AIM: To study the effect of RNA interference on hypoxia-inducible factor-2(HIF-2) in the renal cell cancer in vitro and in vivo.METHODS: HIF-2 RNAi was synthesized and inserted into RNA interference eukaryotic expression vector which was confirmed by sequencing.The vector was transfected into the renal cancer cell 786-0 and positive clone was selected by using G418.The HIF-2 expression was detected by RT-PCR and Western blotting method.The growth of cells was measured by MTT method.Nude mouse xenograft assays were also done.RESULTS: Compared with empty vector group and control group,the amounts of HIF-2 mRNA and protein expression were lower in the HIF-2 RNAi group,the difference was significant(P
2.Association of clinical features with sex hormone levels in 63 azoospermic patients with microdeletion of azoospermia factor C region

Objective:To analyze the clinical features of 63 azoospermic patients with microdeletion of azoospermia factor C region(AZFc) and their relationship with sex hormones.Methods:The clinical data of 63 azoospermic patients with AZFc microdeletion were retrospectively analyzed, including age, height, body weight, body mass index(BMI), testicular volume, and sex hormones.Results:Compared with normozoospermic men, height, body weight, and testicular volume revealed statistical differences in azoospermic patients with AZFc microdeletion( P<0.05). There were significant differences in FSH, LH, testosterone(T), and prolactin levels between two groups( P<0.05). BMI was negatively associated with FSH level( r=-0.251, P=0.04)and positively associated with estradiol(E 2)level( r=0.327, P=0.009). Age had a negative association with T level( r=-0.256, P=0.04). Testicular volume revealed a negative association with FSH level( r=-0.289, P=0.02). There were no association of height with T, E 2, FSH, and LH levels( P>0.05). Conclusion:It is more likely that azoospermic patients with AZFc microdeletion present overweight and earlier decreased androgen level. The adjustment of lifestyle and testosterone supplementation might be beneficial to the long-term quality of life in these patients.
3.Effect of antisense VEGF gene on VEGF expression and growth of human renal cell carcinoma

Objective To study the effect of human antisense VEGF gene on VEGF expression and growth of renal cell carcinoma Methods Reverse transcription-polymerase chain reaction (RT-PCR) for VEGF,VEGF was inserted into eukaryotic expression vector pcDNA3.1 to construct eukaryotic expression vector carrying human VEGF cDNA,then using restrict enzyme to confirm the result.The vector was transfected into renal cell carcinoma 780-0 and positive clone was selected by using G418.VEGF expression was detected by using immunocytochemical technique and the growth curve was detected by using MTT method. Objective To explore clinical characteristics of patients with Klinefelter′s syndrome(KS; 47,XXY).Methods 227 male patients with 47,XXY treated by artificial insemination with donor(AID)were included.Age, education, height, weight, body mass index(BMI), testicular volume, FSH, LH, testosterone(T), prolactin(PRL), estradiol(E2)were analyzed retrospectively.Results Of these patients, their height were(176.4±5.5)cm, weight(74.5±12.7)kg, BMI(23.89±3.66)kg/m2[77 of overweight(33.92%)and 34 of obesity(14.98%)], FSH(38.35±14.33)IU/L, LH(19.40±9.00)IU/L, T(132.00±194.50)ng/dl, E2(23.90±15.00)ng/L, PRL(10.50±8.20)μg/L, E2/T 0.21±0.80.Testicular volume had the positive association with the level of T(r=0.197, P=0.003).BMI had the negative association with the serum concentration of T(r=-0.284, P=0.000), while positive association with the E2(r=0.174, P=0.009)and ratio of E2/T(r=0.323, P=0.000).Age had no association with T, E2, and E2/T(P>0.05), but had negative association with the serum concentration of LH(r=-0.154, P=0.02)nd FSH(r=-0.196,P=0.03).The higher education group were older(P<0.01), while the level of T were lower(P<0.01).Conclusion In patients with Klinefelter′s syndrome(KS; 47,XXY), level of T may associate with testicular volume.T, E2, and the ratio of E2/T seem to associate with their height, BMI, and education level.
