1.Effects of Xinkeshu Capsule on Hemorheology of Rabbits with Experimental Diet ary Atherosclerosis

Objective To study the effects of Xinkeshu capsule (mainly com po sed of Radix Salviae Miltiorrhizae, Radix Puerariae, Fructus Crataegi, Radix Not oginseng, Radix Auklandiae,etc.) on hemorheology of experimental atherosclerosis rabbits. Methods FASCO- 300 viscosity meter was used to detect whole blood v iscosity and plasma viscosity, hematocrit (Wen's tube) and erythrocyte sedimen tation (Wintrobe tube) were tested, and other items of hemorheology were exami ned. 2.Therapeutic Observation of Acupuncture plus Medication for Sudden Hearing Loss

Objective To observe the clinical efficacy of acupuncture plus medication in treating sudden hearing loss.Method Sixty patients with sudden hearing loss were randomized into a treatment group and a control group, 30 cases in each group. The treatment group was intervened by acupuncture plus medication, while the control group was intervened by medication alone. The two groups were treated once a day, 10 d as a treatment course. The pure-tone hearing threshold, plasma endothelin (ET) content, comparative and absolute counting of endothelial progenitor cells (EPCs) were observed before the treatment and after 2 treatment courses, and the clinical efficacies were compared. Result The pure-tone hearing threshold, plasma ET content, comparative counting and absolute counting of EPCs were significantly changed after the treatment in both groups (P<0.05). After the intervention, the pure-tone hearing threshold, plasma ET content, comparative counting and absolute counting of EPCs in serum of the treatment group were significantly different from those of the control group (P<0.05). The total effective rate was 92.6% in the treatment group versus 82.1% in the control group, and the between-group difference was statistically significant (P<0.05).Conclusion Acupuncture plus medication is an effective method in treating sudden hearing loss.
3.The expression and clinical significance of toll-like receptor (TLR) 4 and TLR7 on renal tissue in children with pri-mary nephrotic syndrome

Objective To study the expression and clinical significance of toll-like receptor (TLR) 4 and TLR7 in primary nephrotic syndrome (PNS) of different pathological types in children. Methods Renal expressions of TLR4 and TLR7 were amined and analyzed retrospectively in renal biopsy specimens from 110 PNS patients and 21 healthy controls by immunohisto-chemical method. According to the renal pathologic classification of PNS, the TLR4 and TLR7 expression levels in PNS of dif-ferent types were compared. Results Compared with the renal tissue of healthy controls, the expression level of TLR4 on renal tissue of PNS patients was significantly increased (P<0.01). Among MN, MsPGN and FSGS, the highest expression of TLR4 was observed in MCD (P<0.01). Compared with the renal tissue of healthy controls, the expression level of TLR7 in re-nal tissue of PNS patients was also significantly increased (P<0.01). Among MCD, MN and FSGS, the highest expression of TLR7 was observed in MsPGN (P<0.01). Conclusions TLR4 and TLR7 expression levels are increased in renal tissue of PNS patients and the expression levels may be correlated with renal pathological types.

5.DNA polymerase ? gene mutation in gastric carcinoma and the relationship with H.pylori infection

Purpose:To detect DNA polymerase ? gene (pol?) mutations in human gastric cancer specimens and the relationship between H.pylori infection and pol? mutation. Methods:Extracting total RNA from gastric carcinoma,corresponding cancer specimens tissues and normal tissues, synthesizing cDNA and then using them as templates proceed PCR,The products of PCR were checked by SSCP. Extracting DNA from the specimen, we could detect the H.Pylori from the tissue of gastric carcinoma and the tissue adjacent to them by PCR.Results:There were 7 abnormal SSCP of 32 gastric carcinoma samples, and the mutation rate was 21.9%, but nothing abnormal was found in the tissues adjacent to the tumor. The results of H.Pylori DNA were positive in 15 samples from 32 gastric carcinoma tissues. Positive rate was 46.9(15/32).Detection result of tissues adjacent to tumor was consistent with gastric carcinoma. Comparing pol? SSCP to H.pylori-DNA in gastric carcinoma,we found the positive samples of pol? SSCP correlated with H.pylori-DNA.Conclusions:It is suggested that the pol?gene mutation may be associated with carcinogenesis and development of human gastric cancer. H.pylori infection is possibly related to pol? mutation in gastric carcinoma.
6.Therapeutic Observation of Acupuncture in Treating Subjective Tinnitus

Objective To observe the clinical efficacy of acupuncture in treating subjective tinnitus.Method Seventy subjective tinnitus patients were randomized into a treatment group and a control group, 35 cases in each group. The control group was intervened by conventional medication, while the treatment group was by acupuncture in addition to the medication given to the control. The serum zinc level, copper level, and pure-tone hearing threshold were observed before and after intervention, and the clinical efficacies were compared.Result After intervention, the serum zinc and copper levels, and pure-tone hearing threshold were significantly changed in both groups (P<0.05). The serum zinc and copper levels, and pure-tone hearing threshold of the treatment group were significantly different from that of the control group after intervention (P<0.05). The total effective rate was 93.8% in the treatment group versus 87.9% in the control group, and there was a significant difference between the two groups (P<0.05).Conclusion Acupuncture plus medication is an effective way in treating subjective tinnitus.

Objective To observe the effect of RNAi that silences MTA1 gene on invasion and migration of esophageal carcinoma 9706 cells. Methods The siRNA expression vector that silences MTA1 gene was transfected into EC9706 cells by liposome. MTA1 mRNA and protein expressions were detected through quantitative RT-PCR and Western blot, respectively. The invasion and migration of EC9706 cells were evaluated by scrape wound healing assay and cell invasion assay in vitro. Results MTA1 gene expression significantly decreased. Objectives To observe the expressions ofα-smooth muscle actin (a-SMA) and type III collagen (Col-III) of tubuloin-terstitial ifbrosis(TIF) induced by unilateral ureteral obstruction (UUO) in rat and the intervention effect of supplemental hydrogen sul-ifde (H2S). Methods Ninety-six male Sprague-Dawley rats were randomly divided into 4 groups, sham-operated group, UUO model group, NaHS low-dose group and high-dose group. TIF rat model was established via UUO. After UUO operation, low-dose and high-dose group were intraperitoneally injected twice a day with 1.4μmol/kg and 7.0μmol/kg NaHS, respectively. Sham-operated group and UUO model group were given an equivalent volume of normal saline. Eight rats in each group were killed randomly at 7, 14 and 21 days after UUO operation. The concentration of plasma H2S was detected using deproteinization. Renal tubulointerstitial damage was evaluated with routine Hematoxylin and Eosin staining and Masson staining under microscope. The expressions ofα-SMA, Col-III were measured with immunohistochemistry. Results Compared with sham-operated group, renal tubulointerstitial injury was severer in UUO model group and was alleviated after intervention of NaHS. There was signiifcant difference in tubulointerstitial injury among all groups (P<0.01), but no difference was found between high-dose and low-dose group. Exogenous H2S supplement could down-regu-late the expressions ofα-SMA and Col-III in renal tissues (P<0.05). There was no difference between high-dose and low-dose group in the expressions ofα-SMA and Col-III (P>0.05). Conclusions TIF induced by UUO is associated with decreased level of endogenous H2S. H2S supplementation can ameliorate the development of UUO-associated TIF in part through down-regulating the expressions ofα-SMA and Col-III in renal tissues. However, a dose dependent manner between the two doses of exogenous H2S supplementation was not observed.

Objective To establish human lung adenocarcinoma multidrug resistance cell lines in vitro,observe their biological characteristics,and investigate the mRNA expressions of DNA pol?,mdr 1,mrp1,GST-?,lrp and topo Ⅱ genes.Methods Paclitaxel-resistant cell lines(A549/TXL20) were established in vitro by exposure to stepwise increased concentrations of the drug in a cell culture medium.Biological morphology and cell cycles were analyzed by morphometry and flow cytometry.The chemoresistance indexes of cells were measured by methyl tetrazolium assay.Evaluation of growth and in vitro drug sensitivity were performed.RT-PCR was employed to analyze the mRNA expressions of the DNA pol?,mdr 1,mrp1,GST-?,lrp,and topo Ⅱ genes.Results ① Compared with parent cells,the resistant sublines had a lower confluent density.They were smaller and mixed with giant cells in different sizes and with different numbers of nucleoli,and the growth property of A549/TXL20 did not change significantly compared with A549 cell lines.② The resistant cells,A549/TXL20,were 19.3 times more resistant to paclitaxel and 67.4 times more resistant to cisplatin than the parent cells,and also demonstrated cross-resistance to mitomycin,vinblastine,and 5-fluouracil(5-FU). ③ Compared with the A549 celllines,an unreasonably higher level of drug resistance and lower drug concentration was detected in A549/TXL20 cells after exposure to the drug in the culture medium.④ The mRNA expression level of DNA pol?,mdr1,GST-?,mrp1 andlrp genes in A549/TXL20 cells was significantly higher than that in A549 cell lines(P
