1.Cognitive disorders in patients with obstructive sleep apnea hypopnea syndrome:a review of literature

Patients possibly incur cognitive disorders when they suffer from obstructive sleep apnea hypopnea syndrome (OSAHS). The pathogenesis of OSAHS is mainly associated with turbulence of sleep structure and chronic episodic hypoxia during sleep, and the pathological changes in cerebral areas hypersensitive to hypopnea may be responsible for cognitive dysfunction. It has been preliminarily showed that cognitive disorder in OSAHS is a generalized cognitive impairment and is possibly associated with Alzheimer's disease (AD). Recently, measures for cognitive evaluation have been improved by the application of the neuroimage; however,to build a highly specific,maneuverable evaluation system still demands more exertion. Despite the fact that early remedies for OSAHS may slow down the cognitive dysfunction,there still lacks specialized treatment for cognitive disorders. OSAHS is not rare in children and it can affect their cognitive function, which demands more attention.
2.The Function of SeMNPV IAP3 in Mammalian Cells

The baculoviral inhibitors of apoptosis play a significant role in infectivity and viral host-range, which make them potential candidates for the engineering and improvement of baculovirus insecticidal. The iap3 gene of Spodoptera exigua nucleopolyhedrovirus (SeMNPV), amplified by PCR, was 939 bp encoding IAP3. The PCR product was cloned into EcoR I/Bam H I of the plasmid pEGFP-C1. GFP was fused to the N-terminaus of IAP3 to study distribution in HEK293. It was observed that the plasmid expressing IAP3 significantly inhibited apoptosis induced by cisplatin in HEK293 cells. We conclude that the IAP3 of SeMNPV is functional in mammalian cells.
3.Localization and Functional Analysis of SeMNPV IE1 in Mammalian Cells

In this paper, the function of the iel gene from baculovirus Spodoptera exigua multiple nucleopolyhedrovirus (SeMNPV), belonging to group Ⅱ nucleopolyhedrovirus, was studied in mammalian cells.We amplified the SeMNPV ie1 gene and expressed it by fusing to the C terminal of enhanced GFP protein in HEK 293 cells. Confocal microscopy revealed that the IE1-GFP fusion protein was localized in the nucleus of the mammalian cells. The promoter sequences of AcMNPV gp64, SeMNPV F protein and Drosophila hsp70 were also analyzed, to further study the function of SeMNPV IE1. The results showed that, in the absence of the hr sequence, IE1 improved the expression of the F promoter but didn't influence the gp64 promoter significantly, but IE1 moderately stimulated the hsp70 promoter.
4.The clinical significance of lymphocytes activation hi patients with multiple sclerosis

Objective:To analyze the lymphocyte activation from patients with different activities of multiple sclerosis (MS) and its clinical signifiance. Methods: The positive percentage of CD69 and HLA-DR were determined on peripheral blood (PB) or cerebrospinal fluid (CSF) lymphocytes by flow cytometry from 28 patients with relapsing-remitting MS (relapses n - 20, remission n = 8) , 12 patients with relapse MS and 11 patients with relapse MS after glucocorticoid treatment. ResultS:The percentage of HLA-DR+ and CD3+ /HLA-DR+ on PB lymphocytes in relapses MS group were higher than in remission MS group and controls. The percentage of CD3+ /HLA-DR+ on PB lymphocytes in remission MS group were higher than in controls. The percentage of CD69+ , HLA-DR+ and CD3+ /HLA-DR+ were higher in CSF than in PB of patients with relapse MS. There was no significant correlation between the levels of CD69+ , HLA-DR+ , CDS+ /HLA-DR+ and the time of onset of clinical relapse, duration, clinical disability scale score(EDSS) . In patients with relapse MS, there was significant positive correlation between CD69+ expression on CSF lymphocytes and blood brain barrier damage, intravenous glucocorticoid treatment caused a significant changes in the positive percentage of HLA-DR on PB lymphocytes. Conclusion:The findings support that lymphocyte activation involvement of MS pathogenesis. Lymphocyte activation may be as a marker of disease activity in MS.
5.Laparoscopic cholecystectomy combined with duodenoscopic procedures for selective common bile duct stones

Objective To investigate the value of combined use of laparoscopic cholecystectomy (LC) and duodenoscopic procedures in the treatment of selective common bile duct stones. Methods We retrospectively analyzed clinical data of 32 cases of selective common bile duct diseases treated with laparoscopic cholecystectomy combined with endoscopic retrograde cholangiopancreatography (ERCP) from January 2002 to December 2003. Results Of 24 cases receiving ERCP before LC, 19 cases were found having common bile duct stones, 3 cases having stricture at the lower end of the common bile duct, and 2 cases normal. These cases underwent ERCP and endoscopic sphincterotomy (EST) following by LC. Of 8 cases receiving ERCP after LC, 4 cases were found having common bile duct stones, 3 cases having stricture at the lower end of the common bile duct, and 1 case normal. Except for 1 case of conversion to open surgery because of a failed stone removal, all the cases were cured with EST. Conclusions combined use of LC and ERCP is a highly useful approach in the treatment of selective common bile duct stones, which has broadened the utilization of minimally invasive treatment for bile duct diseases.
6.Expression of ERCC-1 protein and its clinical implication in locally advanced cervical cancer

Objective To examine the protein expression of the nucleotide excision repair gene (ERCC-1) in patients with locally advanced cervical cancer and its relationship with the efficacy of radiotherapy and chemotherapy.Methods The expression of ERCC-1 protein in 88 patients with locally advanced cervical cancer treated in our hospital between 2007-2011 was measured using immunohistochemistry (IHC).The patients were divided into high-expression group (n=48) and low-expression group (n=40) based on the fluorescence intensity on the IHC staining.All patients received cisplatin (40 mg/m2 per week) during radiotherapy.The relationship between ERCC-1 protein expression and the clinicopathological factors of cervical cancer was analyzed using the chi-square test.Survival was calculated using the Kaplan-Meier method and compared by the log-rank test.Multivariate prognostic analysis was performed using the Cox model.Results The overall response rate (CR+PR) was 81%(39/48) in the high-ERCC-1 expression group and 85%(34/40) in the low-ERCC-1 expression group (P=0.641).ERCC-1 protein expression was associated with recurrence and metastasis (P=0.043,0.043).The 5-year survival rate was significantly higher in the low-ERCC-1 expression group than in the high-ERCC-1 expression group (65% vs.42%, P=0.029).Conclusions Patients with high ERCC-1 protein expression are more likely to have local recurrence and distant metastasis than those with low ERCC-1 protein expression.ERCC-1 protein expression may be a clinically significant biomarker for predicting the prognosis of cervical cancer patients.
7.Differentiation of CEA and CEA-Related Substances by Western Blot Technique in Biliary and Gastric Diseases

Objective To eliminate the interference of CEA-related substances in CEA measurement and increase the specificity of CEA in the detection of malignant digestive diseases.Methods CEA level of peripheral blood and digestive juice(bile,gastric juice) from patients with benign or malignant digestive diseases was measured by ELISA,and semi-dry electrophoretic transfer method of Western blot technique to distinguish CEA and CEA-related substances.Results In malignant diseases,the CEA level of digestive juice was significantly higher than that in the blood,and there was no difference of CEA level in digestive juice and blood in benign diseases.Meanwhile,the CEA level of digestive juice and blood in malignant diseases were significantly higher than that in benign diseases.A specific band(molecular weight about 210?10~3) was detected in all malignant diseases except four cases whose CEA level was too low(less than 5 ?g/L),whereas no one of benign diseases had this specific band no matter how high or low the CEA level was.Conclusion The specificity of CEA detection in malignant digestive diseases can be improved by using digestive juice as sample and combining with Western blot technique.
8.Evaluation of matrix-assisted laser desorption/ionisation time-of-flight mass spectrometryfor identification of clinical yeasts

Objective To evaluate the ability of MALDI-TOF MS in identification of the yeast isolated from clinical specimens.Methods 387 strains of yeasts were tested.All the strains were from clinical specimen and identified by VITEK MS and API ID 32C system at the same time.Sequencing of the internal transcribed spacer (ITS) regions of ribosomal DNA was used as the reference method in the analysis of a total of 387 yeast isolates.The accuracy of detected results was analyzed by the X2 test.Results The identification accuracy rates of VITEK MS, API ID 32C were respectively 97.7% and 93.0%.The difference was statistically significant (X2=9.439, P=0.002).There was no statistical significance between MALDI-TOF and ITS sequencing.Conclusion MALDI-TOF is a rapid simple and accurate method which is identified to species level in identification of yeasts.
9.A comparative study of motor conduction velocity and F-wave parameters between diabetes mellitus and healthy subjects

(4)75 2% of all the patients in DM group showed one or more abnormalities in MCV or F wave parameters, and electrophysiological abnormalitis correlated with, or precede to clinical findings in 175 of 190 patients (92 1%). Conclusion MCV and F wave parameters are sensitive measures for detection of DPN. Among these parameters, FwPL, F MIPL from all the four nerves are the most sensitive. To improve diagnostic accuracy, all the MCV and F wave parameters recorded should be considered.
10.Effect of endothelin-1 on differentiation of cardiomyogenic cells induced from rabbit bone mesenchymal stem cells

AIM: To investigate biological effect of endothelin-1 (ET-1) on differentiation of cardiomyogenic cells (CGCs) induced from rabbit bone mesenchymal stem cell (BMSCs) in vitro. METHODS: The BMSCs from shaft of femur of Japan rabbits were isolated and propagated for cell culture. Cultured BMSCs were randomly divided into six groups: Group Ⅰ: the control group (uninduced group); Group Ⅱ: the ET-1 group, added ET-1 (30 nmol/L) into culture medium; Group Ⅲ: 5-aza group, added 5-aza (10 ?mol/L) into culture medium; Group Ⅳ: 5-aza+ET-1 group, after inducing with 5-aza for 3 weeks, ET-1 was added into culture medium. This group was divided into three parts-Ⅳ 1、Ⅳ 2、Ⅳ 3, and separated to add 10、 30、 50 nmol/L ET-1. During the cell culture, growing and differentiation of BMSCs were observed. After inducing for 4 weeks, the differentiation rate and the diameter of cardiomyogenic cells were calculated; Western-blot was employed to analyze the expressions of GATA-4 protein and phosphorylation level. The expression of ?-MHC mRNA was assessed by RT-PCR. Immunohistochemistry staining of Troponin-I and ultrastructure observation of induced cardiomyogenic cells were also completed simultaneously. RESULTS: The cell diameters of CGCs in group Ⅳ 2 were enlarged significantly (P0.05).
