Objective To observe the distributions of epidermal growth factor receptor(EGFR),transforming growth factor ?(TGF ? 1,? 2,? 3)and thyroid transcription factor 1(TTF 1)in developing lungs,and to discuss their functions and relationships each other. Methods The expression of EGFR and TGF ? was examined in mouse fetal lungs by immunohistochemistry.TIGER image analysiser was used in TTF 1 quantitative study. Results 1.In the early developmental phase,EGFR localized in the epithelium of airway surface,TTF 1 was expressed predominantly in the nuclei of distal lung buds and TGF ? was expressed in the bronchioles with localization patterns.2.At the late developmental phase,the positive reactions of EGFR and TTF 1 were mainly detected in alveolar cells.TGF ? expressed in mesenchymal at high levels.3 The average optical density(A A )of TTF 1 in the distal epithelial cells increased gradually during the development,while their volume(VOL)and volume integrated optical density(VI A )all decreased.Conclusion\ The functions of EGFR,TGF ? and TTF 1 might be respectively different at different developmental stages.They not only stimulate the process of branching morphogenesis,but also modulate epithelial maturation and differentiation.\;[
2.Performance validation of ABI ViiA 7 Taqman HBV-DNA detecting system

Objective To verify the performance of the ABI ViiA 7 Taqman HBV-DNA detecting system for confirming its sta-bility,accuracy and reliability.Methods According to the evaluation protocols of Clinical and Laboratory Standards Institute (CLSI),the performance of ABI ViiA 7 Taqman HBV-DNA detecting system was assessed in the aspects of precision,accuracy,lin-earity and comparability;the quantitative detection limit validation experiments was performed by diluting specimen until quantita-tive detection limit is lower than the lower limit of detection,and the detection results were compared with the quality target re-quirements and the analysis capability declared by manufacturers.Results The CV in within-run precision of this detection system was 1.485%,1.990% and 0.932% respectively;the total CV was 1.876%,3.361% and 1.891%,respectively;the maximum devia-tion of accuracy was -6.8%;the linear correlation coefficient was 0.998 3;the regression equation was Y =0.974 8X +0.050 7. The linear range was 1.00E2 - 2.00E8;the quantitative detection limit was 100 IU/mL;the comparability of ABIViiA7 and ABI7500:P =0.115,r2 =0.994,the linear regression equation was Y =0.987 2X +0.051 7.Conclusion The ABI ViiA 7 Taqman HBV-DNA detection system has excellent precision,accuracy,sensitivity and linearity and has a good correlation with ABI7500, which can be used for the detection of clinical specimens.
3.Quantitative analysis of β thromboglobulin level in patients with coronary heart disease complicated dif-ferent complications

Objective:To compare serum level of β thromboglobulin in patients with coronary heart disease (CHD) complicated different complications.Methods:According to their complications,a total of 398 patients with unsta- ble angina pectoris (UAP)were divided into pure UAP group (UAP control group,n=82),hypertension group (n=89),diabetes mellitus (DM)group (n=133)and brain infarction group (n=94).Serum level of β thromboglobu- lin were measured and compared among four groups 6h after onset and before discharge.Incidence of myocardial in- farction within six months were followed up in four groups.Results:On 6h after onset,the serum level of β throm- boglobulin of brain infarction group,DM group,hypertension group,UAP control group was (61.13±3.32)ng/ml,(59.77±3.15)ng/ml,(52.12±3.27)ng/m, (48.55±3.14)ng/ml respectively,in which the level of brain infarction group was the highest,the difference between any two groups were significant (P<0.01 all);Compared with 6h after onset,there were significant reductions in serum levels of β thromboglobulin of four groups before dis- charge P<0.01 all,their ordering and difference significant degree were no change.The incidence of myocardial infarction (MI)in brain infarction group,DM group,hypertension group,UAP control group was 11.7%,6.0%, 3.4%,2.4% respectively,the MI incidence of brain infarction group was significant more than that of UAP con- trol group,the differences among other groups was no significant,P>0.05.Conclusion:β-thromboglobulin level during UAP onset is significant higher than that of remission period,and it rises most significantly in brain infarction group,and in this group the percentage of myocardial infarction occurred within six months is highest
4.Changes of serological indexes and prognosis of acute coronary syndrome patients after percutaneous coronary intervention

Objective To investigate the levels of N-terminal probrain natriuretic peptide(NT-proBNP),thrombolomodulin(TM),C-type lectin-like receptor 2(CLEC-2),galectin-3(Gal-3)and their relationships with prognosis in patients with acute coronary syndrome(ACS)after percuta-neous coronary intervention(PCI).Methods A total of 80 patients with ACS were selected as study objects.The patients were followed up for 6 months by outpatient visit or telephone follow-up,and the occurrence of major adverse cardiovascular events(MACE)was recorded in the patients.According to the occurrence of MACE,patients were divided into MACE group(26 cases)and control group(54 cases).The changes of serological indexes were compared between the two groups.Results The expression levels of NT-proBNP,CLEC-2,TM and Gal-3 in serum of patients in the MACE group were significantly higher than those of control group(P<0.05).Multivariate Logistic regression analysis showed that NT-proBNP,CLEC-2,TM,and Gal-3 were all influential factors for the occur-rence of MACE after PCI in ACS patients(P<0.05).Conclusion NT-proBNP,CLEC-2,TM and Gal-3 are influencing factors for MACE in patients with ACS after PCI,and have a good predictive role in evaluating the occurrence of MACE.
5.Study on the mechanism of long non-coding RNA NUTM2A-AS1 targeting microRNA-129-5p in regulating oxidized low density lipoprotein-induced vascular endothelial cell damage

Objective To explore the effect of long non-coding RNA(lncRNA)NUTM2A-AS1 on the damage of vascular endothelial cells induced by oxidized low density lipoprotein(oxLDL)and its molecular mechanism.Methods Human umbilical vein endothelial cells(HUVECs)were cultured in DMEM medium.The HUVECs treated with 100 μg/mL oxLDL were assigned to oxLDL group,while those cultured under normal conditions were assigned to Con group.After transfection of the ln-cRNA NUTM2A-AS1 interference expression vector and negative control,microRNA-129-5p mimic and negative control into HUVECs,the cells treated with 100 μg/mL oxLDL were assigned to oxLDL+si-NUTM2A-AS1 group,oxLDL+si-NC group,oxLDL+miR-129-5p group,and oxLDL+miR-NC group,respectively.After co-transfection of the lncRNA NUTM2A-AS1 interference expression vector and miR-129-5p inhibitor or negative control into HUVECs,the cells treated with 100 μg/mL oxLDL were assigned to oxLDL+si-NUTM2A-AS1+anti-miR-129-5p group and oxLDL+si-NUTM2A-AS1+anti-miR-NC group.The levels of malondialdehyde(MDA)in cells,as well as the activities of su-peroxide dismutase(SOD)and glutathione peroxidase(GSH-Px)were measured using kits.Cell apoptosis was detected by flow cytometry.Protein expression was detected by Western blot.The tar-geting relationship between NUTM2A-AS1 and miR-129-5p was detected by dual luciferase reporter assay and RNA pull-down experiments.Results Compared with the Con group,the expression lev-el of lncRNA NUTM2A-AS1 was increased,the expression level of miR-129-5p was decreased,the content of MDA was increased,the activities of SOD and GSH-Px were decreased,the apoptosis rate of vascular endothelial cells and the expression levels of cleaved-caspase3 and cleaved-caspase9 were increased in the oxLDL group(P<0.05).After interference of lncRNA NUTM2A-AS1 expression or overexpression of miR-129-5p,the content of MDA was decreased,the activities of SOD and GSH-Px were increased,cell apoptosis was reduced(P<0.05).The damage to vascular endotheli-al cells mediated by knockdown of lncRNA NUTM2A-AS1 under oxLDL conditions can be reversed by downregulation of miR-129-5p.LncRNA NUTM2A-AS1 targeted miR-129-5p for regulation.Conclusion Interference of lncRNA NUTM2A-AS1 expression can inhibit oxLDL-induced vascular endothelial cell damage by targeting miR-129-5p for upregulation.
