1.Expression of Smad4 protein and mRNA in different developmental stages of the rat ovary

0.05). The staining intensity of Smad4 in follicles changed in relation with their development. Its expression in oocytes of antral and mature follicles was significantly decreased, as compared to that of preantral follicles (P0.05). The RT-PCR demonstrated that Smad4 mRNA was expressed in all developmental stages of the rat ovary, and from the 3rd week on, the expression of Smad4 mRNA was significant higher than that of the 1 day postnatal. CONCLUSION: The expression of protein and mRNA of Smad4 in the rat ovary indicate that TGF-?_s may regulate the folliculogenesis by Smad signal transduction model.
2.Experience and improvement in establishing an abdominal working heart transplantation model in rats

Objective To modify the techniques for establishment of an abdominal working heart transplantation model in rats and to sum up the key factors to success.Methods A total of 180 12-week old Brown Norway rats ( donor) and Lewis rats ( recipient) were used in this study:50 BN rats and 50 Lewis rats for pilot experiment, and 40 BN rats and 50 Lewis rats for the formal experiment.The rat model of working heart heterotopic transplantation was adopted and estab-lished by Wiedemann’ s mode.We transplanted the heart from BN rats to Lewis rats and analyzed the survival rate, causes of death and histological changes of the heart ( HE staining) in this experiment.Results After exercise and modification, the survival rate was increased to 77.5%, and the mean total duration of operation was 71 ±11 min, and the mean ische-mic time of the donor hearts was 34 ±5 min.Histological examination ( HE staining) of the cardiac allograft showed a mild inflammatory cell infiltration in the graft at 24 h after transplantation, indicating that the model was reliable.Conclusions A variety of factors may affect the final operation success rate in the establishment of this heart transplantation model.A-mong them, the major affecting factors include: healthy animals, donor heart protection, rapid and effective vascular su-ture, and postoperative animal management.
3.CT Diagnosis of the Cavitating Lung Cancer

Objective To improve the recognization of the cavitating lung cancer in CT scans.Methods From May 1996 to June 2000,37 patients out of total 271 primary lung cancers were confirmed by transthoracic needle aspiration biopsy,operation and pathlogical examination.Results There were 5 pattens cavitaties in the CT manifestations.They were 15 cases cavities with thick irregular walls,14 cases round or ovoid growths with central or eccentric cavitation,4 cases solid lobes containing cavities,2 cases thin-walled cavities resembling cysts and 2 cases fissural cavities.There were 24 cases squamous-cell carcinomas,5 cases adenocarcinomas,2 cases small-cell carcinomas,2 cases alveolar-cell carcinomas.Conclusion The cavitating lung cancer is made misdiagnosis easilier.There is important clinical value in diagnosis with transthoracic needle aspiration biopsy(TNAB).

Objective Amniotic membrane as the carrier to culture the vascular endothelial cells was investigated in this study in order to explore whether the corneal endothelial cells superseded by the vascular endothelial cells is feasible on the account of directing a kind of the original method to handle the bullous keratopathy.Methods The vascular endothelial cells adhering to the sphagitides lumen of the experimental rabbits were digested and gained by means of the perfusion method with 0.25%tripsin plus 0.02%EDTA.Fresh amniotic membrane without the amniotic epithelial cells was cut into the square tissue about 1.5 cm?1.5 cm and spread evenly in the 24-well culture dish.Primary cultured cells were subcultured on the amniotic membrane.We would not transplant the vascular endothelial cells feeding on the amniotic membrane until cells is full of the whole amniotic membrane surface.One to two months after the operation,the change of corneal diaphaneity was observed.Results About 12 days since the cell transplantation was performed,the corneal transparency alteration between the experimental groups and the control one is different.The corneal buttons in the experimental group show the severe edema and opacity,and the anterior chamber couldn't be seen unclearly.But,10 days after the operation,the corneal oedema which begins to extenuate was judged through the indicatrix that the corneal edema in the experimental group has been recovering slowly,among which the anterior chamber tissue of 7 animals was visible through the implant.The corneal edema in the control groups intensified evidently,even,the part of these appeared the ulcerous necrosis as result of the corneal severely edema.There is the existence of difference between two groups(P
6.Effect of soy isoflavone on incretion of female rats

Objective To observe the effect of soy isoflavones on incretion of female rats. Methods 42 fe-male rots of 12 weeks old were divided into 3 groups at random. Low dose group were perfused with 40mg · kg-1·d-1 into stomach per day;high dose group were perfused with 80mg·kg-1·d-1 into stomach per day;control group were perfused with physiological saline into stomach. After 14 days,collected blood via jugular vein and tested 4 inde-xes-FSH, LH,E2 and P with chemoluminescence method. Results In 3 groups of soy isoflavones low doee group,soy isoflavones high dose group and control group,FSH were (0.13±0.021) mIu/ml, (0.12±0.018) mIu/ml, (0.15 ±0.024) mIu/ml respectively; LH were (0.17±0.032) mIu/ml, (0.15±0.043) mIu/ml, (0.18±0.047) mIu/ml respectively, which has no obvious differences (P > 0.05) ; E2 were (0.09±0.03) nmol/L, (0.03±0.03) nmol/L, (0.12±0.04) nmol/L respectively; P were (1.43±0.27) ng/ml, (2.82±0.37) ng/ml, (0.67±0.56) ng/ml re-spectivdy. Compare those 3 groups, E2 activeness of soy isoflavones group decreased obviously; but P activeness of soy isoflavones group increased obviously. Conclusion Soy isoflavones has no obvious effect on hypophysis hormone of rat, but the soy isoflavones of different dosages may measure the secretion of female rat ovary hormones by estrogen ac-tivity and antiestrogen activity.

Objective:To investigate the effect of CpG ODN on the immune responses and immune-contraception induced by ZP~(121-140) synthetic peptide.Methods: BALB/e mice were given an injection into the left tibialis anterior muscle of ZP~(121-140) synthetic peptide with 20 μg CpG ODN or CFA,then the mice were given other injections at 2,4,6 weeks using the same formulation.The mice' s blood was collected before each vaccination and after the last vaccination every 2 weeks.The specific IgG and IgA in sere and non-specific cytokines IFN-γ,TNF-α,IL-10 in vaginal mucosa were measured by ELISA.The ovarial pathological changes were undertaken using hemattxylin and eosin-stained paraffin sections.Results:The specific IgG in sera and IgA in vaginal mucosa induced by ZP~(121-140) synthetic peptide combined with CpG ODN were no more than those of ZP~(121-140) synthetic peptide combined with CFA.There were significant increases in IFN-γ,and TNF-α when CpG ODN was mixed with ZP121-140 synthetic peptide and the increase of CpG ODN was more significant than that of CFA.Otherwise there was a significant decrease in IL-10 when CpG ODN was mixed with ZP~(121-140) synthetic peptide and the decrease of CpG ODN was more significant than thai of CFA.There was no significant difference in the rate of pregnancy between CpG ODN group and CFA group,but the average number of birth mice in CpG ODN group was less than that in CFA group.No pathological changes were found in the ovaries of experimental mice.Conclusion: The adjuvant effect of CpG ODN is more advantageous than that of CFA in contraception vaccine research.
8.An improved method for preparation and evaluation of a porcine model of acute myocardial infarction by minimally invasive surgery and left circumflex artery ligation

Objective To develop a better method for preparation of porcine model of acute myocardial infarction by permanent occlusion of the left circumflex coronary artery and minimally invasive surgery, evaluate its validity and stability, and explore its application in experimental studies of ischemic heart diseases.Methods 25 healthy female 3-month-old Bama minipigs, body weight 25±3 kg, were used in this study.The porcine model of myocardial infarction was established by minimally invasive surgery and the left circumflex artery ligation at the site of OM1 posterior position under general anesthesia.Heart function was assessed by echocardiography at 15 min before surgery, 1 hour and 4 weeks after surgery.Pathological examination was performed at 4 weeks after the left circumflex artery occlusion.The mortality and cause of death were statistically analyzed.Results The 1-hour and 4-week postoperative cardiac function was considerably decreased, showing a decreased ejection fraction from 64.2±4.6% to 48.2±5.3% (1hour after MI) and 49.7±6.1% (4 weeks after MI) (P<0.01).Pathological examination revealed that the ventricular wall was thinner and the amount of collagens was increased in the infracted area.The ventricular fibrillation rate at 1-hour after myocardial infarction was 17.3% and the infarction area was 19.2%.Conclusions A pig model of acute myocardial infarction can be prepared by our modified left circumflex coronary artery ligation at the obtuse marginal artery (OM1) and minimally invasive surgery.This model exhibits advantages such as minimal surgical trauma, high stability of the model, and low mortality, therefore, provides an ideal and economic animal model for experimental studies on acute ischemic heart diseases.
9.Effect of the ioversol on early renal function in patients undergoing cerebral vascular intervention

Objective To investigate the effect of different doses of ioversol on renal function,and to explore early renal injury biomarkers on contrast induced kidney injury and safe ioversol dosage.Methods A total of 158 cases (98 males and 60 females) undergoing cerebral vascular intervention (CVI) in our department was selected with age ranging from 23 to 81 years old (average age 59.70 ± 12.02).Based on ioversol dosage in surgery,patients were divided into three groups:low dose group (≤ 150 ml,n =49),middle dose group (151-200 ml,n =74),and high dose group (>200 ml,n =35).U-κ,U-λ,urinary transferrin (UTRF),urine microalbumin (UMA),urinary immunoglobulin IgG (UIgG),urine beta2-microglobulin (Uβ2-MG),Uα1-MG,urinary N-acetyl-beta-D-glucosaminidase (UNAG),plasma cystatin C (CysC) and Scr were detected by scattering turbidimetry,immune turbidimetry and fully automatic biochemical analysis pre-surgery 24 h and post-surgery 72 h.Contrast-induced acute kidney injury (CI-AKI) was defined as laboratory increase of Scr value≥44.2 μmol/L or ≥25% from baseline measurement at 48 hours after surgery.The relationship in ioversol dosage and various factors was assessed by Single and multiple factors binary logistic regression analysis.Results According to the criterion that Scr increase value were ≥44.2 μmol/L,of 158 cases,3 cases occurred CI-AKI,the AKI incidence was 1.90%.Based on the criterion that Scr increase value was ≥25%,33 cases occurred CI-AKI,the incidence was 20.89%.The concentration of U-κ,UTRF,Uα1-MG,UNAG and plasma CysC were significantly different in high dose group compared to low ioversol dose group (P < 0.05),while the other biomarkers had no significant difference (P > 0.05).Conclusions The contrast media-ioversol could lead to CI-AKI;when the dosage of ioversol was more than 200 ml one-time,the concentration of U-κ,UTRF,Uα1-MG,UNAG and plasma CysC increased significantly.U-κ,UTRF,Uα1-MG,UNAG and plasma CysC could predict the early renal injury in patients who undergoing CVI.The rise of U-κ,UTRF,Uα1-MG,UNAG and plasma CysC are related to the dosage of ioversol.Furthermore,possibility of kidney injury is significantly high when ioversol dosage is more than 200 ml one-time.
