Objective:To identify the effect of ?-synuclein overexpression on mitochondrial membrane structure with atomic force microscopy. Methods:?-syn expression was mediated by AAV (adeno-associated viral vector) and Recombinant AAV/?-syn and AAV/LacZ viral particles were stereotaxically injected in the left side of rat substantia nigra (SN) for rat model of ?-synuclein overexpression. Mitochondria were isolated from rats SN of Brain. Mitochondria were analysis with JC-1 staining, atomic force microscopy and Western blot. Results:By 16 weeks post-infection of AAV-?-syn,the level of ?-syn increased about 2 times in mitochondrial fraction with Western blot and mitochondrial membrane potential (??) decreased with JC-1 staining. Furthermore, mitochondria swelling and porous like structure formed on the mitochondrial membrane with atomic force microscopy. Conclusion:The data suggested that ?-syn could accumulate in mitochondria, might form mitochondrial membrane pores and lead to ?? decreases. ?-syn might lead to mitochondrial dysfunction in Parkinson's disease.

The complex level of constructing biopharmaceutics classification system of Chinese materia medica CMMBCS) was the study of traditional Chinese compound, on the premise of insisting that the multicomponent simultaneous determination, when carrying out the study of intestinal permeability, the primary task was to define the source of the components that was absorbed through the intestinal wall, namely, which medicinal material the components belonged to in traditional Chinese compound. The technology of chemical fingerprint and in vitro everted gut sac model were used in this research to make multicomponent an intuitive source attribution which permeated the intestine in the classic formula Gegen Qinlian decoction, and to lay the foundation for the further qualitative and quantitative research of intestinal permeability.

The Journal of Practical Medicine 2014;(19):3166-3169
Objectives To investigate the prevalance of quinolone resistance in Laribacter hongkongensis and to evaluate the influence of agar dilution and disc diffusion methods on susceptibility testing for the bacterium. Methods Susceptibility to quinolones of L.hongkongensis was tested by agar dilution and disc diffusion methods. The results for both methods were compared. Results L.hongkongensis isolates exhibited different susceptibility rate for quinolones (levofloxacin>ciprofloxacin>norfloxacin>nalidixic acid). All of the fish isolates were susceptible to levofloxacin and more than 90%were susceptible to ciprofloxacin and Norfloxacin. However,by agar dilution, only 62.50%, 37.50%,48.21%, 19.64%of the frogs isolates were susceptible to levofloxacin, norfloxacin, ciprofloxacin, nalidixic acid, respectively. For fish isolates, comparision of susceptibilities between agar dilution and disc diffusion showed a high (> 95%) percentage agreement for levofloxacin and ciprofloxacin and the highest crepancies were observed with norfloxacin (100%) .But it showed a high discrepancy when comparing the two methods for frogs isolates. Conclusions The resistance rate of L. hongkongens to quinolones was high , especially for frog isolates. The results suggested the use of agar dilution on susceptibility testing for quinolones in L. hongkongensis.

The effect of RNA interference-mediated adiponectin gene silence on glucose-lipid metabolism in apolipoprotein E gene knock-out(ApoE~(-/-)) mice was studied. The plasma adiponectin level in ApoE~(-/-) mice with high-fat diet and adiponectin short hairpin RNA (shRNA) adenovirus injection (ADI group) was significantly lower than that in ApoE~(-/-)mice with high-fat diet(HF group), normal-chow(NF group)and naked adenovirus injection (GF, both P<0.01). Fasting blood glucose, fasting plasma insulin, free fatty acids, total cholesterol, triglycerides, low-density lipoprotein-cholesterol (LDL-C), high-density lipoprotein-cholesterol (HDL-C), and body weight were significandy elevated in three high-fat diet groups compared with in NF group, (P<0.01). However, fasting plasma insulin, free fatty acids, total cholesterol, triglycerides, LDL-C, and HDL-C in ADI group were significantly higher than those in HF and GF groups(P<0.01 or P<0. 05). A typical clinical phenotype of glucose-lipid metabolism and insulin resistance in ApoE~(-/-) mice can be induced by high-fat feeding and adiponecting gene silencing.
