1.Effect of preemptive analgesia with rofecoxib in a rat model of acute pain

Objective There are still different news on the preemptive analgesia. We aimed to investigate the preemptive analgesic effect of rofecoxib on the tetrodotoxin-resistant (TTX-R) sodium current in dorsal root ganglion (DRG) neurons m a rat model of acute pain. Methods Seventy-five male SD rats weighing 100-140 g were randomly divided into five groups with 15 animals in each group: (A) sham operation + placebo group; (B) operation control group received placebo orally before operation; (C) preoperatine rofecoxib group received oral rofecoxib 10mg?kg-1 h before operation; (D) and (E) postoperatire, rofecoxib group Ⅰ and Ⅱ received oral rofecoxib 10mg?kg immediately (D) and 1 h (E) after operation. Acute pain was produced by an 1 cm long incision in the plantar surface of the right hindpaw through skin, fascia and muscle. Pain threshold to mechanical stimulation with von Frey filament was measured before (baseline) and 2, 6,12, 48, 96 h after operation. The animals were killed by cervical dislocation before and 24, 48, 96 h after incision. DRGs at L3.5 on the operated side were isolated and minced. DRG neurons were isolated by digestion with collagenase Ⅰ A and trypsin TTX-R Na-current density in DRG neurons were measured by using whole-cell patch clamp technique. Results There was no significant change in pain threshold to mechanical stimulation and Na-current density in DRG neurons during the experiment in group A, The withdrawal threshold to mechanical stimuli was significantly decreased after operation compared to the baseline in the four groups ( B, C, D, E). The decrease in pain threshold was significantly less in group C,D,E than that m group B at 2, 6, 12, 24, 48 h after operation and was the least in group C Current density (pA/pF) was significantly increased at 24 h and 48 h after operation compared to baseline in group B, C, D, E. The increase in current density was significantly less in group C, D, E than in group B and was the least in group C.Conclusion Rofecoxib given before operation has preemptive analgesic effect in a rat model of acute pain. Stronger inhibition of TTX-R Na-current m DRG neurons may contribute to the underlying mechanism.
2.A Novel Method for Extraction of Heart Sounds Envelope Based on Interpolation of Local Peak and Valley Points

Objective To extract envelope of heart sounds exactly,for the purpose of the further analysis of its characteristics.Methods The way that envelope extraction of heart sounds based on key-points was given.The points of local peak and valley were calculated firstly,and then heart sound envelope was gotten by the interpolation of these points.Results Compared with the envelope extracted by Hilbert-transform and mathematical morphology,respectively,the outline of heart sounds was extracted more accurately,and its time-domain characters were acquired by this method.Conclusion The envelope of heart sound is extracted correctly by this method,which is useful for the further analysis.
3.Angiotensin Ⅱ -induced apoptosis of podocyte is meliorated by overexpression of nephrin via PI3K-Akt signaling pathway

ObjectiveTo evaluate the effects of Ang Ⅱ on apoptosis of podocytes and explore the signaling pathwayof nephrin in preventingAng Ⅱ-inducedpodocyte apoptosis.MethodsDifferentiated mouse podocytes were exposed to Ang Ⅱ at different concentrations for 18 h or at 10-8 mol/L for variable incubation times.Undifferentiated mouse pedocytes were transfected using lipofectamine 2000 with the pcDNA3.1-mNPHS1 plasmid and stably transfected cell lines were generated with G418 selection.In separated experiments,untransfected mouse podocytes (MPC) and stably transfected podocytes with pcDNA3.1-neo and PcDNA3.1-mNPHS1 were exposed toAng Ⅱ(10-8 mol/L) or LY294002(a selective Akt inhibitor,50 μmol/L) for indicated times.Apoptosis was evaluated by flow cytometry.The expression of nephrin was assessed by quantitative real-time PCR,immunofluorescence and Western blotting.The phosphorylation level of Akt was determined by Westem blotting.Results(1) AngⅡ promoted podocyte apoptosis in a dose-and time-dependentmanner. PretreatmentwithlosartansignificantlypreventedAngⅡ -induced apoptosis. (2) Nephfin mRNA and protein were obviously decreased in podocytes exposed to 10-8 mol/L Ang Ⅱ for at least 12 h than those in vehicle-treated cells (P<0.05).(3) Ang Ⅱ exposure for more than 15 min inhibited the phosphorylation of AKT in MPC,which was dramatically reversed by pcDNA3.1-mNPHS1 transfection,but not by pcDNA3.1-neo transfection. (4) Podocyte apoptosis was promoted byLY294002. Conversely,Ang Ⅱ-induced podocyteapoptosis was significantly alleviated by pcDNA3.1-mNPHS1 transfection.ConclusionAng Ⅱinduces mouse podocyte apoptosis which is suppressed by overexpression of nephrin through PI3K-Akt signaling pathway.
4.Ultrasonography and magnetic resonance imaging for diagnosis of fetal skull brain deformities

Objective To evaluate ultrasonography combined with magnetic resonance imaging(MRI) diagnosis value for fetal brain deformities.Methods The results of ultrasonography and MRI was analysied in 12 fetuses with deformities,which were compared to postnatal,autopsy or other hospital inspection results and evaluated the accuracy.Results In 8 cases,the diagnoses established by ultrasonography were correct.3 cases were misdiagnosed by ultrasonography,1 case was missed the diagnosis.In 11 cases,the diagnoses established by MRI were correct and 1 case was missed the diagnosis.Conclusion Ultrasonography must be choosed in diagnosising fetal skull brain deformities,MRI was supplement to ultrasonography in complicated pregnancies.The combination of ultrasonography and MRI have an important value in guiding pregnant women to determine whether or not to continue the pregnancy.
5.The effects of a rotating magnetic field in the prevention and treatment of irradiation-induced esophagitis

Objective To observe the effect of a rotating magnetic field in preventing and treating irradiation-induced esophagitis in rats. Methods Forty female Wistar rats were randomized into 5 groups:a non-irradiated control group,an irradiation group,an amifostine treatment group ( amifostine group ),a 90 min magnetic field treatment group (90 min magnetic group) and a 120 min magnetic field treatment group ( 120 min magnetic group),with 8 rats in each group.The esophaguses of all rats except those in the control group were exposed to a single irradiation with 6 MV X-rays from a linear accelerator at a dosage of 43 Gy.Four rats in each group were randomly chosen to be observed 1 and 2 weeks after the irradiation.Blood cytokines were detected in their arterial blood.Any pathological changes of the esophagus were observed with HE staining under a light microscope at the same time. Results Irradiation-induced esophagitis was observed in the irradiation group 7 days after irradiation,with obvious exfoliation and necrosis of the esophagal epithelium mucosae.The submucosa were hyperaemic and dropsical with abundant inflammatory cell infiltration.The pathological changes of the esophagus were similar at 7 and 14 days after irradiation.However,the irradiation-induced esophagitis of rats in the amifostine group,the 90 min magnetic group and the 120 min magnetic group were relatively slighter and the blood leucocytes and neutrophis in those 3 groups were significantly lower than those in the irradiation group,while a tendency toward repair of the mucosa of the esophagus was detected.Serum TNF-α,IL-1 and IL-6 in the 90 min magnetic group and 120 min magnetic group were significantly lower than those in the irradiation group. Conclusions Both a rotating magnetic field and amifostine can help prevent and treat irradiation-induced esophagitis.Their therapeutic efficacy is similar.Exposure to a rotating magnetic field could inhibit the expression of inflammatory factors,and thus lessen the inflammatory reaction of acute irradiation-induced esophagitis.
6.A comparative study of 11C-MET PET with MRI for target volume delineation in postoperative radiotherapy for brain high grade glioma

Objective To evaluate the value of L-(methyl-11C)-labeled methionine positron emissions tomography (MET PET) and MRI in target volume delineation for postoperative radiotherapy for brain high grade glioma (HGG).Methods Thirty-seven patients with supratentorial HGG were included.Both MRI and MET PET scan were performed in the same treatment position for all patients.The consistency to determine residual tumor between MRI and MET PET was analyzed.Imaging data of MET PET and MRI were coregistered using the BrainLAB image fusion software.The extension of the volume with high uptake (VMET) on MET PET were compared quantitately with the enhancing area on MRI T1W gadolinium enhancement (VGd) and the hyperintensity area on MRI T2W (VT2).Results Both MET PET and MRI were positive for 19 patients and negative for 7 patients.The consistency between these two scans was 70.3%.MET PET was integrated with MRI in 30 patients with positive MET uptake.VMET were partially or entirely outside VGd in 29 patients and VT2 in 17 patients, whereas VGd and VT2 were partially or entirely outside VMET in all patients.The maximal distance from the margin of VMET to VGd was ≥ 2.0 cm in 50%patients and the corresponding distance of VMET to VT2 was ≥ 1.0 cm in 33% patients.Conclusions The differences are existing between MET PET and MRI in determination and identification of the location and extension of residual tumor for patients with HGG.The integration of MET PET and MRI can accurately delineate radiation target volume.
7.Study on the mechanism of the annexin II-mediated co-assembly of t-PA and plasminogen.

In order to further investigate the effect of annexin II (Ann-II) on tissue plasminogen activator (t-PA)-dependent plasminogen (PLG) activation and its interactive mechanism, recombinant native Ann-II bound t-PA, PLG and plasmin with high affinity was examined. The flow cytometric assay showed that the ann-II expression rate was higher in the human umbilical vein endothelial cell (HUVEC) (87.65%) than in the HL-60 cells as controls (35.79%). Two irrelevant proteins, bovine serum albumin (BSA) and equine IgG (EIG) had no effect on the production of plasmin. Ann-II-mediated enhancement of t-PA-dependent PLG activation was inhibited by epsilon-aminocaproic acid or by pretreatment of Ann-II with carboxypeptidase B with the inhibitive rate being 77.8% and 77.0%, respectively. It was revealed that the effect of Ann-II on PLG activation was specific for t-PA. Urokinase didn't bind to Ann-II, demonstrating the role of receptor-related lysine residues on activation of PLG, showing that the Ann-II-PLG interaction was dependent upon carboxyl-terminal lysine residues. These findings suggest that annexin II-mediated co-assembly of t-PA and PLG may promote plasmin generation and play a key role in modulating fibrinolysis on the endothelial surface.

8.Clinical features and CT appearances of primary adenosquamous carcinoma of the liver

Objective To investigate the clinical features and CT appearances of primary adenosquamous carcinoma (ASC)of the liver and to improve the understanding of this disease for the sake of misdiagnosis.Methods The clinical features and CT appearances in 7 patients proved by operation and pathology were reviewed,retrospectively.Results All of the patients had dull pain in upper abdo-men,fever,chills and a long history of cholangitis and biliary calculi.In addition,all patients had not the history of hepatitis B and liver cirrhosis.And the serum AFP level was normal.The single lesion was found in every patient including 4 lesions in the left lobe of liver and the other 3 in the right lobe.Plain CT showed all masses with hypodensity,heterogeneity and unclear edge in liver,and multiple irregular and more hypodense areas in lesions were found.Slight heterogeneous honeycomb-like enhancement in the arterial phase was showed.In the venous phase,persistent honeycomb-like enhancement with uneven separations,nodular bulge and hypo-dense necrotic area was found.In the delayed phase,further honeycomb-like enhancement with hypodense necrotic area and obvious-ly enhanced nodular bulge was showed.The bile ducts in the liver and around the mass were dilated and had pneumatosis in company with lithiasis in choledochus and intrahepatic bile duct in 5 patients.Conclusion Primary hepatic ASC has certain clinical character-istics in older patients.The CT characteristic features included:honeycomb-like lesions with persistent,heterogeneous,delayed en-hancement and heterogeneous separation,uneven inner edge and enhanced nodular bulge.
9.Effects of a Baqia Huaihua Liangxue decoction on peripheral blood T helper 17 cells/Tregulatory cells balance and the expression of related cytokines in elderly patients with psoriasis

Objective:To investigate the effects of a Baqia Huaihua Liangxue decoction on peripheral blood T helper 17 cells/Tregulatory cells(Th17/Treg)balance and the expression of related cytokines in elderly patients with psoriasis.Methods:A total of 124 elderly patients diagnosed with psoriasis and receiving treatment at our hospital from September 2018 to December 2019 were divided into the observation group and the control group according to the parity number table, with 62 cases in each group.All patients underwent conventional treatment and physical therapy, and, in addition, the observation group was treated with a Baqia Huaihua Liangxue decoction for 2 months.The blood indexes including levels of Th17, Treg, Th17/Treg, and Th17/Treg-related cytokines such as interleukin-17(IL-17), IL-22 and IL-23 in the two groups were measured before treatment and 2 months after treatment.The psoriasis area and severity index(PASI)was used to evaluate the severity of skin damage in the two groups.The traditional Chinese medicine(TCM)syndrome scores were evaluated and compared between the two groups.Adverse reactions during treatment were monitored and recorded.Results:After 2 months of treatment, the Th17 and Th17/Treg levels were lower[(1.0±0.1)% vs.(2.4±0.9)%, 0.2±0.1 vs.0.6±0.2, all P<0.05]and Treg levels were higher in the observation group than in the control group[(5.3±1.1)% vs.(4.3±1.0)%, P<0.05]. Levels of IL-17, IL-22 and IL-23 were lower in the observation group than in the control group after 2 months of treatment[(22.1±3.5)ng/L vs.(35.6±4.0)ng/L, (44.8±4.4)ng/L vs.(49.9±5.1)ng/L, (43.7±5.2)ng/L vs.(52.5±5.4)ng/L, all P<0.05]. The PASI score and TCM syndrome score were lower in the observation group than in the control group after 2 months of treatment[(2.9±1.0) vs.(6.1±2.3), (3.2±1.9) vs.(7.6±2.1), all P<0.05]. No adverse reaction occurred in the two groups during treatment. Conclusions:The Baqia Huaihua Liangxue decoction has a clear therapeutic effect in elderly patients with psoriasis.It can stabilize peripheral blood Th17/Treg balance, inhibit the expression of related cytokines, reduce the degree of skin damage and has no adverse reaction during treatment.
10.Fatty acid metabolism and tumor occurrence, development and metastasis

In recent years,the relationship between fatty acid metabolism and tumor has become a hot topic and widely recognized.In tumor cell,fatty acid metabolism is important to its growth and metastasis.About synthesis,fatty acid can participate in the synthesis of phospholipids on the membrane of cancer cells and take part in important signal transduction pathways,such as PI3K-AKT-mTOR;in fatty acid catabolism,tumor cells can produce ATP to maintain its growth by β-oxidation;what's more,it could be kept redox homeostasis by producing nicotinamide adenine dinucleotide phosphate hydrogen (NADPH).So,anabolism and catabolism of fatty acid may promote occurrence and development of tumors,but the mechanisms are unclear.This review highlights the effects of fatty acid metabolism on the occurrence,development and metastasis of tumors.
