Objective:To study the expression levels of tumor-associated glycoprotein (TAG72) and glucose-regulated protein 94 (GRP94) in the breast benign and malignant lesions and detect their clinicopathological significance. Methods:EnVisionTM immunohistochemistry was used for assaying expressive levels of TAG72 and GRP94 in conventional paraffin-embedded sections from specimens of breast cancer (n=60) and benign lesions (n=30). The survival analysis was performed by using Kaplan-Meier method.Results:The positive rates and scores of TAG72 and GRP94 were significantly higher in breast cancer than those in benign lesions (TAG72: 68.3% vs 10.0%, 2.53±1.86 vs 0.37±1.13; GRP94: 63.3% vs 13.3%, 2.73±2.23 vs 0.47±1.22; P=0.000). The positive rates and scores of TAG72 and GRP94 were significantly lower in the intra-ductal,histological grade Ⅰ or Ⅱ , with maximal diameter ≤3 cm, without lymph node metastasis, estrogen receptor (ER)-positive, p53 and CA15-3-negative breast cancer than those in the ductal invasive (P<0.05), histological grade Ⅲ (P<0.01), maximal diameter >3 cm (P<0.05), with lymph node metastasis (P<0.01), ER-negative (P<0.05) , p53-positive (P<0.01), and CA15-3-positive (P<0.01) breast cancer tissues. Kaplan-Meier survival analysis showed that the median survival time were significantly shorter in the TAG72-positive, GRP94-positive or TAG72 and GRP94-positive breast cancer patients than those with negative TAG72 and/or GRP94 expression (P<0.01). Multivariate COX regression analysis showed that TAG72 and/or GRP94-positive expression were negatively associated with postoperative survival rate and positively related with death rate. Both of them were independent prognostic factors for breast cancer. Conclusion:The expressive levels of TAG72 and/or GRP94 might have important effects on the carcinogenesis, progression, biologic behaviors and prognosis of breast cancer. The breast cancer patients with over-expression of TAG72 and/or GRP94 might have poor prognosis.

Objective To study the expression and clinicopathological significance of Paxillin and CAⅨ in the benign and malignant lesions of gallbladder.Methods The surgical resected specimens of 108 cases of gallbladder adenocarcinoma, 46 cases of peritumoral tissue, 15 cases of adenomatous polyp and 35 cases of chronic cholecystitis were routinely made paraffin embedded sections.The expressions of Paxillin and CAⅨ were stained with Envision immunohistochemistry.Results The positive rates of Paxillin and CAⅨ expression was significantly higher in gallbladder adenocarcinoma (60.2% and 49.1%) than those in peritumoral tissues (26.1%, x2 =15.00, P <0.01 and 23.9%,x2=8.41,P <0.01), adenomatous polyp (20.0%,x2=8.60,P<0.01 and 20.0%,x2 =4.49,P<0.05) and chronic cholecystitis(14.3% ,x2 =22.89, P<0.01 and 11.4%,x2 =15.63,P <0.01).All the gallbladder epithelia of the benign cases with Paxillin and CA Ⅸ positive expression showed moderate to severe atypical hyperplasia.The positive expression rates of Paxillin and CA Ⅸ were significanctly lower in the cases of well-differentiated, maximal diameter of mass less than 2 cm, no lymph nodes metastasis and no surrounding tissues invasion than those of the cases with poorly differentiated, maximal diameter of mass over 2 cm, lymph nodes metastasis and surrounding tissues invasion.With Kaplan-Meier analysis, it suggested that the survival period after the surgery in Paxillin and CAⅨ positive expression cases was shorter than that of negative expression cases (x2 = 5.65,P<0.05,5.65=5.92, P<0.01).With multivariate Cox regression analysis, it indicated that both Paxillin and CAⅨ positive expression was an important indicator of the poor prognosis in gallbladder adenocarcinoma.Conclusion The expression of Paxillin and CA Ⅸ may be closely related to the carcinogenesis, tumor biological behaviors, and prognosis of gallbladder adenocarcinoma.The positive expression of Paxillin and/or CAⅨ is associated with poor prognosis.

Objective To study the expression of hMLH1, hMSH2 and their association with clinico-pathological characteristics in hepatocellular carcinoma(HCC). Methods The expression of hMLH1 and hMSH2 in 37 HCC tissues was detected by SABC immunohistochemistry. Objective To investigate the relationship between the expression of regulated-upon activation normal T expressed and secreted(RANTES)and microvascular density(MVD)in hepatocarcinoma(PHC)tissues,and explore its clinical significance. Methods The samples of PHC tissues and adjacent tissues in 47 patients were collected.The expression of RANTES in tissues was detected by immunohistochemistry,and MVD was calculated.The relationship among the expression of RANTES,MVD and clinicopathological features was analysed. Results The positive expression rate and expression score of RANTES in PHC tissues were significantly higher than those in adjacent tissues(55.32% vs 19.15%,P<0.01;1.89±1.77 vs 0.77±1.29,P<0.01).MVD in PHC tissues was significantly higher than that in adjacent tissues(67.30±13.68 vs 37.20±10.58,P<0.01).MVD of PHC tissues in patients with metastasis was significantly higher than that in patients without metastasis[(73.50±13.77)/HP vs (64.10±12.68)/HP,P<0.05],while there were no correlations among MVD,expression of RANTES and the other clinicopathological features of PHC.MVD was positively correlated with the expression score of RANTES in PHC tissues(r=0.386,P<0.05). Conclusion RANTES might be closely related to the angiogenesis in PHC.

Objective To study the expression of nuclear factor-kappaB (NF-κB) and the counts of lymph vessels in laryngeal squarnous cell carcinoma and polyps of vocal cord tissues, and explore their clinicopathologic significance and correlation in the course of laryngeal carcinoma. Methods SP immuno-histochemical method was used to detect the expression of NF-κB and the counts of lymph vessels on the routinely paraffln-embedded sections of the specimens from 50 cases laryngeal squamous cell carcinoma and 10 cases of polyps of vocal cord tissues. Results The positive rate of NF-κB and the counts of lymph ves-sels in laryngeal carcinoma[60. 0% ,( 13.3±3.4)/HP]were significantly higher ( P <0. 05 and P <0. 01respectively) than those in polyps of vocal cord tissues[10.0 % ,(6. 1±3. 8)/HP]. The positive rate of NF-κB and the counts of lymph vessels in well differentiated adenocarcinoma and cases without metastasis were significantly lower( P < 0. 05, P <0. 01 ), compared with poor-differentiated adenoearcinoma and ca-ses with metastasis. The counts of lymph vessels in the NF-κB positive cases were significantly higher than thoseinNF-κBnegativecases[(14.9±4.1)/HPvs (9.8±3.1)/ HP, P <0.01] . Conclusions The expression of NF-κB and the counts of lymph vessels might be important markers to be used to monitor the progression, biological behaviors, metastatic status and prognosis of laryngeal carcinoma. NF-κB might pro-mote lympoangiogenesis in laryngeal squnmous cell carcinoma tissues.

ObjectiveTo study the expression of ABCG2, SFRP2, BRMS1 and HPA and detect their clinicopathologicalsignificancesinthebenignandmalignatntlesionsofthegallbladder.MethodsEnVisiom immunohistochemical method for determining the expressions of ABCG2, SFRP2,BRMS1 and HPA was used in paraffin-embedded sections of surgical resected specimens from gallbladder adenocarcinoma (n =108), peritumoral tissues (n =46 ), adenomatous polyp (n =15 ), and chronic cholecystitis ( n =35 ).ResultsThe positive rates of ABCG2 and HPA expression were significantly higher in gallbladder adenocarcinoma than that in peritumoral tissues, adenomatous polyp and chronic cholecystitis (P < 0. 05 or P < 0. 01 ) ; The positive rates of SFRP2 and BRMS1 expression were significantly lower in gallbladder adenocarcinoma than that in peritumoral tissues, adenomatous polyp and chronic cholecystitis(P <0. 05 or P <0. 01 ). The positive cases of ABCG2 and/or HPA as well as negative ones of SFRP2 and/or BRMS1in the benign lesions showed moderately-or severely-atypical hyperplasia of gallbladder epithelium. The frequency of samples with positive staining for ABCG2 and/or HPA in cases with small tumor volume (diameter < 2 cm), no lymph node metastasis, and no invasion into surrounding tissues was significantly lower than that in cases with larger tumor volume (diameter> 2 cm ), lymph node metastasis, and invasion into surrounding tissues ( P < 0.05 or P < 0. 01 ). However, compared with ABCG2 and/or HPA, the expression of SFRP2 and/or BRMS1 showed an opposite correlation in these cases ( P < 0. 05 or P <0. 01 ). Unitivariate Kaplan-Meier analysis showed that increased expressions of ABCG2 (P =0. 019) and HPA ( P =0. 016) or decreased expression of SFRP2 ( P =0. 019) and BRMS1 ( P =0. 008 )were associated with poorer overall survival, respectively. Multivariate Cox regression analysis showed that increased expression of ABCG2 (P =0. 018 ) and HPA ( P =0. 019) and/or decreased expression of SFRP2 (P =0. 015 ) and BRMS1 ( P =0. 011 ) were independently poor-prognostic predictors in gallbladder adenocarcinoma.ConclusionsThe expression of ABCG2, SFRP2, BRMS1 or HPA might be closely related to the carcinogenesis, clinical biological behaviors, and prognosis of gallbladder adenocarcinoma.
