1.Comparison of formation and morphologic features of digestive vacuoles and haemozoin between chloro-quine-sensitive and chloroquine-resistant strains of Plasmodium berghei

Objective: To understand the morphologic foundation for differences in drug-resistance, virulence and immunity between chloroquine-sensitive (N) and chloroquine-resistant (RC) strains of Plasmodium berghei. Methods: The RC strain and the N strain were compared concerning the formation and morphologic features of digestive vacuoles and haemozoin with transmission electron microscope. Results: There was a single large digestive vacuole and multiple micro-single-mem-braned vacuole-like structures in the trophozoites of the N strain, and haemozoins centralized and fused during their schizo-gony were situated under the plasma membrane. Whereas there were few digestive vacuoles in the trophozoites of the RC strain, but with multiple micro-single-membraned vacuole-like structures instead. The RC strain formed obviously less hemo-zoins than that of the N strain and the hemozoins were not centralized and fused during the schizogony. Conclusion: The RC strain forms multiple single-membraned food vacuole-like structures in the trophozoites, and has different mechanism for detoxifying free heme with N strain and the features may be the foundation for the difference in drug-resistance, virulence, immunity between RC strain and N strain of Plasmodium berghei.
2.The Clinical Study of the H1N1 Influenza Virus with Fanggan Decoction

[Objective]To discuss the clinical effect of Fanggan decoction on H1N1 Influenza virus.[Method]Randomly divided 95 cases into 2 groups,treatment group 48 and control one 47.Treatment group takes Fanggan decoction,the control one uses reducing fever tablet or anti-virus drugs if needed.[Result]The effective rate was 89.6% for treatment group and 85.1% for the control one.The treatment group was better than the control one on the reducing fever time,cough duration and cold symptom time.[Conclusion]Fanggan decoction has a good effect on H1N1 influenza virus.It also can be one of the therapies to relieve influenza patients in Zhejiang Province.
3.Comparative studies on codon usage bias of Ganoderma lucidum based on analysis of genomic and transcriptomic data.

Codon usage bias is an important characteristic of genetic information transfer in organisms. Analysis of codon usage bias of different species is important for understanding the rules on genetic information transfer. The previous method for analysis of codon usage bias is mainly based on genomic data. However, this method is greatly limited, because the genome sequences of higher organisms are still not available up to now. In this study, we found that we could obtain the same optimal codons of Ganoderma lucidum (Curtis: Fr.) P. Karst based on its whole genomic data or large-scale transcriptomic data from its liquid-cultured hyphae, primordium and fruiting body, separately. This result indicated the feasibility to understand the codon usage bias based on the large-scale transcriptomic data. By calculating the proportion of rare codons of Escherichia coli and Saccharomyces cerevisiae in 26 terpene synthases (TS) of G. lucidum, we found that the rare codons of S. cerevisiae have a higher proportion in TS genes, while the rare codons of E. coli have relatively lower, suggesting that the TS genes of G. lucidum are possibly more difficult to be expressed in S. cerevisiae than in E. coli. Chemical synthesis of TS genes according to the yeast optimal codons will be an effective way to solve the problem on the mismatch of gene codon bias between the foreign genes and the host strain.
4.Prognostic value of initial arterial lactate and 6-h lactate clearance rate in acute paraquat poisoning

Objective To investigate the correlation of the initial arterial lactate (Lac) and 6-h lactate clearance rate (LCR) with prognosis in patients with acute paraquat poisoning (APP).Methods The included 132 APP patients (70 males and 62 females,age ranging 15-71,30 (18) [M (IQR)]treated at a single center between January 2009 and December 2013.Patients were divided into two groups:group A,survivors > 28 days after admission (n =64); and group B,those died ≤ 28 days after admission (n =68).The prognostic values of arterial Lac and 6-h LCR during the acute stage of poisoning were evaluated.Results Paraquat dose ranged from 5-200 mL,20 mL (37.25 mL) [M (IQR)].The average time from poisoning to arrival at the emergency department was ranging 4-312 h,6 h (8.75h) [M (IQR)].Total mortality was 51.51%.There were no differences in age,gender,and length of time elapsed from poisoning to diagnosis between two groups.Survivors had a significantly lower dose of paraquat ingested compared with nonsurvivors (P < 0.05).An ROC curve analysis determined that the dose had an area of 0.86 (95% CI:0.80-0.92) and the volume cut-off point was 27.5 mL to predict the prognosis in patients with acute paraquat poisoning (75% sensitivity,85.9% specificity,Youden index 0.609).The initial arterial lactate level was higher in nonsurvivors than that in survivors.The ROC curve analysis indicated that the initial arterial lactate level had an area of 0.99 (95% CI:0.99-1.00) and the concentration cut-off point was 5.050 mmoL/L to predicti prognosis in patients with acute paraquat poisoning (sensitivity 98.5%,specificity 100%,Youden index 0.985).The 6 h LCR was lower in nonsurvivors than that in survivors.ROC curve analysis showed that the 6-h LCR had an area of 0.99 (95 % CI:0.97-1.00) and the concentration cut-off point was 17.28% to predict prognosis in APP patients (sensitivity 100%,specificity 97.1%,Youden index 0.971).Conclusions In the early stages of APP,initial arterial Lac and 6-h LCR are closely related to prognosis and may serve as prognostic factors.
5.Establishment of a transgenic zebrafish line with tdTomato expression in the ultraviolet-sensitive cone photoreceptors

Objective To better understand the mechanisms of cone opsin transport , we set to generate a trans-genic zebrafish line with red fluorescence proteins expressing in the cone photoreceptors .Methods We used sws1 promot-er to drive the expression of a chimerical protein , in which the last 44 amino acids of rhodopsin of Xenopus laevis were fused to the C-terminus of tdTomato to restrict its localization to the outer segment of photoreceptors .Results We successfully i-solated such a transgenic zebrafish line and confirmed the localization of tdTomato by immunostaining analysis .Conclu-sions This transgenic zebrafish line will help us to better understand the transport mechanisms of cone opsins , especially the transport in live photoreceptors .
6.The role of 5-HT in antinocieption produced by intrathecal indomethacin in mice

AIM To study the role of 5-HT in antinocieption produced by intrathecal indomethacin (Ind) in mice. METHOD The antinocieption of indomethacin was investigated on the tail immersion test in mice;the contents of 5-HT, 5-HAA were assayed with fluorescent method. RESULT Dose-dependent antinocieption was observed following intrathecal administration of Ind 1.8 mg*kg-1, the effect could be obliterated when the animals were pretreated with cyproheptadine. After intrathecal administration of Ind, the content of 5-HIAA in the spinal cord of mice was significantly increased, but PG had no effect. CONCLUSION The result imply that intrathecal indomethacin could produce antinocieption; this effect is mediated by 5-HT. PG does not participate in the action of 5-HT.

8.Methods on enhancing success rate and survival rate of rat model of myocardial infarction

Objective To investigate the effects of different ligation position,vena coronaria variation and lidocaine on myocardial infarction size and mortality in rats Methods Totally 120 male SD rats were divided into four groups with 30 rats in each group: low ligation with invisible vena coronaria,low ligation with visible vena coronaria,high ligation,and high ligation plus lidocaine Results MI was successfully created in 930% rats of high ligation groups and in 673% rats of low ligation groups The mortality was 433% in rats of high ligation groups,only 133% in rats when lidocaine in use Conclusion High ligation increases the success rate of rat model of myocardial infarction and the application of lidocaine decreases the mortality of the rat model
9.Risk factors complicated cardiovascular and cerebrovascular diseases in patients with refractory hypertension

Objective: To explore risk factors for complicated cardiovascular and cerebrovascular diseases in patients with refractory hypertension (RH).Methods: Clinical data of 536 RH patients, who were treated in our hospital from Jun 2010 to Jul 2015, were retrospectively analyzed.According to presence of cardiovascular and cerebrovascular diseases or not, patients were divided into complication group (n=217) and pure RH group (n=319).Results: Compared with pure RH group, there were significant rise in percentages of drinking and smoking, hypertension course, mean arterial pressure (MAP), mean systolic blood pressure, pulse pressure, serum uric acid (SUA), creatinine (Cr), total cholesterol and low density lipoprotein cholesterol (LDL-C), and significant reductions in high density lipoprotein cholesterol (HDL-C) mean diastolic blood pressure and mean heart rate in complication group, P<0.01 all.Multi-factor unconditional Logistic regression analysis indicated that hypertension course, SUA,MAP, Cr and LDL-C levels were independent risk factors for complicated cardiovascular and cerebrovascular diseases in RH patients (OR=1.014~3.217, P<0.05 or <0.01).Conclusion: Hypertension course,MAP,Cr,SUA and LDL-C are independent risk factors for refractory hypertension.Enhancing their monitor, prevention and treatment are important to reduce incidence rates of cardiovascular and cerebrovascular complications in patients with refractory hypertension.
10.Study progress on antitumor activities of artemisinin and its derivatives

The antitumor activities of artemisinin and its derivatives has become a hot topic in recent years.Artemisinin and its derivatives have been reported to exert antitumor effects mainly by blocking the cell cycle, promoting tumor cell apoptosis, inhibiting angiogenesis and so on.Also, their influence on signaling pathways has attracted considerable interest of many scholars.Currently, in order to discover some compounds with better therapeutic potentials, many studies dedicated to the synthesis and development of new artemisinin derivatives.In addition, the effects of artemisinin and its derivatives on multi-drug resistantace in tumor cells also become a trend of the future research.In this paper, we review the research advances in the antitumor activities of artemisinin and its derivatives during the past five years to provide a reference for future study .
