1.Acupuncture combined with local anesthesia for percutaneous vertebroplasty.

OBJECTIVETo observe the analgesic and sedative effects of acupuncture combined with local anesthesia for percutaneous vertebroplasty (PVP).
METHODSSixty patients of single segmental osteoporotic vertebral compression fractures who were prepared to receive PVP were randomly divided into an observation group, a control 1 group, a control 2 group, 20 cases in each group. The patients in the observation group were treated with electroacupuncture (EA) at Hegu (LI 4), Neiguan (PC 6) and Zusanli (ST 36) 20 min before operation; during operation, EA was given combined with regular anesthesia. The patients in the control 1 group were treated with intramuscular injection of parecoxib sodium (40 mg), combined with regular anesthesia. The patients in the control 2 group were treated with intravenous injection of dezocine (5 mg), combined with regular anesthesia. Visual analogue scale (VAS) and Ramesy sedation score were compared among the three groups.
RESULTSIn the observation group and control 2 group, the VAS during puncture and bone cement placement was higher than that before acupuncture (all <0.01); the VAS during bone cement placement was higher than that before puncture (<0.05, <0.01); the VAS after operation was lower than that during puncture and bone cement placement (<0.05, <0.01). In the control 1 group, the VAS during puncture and bone cement placement and after operation was higher than that before acupuncture (<0.01, <0.05), the VAS after operation was lower than that during puncture and bone cement placement (<0.05, <0.01). There was no significant difference in VAS and Ramesy score among three groups at all time points (all >0.05).
CONCLUSIONCompared with local anesthesia and analgesics, acupuncture combined with local anesthesia has similar analgesic and sedative effect for PVP, which could be considered a better method for PVP anesthesia.
2. Preparation and pharmacokinetics of gambogic acid long-circulating liposomes

Objective: To optimize the preparation process of gambogic acid (GA) liposomes and study the in vitro and in vivo release. Methods: The detection method of GA was established, using the Box-Behnken experiment design to optimize liposomes formula, GA liposomes were obtained with the highest encapsulation efficiency; Using scanning electron micrographs (SEM) to observe liposome surface morphology, using the dialysis method to study the liposome release in vitro, we also measured the stability of liposome in 15 d; Male Wistar rats were injected with GA or GA liposomes (1 mg/mL) via tail vein, UPLC-MS/MS method was used to determine the drug concentration, and differences in pharmacokinetic parameters of the two drugs were compared. Results: After Box-Behnken optimization, the encapsulation efficiency of liposomes was 92.3%, and the optimized liposomes formula is cholesterol of 440 mg, egg phosphatidylcholine of 1823 mg,,and istearoyl phosphoethanolamine-PEG 2000 of 705 mg, liposomes had uniform particle size and smooth surface; In vitro release results showed that the liposomes could be gentle and slowly release and had a long- term effect. The liposomes were stable keeping in 4 ℃ within 15 d; In in vivo study, the half-life of GA liposome was 9.97 h, 4.43 times of GA; AUC0-24 h of GA liposome was 22.55 μg∙h/mL, 4.73 times of GA. Conclusion: Compared with GA, GA liposome has the characteristics of long-circulating, high blood drug concentration, and could release smoothly.
3. Research progress of Hedgehog signaling pathway in renal fibrosis

Renal fibrosis is a common pathological manifestation of most chronic kidney disease (CKD) and a major pathological change leading to end-stage renal disease. Recent studies have found that the Hedgehog (Hh) signaling pathway plays an important role in renal fibrosis. This pathway is different from the TGF-β and Wnt pathways that have been studied in the past. Its activation mainly directly targets the central event of renal fibrosis-activation, proliferation and transformation of myofibroblasts. It suggests that this pathway plays an important role in the process of renal fibrosis, and this pathway interacts with other pathways to promote the development of renal fibrosis. Further research into this pathway may provide new ideas for the treatment of renal fibrosis. This article reviews the relationship between Hh signaling pathway and renal fibrosis.
4. Application of flipped classroom based on Moodle in the teaching of gynecology and obstetrics nursing

The traditional teaching method of the medical curricculum, gynecology and obstetrics nursing has many problem. The flipped classroom subvetted the traditional teaching structure, and realizes the advance of knowledge transfer and optimization of knowledge internalization. The research group made design, recorded micro-course video and used the free and open source of Moodle course management platform to implement flipped classroom. The interactive function of the platform released learning tasks before class, had discussion in class and answered after-class quesstions. The subjectivity of students was highly emphasized. The results showed that the flipped classroom can improve students' academic performance, develop students' good study habits and learning interest, and also bears significance for the cultivation of autonomic learning ability and critical thinking.
5.Effect and safety analysis of Allergovit standardized mite allergen immunotherapy in patients with allergic rhinitis.

OBJECTIVE:
To evaluate the effect and safety analysis of Allergovit standardized dust mite subcutaneous immunotherapy in patients with allergic rhinitis.
METHOD:
From Jan 2006 to Jan 2009,425 patients with persistent were treated with Allergovit standardized dust mite allergen in 3 years, which data were retrospectively analyzed. The scores of the rhinoconjunctivitis quality of life were collected per-treatment and three years treatment. VAS (visual analogue scale) of all nasal symptoms and drug use scores were evaluated before treatment and every 6 weeks. The frequency of local and systemic reactions were recorded in the duration of dose escalation and maintenance.
RESULT:
The Overall QOL were significantly improved after 3 years treatment [pre-treatment: 3.8 (2.9; 4.5); post-treatment: 1.1 (1.0; 2.0), P < 0.01]. VAS of all nasal symptoms and drug used scores decreased dramatically after escalation period [pro-treatment: 23. 7 (20.5; 27.8), 3.9 (3.2; 4.7); post-treatment: 2.1 (1.6; 2.2), 0(0;0), P < 0.01]. Local reaction's rate was 3.15%, the frequency systemic reaction's rate was 0.90%.
CONCLUSION
The standardized specific allergen immunotherapy for allergic rhinitis is safe and effective.

6. A follow-up study of abnormal mutation in neonatal deafness gene screening

Objective:
To screen, diagnose and follow up the abnormal mutation in the gene screening of neonatal deafness.
Methods:
A total of 24161 newborns born in Zhuhai Maternal and Child Health Hospital from February 1, 2015 to January 31, 2008 were screened for hearing and deafness genes, and audiological screening, diagnosis and 1-3 years follow-up were carried out for the newborns with positive gene screening.
Results:
There were 991 cases of deafness gene mutation (533 males and 458 females), and the rate of abnormal mutation was 4.10%(991/24 161). Among them, 921 cases were single heterozygous mutation, 130 cases were failed in primary hearing screening, 11 cases were failed in secondary hearing screening, 8 cases were abnormal in audiological diagnosis finally. In these 8 cases, 3 were diagnosed as otitis media and passed audiological follow-up after cure, 2 cases of single ear sensorineural injury caused by high-risk factors, passed after close audiological follow-up, and the other 3 cases were closely audiological follow-up while none of them were successfully sequenced. 7. Molecular epidemiological study on hepatitis C virus among drug users in Zhuhai, China

Objective:
To investigate the molecular epidemiology of hepatitis C virus (HCV) among drug users in Zhuhai, China.
Methods:
Anti-HCV and HCV RNA were detected by using ELISA and real-time RT-PCR respectively. HCV genotypes and subtypes were determined by direct sequencing of amplicons in partial NS5B regions and BLAST GenBank, followed by phylogenetic analysis.
Results:
Of 435 blood plasma from drug users, 242 was negative for anti-HCV and HCV RNA, 147 were positive for anti-HCV and HCV RNA, 46 positive HCV RNA or anti-HCV. The infection rate of HCV was 44.37%. Three genotypes and five subtypes of HCV were detected in 78 samples of positive HCV RNA. Subtypes 6 a, 3b, 3 a, 1b and 1 a, were detected at frequencies of 60.26%, 15.38%, 14.10%, 6.41% and 3.85%, respectively. Compared with strains from Shenzhen, Foshan, Guangzhou, Shanwei, Hong Kong, Denmark, Canada, USA, United Kingdom, India and Nepal, HCV among drug users in Zhuhai were homophylic.
Conclusions
There were homology among HCV in drug users from Zhuhai and strains from some regions and countries. Subtype 6 a was the most predominant. Subtype 1 a and 3 a were discovered in Zhuhai.
8. Bacteriostatic effect of ε-polylysine and its effect on urine lead level

OBJECTIVE: To evaluate the bacteriostatic effect of ε-polylysine( ε-PL) on four common putrefactive bacteria including Staphylococcus aureus,Enterococcus faecalis,Pseudomonas aeruginosa,and E. coli and its effect on urine lead level. METHODS: Broth dilution method was used for the determination of the minimum inhibitory concentration( MIC) ofε-PL on the four kinds of putrefactive bacteria; the inhibitory effects of ε-PL with final mass concentration of 40. 000 mg / L on the urine sample were observed; graphite furnace atomic absorption spectrometry was used for determining the lead level in the 40. 000 mg / L( mass concentration) ε-PL solution and the urine lead level in normal healthy groups; the bacteriostatic effects of ε-PL and nitric acid were compared. RESULTS: The MIC of ε-PL on Staphylococcus aureus,Enterococcus faecalis,Pseudomonas aeruginosa,and E. coli was 40. 000 mg / L. There was no bacterial growth in the urine sample with40. 000 mg / L( mass concentration) ε-PL when urine was kept at room temperature for 24 hours to 15 days. The lead level was < 2. 0 μg / L in the 40. 000 mg / L( mass concentration) ε-PL solution. When the ε-PL with final mass concentration of 40. 000 mg / L and the nitric acid with a volume fraction of 1. 0% were respectively used as the antiseptics,the descending rates of the lead levels in the urine samples were similar,and after the urine sample was preserved for 15 days,the descending rates of the urine lead were both smaller than 10. 0% after be stored for 15 days. CONCLUSION: ε-PL can substitute nitric acid as a new natural preservative for preservation of samples for urine lead determination.
9.Establishment of a quantitative real-time PCR for rapid detection of Panax quinquefolius.

In this study, the specific primers and probes of Panax quinquefolius were designed for a quantitative real-time PCR, and the rapid identification method of P. quinquefolius was established by optimizing conditions. The method was used to validate 43 samples of the traditional Chinese medicine,and the results showed that 22 samples of P. quinquefolius were identified accurately. The limit of detection of the method can be reach to 1×10⁻⁴ ng. The method is accurate, fast, sensitive and specifically.
