Objective To observe the effect of low frequency vibration (LFV) on bone metabolism, bone mass density (BMD), bone histomorphometric and biomechanical characterization in rats with secondary osteoporosis (OP) after spinal cord injury (SCI).MethodsSCI model was established with 50 SD rats by transecting spinal cord completely at the level of tenth thoracic vertebra, and the animals were randomly divided into 5 groups: SCI 6-week control group, SCI 12-week control group, vibration 6-week (Vi 6w) group, vibration 12-week (Vi 12w) group and vibration 6-12 week (Vi 6-12w) group. All rats of vibration groups accepted LFV (frequency=20 Hz, acceleration=0.15 g) with 10 minutes once, twice per day, and 6 days per week. The LFV was given from the forth day postoperation to the end of the sixth week in Vi 6w group, and to the end of twelfth week in Vi 12w group, while from the seventh week postoperation to the end of twelfth week in Vi 6-12w group. All animals were sacrificed when LFV test finishied. The serum alkaline phosphatase (ALP), bone glaprotein (BGP) and serum calcium (Ca2+)/BMD of femur and the fifth lumbar vertebra (L5)/bone histomorphometric characterization of tibia/bone biomechanical characterization of femur and L5 were measured.ResultsCompared with control groups, the serum Ca2+ and BMD of promixal femur improved significantly in rats of Vi 6w group ( P<0.05), while the maximal load of L5 increased significantly in rats of Vi 12 group ( P<0.05). There were no changes in rats of Vi 6-12w group ( P>0.05).ConclusionThe early treatment of LFV may improve the secondary OP of rats with SCI, but the effect is different in different sites of rats' body. The late treatment of LFV can't improve the secondary OP, but may keep rats with SCI from deteriorating.

BACKGROUND:In recent years, a lot of studies have reported the feasibility and efficacy of transplanting seed celsvia tissue-engineered cardiac patches into specific infarcted myocardial area, helping regeneration of damaged heart tissue and improving heart functions.
OBJECTIVE:To review the methods and strategies of fabricating tissue-engineered cardiac patches and to analyze the structure and functional characteristics of cardiac patches prepared by different methods.
METHODS: The first author retrieved relevant articles about the fabrication methods of tissue engineered cardiac patches by searching PubMed and Web of science data bases. The key words were “cardiac infarction, cel death”.
RESULTS AND CONCLUSION:The fabrication methods and strategies of tissue-engineered cardiac patches are as folows: (1) obtaining multi-layer cel sheets by overlapping monolayer cel sheets; (2) combing gelatin with cels to construct multi-layer cel sheets; (3) seeding cels on scaffolds; (4) homogeneous arrangement of cels. Recent studies about tissue-engineered cardiac patches have demonstrated that the biological activities of seed cels, such as retention, survival and paracrine effects, greatly influence the effect of cardiac patches on the repair and regeneration of infarcted myocardial tissues. Temperature-sensitive method is the most mature and wide-spread used constructing technique; besides, other strategies also successfuly improve the biological activities of seed cels to different extents, and they also increase the mechanical strength and thickness of cel sheets in order to have better operability. Tissue-engineered cardiac patches are usualy prepared by different combined strategies on purpose to obtain better biological and mechanical properties.

[ ABSTRACT] AIM:To investigate the role of chronic psychological stress on periodontitis and the effects of hy-perbaric oxygen ( HBO) on periodontitis with psychological stress in rats.METHODS:Male special pathogen-free Wistar rats (n=80) were randomly divided into 4 groups:(1)normal control group;(2)experimental periodontitis group:the pe-riodontitis model was induced by wrapping 3/0 silk ligature inoculated with Porphyromonas gingivalis around the left maxil-lary second molar of the rats; ( 3 ) psychological stress stimulation group; ( 4 ) periodontitis model with stress stimulation group.Psychological stress was removed at the 9th week after ligature, and 4 rats from each experimental group were ran-domly chosen for HBO treatment. The rats were sacrificed at the 2nd, 4th, 8th and 10th weeks after ligature. The levels of blood glucose, adrenocorticotropic hormone ( ACTH) , corticosterone and adrenaline were measured as the stress markers. The histological changes of periodontal tissues were observed under microscope with HE staining.RESULTS:The levels of blood glucose, ACTH, corticosterone and adrenaline in psychological stress stimulation group and periodontitis with stress group were significantly higher than those in control group and experimental periodontitis group at the 2nd and 4th weeks af-ter ligature (P<0.05).The levels of the stress markers were significantly lower than those in untreated groups in the 10th week after HBO (P<0.01).The sites of gingival attachment were normal in control group and psychological stress stimu-lation group.Periodontal pocket, and periodontal attachment loss ( AL) were observed in experimental periodontitis group. The tissue damage was much heavier in periodontitis model with stress stimulation group as the furcation of tooth was ex-posed and the tissue damage was observed on both sides of the adjacent teeth. No significant difference of AL between psy-chological stress stimulation group and normal control group during the experiment was observed. The AL in periodontal model with stress stimulating group was significantly higher than that in experimental periodontitis group at the 2nd, 4th and 8th weeks (P<0.01).The level of AL was attenuated at the 10th week after HBO (P<0.01).No difference of histologi-cal change in periodontal tissues was observed between control group and psychological stress stimulation group.Severer in-flammatory changes and alveolar bone destruction were observed in periodontitis with stress group than those in experimental periodontitis group. The levels of inflammation reduced at the 10th week after HBO.CONCLUSION:Stress stimulation is one of the inducing factors of periodontitis in rats, which aggravates periodontitis.HBO may represent a useful way in trea-ting psychological stress periodontitis.

Objective To get the maximum information from computed radiography (CR) output images by changing post-processing parameters. Methods Six experimental photos of polypropylene balls of 2.0 mm in diameter were taken by means of one time X-ray exposing on an imaging plate (IP) with different post-processing parameters including rotation amount (GA), gray gradation type (GT), rotation center (GC), shifting amount (GS), frequency rank (RN), frequency type (RT), and degree of enhancement (RE). 6 photos were viewed by three students and one radiologist on a 6 000 lx illuminance viewbox. Receiver operating characteristic (ROC) curves were made by means of 5-value-differentiation method. Results The largest mean area value (Az) below ROC curves of a low contrast experimental photo with post-processing parameters GA=1.0,GT=A, GC=1.6, GS=0.3, RN=4.0, RT=R and RE=3.0 was 0.96, and the maximum information was obtained. The smallest mean area value (Az) was 0.78 with changed post-processing parameters GA=0.8, GS=-0.2 and RE=0.5 while other parameters were not changed. The minimum information was obtained from this photo. Conclusion In order to get the maximum information from a CR output image, the post-processing parameters should be suitably selected.

ObjectivesTo study the relationship between the allele frequencies and genotype distribution of transforming growth factor-beta 1(TGF-β1) gene promoter polymorphisms in Chinese patients with heptocellular carcinoma(HCC),and to analyze the association of the serum levels and genotype of TGF-β1 with HCC.MethodsThe polymorphisms of TGF-β gene, including polymorphisms of TGF-[β1 gene -800G/A、-509C/T,were analyzed by the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP)methods in 102 patients with HCC and 110 healthy controls, and the serum level of TGF-β1 was determined by enzyme-linked immunosorbent assay (ELISA). ResultsThe HCC group showed significantly higher serum levels of TGF-β1 than control group [(51.06 ± 9.74)μg/L,(22.12 ± 8.67 )μg/L,t=22.884, P<0.01], The distributions of TGF-β gene -800G/A polymorphisms were not different significantly between HCC group and control group, but TGF-β1 -509C/T gene polymorphism was significantly different. The relative risk suffered from HCC of C allele was 1.822 times of the T allele (OR=1.822,95 %CI:1.238-2.682,t=22.884,P<0.01), the serum level of TGF-β1 T allele carriers was significantly higher than that of no carriers [(53.52:±:10.07)μg/L,(43.57±9.89)μ.g/L,t=3.898, P<0.01]. ConclusionTGF-β1 gene -509C/T polymorphism is associated with HCC, and T allele may be a risk factor for HCC, in which the TGF-β1 T allele carriers may have increased risk by enhancing the TGF-β1 expression in the pathogenesis of HCC.
