclusion The prevalence of hypertension among adult residents in Mongolian region of Hulunbeier is relatively higher.
9.The structure characteristics of prophages of foodborne Enterococcus hirae R17 and their interaction relationships with host bacterium

Objective This study was to understand the structure characteristics of prophages in the genome of Enterococcus hirae R17,and also to analyze their interaction relationships with the host bacterium.Methods The gene distribution and gene encoding characteristics of prophages in the genome of Enterococcus hirae R17 were identified using the PHAST software.The virulence gene,antimicrobial resistance genes,and environmental resistance genes in the prophages were also analyzed.Results Three prophages were found on the chromosome of Enterococcus hirae,including two incomplete prophage elements (Prophage-1 and Prophage-2) and one complete prophage (Prophage-3).Some function genes of bacteria were found in the sequence of three prophages,including nucleotide transportation and metabolism related genes.One incomplete prophage carrying erythromycin-and bacitracin-resistance genes was identified in the plasmid,which suggested that prophage induced gene horizontal transfer caused erythromycin-and bacitracin-resistance of Enterococcus hirae R17.Conclusion This study laid a solid foundation for the diversity analysis of prophages of Enterococcus hirae.Prophages played an important role in promotion of antimicrobial resistance of enterococci.Scientists should pay more attention to the spread of antimicrobial resistance and pathogenicity induced by prophages.

Endothelial dysfunction was closely related with AS , NO bioavailability ( production and utilization of endothelial NO ) was decreased by oxidative stress , lipid infiltration , inflammatory factor expression , vascular tone alteration and so on , which play an important role in endothelial dysfunction .Enhanced arginine , activityand asym-metric dimethylarginine together with increased hyperhomocysteinemia all promote AS by intervening NO bioavail -ability.Diabetes mellitus, obesity, chronic kidney disease , smoking and so on also involved in AS via influencing NO bioavailability and NO level .
