1.Research progress of platelet-rich plasma in cartilage regeneration

Cartilage is vulnerable to traumatic injury and unable to facilitate a satisfactory healing response due to its poor vascularity and inability to access mesenchymal stem cells.Continuous defects in the joint surfaces cause pain,swelling,and mechanical symptoms that result in functional impairment and limitation of athletic participation.Commonly used repair techniques include marrow stimulation,structural osteo-articular autografts or chondrocyte implantation.Platelet-rich plasma (PRP) is a concentrate extract of platelets from autologous blood,which is rich in growth factors and other cytokines and provides local environment for tissue regeneration and lends a possible option for the stimulation and acceleration of cartilage regeneration.This review gives summarization on the current state of the use of PRP for cartilage regeneration.
2.Comparison of the outcomes of monopolar and bipolar radiofrequency ablation in surgical treatment of atrial fibrillation.

OBJECTIVETo compare the therapeutic effects and safety of monopolar and bipolar radiofrequency (RF) ablation used during cardiac surgery to treat atrial fibrillation.
METHODSWe retrospectively studied a total of 81 patients with chronic atrial fibrillation who underwent open cardiac surgery with concomitant RF ablation between January 2007 and March 2011. Fifty-eight patients received bipolar RF ablation and 23 received monopolar RF ablation, respectively. The sinus rhythm restoration rate, the procedural duration, the frequency of severe perioperative complications, and mortality were compared between the two groups.
RESULTSThe sinus rhythm restoration rate did not differ significantly between the two groups after follow-up of 15.1 ± 12.6 months (P=0.199). The frequencies of severe perioperative complications and mortality were also similar in the two groups. The total procedural time using bipolar RF ablation was significantly shorter than that using monopolar ablation (19.7 ± 4.6 minutes vs. 28.1 ± 8.5 minutes, P< 0.001).
CONCLUSIONSBoth monopolar and bipolar RF ablation are safe and effective in treating chronic atrial fibrillation patients during open cardiac surgery, but bipolar RF ablation is more convenient in practice.

3.Relation among Ultrasound Findings,Clinical Significance and Expression of FosB in Invasive Breast Cancer

【Objective】To investigate the expression of FosB in invasive breast cancer and its correlation to the ultrasound findings and clinical significance.【Methods】Immunohistochemical and ultrasound examination were performed for 155 cases with invasive breast cancer,36 cases with paracancerous tissue and 30 cases with normal breast tissue. To investigate the correlations between the different expression of FosB with histological grade ,lymph nodes,and ultrasound findings.【Results】①The expression of FosB in paracancerous tissue and normal breast tissue was higher than that in invasive breast cancer(P<0.01). ②Based on immunohistochemical staining,high positive expression rate of FosB was associated with low histological grade,ER positive and PR positive(P<0.05). There was no correlation between FosB and age,tumor diameter,lymph nodes,Her2 and clinical stages(P>0.05)③The expression of FosB was higher in microcalcification positive and lymph nodes bigger tissues(P<0.05). There was no correlation between FosB and tumor size, regular shape,boundary,posterior attenuation ratio and blood flow(P>0.05).【Conclusion】The expression of FosB in invasive breast cancer was lower than that in paracancerous tissue and normal breast tissue. The different expression of FosB was correlated with histological grade,ER positive and PR positive. To study the expression of FosB in invasive breast cancer may be helpful for differential diagnosis and targeted treatment. The different expression of FosB was correlated with microcalcification,lymph nodes,and combined FosB with ultrasound findings may contribute to prognostic evaluation of breast cancer.
4.Ureteroscopic holmium laser lithotripsy in treatment of patients with acute obstructive renal impairment

Objective To evaluate the efficacy and safety of holmium laser for treatment ureteric stones in patients with acute obstructive renal impairment.Methods Thirty-two patients were included in this study.None of the patients had a ureterie stent or nephrostomy tube before the ureteroseopy.All patients were treated with holmium laser.Results 30 patients with ureter stones in middle and inferior segment were free of stones by ureteroscopic lithotripsy.The success rate for treatment of ureteral stones lithotripsy and calculus removal was 93.7%.Ureter stones located in superior segment in two patients were sent back pelvis.Extracorporeal shock-wave lithotripsy were performed.The two patients were free of any stone fragmens a month later.In all patients, including the five with obstructive anuria,the renal impairment resolved or improved as evidenced by normalization or fall in blood urea and creatinine.100% of the patients were free of any stone fragments postoperatively.Conclusion A holmium laser was a safe and effective modality of ureteroscopic lithotripsy in patients with significant renal impairment or even obstructive anuria.It also had merits of small wound and fast postoperation recovery.Treatment of both-side ureteral stones could be handled at the same time.The use of holmium laser by ureteroacopy could be considered the first choice in patients with acute obstructive renal impairment.
5.Changes of free radicals and antioxidative system of prostate tissue in the male rats with short-term diabetes

AIM: To study the dynamic changes of free radicals and antioxidative system of prostate in rats with short-term diabetes. METHODS: 48 Wistar male rats were divided into 6 groups at random (8 rats in each group). One group were injected(ip) with sodium citrate buffer to be the control group, the other 5 groups were injected (ip) with steptozocin(STZ) 60mg/kg body weight which were called diabetes group. Rats were killed after 1, 3, 5, 7 and 14 days; superoxide dismutase(SOD), malondialdehyde(MDA), glutathione(GST), glutathione s-transferase(GST), glutathione peroxidase(GSH-px), nitric oxide (NO) and nitric oxide synthase(NOS) of prostate were examined. RESULTS: The levels of MDA, GSH-px and NO in prostate homogenate of diabetes group were higher evidently as compared with control levels. The activities of SOD, GST, NOS and the levels of GSH in prostate homogenate were increased obviously after injection of STZ with control levels, then returned near to control values. CONCLUSION: The evidences mentioned above indicate that high-level of free radicals and decrease in antioxidative system rendere the prostate cells to be in oxidative stress.
6.Package and titer assay of recombination adeno-associated virus with c-Met short hairpin

Objective To package the recombination adeno-associated virus with human U6 promoter and c-met short hairpin in order to establish foundation for research on inhibiting the expression of c-Met and cancer gene therapy.Methods c-Met short hairpin was synthesized and linked to the down stream of U6 promoter by PCR.Adeno-associated viral vectors pSNAVUshMet(1,2) were constructed and transfected into BHK cells.Positive cell clones were selected by G418.Adeno-associated virus with U6shMet were packaged by adding rHSV1-repcap.The virus purity was analysed by SDS-PAGE and titer was assayed by dot blot.Results Two fragments(U6shMet1 and U6shMet2) with c-Met short hairpin were obtained and two recombination adeno-associated virus(rAAVUshMet1 and rAAVUshMet2) with U6shMet were packaged successfully.SDS-PAGE analysis showed that the purified virus appeared clear characterized bands.The virus titer was 4?1012mg?L-1.Conclusion The recombination adeno-associated virus(rAAVUshMet1 and rAAVUshMet2) with U6shMet are constructed successfully.The virus have high titer and good purity and could act as the effective vehicle of inhibiting the expression of c-Met and cancer gene therapy.

8. Advances in biomarkers predicting the efficacy of sorafenib in advanced hepatocellular carcinoma

Hepatocellular carcinoma (HCC) is a common kind of cancer in human beings, and ranks the third in cancer-related death. Nowadays, sorafenib is the only approved molecular targeted agent for the treatment of advanced HCC. However, the efficacy of sorafenib varies among individuals, which is unsatisfactory. People have researched some biomarkers predicting the efficacy of sorafenib in order to optimize treatment scheme, reduce health care expenses and improve the quality of life for advanced HCC patients treated with sorafenib. The studies show that the expressions of phosphorylated extracellular signal regulated kinase (pERK), phosphorylated c-Jun at the downstream of c-Jun N-terminal kinase (JNK) signal pathway, vascular endothelial growth factor receptor-2 (VEGFR-2) in tumor tissues, alpha-fetoprotein (AFP), transforming growth factor (TGF) and vascular endothelial growth factor (VEGF) in peripheral blood, as well as pERK/phosphorylated Akt (pAkt) expression in circulating tumor cells, are important factors to predict the efficacy of sorafenib, so these molecules have the potential of acting as predictive biomarkers for sorafenib treatment. This paper summarizes the recent clinical cohort studies on this topic.

10.The destructive role of soluble Robo4 secreted by the M1-polarized-microglia during cerebral ischemia-reperfusion in blood-brain barrier integrity.

This project was aimed to investigate the role and the underlying mechanism of microglia polarization on blood-brain barrier (BBB) during cerebral ischemia-reperfusion. After construction of the mouse model of cerebral ischemia-reperfusion, upregulated IL-6 and TNF-α in peripheral blood and increased IL-6 and iNOS in ischemia tissues were confirmed. The supernatant expression of TNF-α and IL-6, as well as IL-6, iNOS and CD86 mRNA, was significantly increased in the of Bv-2 cells after oxygen-glucose deprivation/reoxygenation (OGD/R) model was constructed in vitro. For further understanding the expression pattern of RNAs, the next-generation RNA sequencing was performed and upregulation of Robo4 (roundabout guidance receptor 4) was found both in M1-polarized and OGD/R treated Bv-2 cells, which was also confirmed by RT-qPCR. Extracellular soluble Robo4 (sRobo4) protein also increased in the supernatant of M1-polarized and OGD/R treated Bv-2 cells. Treating bEND3 cells with the Robo4 recombinant protein, M1-polarized Bv-2 cell supernatant or OGD/R Bv-2 cell supernatant decreased trans-endothelial electrical resistance (TEER), suggesting the injury of BBB. In addition, Robo4 was also highly expressed in the serum of patients who experienced acute ischemia stroke and mechanical thrombectomy operation. All the results suggest that increased secretion of Robo4 by M1-polarized-microglia during cerebral ischemia-reperfusion is most likely one of the causes of BBB injury, and Robo4 may be one of the therapeutic targets for BBB functional protection.
