A new approach, combining microwave digestion and sensitive determination of selenium by a normal-phase high performance liquid chromatography after derivatization with 2,3-diamino-naphthalene was proposed to the measurements of trace selenium concentration in natural waters. Two digestions systems, I.e. HNO3 and H2SO4-H2O2, were evaluated and their working conditions were optimized. Under optimal conditions, this approach provides a detection limit of 7.2 pg (with HNO3） or 10.4 pg (with H2SO4-H2O2）, a recovery of 99.1%～103.4%, as well as a linear range of 0～1.5 ng Se(Ⅳ). The approach was applied to determine Se concentration in 4 different types of environmental waters (sample volume: 2 mL) and the concentration levels of 0.20～0．90　μg/L were found, with recoveries of 99.1%～103．4% by standard addition.

Objective To evaluate the effects of two different chronic glaucoma models on intraocular pressure elevation and retinal structure changes in rat.Methods Two different chronic ocular hypertension (COH) models were made by three episcleral vein cauterization or latex microspheres injection into anterior chamber,6 cases of each model.IOP measurements of right eyes (COH eye) and left eye (control eye) were taken weekly by TonoPen (an applanation tonometer).Retinal ganglion cells (RGCs) were retrogradely labeled with 1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI) seven weeks later.Retina structure was observed by immunofluorescence.Results IOP was elevated at postoperative 1-8 weeks,and the mean IOP in the episcleral vein cauterization group was (27.20 ± 1.83) mmHg (1 kPa =7.5 mmHg),whereas the control group was (19.80 ± 1.35) mmHg (P =0.001,n =6).The mean IOP in the latex microspheres injection group was (27.40 ± 1.88) mmHg,whereas the control group was (19.40 ± 1.00) mmHg (P =0.000,n =6).Compared with control rat at postoperative 8 weeks,RGCs loss in episcleral vein cauterization group were 37.9％,39.6 and 33.5％ (all P =0.000,n =6),latex microspheres injection group were 37.3％,39.4％ and 33.5％ (all P =0.000,n =6).There was no statistical difference between episcleral vein cauterization group and latex microspheres injection group (P =0.855,0.949,0.634,n =6).Compared with control rat at postoperative 8 weeks,GCL thickness in both COH models were also significant reduced,but there was no statistical difference in GCL thickness among control group (7.32 ± 0.39) μm,episcleral vein cauterization group (4.97 ±0.33) μm,latex microspheres injection group (5.00 ±0.31) μm.Misoperation or careless operation may lead to microspheres particle residual on flat-mounted rat retina.Conclusion The episcleral vein cauterization or latex microspheres injection into anterior chamber can all increase the IOP.However,there are some advantages in episcleral vein cauterization such as few costs than latex microspheres injection and no microsphere contamination.

Objective To compare the effects of mild moxibustion at different frequencies on sub-health state, and to discuss the effectiveness and dose-response relationship of moxibustion in treating sub-health state. Method Totally 108 subjects were divided by using random number table into group Ⅰ (36 cases), group Ⅱ (36 cases) and group Ⅲ (36 cases). Same acupoints were selected for the three groups, including Guanyuan (CV3), bilateral Zusanli (ST36), and bilateral Sanyinjiao (SP6). A moxa stick was ignited at one end to treat the corresponding points, 3-4 cm away from the skin, making the topical area hot but without burning pain. Each point was treated for 10 min, till the skin flushed. The 3 groups were respectively treated once a day, once every other day, and once a week, 1 month as a treatment course, totally for 2 courses. The Sub-health Symptoms Scale, immunoglobulins IgA, IgM and IgG, and superoxide dismutase (SOD) were evaluated and statistically analyzed before and after intervention. Result The total effective rate was 87.9% in group Ⅰ, 84.8% in group Ⅱ, and 34.3% in group Ⅲ. The paired comparisons showed that there was no significant difference between group Ⅰ and Ⅱ (P>0.05), while group Ⅰ and group Ⅱ were both significantly different from group Ⅲ (P<0.05). After intervention, the levels of IgA, IgM, IgG, and SOD were significantly increased in group Ⅰ and Ⅱ (P<0.05), but the increases in group Ⅲ were statistically insignificant (P>0.05). Conclusion The therapeutic efficacy of mild moxibustion for sub-health is related to the treatment frequency, and frequencies at once a day and once every other day both can produce significant efficacies, which are superior to that of moxibustion at once a week.

Objective To analyze the clinical effects of internal radiation of iodine-125 seed implantation in the treatment of advanced pancreatic carcinoma.Methods 22 patients with unresectable and advanced pancreatic carcinoma were treated by the internal and interstitial radiation using iodine-125 seed implantation.Results Survival of 20 months was observed in 5 patients,10 ～ 20 months in 9 patients,2 ～10 months in 8,patients with an average of (13.47 ± 8.12) months.Among the 22 patients,complete response was obtained in 5 patients,partial response in 8 patients,no response in 7 paticnts,and the other showing PD.The response rate( CR + PR) was 59％.Conclusions Intra - operative internal radiation by iodine - 125 seed implantation does show some therapeutic effects for advanced pancreatic carcinoma.

To investigate the effects of electroacupuncture (EA) at Neiguan (PC6) and Xinshu (BL15) on the nerve electrical activity in spinal dorsal root and norepinephrine (NE) and dopamine (DA) concentrations in the paraventricular nucleus of the hypothalamus in rats with acute myocardial ischemia (AMI).

OBJECTIVE: To explore and summarize the operation method and operation stage for cochlear implant with chronic suppurative otitis media, to provide the clinical reference for cochlear implant with chronic suppurative otitis media. METHOD: The clinical data of 6 cases of cochlear implant with chronic suppurative otitis media from Jun 2006 to Mar 2009 in our hospital was analyzed retrospectively. The operation stage. surgical skill, possible risk and prognosis was analyzed and summarized. RESULT: 3 of 6 cases received single stage subtotal petrosectomy and cochlear implant. 3 of 6 cases received subtotal petrosectomy, they received staged cochlear implant 1 to 6 months later. No complications occurred, all of the cochlear implanted had good open set speech perception. CONCLUSION Staged operation was the first choice for cochlear implant with chronic suppurative otitis media. Single stage operation took potential risks, it should be done cautiously. The key points for the operation was the clearance of the pathological tissue totally, this required good surgical skill and operation instrument.

Objective To provide reference for effective scientific literature novelty assessment by investigating its recognition in medical and health staff of a military area command and their use of information resources and need of information .Methods Knowledge of scientific literature novelty assessment , satisfaction of scientific literature novelty assessment report, need and use of information resources were investigated in 149 staff of 9 hospitals with self-de-signed questionnaire .Results The general recognition and satisfaction levels of scientific literature novelty assess-ment were good in medical and health staff of a military area command .However , the information resources were insufficiently used , which was related with the scientific research level and scientific literature novelty assessment popularization .Conclusion Scientific literature novelty assessment should be popularized , staff engaged in scientific literature novelty assessment should be trained, sharing of information resources should be strengthened , and infor-mation service should be expanded and deepened in military area commands.

Objective To investigate the influence of silencing Med19 on metastasis and invasion of human breast cancer MCF-7 cells. Methods lentivirus expression vector delivering small hairpin RNA (shRNA) against Med19 gene was constructed, then transfected of human breast cancer MCF-7 cell line. There were three experimental groups: non-infected (CON) group, Lv-NC-infected (NC) group, and Lv-shMed19-infected (KD) group. To determine the lentiviral infection efficiency, expression of GFP was detected with fluorescence microscopy. The mRNA and protein levels of Med19 in three groups MCF-7 cells were detected by real-time PCR and Western blotting. Scratching and transwell tests were employed to determine the ability of metastasis and invasion of cells. Results The highest infection efficiency was obtained, resulting fluorescent expression identified in more than 90%of MCF-7 cells 120 h after infection. The expression of Med19 mRNA in the KD group was dramatically decreased by 72.3%compared with the NC group, and by 72.1% compared with the CON group, respectively (P < 0.05). The Med19 protein expression in the KD group was significantly lower than that of the NC group and the CON group, with a reduction of 85.4%and 85.3%, respectively (P<0.05). No statistical significance was detected between the NC groups and the CON groups. Thus, the constructed Lv-shMed19 was demonstrated to be active and specific in inhibiting the expression of Med19. Moreover, the invasive distances of KD group, NC group and CON group MCF-7 cells were compared at 6 hour, 12 hour and 24 hour. The invasive ability of MCF-7 cells decreased significantly with the extension of time (P<0.05). The cells passed through polycarbonate membrane in KD group, NC group and CON group were 23.8 ± 4.32, 43.4 ± 3.65 and 45.8 ± 5.81 respectively. The metastatic ability of KD group was significant reduced (P<0.05). Conclusion Silencing of Med19 gene significantly decreases the ability of metastasis and invasion of human breast cancer MCF-7 cells, and then suppresses the malignant biological behavior of breast cancer.

BACKGROUND:Hidden blood loss is one of most important complications after total knee arthroplasty, but the mechanism and influential factors are not yet clear. OBJECTIVE:To analyze the relative influential factors for hidden blood loss in primary unilateral total knee arthroplasty. METHODS:Data of 235 patients who had undergone primary unilateral total knee arthroplasty from April to September 2014 were retrospectively studied. There were 38 males and 197 females aged from 48 to 82 years old with a mean age of 66 years. The Gross formula was used to calculate the amount of hidden blood loss. The effects of gender, age, height, body weight, body mass index, anesthesia method, administration of tranexamic acid, postoperative anticoagulation method, typeof prosthesis, tourniquet time and pre-operative coagulation function on the postoperative hidden blood loss and total blood loss after total knee arthroplasty were analyzed. RESULTS AND CONCLUSION:(1) Significant differences in hidden blood loss and total blood loss after total knee arthroplasty were detected between male and female patients (P< 0.01). Significant differences in hidden blood loss and total blood loss were found between tranexamic acid and non-tranexamic acid groups (P< 0.05,P< 0.01).(2) Multivariate linear regression analysis showed that preoperative hemoglobin level and heightwere important factors influencing the blood loss after arthroplasty. Hidden blood loss and total blood loss were not correlated with age, body mass index, anesthesia method, postoperative anticoagulation method, type of prosthesis, tourniquet time and preoperative coagulation function. (3) Results indicate that gender and administration of tranexamic acid affect hidden blood loss and total blood loss after total knee arthroplasty. However, age, body mass index, anesthesia method, postoperative anticoagulation method, type of prosthesis, tourniquet time and preoperative coagulation function do not greatly affect hidden blood loss.

Objective:To build the model of the gene FKBP38 (FK506 binding protein 38) conditional knock out in liver.Methods:Transgenic mouse whose FKBP38 gene was flanked with loxP was constructed by embryo microinjection.The FKBP38 gene was deleted by breeding mice harboring two loxP sites in FKBP38 (FKBP38fl/fl) with the mice bearing the expression ofCre recombinase mice driven by an album promoter.Afterward,the genotype of FKBP38 conditional knockout mice was analyzed.Results:①Relative hepatic FKBP38 mRNA levels showed significant difference between FKBP38 conditional knockout mice (FKBP38-/-) and wild type(P＜ 0.001).②Relative hepatic FKBP38 protein expression levels of FKBP38 conditional knockout mice (FKBP38-/-) were significantly different with wild type(P＜0.001).③Relative phosphorylation of hepatic p70 S6K and 4E-BP-1 protein of FKBP38 conditional knockout mice (FKBP38-/-) showed no significant difference,with slight decrease in phosphorylation of 4E-BP-1,compared with wild type.④No significant difference in expression of hepatic Bcl-2 between FKBP38-/-and wild type.Conclusions:The mouse model of the gene FKBP38 (FK506 binding protein 38) conditional knock out in liver is successfully built.
