Objective To investigate clinical efficacy of different ventilator parameters on acute left heart failure.Methods One hundred and nineteen patients with acute left heart failure were randomly divided into A,B,C,D group.Patients in A,B,C,D groups were treated separately with higher air pressure,high pressure,medium pressure or low pressure respectively.In each group,clinical efficacy,heart rate (HR),arterial partial pressure of oxygen (PaO2),pulse oxygen saturation (SpO2),partial pressure of carbon dioxide (PaCO2) and pH were measured.Results There were significant differences among the four groups in terms of HR,PaO2,SpO2,PaCO2 and pH (F =32.21,29.46,12.34,36.47,18.38 ; P ＜ 0.05).Compared with that in A,C and D group,the levels of HR,PaO2,SpO2,PaCO2 and pH in B group were significant difference (P ＜ 0.05).Meanwhile those indices in B group were difference among before and after treatment (t =12.25,13.46,8.75,9.83,2.58 ;P ＜ 0.05).The efficacy rates of four groups were significant different (x2 =0.97,P =0.012),and in B group was 93.1％,higher than that of other 3 groups(71.0％,65.6％,51.8％ ;P ＜0.05).Condusion The ventilator parameters(10-15 cmH2O,55％-75％ FiO2 at beginning and later revised the FiO2 to 30％-40％) was proved to be worthy in treating acute left heart failure.

Objective To study the seasonal change of antibodies levels in the serum of asthma patients and Der P1 concentration in the dust samples collected from their bedrooms. Methods 36 asthma patients who were allergic to mites were treated as the patients group,21 male and 15 female,aged 43 years of average age. 20 healthy individuals were treated as the control group,11 male and 9 female,with the same age as the patients group. In September and December in 2005,and Match and June in 2006,the dust samples in the bedrooms of patients and the venous blood were collected,the samples of the control were collected only in September. Der P1 concentrations in dust samples and the levels of total IgE,mite specific IgE (s-IgE),s-IgG1,s-IgG2,s-IgG3 and s-IgG4 were detected by ELISA assay. Objective To investigate the expression of kallikrein 9 (KLK9) in liver cancer and to determine its significance.Methods The expression of KLK9 in liver cancer was detected by immunohistochemistry and RT-PCR techniques.Results The rate of expression of KLK9 protein in liver cancer tissues was significantly higher than paracarcinoma tissues and normal liver tissues (P ＜ 0.05).The expression of KLK9 mRNA in liver cancer cells was significantly higher than normal liver cells (P ＜ 0.05).The expression of KLK9 was related to metastasis,size of tumors,degree of malignancy and clinical staging of the liver cancer (P ≤ 0.05),but there was no associated with age,HbsAg and sex (P ＞ 0.05).Conclusions KLK9 may play an important role in the occurrence and development of liver cancer.It may be used as a tumor marker and a prognostic factor.It may also provide a theoretical basis for the diagnosis and biological targeted therapy of liver cancer.

Aim To study the protective effect of 6-Hydroxy-1 H-indazole via inhibition of Tau phosphoryla-tion on MPP +-induced SH-SY5Y cells apoptotsis. Methods An apoptotic model induced by 1 -methy-4-phenylpridnium ion (MPP +)was established in cul-tured human neuroblastoma SH-SY5Y in vitro.MTT a-nalysis was used to evaluate the protective effect of pre-treatment of cells with 6-Hydroxy-1 H-indazole for 2 h. Hoechst33258 staining was used to observe apoptotic situation.Immunohistochemistry was used to detect the p-Tau (Ser396)expression.Results The viability of cells exposed to 200 μmol · L -1 MPP + for 48h was (47.80 ±0.84)% (P <0.01 ),MPP + induced phos-phorylation of Tau at Ser396 and neuronal apoptosis, while 6-Hydroxy-1 H-indazole inhibited MPP +-induced cellular apoptosis,increased the number of TH-positive cells via inhibiting Tau phosphorylation.Conclusion These results indicate that Tau may be a new target used to cure neurodegenerative disorders such as PD.

The fused gene (PTH-HSA) of parathyroid hormone (PTH) gene and Human Serum Albumin(HSA) gene was amplified without linker by Overlapping PCR technology. The spliced gene was clone into Pichia pastoris secretory vector pPIC9K. With the help of promoter AOX1 and mat ? signal peptide, the PTH-HSA gene was designed to secretory expression.Linearized by restriction enzyme SalI, The recombinant plasmid pPIC9K/PTH-HSA was transformed into Pichia pastoris KM71 by electroporation. The recombinant strains which were identified by G418 and PCR analysis were induced by methanol to express protein PTH-HSA. The target protein was expressed in fermentation supernatant. Western blot analysis of the fusion protein showed that the expressed fusion protein PTH-HSA had the antigenicity of HSA.adenylate cyclase assay proved that the fused protein exhibited the bioactivity to stimulate cAMP synthesis The specific activity of broth was about 318IU/ml.

The content of bicarbonate ( HCO-3 ) and carbonate ( CO2-3 ) ions in groundwater and lake water reflects a broad set of carbon cycling reactions associated with decomposition or synthesis of organic compounds with mineral dissolution or precipitation, which indicates the local geochemical environment. However, the content of HCO-3 and CO2-3 changes easily under the influence of pH, temperature, atmosphere pressure in the process of sampling, transportation and storage, so it has been a worldwide problem to determine the real content of HCO-3 and CO2-3 ions in groundwater and lake water. This article proposed a new way to solve the problem by fast field detection of HCO-3 and CO2-3 ions through the use of pH electrode combined with carbon dioxide electrode. Studies showed in the base solution of pH=4. 8 ± 0. 1, the detection range of HCO-3 ion was 0. 027-570 mg/L and that of CO2-3 was 1. 25 ×10-8-39. 7 mg/L. In the most case, the coexisting ions and weak acid ( K+, Na+, Mg2+, Cl-, SO2-4 <100 mg/L;HSO-3 , NO-2 , HOAc<50 mg/L) did not interfere with the analysis. The method was validated for real water samples and the recoveries were in the range of 95. 2%-99. 2% with the relative standard deviations (RSDs) of 2. 6%-3. 7%. Compared with the acid-base titration method, the accuracy of this method had proved to be good. However, the method could be affected by temperature, so the standard solution and samples should be measured at the same temperature. Above all, this method is suitable for fast field analysis for HCO-3 and CO2-3 ions in the nature water as it is sensitive, fast, economical, and the electrodes are easy to carry and operate. It has been successfully applied in the determination of HCO-3 and CO2-3 in groundwater and lake water in Qinghai Province. Experiment showed that the pH of the groundwater samples from Haidong district was 6 . 4-7 . 4 , with 234-4096 mg/L HCO-3 and 0. 16-1. 89 mg/L CO2-3 . The pH of the lake water samples was about 8. 7, with 1. 36-1. 86 g/L HCO-3 and 32. 3-43. 9 mg/L CO2-3 , which was consistent with the previous results.

Objective:To study the clinical efficacy of adjuvant therapy with thymosin alpha -1 in patients with severe peritoneal cavity infection(sIAI),and to explore its effect to celluar immune function and T-like receptor.Methods: A total of 60 patients with sIAI,who received treatment in the intensive care unit of our hospital in January 2012-December 2014,were divided into observation group and control group.Cases in control croup received routine treatment ,and cases in observation group received routine treatment besides thymosin alpha-1.T-lymphocyte subsets were determined by flow cytometry and expression of Toll-like receptor (TLR)2 and TLR4 in the peripheral blood mononuclear cells were determined by real time PCR before treatment and in the 2nd week after treatment.The scores of acute physiology and chronic health evaluation ( APACHE)Ⅱscore and gastrointestinal function were recorded before treatment and in the 2nd week after treatment.The rate of systemic inflammatory response syndrome (MODS),the rate of death in hospital stays,intestinal function recovery and hospital stays in both groups were compared .Results: (1) CD3+,CD4+and CD4+/CD8+of cases in observation group significantly increased ( P<0.01 ) , and CD8+significantly decreased ( P<0.01 ) compared with those in control group.(2) The expression levels of TLR2 and TLR4 mRNA in PBMCs of cases in observation group significantly decreased compared with those in control croup (P<0.01).(3)The scores of APACHEⅡ and gastrointestinal function in observation group significantly decreased compared with those in control group ( P<0.01 ) .( 4 ) The recovery of intestinal function , vanishment of SIRS,hospital stays and the rate of MODS in observation group significantly decreased compared with those in control group (P<0.01), the rate of death in hospital stays had the trend of reduction with no statistic significance (P>0.05).Conclusion:Adjuvant therapy with thymosin alpha-1 in patients with sIAI has good clinical efficacy ,and can improve the cellular immune function ,decrease the expression of Toll-like receptor.
