1.Occupational exposure of medicine and safety protection measures

With reference to medical personnel's infection during the prevalence of SARS, the paper offers some general information on the risks of occupational exposure of medicine. The commonly seen risks fall into four major categories, including biological risks, such as infection of hepatitis B, hepatitis C, HTV, etc.; chemical risks, such as those from antineoplastic drugs, cleaning agents, disinfectants, narcotics, dusts; physical risks, such as radiation, injury by sharp instruments, injury from heavy burden, forced body positions, etc.; and psychological risks. The paper also puts forward some proposals for safety protection: intensifying training in occupational safety of medicine, reinforcing the protection of high-risk groups of people and high-risk body parts, augmenting the supply of protective materials and the improvement of protective facilities, and setting up and perfecting long-acting protection and supply mechanisms.
2.Real-time fluorescent quantitative PCR assay for detection of human Bocavirus

Objective To establish the method for detecting human Bocavirus (HBoV) by a real-time fluorescent quantitative PCR (FQ-PCR) and to assay HBoV from nasopharyngeal samples of children with acute respiratory tract infections.Methods The specific primers and Taqman probes of targeted HBoV NP1 gene were designed.The aimed fragment of NP1 gene was amplified with PCR and ligated into a pCEM-T Easy vector to set up the standards.The method for detection of NP1 gene using FQ-PCR was established.The sensitivity and specificity of the method had been tested.Then 195 clinical specimens were subsequently tested.Results The specificity of this method was excellent.The linear amplification of the assay ranges from 10 copies/μl to 10~8 copies/μl.Twelve specimens were tested positive for HBoV in 195 clinical specimens (6.2%).Conclusions The method for detection of HBoV NP1 geneby real-time fluorescent quantitative PCR was established successfully.It can offer fast and reliable diagnosis for HBoV infection.
3.Rapid on-site evaluation in pulmonology

Rapid on- site evaluation (ROSE) had gradually become an essential tool in interventional section, especially in pulmonary diseases. People were no longer limited to routine endoscopic operation. ROSE combined with fine-needle aspiration (FNA) enhanced the diagnostic efficiency and diagnostic accuracy of routine bronchoscopes, endobronchial ultrasound- guided transbronchial needle aspiration (EBUS-TBNA) and percutaneous lung biopsy. ROSE can read out the specimen on site , give preliminary diagnosis, and provide help for next operation. The ROSE included cytological ROSE (C-ROSE) and microbiological ROSE (M-ROSE). Now in clinic, C-ROSE was used more broadly. C-ROSE expanded the check range of routine endoscopic operation, and it had been proven to be safe and useful tools.
4.Clinical application of high resolution melting curve assay in bacteria identification and antimicrobial susceptibility testing

High resolution melting ( HRM) analysis is a real-time PCR based technique which can detect DNA sequence polymorphism .Immediately after routine PCR with saturated fluorescent dye , melting transition of the amplicon was monitored and fluorescence was collected .According to the different melting curve, single base differences can be distinguished .HRM is a low -cost, single -step, closed -tube, accurate and rapid method , which has aroused general concern .Rapid and sensitive detection of the clinical pathogens is critical to diagnosis of infectious diseases .However , clinical research on application of HRM in infectious disease pathogen identification and drug resistance , especially melting curve analysis based bedside technology integration will provide strong support for the diagnosis and treatment of infectious diseases.

Objective To investigate the expression of matrix metalloproteinases 9 (MMP 9) and tissue inhibitor of metalloproteinases 1(TIMP 1) mRNA in the endometrium during mid luteal phase in women with unexplained infertility and its steroidal regulation Methods In situ hybridization method was utilized to detect the location of MMP 9 and TIMP 1 mRNA. Reverse transcription polymerase chain reaction (RT PCR) method was utilized to detect and quantitate MMP 9 and TIMP 1 mRNA expression levels in the endometrium during mid luteal phase. Serum concentration of estradiol (E 2) and progesterone (P) were measured by radioimmunoassay. Thirty eight patients with unexplained infertility were included as study group, and 20 women with male factor infertility or normal volunteers were selected as control group. Results MMP 9 and TIMP 1 mRNA were diffusely distributed in the cytoplasm and neuclei of glandular and stromal cells. Their staining in control group were more abundant than those in study group. 10.Expressions of ubiquitin and apotosis-related protein in diabetic rat brain

Objective To explore the effect of continuous hyperglycemia on learning and memory capacity in rats, and to observe the changes of expressions of ubiquitin and apotosis-related proteins, so as to provide a relative basis for diabetic encephalopathy. Methods The male SD rats were randomly divided into normal control group and diabetes mellitus (DM) group. The DM rats was made by intraperitoneal injection of streptozocin (STZ). The capacities of learning and memory were tested by Y-maze. The structure changes in frontal lobe cortex and hippocampus were observed by Nissl's staining and immunohistochemistry. The apoptosis cell count was measured by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL). The expressions of Bcl-2 and P53 were assessed by Western-blot analysis. The expression of ubiquitin mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results In DM rats, the learning and memory capacities were worse than in control rats (P<0. 05), and the apoptosis cell counts in frontal lobe cortex and hippocampus were higher than in control rats, the expression of Bcl-2 gene was reduced while the expressions of ubiquitin and P53 gene were increased (P<0. 05). Conclusions In the frontal lobe cortex and hippocampus, the expression of Bcl-2 gene is reduced while the apoptosis cell numbers and ubiquitin P53 gene expression are increased, which may contribute to neurodegeneration in DM rats.
