1.Application value of blood culture combined with serum procalcitonin and CRP in diagnosing infectious fever patients

Objective To evaluate the application value of the blood culture combined with the detection of serum procalcitonin and C-reactive protein(CRP)in diagnosing infectious fever patients.Methods The PCT and CRP detection results on the blood sampling day in 126 cases of fever with positive blood culture and 123 cases of fever with negative blood culture in the emergency department were retrospectively analyzed,and the two sets of data were compared and statistically analyzed by the SPSS 16.0 soft-ware.Results The positive rates of CRP and PCT in 126 cases of positive blood culture were 100% and 100%,the detection values were 26.9-256.8 and 0.25-98.6,the positive rates of CRP and PCT in 123 cases of negative blood culture 95.5% and 15.4% re-spectively,the detection values were 1 .2-126.8 and 0.02-0.98.The PCT and CRP detection values in the positive blood culture group were significantly higher than those in the negative blood culture group,the difference was statistically significant(P <0.01). Conclusion PCT and CRP can be served as the monitoring indicators for blood bacterial infections,the blood culture combined with PCT and CRP detection has the guidance significance for early diagnosing bacterial infection.
2.Effect of Electroacupuncture on Motilin and Somatostatin inRats with Diabetes Gastroparesis

Objective To investigate the effect of electroacupuncture on motilin (MOT) and somatostatin (SS) in rats with diabetes gastroparesis (DGP). Methods One hundred clean healthy SD rats with negative urine glucose were chosen. After they were fed for three days and into a stable state, 30 rats were randomly selected as a control group. The other 70 rats were used as an animal model of insulin-dependent diabetes made with streptozotocin. At 10 weeks after model making, rats as successful models were randomly allocated to model and treatment groups, 30 rats each. The model group received no treatment and the treatment group, electroacupuncture at points Zusanli, Zhongwan and Weishu. Serum and antral MOT and SS were measured in every group before and after treatment. Results There were statistically significant differences in body weight and gastrointestinal propulsion rate between the treatment or model group of rats and the control group (P<0.01), and between the treatment and model groups after treatment (P<0.01). There was a statistically significant difference in blood glucose value between the treatment or model group of rats and the control group after model making and at 10 weeks and the end of treatment (P<0.01). There was a statistically significant difference in blood glucose value between at the end and 10 weeks of treatment in the treatment group (P<0.01). There were statistically significant differences in serum and antral MOT and SS contents between the treatment or model group and the control group (P<0.01), and between the treatment and model groups after treatment (P<0.01). Conclusions Electrocupuncture can markedly increase serum SS and antral MOT and decrease serum MOT and antral SS to ameliorate high blood motilin and somatostatin in DGP rats.
3.Effect of Menthol compound decoction on the growth and metabolism of cariogenic bacteria in vitro

Objective To investigate the effect of menthol compound decoction on the growth and acid metabolism of cariogenic bacteria and to find out the anticariogenic mechanism of Menthol compound decoction .Methods Streptococcus mutans ,Streptococ‐cus sanguinis and Porphyromonas gingivalis were chosen as the experimental bacteria .Firstly ,slip diffusion method was used to ex‐amine the inhibitory effect of menthol compound decoction on cariogenic bacteria .Secondly ,observing the effect of menthol com‐pound decoction of different concentration on acid metabolism .Results Menthol compound decoction could inhibit the growth of Streptococcus sanguinis and Porphyromonas gingivalis .And it also inhibited the acid production of cariogenic bacteria ,but the con‐centration of menthol compound decoction were different .Conclusion Menthol compound decoction may have an inhibitory effect on the growth and acid metabolism of cariogenic bacteria .
4.Infection distribution and drug resistance of Acinetobacter baumannii

Objective To study the clinical distribution and drug resistance of Acinetobacter baumannii (AB) .Methods Clinical distribution and drug resistance of 1 190 strains of AB ,isolated in 2012 and 2013 ,were retrospectively analyzed .Results Most of AB strains were isolated from sputum ,and mainly from Intensive Care Unit ,Department of Neurosurgery and Department of Re‐spiratory .Resistance rates of AB to most antimicrobial agents were 60% -80% .Resistance rate to tobramycin ,piperacillin/tazobac‐tam and imipenem was increased significantly .Resistance rate to cefoperazone/sulbactam was decreased .Conclusion Drug resist‐ance of AB might be serious ,with increasing tendency .
5.Study of bacterial contamination associated with bandage contact lenses after photorefractive keratectomy

Objective To evaluate the occurrence rate and agents of the bacterial contamination of bandage contact lenses after photorefractive keratectomy(PRK) .Methods In a prospective study ,50 patients (100 eyes) underwent PRK .Conjunctival sac se‐creta were placed onto chocolate agar before PRK .All patients accepted bandage contact lenses and anti‐inflammatory drug therapy . Contact lenses and conjunctival sac secreta were placed onto chocolate agar after PRK .Corrected visual acuity ,intraocular pressure and corneal thickness were compared in the 2 groups .Results Among 100 pieces of cornea contact lens ,3 pieces (3% ) were tested positive for bacteria detection and bacteria were staphylococcus epidermis .All conjunctival sac secreta of preoperative and postoper‐ative were not detected bacteria ,postoperative eye infection was not found .Between the positive and negative groups ,tear secretion , may be related to cultivate positive correlation .Conclusion Bacterial contamination is possible when using bandage contact lenses after PRK ,specially for patients with less tear secretion .
6.Clinical Study on Kidney-Nourishing and Governor Vessel-Regulating Acupuncture Therapy for Apoplexy Sequela

To observe the clinical efficacy of kidney-nourishing and governor vessel-regulating therapy for apoplexy sequela.Methods:Sixty subjects were equally randomized into observation group and control group,and respectively treated for 35 days.The scores of survival quality,interleukin-6(IL-6),interleukin-8(IL-8)and tumor necrosis factor (TNF) were measured,and the clinical efficacy were compared between two groups.Results and Conclusion:After treatment.all the observational items were improved,with better results in observation group than in control group.This therapy has better effects than regular method in the treatment of apoplexy sequela.
7.Application of PCR-reverse dot blot hybridization in the diagnosis of vulvovaginal candidiasis and the detection of Candida albicans drug-resistant genes

Objective To evaluate the application value of PCR-reverse dot blot hybridization in the identification of Candida and the detection of Candida albicans drug-resistant genes.Methods The vaginal secretion samples from 285 patients with candidal vaginitis and 50 healthy women were collected.The identification of Candida species and their drug susceptibility were detected by the bioMérieux Yeast identification cards and MIC method(Zhengzhou Antu kit),respectively.The identification of Candida species and the mutation of Candida albicans,drug-resistant genes were also detected by the Shenzheng Yaneng test kit(PCR-reverse dot blot hybridization).The drug-resistant genes were also identified by PCR and nucleic acid sequencing.Based on the culture identification,MIC method and nucleic acid sequencing as the contrast methods,the sensitivity,specificity and accuracy of PCR-reverse dot blot hybridization in the identification of Candida species and the mutation detection of Candida albicans drug-resistant genes were evaluated.Results Compared with the bioMérieux Yeast identification method,the sensitivity,specificity,positive predictive value,negative predictive value and total coincidence rate of PCR-reverse dot blot hybridization for detecting six kinds of Candida species,including Candida albicans,Candida glabrata,Candida tropicalis,Candida parapsilosis,Candida krusei and Candida guilliermondii,were above 95%,96%,96%,98% and 97%,respectively.There was no significant difference in detecting six kinds of Candida species between the two methods (x2 =0.44,0,0,0,0 and 0,respectively,P > 0.05),and there was good consistency between them (Kappa > 0.9).Compared with the MIC method,the sensitivity,specificity,positive predictive value,negative predictive value and total coincidence rate of PCR-reverse dot blot hybridization for detecting the drug resistance of Candida albicans were 98%,88%,98%,88% and 96%,respectively.There was no significant difference in detecting the drug resistance of Candida albicans between the two methods (x2 =0.17,P > 0.05),and there was good consistency between them (Kappa > 0.8).The results of PCR-reverse dot blot hybridization in detecting the mutation sites of six kinds of Candida albicans drug-resistant genes were 100% of coincidence with that of the nucleic acid sequencing method.Conclusion The PCR-reverse dot blot hybridization has high consistency with the culture method and the nucleic acid sequencing method in the identification of Candida species and the mutation detection of Candida albicans drug-resistant genes,which is more early and rapid than the traditional detection methods,and may be applied to the auxiliary diagnosis of vulvovaginal candidiasis (VVC).
8.Surveillance of Antimicrobial Resistance in Staphylococcus spp and Enterococcus spp

OBJECTIVE To analyze the resistant rates of Staphylococcus spp and Enterococcus spp isolated from clinical infections to antibiotics,and to provide reference method for effective control infections of Staphylococcus.METHODS The Staphylococcus spp and Enterococcus spp were identified with VITEK-32 automicrobiology system(AMS) and GPI card,drug resistance was detected with VITEK-32 AMS and GPS-107 card.Laboratory data were analyzed by WHONET-5 statistic software.RESULTS Among 1 445 Staphylococcus spp and Enterococcus spp strains isolated from clinical samples,330 strains(22.8%) were Staphylococcus aureus,872 strains(60.3%) were coagulase negative Staphylococcus,213 strains(14.7%) were Enterococcus faecalis,and 30 strains(2.1%) were E.faecium.From S.aureus 223 strains(67.6%) were MRSA,718 strains(82.3%) of coagulase negative Staphylococcus were MRCNS.The detectable rates of MRSA and MRCNS in 2004 were 75.3% and(82.3%,) which were higher than those in 2003(48.4% and 78.4%).Neither strains of S.aureus nor strains of coagulase negative Staphylococcus were found resistant to vancomycin.MRSA and MRCNS resistant rates were found(higher) than MSSA and MSCNS.From the isolated strains of E.faecalis in 2004,the resistance rates to(ciprofloxacin,) nitrofurantoin,gentamicin-500,levofloxacin,and penicillin G were found higher than that in 2003.(E.faecium)(resistant) rates were found significantly higher than E.faecalis.CONCLUSIONS Staphylococcus spp and Enterococcus spp are the main pathogens leading to clinical infections.The findings of these(surveillance)(studies) will enhance our knowledge regarding the problem of antimicrobial resistance and will serve as a basis for future policies and practice styles.
9.Determination and analysis of serum specific IgE in patients with allergic diseases in Shenzhen area

Objective To analyze the specific IgE test results and the distribution of allergens in patients with allergic diseases in Shenzhen area .Methods Western blot was used to detect inhaled and food specific IgE in 2154 patients with allergic diseases ,and the types and distribution of allergens were analyzed .Results The total positive rate of allergen specific IgE was 48 .88% .More than 20% of the patients were allergic to more than two allergens .The most common allergies were dust mites ,freshwater fish and sea fish .The posi-tive rate of IgE in male allergens was higher than that in females ,and the difference was statistically signifi-cant (P<0 .05) .The positive rate of allergen specific IgE in different age groups was statistically significant (P<0 .001) .The positive rate of dust mites increased year by year before the age of 18 ,and then decreased year after year .The positive rate of chicken protein and milk showed a significant downward trend after 7 years old .The positive rate of allergen IgE in patients with different diseases was statistically significant (P<0 .001) .The positive rate of dust mite combination in rhinitis patients was the highest ,49 .94% .Conclu-sion The main allergen of allergic diseases in Shenzhen area is dust mites combination ,freshwater fish com-bination and sea fish combination .The main food allergen of children is egg and milk .The main allergen of a-dult is dust mites combination and fish ,and the patients with rhinitis are high sensitized people of dust mites combination .
10.Progress in subclinical research of kidney xenotransplantation

Xenotransplantation is an efficient pathway to solve the problem of transplant organ source deficiency in clinical settings. With the increasing progress of gene editing technique and immune suppression regimen, important development has been achieved on researches regarding pig to non-human primate kidney xenotransplantation, which provides a good condition for the introduction of the technique in the clinical application. In view of the substantial difference between human and non-human primate, and to meet the needs of current ethic requirements, it is necessary to perform subclinical studies for pig to human kidney xenotransplantation. In recent years, such subclinical studies with regard to the genetically modified pig to brain death recipient kidney xenotransplantation had been performed, indicating that kidney xenotransplantation gradually began to transit to the clinical development stage. However, donor/recipient selection and immune suppression regimen has not reached a consensus yet, and has to be clarified in subclinical studies. In this article, the current status and confronted problems of donor/recipient selection, immune suppression regimen and post transplantation management in the subclinical studies of kidney xenotransplantation were reviewed, aiming to promote the clinical transformation of kidney xenotransplantation to the clinical application.
