MicroRNAs (miRNAs) are closely associated with cancer initiation,development and clinical progression.Detecting miRNA levels in different body fluids,such as sputum,pancreatic juice,saliva,urine,blood and so on,may play some roles in cancer classification,staging,early diagnosis,prognostic prediction and drug efficacy evaluation.
2.Comparison of life quality between surgical castration and drug castration in advanced prostate cancer

Objective To explore the effects of surgical castration and intermittent androgen blockade (IAB) in patients with advanced prostate cancer.Methods 115 cases of advanced prostate cancer were selected including 48 cases with drug castration receiving IAB and 67 cases with surgical castration.Before the treatment, 6 months and 1 years after the treatment, the quality of life of the two groups was investigated by the quality of life questionnaire (EORTC QLQ-C30 +QLQ-PR25).Results Compared with before treatment, the general health status of the patients in the drug castration group and the surgical castration group were significantly improved (P<0.01), the whole body and the local pain were relieved (P<0.01) and the micturition symptoms were controlled ( P<0.01 ).The overall quality of life of drug castration group and surgical castration group were significantly decreased (P<0.05), and the degree of sexual function was particularly affected (P<0.01), but the body function of patients in drug castration group restored to a certain extent in drug castration group after 1 year’s treatment(P<0.01), while the quality of life in patients of surgical castration did not reverse.Conclusion Both of surgical castration and androgen deprivation therapy can reduce the quality of life of patients with advanced prostate cancer, but the adverse effects of drug treatment on the quality of life can be recovered in treatment intermission.
3.Clinical Efficacy of Long-pulsed 1064 nm Laser Combined with Amorolfine Hydrochloride in the Treatment of Patients with Onychomycosis

Objective:To investigate the clinical efficacy of long-pulsed 1064nm laser combined with amorolfine hydrochloride in the treatment of patients with onychomycosis. Methods:Totally 106 cases of patients with onychomycosis were randomly divided into the observation group (n=53) and the control group(n=53) according to the random number table. The control group was given 5%amorolfine hydrochloride liniment, while the observation group was treated with long-pulsed 1064nm laser additionally. The course of treatment was 6 months. The clinical efficacy, length of deck in ward areas, length of deck in health zone, clearance of mycology and adverse reactions were compared between the groups. Results:The total effective rate of the observation group(90. 57%) was signifi-cantly higher than that of the control group(75. 47%, P<0. 05). After the treatment, the length of deck in ward areas in both groups were significantly decreased, while the length of deck in health zone were significantly increased (P<0. 05). The length of deck in ward areas in the observation group was significantly lower than that in the control group, while the length of deck in health zone was significantly higher than that in the control group(P<0. 05). The clearance rate of mycology in the observation group (86. 79%) was significantly higher than that in the control group (67. 92%, P<0. 05). There were no serious adverse reactions during the treatment course. Conclusion:Long-pulsed 1064nm laser combined with 5% amorolfine hydrochloride liniment in the treatment of patients with onychomycosis has remarkable, safe and reliable clinical effects, which shows important study value.
4.Up/down-regulation of miR-21 changes biological function of colon can-cer cells and sensitivity to cetuximab

AIM:To explore the effects of miR-21 on biological behavior of colon cancer cells and their sensi-tivity to epidermal growth factor receptor monoclonal antibody cetuximab .METHODS:Lentiviral vectors were constructed to generate up-and down-regulations of miR-21 lentiviruses (LV-miR-21 and LV-anti-miR-21, respectively), and the cor-responding negative control viruses (LV-miR-21 NC and LV-anti-miR-21 NC, respectively) were also constructed.The vi-ruses were used to infect human colon cancer RKO cells .The changes of the miR-21 expression level , the cell prolifera-tion, the colony-forming ability, the cell apoptosis and the sensitivity of the cells to cetuximab were detected by real -time PCR, MTT assay, soft agar colony assay , flow cytometry and CCK-8 assay.RESULTS: The lentivirus titers of LV-miR-21, LV-miR-2 NC, LV-anti-miR-21 and LV-anti-miR-21 NC were 3.0 ×1012 TU/L, 6.0 ×1011 TU/L, 2.0 ×1012 TU/L and 8.0 ×1011 TU/L, respectively.The infection efficiency was over 80% by the observation of green fluorescence .The miR-21 expression level , the cell proliferation , and the colony-forming ability in LV-miR-21 group were significantly higher than those in LV-anti-miR-21 group.The early apoptotic rate and the inhibitory rate of cetuximab for the cells in LV-anti-miR-21 group were higher than those in LV-miR-21 group.CONCLUSION: miR-21 promotes the proliferation of colon cancer cells.Down-regulation of miR-21 enhances the sensitivity of the colon cancer cells to the targeted therapy drug cetuximab.
6.phVEGF165 gene transfer for prevention of stent restenosis after TIPSS

Objective To test the hypothesis that locally direct gene transfer of human vascular endothelial growth factor(phVEGF) 165 could passivate hepatic venous metallic stents by accelerating endothelialization and augmenting biocompatibility of endovascular stent. Methods The complexes of pAdTrackCMV and lipofectamine was smeared homogeneously to the surface of stent coating with poly-1-lysine. Bare stainless steel stents were used as controls. All stents were implanted into the hepatic right vein by the procedure of transjugular intrahepatic venous stent deployment. Results At the end of 1 week after implantation, the site -specific expression of phVEGF was detected by RT-PCR. Green fluorescence and the expression of phVEGF gene were detected in the transfered stented vessel of the treatment group. These phenomena were not observed in the control group. Scanning electron microscopy revealed that the endothelialization of stent was more pronounced in the treatment group than that in control group. At the end of 8 weeks after implantation, quantitative angiography analysis showed the degree of internal diameter stenosis was less severe in the treatment group compared with the control group. Mean neointima thickness, mean neointima areas and percent cross-sectional area narrowing in treatment group was significantly fewer than that in the control group, respectively. Immunohistochemistry analysis revealed that the proliferation of vascular smooth muscle cells was more active in control group than that in the treatment group. Conclusions Local gene transfer of phVEGF165 can passivate endovascular stents by accelerating stents endothelization and enhancing their biocompatibility in hepatic vein, resulting in reducing thrombus formation and attenuating intimal hyperplasia.
7.Cloning,Expression and Activity Detection of Vascular Endothelial Growth Factor

【Ojective】To clone,express and detect activity of VEGF165.【Methods】Using human heart cDNA library as template,amplified the VEGF gene by PCR.The PCR product was ligated to PUC19 plasmid and sequenced.A eukaryotic expression plasmid AdtrackCMV harbouring VEGF165 was constructed.,then transformed into 293 cells.RNA dot blot and Western blotting were performed to demonstrated whether the tranfermants expressed VEGF165 at mRNA and protein level respectively.Furthermore the bio-activity of VEGF165 was preliminarily detected with Miles test.【Results】Sequence of 582bp VEGF165 cDNA was proved correct by sequencer analysis.The expression of VEGF165 mRNA was identified by RNA dot blot,Western blotting indicated that the molecular weight of VEGF165 protein was 22ku.VEGF165 also is of vascular permeability.【Conclusion】VEGF165 gene has been successfully cloned and expressed,which makes a basis for the further study in vivo.
8.Study of Concentration of Urinary Cadmium and Serum Sex Hormone in Cadmium-exposed Male Workers

Objective To evaluate the concentration of urinary cadmium and serum sex hormone in cadmium-exposed male workers and explore the affecting factors and related mechanism.Methods The individuals exposed to cadmium from three workshops 40 in each, in a cadmium rechargeable battery manufacture plant were selected as the exposure groups according to concentrations of cadmium oxide in the workplace in November 2008, namely assembly, charging and butt-weld workshop for low, moderate and high dosage group.Another forty male workers without cadmium exposure of the same plant were selected as the control group.Urinary cadmium, serum testosterone,FSH and LH concentrations were measured and urinary cadmium concentration abnormity rate was calculated for the following analysis.Results Compared with the control group, the urinary cadmium concentration and its abnormity rate of the low and high dosage groups were all significantly increased(P9 years groups were all significantly increased(P
9.Determination of glaucocalyxin A by HPLC and its protein binding rate in plasma of rat

Objective To study the protein binding rate of glaucocalyxin A in plasma of rat. Methods The plasma balance dialysis was used. An HPLC for the quantitative determination of glaucocalyxin A was presented and used to calculate the protein binding rate in plasma of rat. Results There were protein binding rate results of 74.46%, 77.87%, and 75.29% at three various concentrations 20, 10, and 1 ?g/ mL of glaucocalyxin A in plasma of rat. Conclusion The HPLC method used to determine glaucocalyxin A is simple, rapid, and sensitive with good specificity, precision and accuracy, and glaucocalyxin A has medium capacities in protein binding rate in plasma of rat.
10.Experimental study of Radix Saposhnikoviae and Feutus Trbuli on antagonizing type Ⅰ allergy

AIM: To study the effects of Radix Saposhnikoviae(RL) and Feutus Trbuli(FI) used singly and combinedly on antagonizing type Ⅰ allergy,such as passive cutaneous anaphylaxis(PCA),released model from mast cell granules,Schulz-Dale respond. METHODS: Mouse and rat models mainly induced by IgE were used in experiments. RESULTS: RL and FI used singly and combinedly could significantly inhibit PCA of same species of mice,the degranulation of mast cells of skull in rats and Schulz-Dale response. CONCLUSION: Both RL and FI respectively have antagonistic effect on type Ⅰ allergy and RL in combination with FI will possess more satisactory effect
