1.Influence of PD-1 inhibitor pembrolizumab on T lymphocyte subsets and NK cells in patients with late stage non-small cell lung cancer

Objective To observe the changes of T lymphocyte subsets and natural killer (NK) cells in patients with late stage non-small cell lung cancer (NSCLC) before and after treatment with PD-1 inhibitor and its clinical effect.Methods Totally 23 patients with NSCLC in Guangzhou Modern Hospital from January 2015 to January 2016 were collected.All patients were given 6 cycles of PD-1 inhibitor treatment after chemotherapy or targeted drug treatment failure.Peripheral venous blood was collected before and after treatment to detect the percentage of CD3 +,CD4 +,CD8 + and NK cells in peripheral blood lymphocytes.The curative effects were evaluated by chest CT after treatment of 2,4,6 cycles.Results Compared with before treatment,the proportions of CD3+ (69.56% ±7.81% vs.63.91% ±6.43%,t =2.679,P =0.005),CD4+ (39.01% ±4.98% vs.36.09% ±4.77%,t =2.031,P =0.024) and CD4+/CD8+ (1.82 ±0.48 vs.1.49 ± 0.32,t =2.743,P =0.004) were increased after treatment,with significant differences.While compared with before treatment,the proportions of CD8 + (24.08% ± 5.13% vs.26.04% ± 6.44%,t =1.142,P =0.130) and NK cells (22.68 % ± 9.56% vs.21.45 % ± 10.01%,t =0.426,P =0.337) had little changes,with no significant differences.There were 3 patients with complete remission,10 patients with partial remission,8 patients with stable disease and 2 patients with progressive disease when completing 6 cycles of PD-1 inhibitor treatment.Ten patients showed untoward effects such as mild sleepiness,thirst,tussis,pruritus and rash,and they were well tolerable.Conclusion PD-1 inhibitor can improve the patient's cellular immune function,and can achieve a more satisfactory short-term efficacy and acceptable adverse reactions,which maybe bring new hopes for patients with NSCLC.
2.Microstructural Abnormalities of Basal Ganglia and Thalamus in Bipolar and Unipolar Disorders: A Diffusion Kurtosis and Perfusion Imaging Study.

OBJECTIVE: Bipolar disorder (BD) is often misdiagnosed as unipolar depression (UD), leading to mistreatment and poor clinical outcomes. However, little is known about the similarities and differences in subcorticalgray matter regions between BD and UD. METHODS: Thirty-five BD patients, 30 UD patients and 40 healthy controls underwent diffusional kurtosis imaging (DKI) and three dimensional arterial spin labeling (3D ASL). The parameters including mean kurtosis (MK), axial kurtosis (Ka), radial kurtosis (Kr), fractional anisotropy (FA), mean diffusivity (MD), axial diffusivity (Da), radial diffusivity (Dr) and cerebral blood flow (CBF) were measured by using regions-of-interest analysis in the caudate, putamen and thalamus of the subcortical gray matter regions. RESULTS: UD exhibited differences from controls for DKI measures and CBF in the left putamen and caudate. BD showed differences from controls for DKI measures in the left caudate. Additionally, BD showed lower Ka in right putamen, higher MD in right caudate compared with UD. Receiver operating characteristic analysis revealed the Kr of left caudate had the highest predictive power for distinguishing UD from controls. CONCLUSION: The two disorders may have overlaps in microstructural abnormality in basal ganglia. The change of caudate may serve as a potential biomarker for UD.

Objective To analyze the effects of Hual qi huang granules on children with mycoplasma pneumoniae pneumonia.Methods A randomized,multicenter parallel controlled clinical trial was carried out.A total of 3 000 cases of hospitalized children with mycoplasma pneumoniae pneumonia were selected.All of them were given treatment for mycoplasma pneumoniae pneumonia with macrolide antibiotics and symptomatic treatment.They were randomly divided into 2 groups:research group and control group.The children of research group were give oral Huai qi huang granules for three months.According to the classification of pneumonia,these two groups were divided into:lobar pneumonia research group,lobar pneumonia control group,lobular pneumonia research group,lobular pneumonia control group.The hospitalization duration of fever,length of hospital stay,the absorption area of lung inflammation and pneumonia severity sores were observed.The frequency of upper respiratory infections,bronchitis,pneumonia were observed in 3 months after discharge.Results 2 378 cases were investigated.The hospitalization duration of fever,length of hospital stay of research group were significantly shorter than that of in control group (P < 0.001).The children with lobar pneumonia,2 weeks after treatment,the absorption of consolidation of the lobar pneumonia research group is significantly better than lobar pneumonia control group (P <0.001).After two weeks treatment,the pneumonia scores of lobar pneumonia research group is lower than lobar pneumonia control group (P < 0.05).Followup of 3 months after hospital discharge,frequency of upper respiratory infection and bronchitis of research group,were significantly lower than that of control.In addition,appetite increased significantly in research group than control (P < 0.001).There are 21 cases with drug associated adverse reactions (mild diarrhea),including 12 cases of research group,9 cases of control group,and there was no statistical significance (P >0.05).Conclusion Standard treatment combined with oral Huai qi huang granules in the treatment of mycoplasma pneumoniae pneumonia,can significantly shorten hospitalization duration of fever,length of hospital stay and reduce the severity score of pneumonia.Three months oral Huai qi huang granules can significant reduce the frequency of respiratory infections and bronchitis,also can increase patients appetite,and be safe.
4.Current research status of hepatitis D virus RNA detection

Hepatitis D caused by hepatitis D virus (HDV) infection is the most serious type of viral hepatitis. The prevalence rate of HDV has been seriously underestimated due to the lack of accurate HDV RNA detection methods. HDV RNA is the gold standard for the diagnosis of HDV infection and is of great significance in the diagnosis, screening, and treatment guidance of HDV. However, the multiple genotypes and strong secondary structure of HDV have led to great difficulties in HDV RNA detection. This article reviews the advances in HDV RNA detection methods and elaborates on the development from qualitative to quantitative detection methods, in order to provide new ideas for understanding the significance of HDV RNA detection and promoting the research and development of new HDV RNA detection methods.
5.Effect of CAR⁃NK⁃92 targeting MSLN on ovarian cancer

Objective :
To explore the killing effect of chimeric antigen receptor NK⁃92 (CAR⁃NK⁃92) cells targeting mesothelin (MSLN) on ovarian cancer cells.
Methods :
The expression of MSLN in primary ovarian cancer tissues and ovarian cancer cell lines was detected by immunohistochemical analysis and immunofluorescence, respectively. The CAR⁃NK⁃92 cells targeting MSLN were constructed by lentiviral transfection, the transfection efficiency was detected by flow cytometry, and the killing effect of CAR⁃NK⁃92 on ovarian cancer was verified in vivo and in vitro.
Results :
MSLN was highly expressed in primary ovarian cancer tissues and ovarian cancer cell lines. Flow
cytometry showed that the purity of CAR⁃NK⁃92 targeting MSLN was about 70% . The experiments found that MSLN⁃CAR⁃NK⁃92 cells had a strong anti⁃ovarian cancer effect in vivo and in vitro , and could release more cytokines interferon⁃γ and tumor necrosis factor⁃α .
Conclusion
MSLN⁃CAR⁃NK⁃92 has strong anti⁃ovarian cancer effects in vivo and in vitro, providing a new potential treatment option for ovarian cancer patients.
