1.Three Dimensional Contrast Enhanced Subtraction MRA:A New Technique in theDiagnosis of Body and Lower Extremity Blood Vessel Diseases

Objective To study the value of three dimensional contrast enhanced subtraction MRA(3D CES MRA)in the diagnosis of body and lower extremity blood vessel disease Methods Eighteen cases were studied by 3D ECS MRA and proved by operationResults 3D CES MRA images were of diagnostic quality without ghost for all 18 patients.3D CES MRA clearly showed normal vascular anatomy and various disorders.According to surgery,the sensitivity and specificity of 3D CES MRA in the diagnosis of body and lower extremity blood vessel diseases were both 100%.Conclusion 3D CES MRA is a new technique in diagnosing of body and lower extremity blood vessel diseases.It is proved to be a simple,noninvasive and commercial method in diagnosing blood vessel diseases.
2.The effects of estradiol replacement therapy on novel-object recognition and extinction of conditioned fear memory in ovariectomized rats

Objective To investigate the effects of estradiol replacement therapy on novel-object recognition and extinction of conditioned fear memory in ovariectomized (OVX) rats.Methods Sixty sexually mature (>90 days) female SD rats weighing 250g-300g were used as subjects.The rats were randomly divided into 7groups as following:OVX with low dose of estradiol replacement (OVX + LD,0.1 mg · kg-1),medium dose (OVX + MD,0.3mg · kg-1) and high dose (OVX + HD,0.9mg · kg-1),estradiol replacement with medium dose at 2 weeks after OVX (MD + 2W) and 4 weeks after OVX (MD + 4W),OVX group and Sham group (SH).According to the different dose and time schedules,each group was administrated estradiol or vehicle via subcutaneous injection on the dorsal side of the rat.Estradiol maintained for one month after the surgery,and then the novel-object recognition and extinction of conditioned fear memory were tested.Results (1) The rats after OVX (5.83 ±4.5)s showed access-dependent impairments on novel-object recognition ((5.83 ± 4.5)s vs(27.14 ± 6.6)s,P <0.01) and the estradiol replacement therapy can reverse this impairment,but not in the HD group.There was an increase of the ability on novel-object recognition in group LD and MD + 2W as compared with group OVX,but the difference was not statistically significant (P > 0.05).Moreover,group MD (27.86 ± 2.6)s had a significant difference compared with group OVX (P < 0.05).(2) The conditioned fear memory of rats after OVX (21.67 ± 2.0) %showed significantly lower than group SH (56.81 ± 4.6) % (P < 0.01).Chronic administration with estradiol enhanced acquisition of context fear,respectively (P < 0.01),but except group HD and group MD + 4W.The group HD(18.43 ± 1.1)% was negatively enhanced and the group MD +4W(25.25 ±2.5)% was no difference compared with group OVX.(3) On extinction of conditioned fear memory,the OVX rats produced significantly less freezing by fear context (F =3.337,P < 0.01),and estradiol treatment (except group MD + 4W)also facilitated the extinction of conditioned fear memory,respectively (P < 0.01).Conclusion These results demonstrate that estradiol may have a beneficial effect on the cognition function,and the improving effect is better when estradiol replacement is given at once depriving of sex hormone.

MicroRNA (miRNA for short) can induce mRNA cleavage or inhibit mRNA translation and regulate gene expression in the post-transcriptional level, which involves cell development, proliferation, differentiation, apoptosis, and a series of important biological processes. Recent studies have found that the abnormal regulation of miRNA's target genes may be involved in tumor resistance. And it is expected to become important tumor resistance-associat-ed molecular markers and therapeutic targets. Mechanism of traditional Chinese medicine (TCM) and its active ingre-dients can affect miRNA to regulate target proteins and target genes mediated tumor multidrug resistance. It can pro-vide new ideas for the mechanism of reversing multidrug resistance by TCM.
6.The factors influencing the transfer of porcine endogenous retroviruses across the membrane in bioartificial livers

ObjectiveTo identify the factors influencing the transfer of porcine endogenous retroviruses across the membrane of a new bioartificial liver (BAL),which is a necessary caution to consider for BALs carrying porcine hepatocytes.MethodsA novel porcine BAL composed of two circuits was constructed using a plasma filter with membrane pore size of 500 nm and a plasma component separator with membrane pore sizes 10 nm,20 nm,30 nm,and 35 nm.Co-cultured cells of porcine hepatocytes and mesenchymal stem cells or single porcine hepatocytes were incubated in the bioreactors.The BAL was continuously worked for 72 hours during which the supernatant was collected from the internal and external circuits every 12 hours.PERV RNA,reverse transcriptase (RT) activity,and in vitro infectivity from the supernatant were detected.ResultsIn the plasma filter group,the PERV RNAlevels were the same in both circuits,suggesting little to no effect of the plasma filter on the PERV RNA's crossing.With plasma component separators,PERV RNA was found in the external circuits at different times without positive RT activity and HEK293 cell infection,but its level was reduced significantly.The larger the membrane pore size was,the earlier and the more RNA was detected.ConclusionsThe membrane pore size,the treatment time and the viral level in the internal circuit are contributing factors influencing the transfer of PERV RNA across the membrane in a BAL.
