1.The effect of ulinastatin on NT-proB-NP and Picco in patients with severe sepsis

Objective To observe the effect of ulinastatin on NT -proB -NP and Picco in patients with severe sepsis.Methods 50 patients with severe sepsis were randomly divided into the control group and observation group,25 patients in each group.The control group received the routine treatment,and the observation group received ulinastatin additionally.The venous blood was collected to determine cTn -I,NT -proBNP,TNF -αand Lac before treatment and 7 days after treatment.The APACHE Ⅱ and CFI were observed before treatment and 7 days after treat-ment.Results The differences of CFI,cTnI,NT -proBNP in the two groups before treatment were not significant (t =0.419,0.321,0.335,all P >0.05),after treatment,cTnI and NT -proBNP decreased more significantly(t =2.976,2.910,all P <0.05),while lifting more apparent CFI[(3.98 ±0.95)vs.(2.39 ±0.85),t =6.237,P <0.05]in the observation group than in the control group.The differences of TNF -αand Lac in the two groups before treatment were not significant (t =0.602,0.848,all P >0.05 ),after treatment,TNF -αand Lac decreased significantly faster in the observation group than in the control group(t =8.478,3.104,all P <0.05).After treat-ment,APACHE Ⅱ score decreased significantly in observation group than in the control group(t =2.321,P <0.05). Conclusion Ulinastatin had protective effects on heart function in patients with severe sepsis.
2.Expression of tumor suppressor gene-p33/ING1 in colorectal carcinoma

Objective To explore the correlation between the expression of p33/ING1 and the clinicopathological features in patients with colorectal cancer, and to understand the possible mechanism of p33/ING1 in the oncogenesis of colorectal neoplasms. Methods All samples of normal mucosa and cancer tissues from 52 patients with colorectal cancer were detected for their expression levels of p33/ING1 by RT-PCR. Results A significant decrease in p33/ING1 mRNA expression was found in 30 out of 52(57.7%) colorectal cancer tissues. The results also showed that repression of p33/ING1 expression markedly related to both the Duke's stage and metastasis. Conclusion Down-regulation of p33/ING1 may play an important role in the oncogenesis and development of colorectal carcinoma.
3.Expression and mutation of ING1 in colorectal carcinoma and their clinical significance

ObjectiveTo study the expression of ING1 mRNA and its mutation status in human colorectal cancer. MethodsSamples of normal mucosa and cancer tissues from 52 patients with colorectal cancer were detected for their expression of ING1mRNA with RT PCR, and screened for its mutation with PCR SSCP. The expression of p53 protein was detected with immunohistochemical staining.ResultsThe impaired expression of ING1mRNA in colorectal cancer was associated with an advanced Dukes′ stage and metastasis of lymph nodes ( P ≤ 0 01). The expression of ING1mRNA positively correlated to that of p53 protein ( P ≤0 01). And a infrequent mutation of ING1 was also observed. Conclusion Our results suggest that ING1 contributes to the tumorigenesis and progression of colorectal cancer through down regulating its expression rather than through mutation.
4.Expression and mutation of cancer suppressor gene ING1 in colorectal cancers

BACKGROUND: Overexpression of cancer suppressor gene ING1 may cause apoptosis of many kinds of cancer cells and results in cell cycle arrest in G1 stage. ING1 is involved in p53 signaling pathway and regulates the activity of p53 as a transcription-activating factor.OBJECTIVE: To explore the association of ING1 gene expression and mutation with the clinicpathological features of colorectal cancer.DESIGN: A controlled observation of the pathological samples.SETTING: Department of General Surgery, Southwest Hospital, Third Military Medical University of Chinese PLA.PARTICIPANTS: Totally 52 fresh colorectal cancer samples were obtained from patients hospitalized in the Southwest Hospital, Third Military Medical University of Chinese PLA from October 1998 to October 1999. Normal mucosal tissues were also collected 10 cm from the margin of the tumor during the operation. The patients included 29 male and 23 female patients with an average age of 50.28 years.METHODS: ING1mRNA and protein expressions were detected in the 52 colorectal cancer tissue samples. The ING1 gene mutation was detected by analyzing single-stranded DNA polymorphism. The expression of p53 protein in the colorectal cancer tissue was detected immunohistochemically.tion-single-strand conformation polymorphism.was obviously decreased in comparison with that in the normal mucosa(0. 626 ±0. 382 vs 1. 166 ±0. 245, P < 0. 001) . The expression of ING1 in the tumor tissue of patients with lymphatic metastasis was significantly lower than that in the tumor tissues from patients without lymphatic metastasis (0. 393 ±0. 243 vs 0. 960 ±0. 299, P <0.01). The expression of p53 protein was positively correlated with ING1 mRNA expression( P < 0.01). Only one of the 52 samples was positive for ING1 gene mutation.CONCLUSION: ING1 gene may participate in the occurrence and progression of colorectal cancer and its decreased expression inhibits the action of tumor suppressor genes. The decrement in ING1 mRNA and protein expression without gene mutation indicates that ING1 participates in the pathogenesis of colorectal cancer in a manner quite different from that of the classic tumor suppressor genes.
5.Comparative study of bovine and porcine derived materials in hydrolysate samples by real-time fluorescence quantitative PCR and general PCR

Objective To compare real-time fluorescence quantitative PCR with general PCR in detecting bovine and porcine derived materials in hydrolysate samples.Methods DNA were extracted from hydrolysate samples which prepared by different steps by real-time fluorescence quantitative PCR and general PCR.Results DNA of bovine and porcine could be detected by real-time fluorescence quantitative PCR and general PCR in samples prepared in the processes before enzymolysis solution, but not detected in samples from supermatant to the fourth ultrafiltrate.Conclusion Both real-time fluorescence quantitative PCR and general PCR can be applied to detect the fragments in hydrolysate samples.And real-time fluorescence quantitative PCR has the advantage such as rapid,convenient, non-environment-polluted, good repeatability, which improves the quality and efficiency.
6.Assessment of low quantitative ultrasound values of calcaneus in Chinese southern healthy females

Objective To study the pattern of ultrasonic bone mineral density of calcaneus with age, height and body weight, and to establish a normal reference value of stiffness index (SI) of females in Southern China for providing scientific foundation for osteoporotic diagnosis and prevention. Methods SI for calcaneus in 2498 healthy people 10~87 years old was measured with Achilles Express ultrasound apparatus made in USA. They were divided into groups according to sex and age. One group for 10 years each and the records beyond 69 years were classified into one group, total 7 groups. Results The SI showed a characteristic mild rise-then-fall pattern with increasing age. And the peak value of SI presented in 20~29 age group. Pearson correlation analysis showed negative correlation between SI and age and a positive correlation between SI/ height and weight. The prevalence of osteoporosis gradually increase with age. Conclusion There are significant correlations in SI with age, height and weight; the values of SI will provide an important data reference to establish the normal values and diagnostic]standard of osteoporosis.

Objective To investigate the correlation between IL‐6 ,hs‐CRP and blood lipids ,blood glucose in type 2 diabetes mel‐litus(T2DM) patients complicated with coronary heart disease .Methods 64 outpatients first diagnosed T2DM complicated with coronary heart disease were selected ,56 T2DM patients and 58 health examination were as compare from 2014 January to November in my courtyard .Interleukin‐6(IL‐6) ,high sensitivity C reactive protein(hs‐CRP) and total cholesterol(TC) ,low density lipopro‐tein‐C(LDL‐C) ,blood glucose and HbA1c were detected in 3 groups of person .Results T2DM group and T2DM complicated with coronary heart disease with fasting glucose ,HbA1c ,TC and LDL‐C was significantly higher than normal group ,the difference was statistically significant(P<0 .05);The level of IL‐6 ,hs‐CRP in patients T2DM with coronary heart disease complicated was signifi‐cantly higher than that of T2DM group ,and T2DM group was higher than that of healthy group ,the differences were statistically significant(P<0 .05) .Conclusion IL‐6 and hs‐CRP can be as a specific index to predict the disease process of T2DM complicated with coronary heart disease .

Objective: To study the curative effect of Shengmai injection(生脉注射液) and composite Danshen injection(复方丹参注射液) on acute septic cardiomyopathy(ASC).Methods: Sixty patients(clinically) diagnosed as ASC were randomly divided into two groups: treatment group and control group.All the patients received western routine treatment,and Shengmai injection and composite Danshen injection were added to the patients in the treatment group once per day for 14 days.Cardiac troponin(cTn),electrocardiogram(ECG),clinical synthetic curative effect and amelioration rate of traditional Chinese medicine syndrome scores as well as inflammatory indexes including C-reactive protein(CRP),procalcitonin(PCT) and white blood cell count(WBC) were observed after treatment.Results: The levels of cTn,ECG,clinical synthetic curative effect and amelioration rate of traditional Chinese medicine syndrome scores were more improved in the treatment group than those of the control group(all P

Objective To explore the molecular mechanism of renal function defects after urinary obstruction and investigate the effect of sirolimus on the expression of γ-ENaC,Na + K + ATPase and AQP2,and its mechanism of renal Water-Electrolyte imbalance following bilateral ureteral obstruction (BUO) in rat kidneys.Methods Forty-eight rats were randomly divided into a sham operation group ( sham group),a BUO group,and a sirolimus treatment after BUO group.Bilateral ureters were exposed and occluded with ligature in the BUO and sirolimus treatment groups.Twenty-four hours later,the obstructed ureters were decompressed by removal of the ligature.The sham animal group underwent identical surgical procedures,but the ureter was simply dissected without removal of the ligature.The sirolimus treatment groups was given sirolimus intragastricly 0.4 ml per day (2 mg/kg · d) from the day before surgery until the rats were scari fled.The sham and BUO groups were given the same volume of intragastric saline.The urine and blood were collected at 4 d,7 d after surgery,and the functional data were observed.The expression of γ-ENaC,Na+K + ATPase and AQP2 were examined by immnohistochemistry and immunoblotting.Results On day four and seven post ureteral obstruction release,urine volume in the BUO group were (85.31 ± 13.15,66.39 ±10.56 ml),significantly higher than that of sham operation (35.36 ± 7.74,33.90 ± 8.03 ml) and sirolimus treatment groups (69.81 ± 10.70 ml,48.57 ± 9.01 ml) (P < 0.05 ).Urine sodium concentrations in the BUO group were (42.17 ± 7.35 mmol/L,43.63 ± 18.39 mmoL/L),significantly lower than that of sham operation ( 170.56 ± 18.39 mmoL/L,172.52 ± 7.35 mmol/L) and sirolimus treatment groups (76.18 ± 13.20 mmol/L,134.28 ± 13.20 mmol/L),P < 0.05.Immunoblotting assay showed that,on day four and seven post rats ureteral obstructions were released,integral optical density of γ-ENaC (2.09 ±0.32,2.27 ±0.35),Na+ K+ATP enzyme (2.41 ±0.48,2.67 ±0.43) and AQP2 (2.17 ±0.45,2.63 ±0.28) in the sirolimus treatment group were significantly higher than those of BUO group ( 1.28 ± 0.21,1.45 ±0.17) (1.99 ±0.28,2.18±0.24) (0.93 ±0.22,1.31 ±0.16),but still lower than the sham group (2.58±0.51,2.60±0.56) (2.89±0.53,2.97 ±0.66) (3.05 ±0.63,3.10±0.67).There were significant differences among all the three groups ( P < 0.05 ).Conclusions The downregulation of γ-ENaC,Na + K + ATPase and AQP2 expression after BUO may contribute to the impaired renal tubular sodium reabsorption,decreased urinary concentration,and postobstructive polyuria.Sirolimus treatment significantly prevents impairment in renal function and also prevents downregulation of y-ENaC,Na + K+ ATPase and AQP2during BUO,demonstrating a marked renoprotective effect of sirolimus treatment in conditions with urinary tract obstruction.
10.A clinical analysis of morning blood pressure surge in middle and old aged male hypertensive patients

ObjectiveTo investigate the morning blood pressure surge(MBPS) and its relationship with 24-hour blood pressure variability and anti-hypertensive drugs in middle-aged and elderly hypertensive patients.Methods A total of 521 middle-age and elderly men were surveyed with ambulatory blood pressure monitoring and ambulatory electrocardiograms recordings as well as questionnaire investigation from January 2009 to December 2010.Subjects were divided into MBPS positive group and MBPS negative group according to the level of MBPS[ >35 mm Hg (1 mm Hg=0.133 kPa) or ≤35 mm Hg].ResultsIn all the cases,the prevalence of MBPS was 19.4%,of which the elderly and very elderly had higher prevalences ( 18.9% and 21.8%,respectively) than the middle-aged (5.6%,both P <0.01 ).Significant differences could be found in age [ ( 81.6 ± 6.4 ) years vs ( 78.7 ± 9.7 ) years ],day mean systolic blood pressure [ ( 132.8 ± 13.3) mm Hg vs ( 128.8 ±13.3) mm Hg],fasting blood glucose [ (5.96 ± 1.59) mmol/L vs (5.68 ± 1.22) mmol/L] and 24-hour blood pressure variability between the two groups ( all P < 0.05 ).Significant difference could be observed in the prevalence of MBPS between the diuretics-taking group and non-taking group (27.4% vs 17.6%,P <0.05 ).ConclusionsThe elderly hypertensive patients are prone to appear MBPS phenomenon.Fasting blood glucose level,24-hour blood pressure variability may be associated with MBPS.Diuretic antihypertensive treatment may not be conductive for MBPS control.
