3.Curcumin inhibits the adherence of leukocytes to BMECs induced by TNF-? in vitro

Aim To investigate the effects of curcumin on the adherence between isolated rat brain microvascular endothelial cells(BMECs)and leukocytes induced by TNF-? in vitro.Methods The adherence ofleukocytes to BMECs and the effects of curcumin were determined by myeloperoxidase.The expression of ICAM-1 was detected by RT-PCR and immunoblotting.Results Curcumin inhibited the TNF-? stimulated adherence of leukocytes to BMECs.Pretreament of curcumin also inhibited TNF-?-induced increases in the mRNA and protein levels of ICAM-1 in BMECs.Conclusions Curcumin could protect the endothelial cells against damage caused by TNF-?.The protective effects of curcumin may be mediated through downregulation of the expression of ICAM-1.
4.Effect of aspirin on learning and memory in Alzheimers disease rats and its molecular mechanism

AIM To observe the effect of aspirin on a model of Alzheimers disease in rats and its molecular mechanism. METHODS The AD pathological model was made by hippocampal CA 1 lesions with stereotaxic mini-injection of quinolinic acid;the rats learning and memory was observed by Y-maze; the apoptosis of hippocampal cells were detected by using flow cytometry and electronic microscope; the content of calcium in AD rats hippocampus was determined by atomic absortion spectrophotometer. RESULTS Aspirin was shown to improve learning and memory deficiency in rats with bilateral hippocampal lesions induced by quinolinc acid, to decrease apoptosis rate in hippocampal cells significantly and to reduce the concentration of calcium in hippocampus. CONCLUSION The protective effects of aspirin on a model of AD in rats may be involved in antagonism of calcium and inhibition of apoptosis in hippocampal cells induced by quinolinic acid.
5.Diagnosis and treatment of Mirizzi syndrome

Objective To investigate the diagnosis and treatment of Mirizzi syndrome.Methods 6 cases of Mirizzi syndrome were analyzed with review of the latest literatures on its diagnosis and treatment.Results None of the 6 cases were definitely diagnosed by ultrasonography and CT.All of them underwent operations,five of them were cured and one died.Conclusions Ultrasonography and CT can give a hand to diagnosis.Definite diagnosis depends on ERCP and PTC.The specific conditions of the patients and the experience of surgeon decide the selection of treatment.Subtotal cholecystectomy and hepaticojejunostomy are the basic choices.
6.Neuroprotective Actions and Possible Underlying Molecular Mechanisms of Valproate

Valproate (VPA) has long been used for the treatment of bipolar mood disorder. VPA is effective in control of mania and depression. Recent studies have demonstrated that VPA has profound neuroprotective effects in against various apoptotic stimuli. Moreover, VPA can promote neurogenesis, neuronal proliferation and differentiation. Although intensive research has been dedicated to VPA′s neuroprotection, the molecular mechanisms by which VPA protects neurons are still not fully understood. In this paper, recent progresses in the study of VPA′s neuroprotection and underlying mechanism are reviewed.
7.Study on the Relationship between Feature of Pharynx Flora and the TCM Syndrome Type of Acute Upper Respiratory Infection

Objective The purpose of this study was to determine the relationship between microecological balance and the syndrome type of acute upper respiratory infection.Methods The nature and quantity of pharyngeal flora were assayed in 31 cases of wind-cold acute upper respiratory infection and 36 cases of wind-heat acute upper respiratory infection,with 30 healthy cases served in control group.Results There was a significant rise of concentration of pharynx flora in acute upper respiratory infection than that in control group,whereas there was a significant decrease of diversity of pharynx flora in acute upper respiratory infection than that in control.Conclusion Imbalance of pharyngeal micro-ecology is one of the major factors leading to acute upper respiratory infection,manifested as insufficiency of genuine Qi failure in guarding.

9.Study on the liver injuries induced by cisplatin and carboplatin in mice

Objective To study the time the liver injuries appeared,the different level of liver injury induced by injecting Cisplatin and Carboplatin in mice and the possible mechanisms of the liver injury.Methods Mice were intravenously given,Cisplatin(17 mg/kg),Carboplatin(67 mg/kg) respectively.In the 3th,7th,11 th and 14th day after injecting the medicines,the serum biochemistry parameters,oxidation and antioxidation parameters of liver and histopathologic change in hepatic tissue were observed.Results HE stain showed that fatty degeneration of hepatocyte in the two experiment groups in the 3th day after injecting medicines,and hepatocellular spotty necrosis,and infiltration of inflammatory cells arroud the necrotic liver were found in,Cisplatin group.The level of aspartate aminotransferase and alkaline phosphatase in the two experiment groups increased significantly compared with those in the control (group P<0.01),and alanine aminotransferase in Cisplatin group increased significantly(P<0.01),compared with that in the control group and Carboplatin group.Conclusion Hepatic injury was observed in the 3th day after injecting medicines,the level of liver injury in Cisplatin group was heavier than that in Carboplatin group.
10.Expression and significance of T cell immunoreceptor with Ig and immunoreceptor tyrosine-based inhibitory domains (TIGIT) on peripheral CD4+ T and CD8+ T cells from patients with rheumatoid arthritis

Objective To investigate the expression of T cell immunoreceptor with Ig and immunoreceptor tyrosine-based inhibitory domains (TIGIT) on peripheral CD4+ T and CD8+ T cells from patients with rheumatoid arthritis (RA) and its significance in order to clarify its role in the development of RA.Methods Peripheral blood samples were collected from 81 patients with RA and 33 healthy controls (HC).Expression of TIGIT on the surface of peripheral blood leukocytes was detected by flow cytometry.Differences in TIGIT expression between RA and HC groups were comparatively analyzed.Correlations of TIGIT expression on CD4+ T and CD8+ T cells with several laboratory indexes were analyzed.All data were statistically analyzed.Results (1) The percentage of TIGIT-expressing CD3+ T cells in patients with RA was significantly higher than that in HC (P<0.01).Moreover, the median fluorescence intensity (MFI) of TIGIT on CD3+ T cells was significantly elevated in patients with RA as compared with that in HC (P<0.01).No significant difference in the expression of TIGIT on B cells, monocytes or neutrophils was observed between RA and HC groups.(2) The percentages of TIGIT-expressing CD4+ T and CD8+ T cells were significantly elevated in patients with RA as compared with those in HC (P<0.01).Moreover, the MFI of TIGIT on CD4+ T and CD8+ T cells were significantly elevated in patients with RA as compared with those in HC (P<0.01).(3) The percentages of both TIGIT-expressing CD4+ T cells and TIGIT-expressing CD8+ T cells in patients with RA were positively correlated with erythrocyte sedimentation rate (ESR) (rs=0.355, P<0.01;rs=0.277, P=0.013).(4) The percentages of both TIGIT-expressing CD4+ T cells and TIGIT-expressing CD8+ T cells in patients with RA were positively correlated with rheumatoid factor (RF) (rs=0.265, P=0.017;rs=0.366, P<0.01).The MFI of TIGIT on CD4+ T cells was positively correlated with RF in RA group (rs=0.226, P=0.043).The percentage of TIGIT-expressing CD4+ T cells was positively correlated with anti-cyclic citrullinated peptide (anti-CCP) antibody in patients with RA who were positive for anti-CCP antibody (rs=0.324, P=0.012).(5) The percentage of TIGIT-expressing CD4+ T cells as well as the MFI of TIGIT on CD4+ T cells in patients with RA was positively correlated with Disease Activity Score-28 (DAS28) (r=0.232, P=0.038;r=0.343, P<0.010).Conclusion The expression of TIGIT on T cells is elevated in patients with RA and correlated with inflammatory markers, antibody production and disease activity.
