1.SAHA affects cell cycle of MCF-7 breast cancer cells byregulating acetylated levels of p21WAF1/CIP1 promoter

Aim To study the regulation mechanisms of deacetylase inhibitor SAHA in p21WAF1/CIP1 promoter acetylation in breast cancer MCF-7 cells.Methods We used quantitative real-time PCR, Western blot and DNA-ChIP to determine the effects on the regulation of cell cycle with SAHA treatment in MCF-7 cells.By DNA-ChIP, we assessed the acetylation level of p21WAF1/CIP1 promoter.Results SAHA significantly affected the expression of cell cycle-related factors, and induced the mRNA and protein expression of p21WAF1/CIP1.SAHA could adjust the acetylation level of p21WAF1/CIP1 promoter.Conclusion SAHA regulates the cell cycle progression by adjusting the acetylation level of p21WAF1/CIP1 promoter in MCF-7 cells.

Objective To develop developer with fast determining stopped spot of electrolyte electrode in wartime.Methods According to certain proportion configuration,the developer was prepared by using relative reagent and distilled water.Through injector to take 1ml developer and curve syringe needle,then straight insertion the type needle of electrolysis meter and pour into the developer to fill suction tube.Results The reagent can be seen various electrodes change and fast display the electrode breakdown anywhere.Conclusion The color developing reagent is low-cost,efficient and convenient to use.
3.Influence of Baicalin on TNF-? and Soluble Intercellular Adhesion Molecule-1 in Rats Infected with Pneumocystis carinii

Objective To explore the effect of baicalin on tumour necrosis factor-?(TNF-?) and soluble inter-cellular adhesion molecule-1(sICAM-1) of immunosuppressed rats infected with Pneumocystis carinii.Methods Forty-nine SD rats were randomly divided into 7 groups:normal control(A), immunosuppressed control(B), SMZ/TMP control(C), baicalin prevention(D), low dose(E), moderate dose(F) and high dose(G).Rats of group D received six injections at three-day intervals with 3.5 mg dexamethasone sodium phosphate for 3 weeks, while groups B, C, E, F and G received same immunosuppressor but for 6 weeks.Rats in group D were given 100 mg/kg baicalin daily for 5 weeks, and groups C, E, F and G were given 250 mg/kg SMZ+50 mg/kg TMP, low dose 100 mg/(kg?d), moderate dose 200 mg/(kg?d), and high dose 400 mg/(kg?d) baicalin daily for 2 weeks, respectively.At the end of 8th week after immunosppression, the contents of TNF-? and sICAM-1 in peripheral blood were detected by radioimmunoassay(RIA) and ELISA, respectively.The pathological change of lung was observed by lung imprint smear with gormori methenamine silver(GMS) staining and lung section with hematoxylin-eosin(HE) staining.Results The content of TNF-? in group D(2.14?0.14) ng/ml, group E(2.57?0.15) ng/ml, group F(1.46?0.14) ng/ml, group G(1.12?0.13) ng/ml andgroup C(1.59?0.14) ng/ml were higher than that of group A(0.70?0.21)ng/ml(P

6.Protective effect of alkaline phosphatase on acute lung injury after orthotopic autologous liver transplantation in rats

AIM:The complications following liver transplantation consist of the liver damage and the function failure of other organs,among which lung is the most prone and the earliest affected lesion.By establishing an autologous orthotopic liver transplantation(OLT) model in rats,this study was to investigate the acute lung injury after liver transplantation and the role of alkaline phosphatase(AP) in protection of the injury.METHODS:①The experiments were performed in the Test Animal Center and Digestion Laboratory,Nanfang Hospital,Southern Medical University from May to October in 2007.All the proposals were in accordance with the ethical standard of animals.② Twenty-four SD rats were randomly divided into three groups:sham-operation control group,autologous OLT group and AP group,each containing 8 animals.Autologous OLT models were induced in OLT group and AP group,while control group,the abdomen was only opened and then closed besides the liver lobes were isolated.Lingual vein injection of AP was given in AP group 5 minutes before the blood supply of the liver restored.③The concentration of myeloperoxidase(MPO),malondialdehyde(MDA) and surfactant protein A(SP-A) in lung tissue were respectively examined in each group,lung dry-to-wet weight ratio was also measured.Furthermore,hepatic and lung pathological sections were observed.RESULTS:All the 24 rats were involved in the result analysis.①Compared to control group,the concentration of MPO and MDA in lung tissue were significantly increased in OLT group and AP group(P
7.Research progress in the diet of diabetes

It is widely accepted that diabetes is intimately linked to diet. The latest researches showed that diet-microbiota interactions acted as moderators of human diabetes. Gluten-free diet and hydrolyzed protein decreased the incidence of type 1 diabetes mellitus (T1DM)by the mechanism of changes in gut microbiota and immune micro-environment. Consuming more dairy products decreased the incidence of type 2 diabetes mellitus (T2DM). High fat diet can induce insulin resistance (IR),thus increasing the incidence of T2DM. Parental dietary affected on offspring diabetes by the epigenetic mechanism. Low carbohydrate diet,dietary fiber and legumes were effective to the treatment of diabetes. Prediction of postprandial glucose level and treatment based on gut microbiota contributed to control the glucose of diabetic patients. An in-depth understanding of these mechanisms provides new ideas for the individualized and precise treatment of diabetes.
8.The levels of serum tHcy and Lp(a) and their relation with insulin resistance in patients with combined type 2 diabetes and CHD

Objective To investigate the levels of serum tHcy and Lp(a)and their association with insulin resistance in patients with type 2 diabetes and CHD. Methods T2DM patients were divided into two groups with CHD and without CHD in addition to normal control.The fasting serum levels of tHcy,Lp(a),FPG and FIns were detected, ISI were calculated,and the correlations among them were analyzed. Results The serum levels of tHcy,Lp(a),FIns,FPG of T2DM plus CHD were significantly higher than those of T2DM without CHD and controls(P<0.01~0.05).ISI was significantly lower in T2DM cases with CHD than those of T2DM cases without CHD and controls(P<0.01). ISI was negatively correlated with tHcy and Lp(a)in T2DM plus CHD group(P<0.05)but not correlated with those in T2DM case without CHD group(P>0.05). Conclusions Detections of the serum levels of tHcy and Lp(a)of T2DM play significant roles in knowing patient condition,guiding clinical treatment and earlier preventing and treating vascular complication of diabetes.
9.Effect of berberine on positive transcription elongation factor b expression in diabetic rat retinas

AIM To inVestigate the positiVe transcription elongation factor b(P-TEFb)expressjon and the effect of berberine on diabetic retina of the rat.METHODS Type 2 diabetes mellitus rat models were diabetic control rats(group A)that neither received STZ nor the high-carbohydrate/high-fat diet;16-wk diabetic rats without any drug treatment(group B);diabetic rats treated with berberine at a dose of 75,150 or staining and P-TEFb(cyclin-dependent kinases 9(CDK9)and cyclin T1)protein expression was detected by immunohistochemjstry.RESULTS The retinas of control group were thicker than those of other 6 groups.After thickness but no difference in retinal structure among all groups.Middle-.high-dose berberine and rosiglitazone fenofibrate showed no effect on CDK9 and cyclin T1 expression.CONCLUSION Berberine modulating P-TEFb level in diabetic retina may probably be one of the mechanisms to ameliorate retinopatby induced by STZ and the high-carbohydrate/high-fat diet.
10.Effect of berberine on cyclin dependent kinase 9 and cyclin T1 expressions in type 2 diabetic rat kidney

AIM To investigate cyclin dependent kinase 9(Cdk9) and cyclin T1 protein expressions in diabetic rat kidney and the effect of berberine on them. METHODS Type 2 diabetes mellitus (T2DM) rats were induced by injection (ip) with diet for 16 weeks. From week 17 to 32, diabetic rats were given berberine 75, respectively. The kidney tissue structure was observed with hematoxylin/eosin (HE) staining, kidney to body weight ratio was calculated, and Cdk9 and cyclin T1 expressions were examined by immunohistochemistry. RESULTS Compared with control rats, the volume of diabetic model rat glomerulus accreted, some intercapillary cells proliferated and mesangial region expanded, both glomerular basement membrane and renal tubular basement membrane thickened. Treatment with diabetic nephropathy symptom. The diabetic kidney to body weight ratio protein expressions in diabetic kidney to near control level. CONCLUSION Berberine regulates Cdk9 and cyclin T1 protein expressions in diabetic kidney which may partly contribute to ameliorate nephropathy complication induced by STZ and the high-carbohydrate/high-fat diet.
