1.Preparation and in vitro Transdermal Penetration of Capsaicin Liposomes

Objective:To prepare capsaicin liposomes and study the feasibility by in vitro percutaneous penetration test. Meth-ods:Capsaicin liposomes were prepared by a film-ultrasonic method. The best formula was screened by orthogonal test based on single factor studies with the entrapment efficiency as the index. The improved Franz diffusion cells were used to study the transdermal pene-tration of capsaicin suspensions, capsaicin liposomes and capsaicin ointments, and the cumulative penetration amount through the isola-ted rat skin was compared. Results:The optimal formula of capsaicin liposomes were as follows:the ratio of capsaicin to lipids was 1∶5;the amount of Tween-80 was 100 mg;the amount of vitamin E was 50 mg;10 ml dichloromethane was used as the solvent. The op-timal pH value of the external phase was 6. 5 with the volume of 10 ml. The ultrasonic time was 8 min. The order of 12-hour cumulative penetration amount was capsaicin liposomes>capsaicin-PBS suspensions>capsaicin ointments. Capsaicin liposomes had the highest 12-hour cumulative penetration amount and showed obvious sustained-release property. Conclusion:Capsaicin liposomes have high en-trapment efficiency, good percutaneous penetration and sustained-release property, and the preparation technology is simple.
2.Efficacy and Mechanism of Zhuang Nationality Medical Lotus Needle Plus Back Cupping Therapy for Allergic Rhinitis

Objective The effectiveness of Zhuang nationality medical lotus needle plus back cupping therapy ( Zhuang needle-cupping therapy) , Flixonase aqueous nasal spray and cetirizine tablets in treating allergic rhinitis (AR) was compared for the exploration of the therapeutic mechanism of Zhuang needle-cupping therapy. Methods A total of 200 recruited AR patients were randomly divided into four groups in the proportion of 1:1:1:1. The four groups were Zhuang needle-cupping therapy group, cetirizine group, Flixonase group and blank control group. The blank control group had no medication, and the patients of the other three medication groups were given the corresponding treatment. Ten days constituted one treatment course, and interval between two courses lasted one week. After two courses, the therapeutic effect was evaluated. The changes of specific IgE (S-IgE), leukotriene (LT), interleukin 4(IL-4), IL-9 mRNA, interferon gamma (IFN-γ), Thl / Th2 cells, and Th17 cytokine ( IL-17) were observed before and after treatment. Results ( 1) After two treatment courses, Zhuang needle-cupping therapy group had better therapeutic effect than cetirizine group , Flixonase group and blank control group, and the therapeutic effect of cetirizine group and Flixonase group was better than the blank control group (P<0.05). However, the therapeutic effect of cetirizine group was similar to that of Flixonase group ( P>0.05). ( 2) After treatment, the levels of S-IgE, LT, IL-9 mRNA, IL-4 and IL-17 were decreased, and IFN-γ and Th1/Th2 levels were increased in the three medication groups ( P<0.05 compared with those before treatment) . The differ ences of the laboratory indexes in the blank control group were insignificant before and after treatment ( P>0.05). The results of inter-group comparison after treatment showed that Zhuang needle-cupping therapy group had better effect on improving S-IgE, LT, IFN-γand Th1/Th2 than cetirizine group and Flixonase group (P<0.05). (3) During the trial, no adverse reaction was found. Conclusion Zhuang needle-cupping therapy exerts certain therapeutic effect for AR, and the mechanism may be related with the inhibition of S-IgE, LT, IL-9 mRNA and IL-17 expression, and with the regulation of Th1/Th2 imbalance by decreasing TH2 cytokine level and increasing Th1 cytokine level.
3.Determination of three kinds of berberine type alkaloids in Rhizoma Coptidis and Jinqi Jiangtang Tablet by RP-HPLC

AIM: To establish a reversed-phase HPLC method of determining three kinds of berberine type alkaloids(jatrorrhizine,palmatine and berberine) in Rhizoma Coptidis and Jinqi Jiangtang Tablet(Flos Lonicerae japonicae,Rhizoma Coptidis and Radix Astragali) METHODS: RP-HPLC was used for determination.The separation was carried out on Diamonsil C_(18)column(4.6 mm?250 mm,5 ?m) with the mobile phase containing acetonitrile-0.1 mol/L potassium dihydrogen phosphate(30(∶)70) and a detective wavelength was at 345 nm.The flow rate was 1.0 mL/min. RESULTS: The average contents of jatrorrhizine,palmatine and berberine in Rhizoma Coptidis and Jinqi Jiangtang Tablet were 1.34%,1.61%,7.12% and 0.35%,0.64%,1.54% respectively. CONCLUSION: The method is simple,accurate with a good recovery and repeatability and can be used to control the quality of Rhizoma Coptidis and Jinqi Jiangtang Tablet.
4.An Experimental Study on Xiaochaihu Decoction for Endometriosis in Rats

Objective To observe the effect of Xiaochaihu decoction(XCD) on experimental endometriosis in rats.Methods Rat models of endometriosis were established and the effect of XCD were evaluated by measuring the volume of ectopic endometrium (EE) and its ultrastructure with transmission electron microscopy.Results After treatment,moderate and high-dosage of XCD(10g/kg and 15g/kg respectively) could markedly inhibit endometrial explant growth and decrease the volume of EE(P 0.05).Meanwhile,the glandular cells in the endometrium after treatment in the moderate and high-dosage groups showed characteristic features of apoptosis:cellular atrophy,condensation of cytoplasm and nuclear chromatin and increase of their density,occurrence of the intercellular apoptotic bodies as well as degeneration and necrosis of stromal cell.Conclusion XCD has obvious effects in inhibiting ectopic endometrium and its mechanism may be related to the reduction of cell death (including apoptosis and necrosis) by regulating the immune function.
5.Impacts of astragaloside Ⅳ on the proliferation and angioblastic differentiation of human umbilical cord blood-derived mesenchymal stem cells

Objective:To investigate the impacts of astragaloside Ⅳ (AS-Ⅳ) on in- vitro proliferation and angioblastic differentiation of human umbilical cord blood-derived mesenchymal stem cells (hUCBMSCs), providing a basis for further research about the effects of AS-Ⅳ on mesenchymal stem cells (MSCs)-mediated angiogenesis. Methods:The hUCBMSCs were extracted from umbilical cord blood of normal full-term infants and subcultured. Osteoblasts, chondroblasts, and lipoblasts were induced, differentiated and identified. At the same time, the surface antigens CD44, CD73, and CD105 on hUCBMSCs were determined by flow cytometry. The successfully identified hUCBMSCs were cultured and treated with a series concentrations of AS-Ⅳ (0, 50, 100, 200, 300, and 400 mg/L). The optimum concentration of AS-Ⅳ for cell proliferation in hUCBMSCs was confirmed. In another experiment, hUCBMSCs were randomly divided into the experimental group and the control group. The cells in the experiment group were treated with the optimum concentration of AS-Ⅳ, and those in the control group were treated with equal volume of PBS. The impact of AS-Ⅳ on the proliferation of hUCBMSCs was detected using the cell counting kit (CCK-8). Besides, the impact of AS-Ⅳ on the angioblastic differentiation of hUCBMSCs was examined using the matrigel in- vitro tube formation assay. CD31 and von willebrand factor (vWF) expressions were determined using immunofluorescence after hUCBMSCs differentiated towards endothelial cells. Results:Under the light microscope, hUCBMSCs had clear edges and arranged orderly, showing a typical long fusiform structure. Flow cytometry confirmed that hUCBMSCs had surface markers of mesenchymal stem cells. The optimum concentration of AS-Ⅳ for the proliferation of MSCs was 300 mg/L. The OD values of the control and experimental groups were (0.51±0.01) and (0.98±0.05), respectively, with statistical significance ( t=15.96, P<0.05), indicating that the proliferation ability of the experimental group was enhanced. Compared with the control group, the tube density and the length of the tube network in vitro in the experimental group were higher, with statistically significant difference [(629.80±52.94)mm vs (110.36±13.19)mm, P<0.05]. Compared with the control group, the expression of CD31 and vWF increased in the experimental group after AS-Ⅳ induced hUCBMSCs differentiation ( t=13.64, 13.18, P<0.05). Conclusions:AS-Ⅳ has no toxicity to human umbilical cord blood mesenchymal stem cells, and can improve their proliferation function, and induce hUCBMSCs to differentiate into endothelial cells.
6.Epidemiological study on clinical characteristics of syndrome types of laryngeal cough based on the holism concept of TCM

Objective To study the clinical characteristics of syndrome types of laryngeal cough ,in order to provide an epidemiology basis for classification diagnosis. Methods Medical history,clinical signs and symp-toms of patients with laryngeal cough were collected. And questionnaire survey was carried out. Results (1)Of the 4 syndrome types,compared with wind evil invading lung type laryngeal cough incidence,the insecure defen-sive exterior and intrinsic special type showed no statistical difference(P>0.05). And the comparative differences between the rest 2 groups are of statistical differences(P < 0.05).(2)The onset time was analyzed. The results show laryngeal cough incidence from yin deficiency type and fire hyperactivity type showed no statistical difference (P > 0.05). The rest are all of statistical difference(P < 0.05).(3)The clinical course was analyzed. No differ-ence was found between spleen deficient phlegm type ,yin deficiency and fire hyperactivity type laryngeal cough (P>0.05). Differences of the rest 2 groups showed statistical difference(P<0.05).(4)The state of illness was analyzed. The results showed statistical difference between spleen phlegm type and yin deficiency and fire hyperac-tivity type(P<0.05). No statistical difference can be found between the rest 2 groups(P>0.05).(5)The cross-tab chi-square test was carried out on the 4 groups of syndromes on the nasal allergy eye and allergy concomitant incidence. The results showed that there was no significant difference between the 4 groups(P>0.05).(6)On the phlegm part,the results showed difference between wind evil invading lung type and yin deficiency and fire hyper- activity type laryngeal cough(P>0.05). Differences between the rest 2 groups are of statistical significance(P<0.05).(7)Foreign body sensation was analyzed. The results showed no difference between insecure defensive exte-rior and intrinsic special type and wind evil invading lung type ,deficient spleen phlegm type and yin deficiency and fire hyperactivity type(P>0.05). Differences between the rest 2 groups are all of statistical significance(P<0.05). Conclusions The 4 groups of syndrome types,insecure defensive exterior and intrinsic special type and wind evil invading lung type are the most common. Deficient spleen phlegm type is middle ,yin deficiency and fire hyperactivity type come next. Cough caused by wind invading lung mainly occurs in the evening. Its duration is usually within 1 month. It may be accompanied by phlegm with or without occasional pharyngeal paraesthesia. The daily life is usually unaffected or slightly affected. Cough caused by deficiency of vital energy and specific intrinsic quality mainly occurs in the morning. Its duration is usually 3 months to 1 year. It may be accompanied by less phlegm or no phlegm without or with occasional pharyngeal paraesthesia. The daily life is unaffected or slightly affected. Cough caused by Yin deficiency and fire hyperactivity occurs from time to time. It duration is more than 1 year. It may be accompanied by phlegm with pharyngeal paraesthesia. The daily life is unaffected or seriously affected. Cough caused by spleen deficiency and phlegm turbidity mainly occurs in the morning. Its duration is more than 1 year. It may be accompanied by more phlegm with pharyngeal paraesthesia. The daily life is seriously affected.
7.Analysis of the current situation, value implications, and path of the inheritance of red doctor’s culture

8.Arthroscopic treatment of borderline developmental dysplasia of the hip with labral tear: analysis of mid-term outcomes

Objective:To investigate the clinical efficacy of arthroscopic limited incision of the articular capsule to repair the glenoid labrum in the treatment of borderline developmental dysplasia of the hip (BDDH) complicated with labral tear.Methods:A retrospective study was conducted to analyze the data of 18 patients with BDDH complicated with labral tear who had been admitted to Department of Orthopaedics, The Second Hospital Affiliated to Suzhou University from January 2016 to December 2019 (observation group). There were 12 males and 6 females with an age of (41.8 ± 8.5) years. Simultaneously, another 18 patients were selected as the control group whose hip development was normal but age and gender were matched with those in the observation group. There were 9 males and 9 females with an age of (43.5 ± 10.3) years. Both groups were treated by arthroscopic limited incision of the articular capsule to repair the glenoid labrum. The 2 groups were compared in terms of modified Harris hip score (MHHS), Hip Outcome Score Activities of Daily Living Subscale (HOS-ADL), and visual analogue scale (VAS).Results:There was no significant difference in the demographic data like age, gender ratio, body mass index, severity of labral tear or time from injury to operation between the 2 groups, indicating comparability between groups ( P>0.05). The observation and control groups were followed up for (38 ± 7) and (43 ± 6) months, respectively. For the observation and control groups, respectively, MHHS was (97.1 ± 3.3) points and (95.4 ± 4.2) points, HOS-ADL (92.6 ± 2.8) points and (91.4 ± 4.1) points, and VAS (0.6 ± 0.5) points and (1.0 ± 0.8) points, all showing no significant difference between groups ( P>0.05). Conclusion:In the treatment of BDDH patients complicated with labral tear, simple arthroscopic limited incision of the articular capsule to repair the glenoid labrum can lead to the same good medium-term efficacy as it can in those with normal hip development.
