1.The clinical features,curative effect and prognosis factors analysis in 58 patients with advanced ovarian cancer after intraperitoneal metastases

Objective To explore the clinical characteristics and the influence factors of prognosis of ad -vanced ovarian cancer patients after abdominal metastases .Methods We retrospectively analyzed 65 cases dur-ing January 2013 to January 2016 in the First People′s Hospital of Tianmen .The patients were diagnosed clearly with pathological ,diagnosed for the first time and has the complete clinical data including peritoneal metastases in 58 cases,which were analyzed for single factor and multiple factors of influencing factors survival analysis .Results 58 cases of patients with diagnosis of peritoneal metastasis in ovarian cancer that the average is (49.2 ±6.5), from ovarian cancer diagnosis to abdominal metastases for an average time of 11 months,ovarian cancer patients with peritoneal metastasis of the median survival time was 8 weeks,but 7 cases without abdominal metastases that the median survival time was 15 weeks.The single factor survival curves(Kaplan-meier)showed that marital sta-tus,reproductive history,history of breastfeeding,malignant ascites,neoadjuvant chemotherapy,comprehensive treatment(chemotherapy and tumor cells to destroy the loss and laparotomy abdominal hot perfusion chemothera -py) ,peritoneal metastasis tumor number ,residual lesion size ,quality of life in patients with residual lesions ( KPS scores),D-dimer level in plasma and urine trace albumin levels related to the prognosis of patients (P<0.05). Conclusion Patients with advanced ovarian cancer patients with peritoneal metastasis survival time is shorter , and also company with poor prognosis;Neoadjuvant chemotherapy combined tumor cells to destroy the loss and postoperative intraperitoneal hot perfusion chemotherapy ,short-term curative effect is good ,it can not only im-prove the advanced ovarian cancer treatment effectiveness ,but also improve the quality of life in patients of late phase.The D -dimer level in plasma and urine trace albumin levels before treatment have remained in the pa-tients with high level or not reduce ,treatment effect and prognosis was poor .These can be used as a new index of judging prognosis .
2.Proteomic analysis of energy metabolism-related proteins of mice midbrain

Objective To identify energy metabolism-proteins of mice midbrain by using the proteomic technique,and to investi-gate the relationship between these proteins and neural diseases.Methods Two-dimensional gel electrophoresis was used to sepa-rate totally soluble proteins extracted from mice midbrain.Some protein spots on two-dimensional gel electrophoresis gels were ana-lyzed by using matrix assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS).Results 24 protein spots related with energy metabolism were separated by two-dimensional gel electrophoresis and identified by using MALDI-TOF MS successfully.Conclusion The establishment of energy metabolism-related proteins map of mice midbrain lays a foundation for the research on the involvement of these proteins in neural disease pathogenesis.
3.Diagnosis and prognosis of pulmonary hypertension in patients on hemodialysis using spiral CT scanning

Objective To assess the value of multislice spiral computed tomography (MSCT) in diagnosing pulmonary hypertension.Methods One hundred and forty-two patients on hemodialysis were divided into the group with pulmonary artery hypertension and the group without pulmonary artery hypertension.The diagnosis of pulmonary artery hypertension (pulmonary artery systolic pressure,PASP > 35 mmHg) was according to the guideline from the American Society of Echocardiography.All patients were received the check of MSCT and the diameters of the main pulmonary artery,ascending aorta and descending aorta were recorded.PASP and left ventricular ejection fraction were assessed by echocardiography.High sensitivity C-reactive protein and rumor necrosis factor were measured by automatic analyzer and enzyme linked immunosorbent assay.Results There were significant differences between the two groups in systolic blood pressure,hemoglobin,serum albumin,high sensitivity C-reactive protein and TNF-α (P < 0.05); There were significant differences between the two groups in diameters of the maim pulmonary artery,ratio of the diameter of the main pulmonary artery to the diameter of ascending aorta and ratio of the diameter of the main pulmonary artery to the diameter of descending aorta (P < 0.05).In different heart function groups,there were significant differences in diameters of the main pulmonary artery,ratio of the diameter of the main pulmonary artery to the diameter of ascending aorta,and ratio of the diameter of the main pulmonary artery to the diameter of descending aorta,and left ventricular ejection fraction (P < 0.05).Ratio of the diameter of the maim pulmonary artery to the diameter of ascending aorta was positively related to PASP (r=48.77,P < 0.01),and left ventricular ejection fraction was negatively related to PASP (r=-0.40,P < 0.01).In multivariate linear regression,TNF-α,ratio of the diameter of the maim pulmonary artery to the diameter of ascending aorta and ejection fraction were independent factors of PASP (P < 0.01).Conclusions MSCT measurements play an important role in diagnosis of pulmonary hypertension and in evaluation of clinical prognosis in patients on hemodialysis.
4.Effect of He-Ne laser verus low-level CO2 laser irradiation on accelerating fracture healing

BACKGROUND: The effects of low-level laser biostimulation have been proved by various experimental studies and clinical application, which are manifested as tissue repairing, analgesia, antiinflammation, etc.OBJECTIVE: To observe the effects of He-Ne laser and low-level CO2 laser irradiation on fracture healing.DESIGN: A randomized controlled animal experiment. SETTING: Research Room of Laser Medicine, Chengde Medical College.MATERIALS: Forty-eight healthy male New Zealand rabbits, weighing 2.0-2.5 kg, were used.METHODS: The experiments were carried out in the Research Room of Laser Medicine, Chengde Medical College from 1998 to 2003. ①The 48 rabbits were induced into models of experimental fracture of the left radius, and then they were divided into three groups with 16 rabbits in each group: control group, He-Ne laser irradiation group and CO2 laser irradiation group. The fracture areas of the animals in the He-Ne and CO2 laser irradiation groups were respectively irradiated with 28 mW/cm2 He-Ne laser and 150 mW/cm2 CO2 laser for 10 minutes, once a day. ② The animals were killed on the 15th and 35th days after taking roentgenogram respectively. At 15 days, the collagen and calcium contents in callus were detected. At 35 days, the biomechanic anti-torsion strength was examined.MAIN OUTCOME MEASURES: ①Results of roentgenogram in each group; ② Collagen and calcium contents in callus; ③ Results of biomechanic anti-torsion strength.RESULTS: Totally 45 rabbits were involved in the analysis of results. ① On the 15th and day after fracture, the roentgenogram, collagen and calcium contents in callus in the He-Ne and CO2 laser irradiation groups were better than those in the control group, and the collagen and calcium contents in callus were higher in the CO2 laser irradiation group than in the He-Ne laser irradiation group [(341.9±30.1) vs (302.1±28.7) mg/g; (197.1±19.7)vs (156.5±17.6) mg/g, P< 0.05]. ②On the 35th days after fracture, the biomechanic anti-torsion functions in the He-Ne and CO2 laser irradiation groups were superior to that in the control group, and there were no significant difference between the two laser irradiation groups. ③ The results of roentgenogram in the He-Ne and CO2 laser irradiation groups were better than that in the control group both on the 15th and 35th days after fracture, and there was no significant difference between the two irradiation groups.CONCLUSION: Both CO2 laser and He-Ne laser irradiations can accelerate the process of fracture healing, and the curative effect of CO2 laser is better than that of He-Ne laser on the 15th day after fracture, but there is no significant difference clinically between the CO2 laser and the He-Ne laser irradiations on the 35th day after fracture.
5.A Simple Reliable Fluorescent Assay Method for Histone Acetyltransferase (HAT) Inhibitors

Histone acetyltransferases (HATs) are involved in the regulation of gene transcription in eukaryotic cells and their inhibitors could be a promising class of drugs due to their ability to modulate transcription and exert antiviral, anti-inflammatory as well as antioxidant effects. Nonradioactive spectrophotometric HAT assay is an alternative method to the widespread radioactive assay but suffers from drawbacks as lack of sensitivity and accuracy. A simple, non-radioactive fluorescent assay that measures the production of CoASH was established by its facile reaction with O-phthalaldehyde and 2-amino-ethanol. This method gives much higher accuracy compared to spectrophotometric assay, and allows screening of various compounds with potential HAT inhibition. The novel assay should be a valuable tool in transcriptional research and especially drug discovery.
6.The effects of pravastatin on serum adipocytokines in patients with IGT and metabolic syndrome

Objective To investigate the effects of pravastatin on the blood biochemical index,the levels of serum resistin,tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in patients with impaired glucose tolerance (IGT) and metabolic syndrome (MS).Methods 60 patients with IGT and MS were random treated with pravastatin or remedial life-style intervention,and the levels of lipids profiles,insulin resistance index (HOMA-IR),serum resistin,TNF-α and IL-6 were measured before and after treatment.Results The levels of total cholesterol [ (4.45 ±0.60)mmol/L vs (5.58 ±0.96) mmol/L,t =-5.42,P <0.01],HOMA-IR [3.22 ±0.64 vs 3.58 ±0.71,t =-2.05,P <0.05) ],resistin[ (1.97 ±0.72) μg/L vs (2.76 ±0.73) μg/L,t=-4.26,P <0.01 ],TNF-α[ (9.36 ±2.03) μg/L vs (13.87 ±2.30)μg/L,t =-8.06,P <0.01] and IL-6[ (3.50 ±0.99) μg/L vs (6.32 ±1.17) μg/L,t =-10.06,P <0.01 ] in pravastatin group were significantly lower than those in life-style intervention group.Conclusion Pravastatin could improve insulin sensitivity of the patients with IGT and MS,significantly decreased the serum levels of resistin,TNF-α,IL-6,and it had anti-inflammatory effect.
7.Research on the relationship between fructose-1, 6-bisphosphatase and chemosensitivity of ovarian carcinoma

Background and purpose: Epithelial ovarian carcinoma is the most malignant tumor in female reproductive system because of its resistance to chemotherapy. Fructose-1, 6-bisphosphatase (FBP1) is a rate-limiting enzyme in gluconeogenesis used to catalyze the hydrolysis of fructose-1, 6-bisphosphate to fructose-6-phosphate and inorganic phosphate, thereby inhibiting the effect of glycolysis in tumor cells. This study aimed to investigate the association between the expression of FBP1 and chemosensitivity. Methods: The expression level of FBP1 in ovarian cancer patients was measured by immunohistochemistry. Results: According to the results of immunohistochemistry in 209 ovarian carcinoma specimens, the percentage of positive FBP1 expression was about 49.3% (103/209). Loss of FBP1 was a negative factor of survival (42.6 months vs 62.1 months, P=0.003). Besides, patients who were sensitive to chemotherapy displayed significantly higher scores of FBP1 expression than patients who were resistant to therapy (P=0.007). Conclusion: The rate-limiting enzyme FBP1 in gluconeogenesis can be used as a biomarker for predicting the chemoresistance and prognosis of ovarian cancer patients.
8.Risk factors of pulmonary hypertension and analysis of survival in hemodialysis patients

One hundred and forty-two patients on maintenance hemodialysis were enrolled in the study.According to the guideline of American Society of Echocardiography 97 patients (68.3%) were classified as pulmonary artery hypertension [PAH,pulmonary artery systolic pressure (PASP) > 35 mmHg (1 mmHg =0.133 kPa)] and 45 patients (31.7%) as non-PAH (PASP ≤ 35 mmHg).High sensitivity Creactive protein (hs-CRP) and tumor necrosis factor-α (TNF-α) were measured by automatic analyzer and enzyme linked immunosorbent assay (ELISA),respectively.There were significant differences in CRP and TNF-α levels between PAH and non-PAH patients (P < 0.05).Multivariate linear regression showed that TNF-α and interdialytic weight gain were positively correlated with PAH and left ventricular ejection fraction was negatively correlated with PAH (P < 0.01).In the multivariate Cox proportional hazards models,PAH and TNF-α level were independently associated with higher risk for all-cause death (HR =1.06,95% CI:1.03-1.09 and HR =1.31,95% CI:1.17-1.46,respectively).In Kaplan-Meier survival analysis,the risk of all-cause mortality increased in parallel with PASP.The risk of death in patients with PASP >45 mmHg was higher than that in patients with PASP ≤45 mmHg (log-rank test:x2 =6.58,P =0.010),the risk was 2.82-fold (HR =2.82,95% CI:1.38-5.77,P =0.004).
9.Expression of Decidua Cytokines and Co-Stimulating Factor in a Rat Model of Spontaneous Abortion due to Kidney Deficiency

Objective To establish a spontaneous abortion rat model for a syndrome in traditional chinese medicine, kidney deficiency, and observe the changes of physiological indicators and related cytokine expression in the model. Methods 40 female and 20 male rats were used in this study. The female and male rats were mated (mating ratio 2:1). The day of vaginal smear with a large number of sperm was considered as the first day of pregnancy. The rats were randomly divided into control group and model group. The model group received 450 mg/kg hydroxyurea every day. Mifepristone was given on the eighth day in a dose of 3.75 mg/kg. The diet amount, the diameter index of kidney, ovary and embryos were analyzed. The mRNA expression of Th1/Th2 cytokine was detected by RT-PCR, and the expression of co-stimulating factors CD80, CD86, CD28, CTLA-4 were determined by flow cytometry.Results Comparing the model group with control group on the eighth day, there were significant differences between the model and control groups in quantity of food and water intake, and weight increase (P<0.05), and also in the embryonic diameter index, average of abortion rate, Th1/Th2 type cytokines, co-stimulating factor CD80, CD86, CD28, and CTLA-4 (P<0.05). Conclusion A rat model of spontaneous abortion due to kidney deficiency can be successfully established with hydroxyurea and mifepristone. The high expression of Th1 (TNF-α, IFN-γ) may cause abortion and be harmful to pregnancy. Th2 type (IL-4, IL-10) may facilitate pregnancy. The expression co-stimulating factor CD80, CD86, CD28, CTLA-4 may be relevant to the spontaneous abortion.
10.Research on the cytostatic effect of bufalin on cervical carcinoma cells and the related mechanism

Background and purpose:Cervical cancer remains the second leading cause of death in gynecologic malignancies partially because of resistance to chemotherapy. Bufalin, a component of the traditional Chinese medicine Chansu, has been widely used in cancer treatment in China. This study aimed to investigate the effects of bufalin on inhibiting the proliferation of ME180 and C33A and explore its possible mechanism.Methods:The cytostatic effects of bufalin on ME180 and C33A cells were evaluated by CCK8 assay (cell counting kit-8). Glucose levels in ME180 and C33A cells were measured using glucose assay kit. Then the alterations of GLUT1 (glucose transporter 1) and HK2 (hexokinase 2) gene expression were detected by quantitative real-time polymerase chain reaction (qRT-PCR). The expressions of proto-oncogene C-MYC and HIF1α (hypoxia-inducible factor 1α) were determined by Western blot.Results:According to the results of CCK-8, bufalin can significantly inhibit the proliferation of carcinoma cells ME180 and C33A (P=0.027,P=0.018). Test on glycometabolism indicated that glucose uptake in cells treated with bufalin decreased (P=0.034,P=0.036). Results from real-time PCR showed that the expression of glycometabolism related indicators GLUT1 (P=0.019) and HK2 (P=0.016) levels were signiifcantly down-regulated in bufalin treated group. Western blot showed that the expression of C-MYC and HIF1αin cells with bufalin treatment was down-regulated markedly.Conclusion:Bufalin can inhibit the proliferation of the cervical carcinoma cells ME180 and C33A through inhibition of their glucose metabolism.
