BACKGROUND:In vitro experiments suggest that LIM mineralization protein-1 (LMP-1) gene can increase the expression of LMP-1 protein in osteoporotic bone marrow mesenchymal stem cells. OBJECTIVE:To investigate the LMP-1 expression in osteoporotic bone marrow mesenchymal stem cells transfected with RV-LMP-1-GFP in vitro. METHODS:Twelve SD female rats were selected and subjected to bilateral ovariectomy for establishment of osteoporosis models. After 2 months of feeding, bilateral femurs, tibiae, and humeri of rats were taken to isolate and culture bone marrow mesenchymal stem cells. Passage 3 cells were taken and randomly divided into ovariectomized group and LMP-1 transfection group. Another six rats only underwent removal of the same amount of fat tissue around the ovary, and passage 3 cells which were harvested as those in the former two groups served as sham group. RT-PCR and western blot assay were performed to determine the expression of LMP-1 protein and mRNA. RESULTS AND CONCLUSION:Under an inverted fluorescence microscope, transfected bone marrow mesenchymal stem cells from osteoporotic rats showed green fluorescent expression. The three groups al could express LMP-1 at the protein and mRNA levels. The expression levels of LMP-1 protein and mRNA in the LMP-1 transfection group were significantly higher than those in the ovariectomized group and sham group (P<0.05), but there was no statistical difference between the latter two groups (P>0.05). The successful expression of the RV-LMP-1-GFP gene in osteoporotic SD rat bone marrow mesenchymal stem cells was realized at the protein and mRNA levels;moreover, the expression of LMP-1 was increased dramatical y. These findings indicate that LMP-1 gene is successful y transferred into osteoporotic SD rat bone marrow mesenchymal stem cells, and significantly elevates the expression of LMP-1 mRNA and protein.

Objective To explore the value of vascular endothelial growth factor(VEGF) in predicting and mornitoring ovarian hyper-stimulation syndrome(OHSS). Methods 11 cases of OHSS patients and 16 cases of high-responder patients were enrolled in this study, respectively from IVF-ET or ICSI programme. Serum VEGF level before giving gonadotrophin, on the days of hCG administration, OPU and ET, and at the 7th and 14th days after ET as well as the VEGF level of follicular fluid (FF) on the day of OPU were measured. The serum VEGF of hospitalized women with severe OHSS were also measured during the whole therapy. Results Compared with high-responder patients, serum VEGF level on mid luteal and late luteal phases significantly increased in OHSS patients, and FF VEGF level obviously increased as well. Serum VEGF level gradually decreased along with clinical and biological improvement of OHSS. Conclusion FF VEGF level could predict the occurrence of OHSS, and serum VEGF level could monitor the progress of OHSS.

Objective To investigate significance and correlation of free fetal DNA (fDNA) and β-human chorionic gonadotropin(β-hCG) in circulation in pregnant women with high-risk of Down's syndrome (DS). Methods Pregnant women with a male fetus at second trimester screening for Down's syndrome were chosen, including 5 women with a trisomy 21 fetus (DS group), 21 women with DS high-risk pregnant women (DS high-risk group) matched with 22 normal pregnant women as control group. Free fDNA in maternal plasma were extracted. Male DYS14 gene was labled as fDNA, real-time PCR was used to detect fDNA expression. The concentration of β-hCG in maternal serum was detected by chemiluminescence immune assay. The relationship between level of free fDNA and β-hCG concentration was analyzed by Pearson correlation analysis. Results (1) The mean level of free fDNA was (127±58 ) GE/ml in DS group, which was significantly higher than (78±28) GE/ml in DS high-risk group and (48±21 ) GE/ml in control group,respectively (P<0.01). When compared the level of free fDNA between DS high-risk group and control group, it reached statistical difference (P<0.01). (2) The mean concentration of β-hCG was (97±43) kU/L in DS group, which was significantly higher than (58±25) kU/L in DS high-risk group and (38±19) kU/L in control group, respectively (P<0.01). The level of β-hCG in DS high-risk group was also significantly higher than control group (P<0.01). (3) The positive relationship between the level of free fDNA in maternal plasma and β-hCG concentration in maternal serum was observed amongthree groups (r=0.83,P<0.05;r=0.76,P<0.01;r=0.86,P<0.01). Conclusions Free fDNA in maternal plasma might be a candidate marker used for prenatal DS screening. However, its clinical value need to be evaluated because of positive correlation between free fDNA and β-HCG in maternal circulation.

OBJECTIVE:To compare therapeutic efficacy and safety of budesonide or beclometasone combined with terbutanline in the treatment of acute attack of mild and moderate asthma in children.METHODS:A total of 110 children with mild and moderate asthma were randomly divided into group A (55 cases) and group B (55 cases).Based on routine treatment,group A was given Terbutaline sulphate solution for nebulization 1 mL,nebulization inhalation,twice a day+Budesonide suspension for inhalation 2 mL,nebulization inhalation,twice a day.Group B was given Terbutaline sulphate solution for nebulization (same usage and dosage as group A)+Beclomethasone dipropionate suspension for inhalation 2 mL,nebulization inhalation,twice a day.Treatment courses of 2 groups lasted for 7 d.Clinical efficacies,symptoms and signs disappearance time,hospitalization time,asthma relief rate as well as symptoms and signs scores,peak value expiration velocity of flow (PEF),the occurrence of ADR were observed in 2 groups.RESULTS:There was no statistical significance in total response rate,symptoms and signs disappearance time,hospitalization time,asthma relief rate or the incidence of ADR between 2 groups (P＞0.05).After treatment,symptoms and signs scores of 2 groups were significantly lower than before,while the PEF were significantly higher than before,with statistical significance (P＜0.05),but there was no statistical significance between 2 groups (P＞0.05).CONCLUSIONS:Based on routine treatment,budesonide or beclometasone combined with terbutaline sulphate can be used for acute attack of mild and moderate asthma in children.They are similar to each other in therapeutic efficacy and safety.

Objective To explore the relationship between the ultrastructure and gallstone recurrence after cholecystolithotomy.Methods By implanting human cholesterol gallstone into the gallbladder of rabbits, the ultrasructure of the gallbladder was observed with scanning eleetron microscope and transmission electron microscope,3 months before and after cholecystolithotomy.Results ln the stone-implanted group,the epithelium cells were massively damaged,nuclei karyopyknosis,mitochondria destroyed, and phagolysosomes increased. After the gallstone removal, the epitheliune cells did not recover completely where exposed connective tissues and covering surface by mucus. In cytoplasm, the lysosome decreased, and mucus granules secretion increased and actively secreted into the gallbladder lumen.Conclusions The pathologic changes of chronic cholecystitis caused by implanting stones in rabbits is similar to that of the chronic calculous cholecystitis in human.The basic cause of gallstone recurrence after cholecystolithotomy might be that the ultrastructural changes not be restored and the mucus secretion increased obviously in gallbladder.

AIM: To observe the effects of sapindoside on blood pressure,AngⅡ,Ald,ET in the blood plasma and NO in the serum in primary hypertension rat. METHODS: The primary hypertension rats were divided into 5 groups(high,middle and low sapindoside group,control group and captopril group,10 each) in a random fashion and drugs had been given by ig.for 32 days.The blood pressure was messured in the 1,3,7,32 day after administration.At the end of the 32 nd days,AngⅡ,Ald,ET in the blood plasma and NO in the serum were measured. RESULTS: Sapindoside could significantly lower the blood pressure,increase the levels of NO in serum,reduce the concentration of AngⅡ,Ald,ET in the blood plasma. CONCLUSION: Sapindoside plays an important role in decreasing the blood pressure of primary hypertension rat.
