1.Activity Study on Antibacterial Protein Isolated From Musca domestica During Larvae-pupae Metamorphosis

Objective: To research the activity of antibacterial proteins isolated from Musca domestica during larvae-pupae metamorphosis. Methods: The 5-day-old larvae of Musca domestica were injured with a needle. 24h later, the larvae which had changed into pupae were selected. The antibacterial protein was isolated by a four-step protocol including grinding, heating, CM-sepharose F. F. cationexchange chromatography and another CM-sepharose F. F. chromatography. Antibacterial activities of samples were tested by an ultrasensitive radial diffusion assay using Staphylococcus aureus as the indicator organism. The molecular weight of the isolated protein was determined by SDS-PAGE.Results: The molecular weight of this isolated protein was 43kDa, and its antibacterial activity was strong. Conclusions: A kind of protein with strong antibacterial activity can be produced in the Musca domestica during larvae-pupae metamorphosis.
2.Coxsackie-virus and adenovirus receptor expression in lung cancer bronchiole-alveolar carcinoma features and its significance

Objective:This study aims to investigate the protein expression of coxsackie-virus and adenovirus receptor (CAR) in partial subtypes of pulmonary adeno-carcinoma, as well as its expression with clinico-pathological factors and prognosis. Methods:CAR expression was immunohistochemically assessed in 137 cases of lung cancer with bronchiole-alveolar carcinoma (PWBF) fea-tures, and analyzed in relation to various clinico-pathological parameters. All data were analyzed using SPSS statistics software, and Ka-plan-Meier survival curves were constructed. A Log-rank test was also conducted. Results: The CAR positive rates in PWBF, other types of lung cancer, and normal lung tissue were 71.5%, 50.0%, and 13.3%, respectively. The difference was statistically significant (P<0.05). In addition, a statistically significant difference was observed between the positive expression of CAR and other clini-co-pathologic parameters, such as pathological type and histological differentiation. No statistical significance was observed in other pa-rameters, such as gender, age, smoking, and tumor diameter. Patients with CAR positivity were characterized by longer survival times than the others;however, this difference did not reach statistical significance. Conclusion:CAR is related to the occurrence and devel-opment of lung cancer, especially PWBF. The higher expression of CAR in PWBF for gene therapy with adenovirus vectors opened a broader space. The conspicuous regulation of CAR may be reliable evidence and may bright future for the gene therapy of other types of lung cancer.
3.Caspase-8 dependent osteosarcoma cell apoptosis induced by proteasome inhibitor MG132

AIM: To investigate the effects of proteasome inhibitor Z-LLL-CHO(MG132) on human osteosarcoma cell line MG-63 and its possibly mechanism.METHODS: After treated with different concentration of MG132,the morphological change,ultrastructral morphology,cell viability,cell apoptosis,gene transcription and protein expression in MG-63 cells were accessed by fluorescence microscope,electron microscope,MTT assay,agrose gel electrophoresis,FCM,RT-PCR and Western blotting.RESULTS: Proteasome inhibitor MG132 was an effective inducer of apoptosis in human osteosarcoma MG-63 cells.Not only apoptotic changes,but also cell arrest at G2-M-phase,the accumulation of p27kip1,the accumulation of activated caspase-8 and increased ratio of Bax∶Bcl-2 were observed.However,to normal human diploid fibroblast cells,MG132 did not show apoptosis-inducing effect.CONCLUSION: Apoptosis induced by MG132 may be caspase-8,p27kip1and bcl-2-related.
4.Discuss on the application of informatization clinical blood transfusion apps in blood transfusion management under the new normal

Objective To study the value of the informatization clinical blood transfusion apps application in blood transfusion management under the new normal.Methods By retrospective analysis of the audit qualification rate of 16 988 general clinical blood transfusion and informatization clinical blood transfusion apps forms,and both types of analogy each stage time nodes correlation analysis.Results Review the qualification rate of the 8 616 general clinical blood transfusion form is 76.9%,the time of the doctor's advice to send is (20± 8) minutes,the time of the Blood Transfusion Department to receive is (90±48) minutes;review the qualification rate of the 8 372 informatization clinical blood transfusion apps form is 99.2%,the time of the doctor's advice to send is (3± 1) minutes,the time of the Blood Transfusion Department to receive is (2± 1) minutes.Conclusion Informatization clinical blood transfusion application form to adapt to clinical doctor operation,ensuring the safety of blood transfusion by shortening the each stage time nodes for clinical service.
5.Effect of Motor Imagery on Balance Ability in Hemiplegics after Stroke

Objective To study the effect of motor imagery on balance rehabilitation in hemiplegics after stroke.Methods 20 hemiplegic patients who were in chronic stage(above 6 months)were randomly divided into two groups.Normal rehabilitation was performed in the whole stage and motor imagery was added in group A in stage Ⅰ and group B in stage Ⅱ.All patients were assessed with Berg Balance Scale(BBS),blance subscale of the Fugl-Meyer Assessment(FM-B),max affected limb-loading per weight and Functional Independence Measure(FIM)at the beginning and the ending of the every stage,respectively.Results The deference scores of BBS and FIM,as well as max affected limb-loading per weight between in the stage with motor imagery was better than those in the stage without motor imagery(P<0.01),as well as that of FM-B score(P<0.05).Conclusion Motor imagery can improve balance rehabilitation and activity of daily living in chronic hemiplegics.
6.Effect of Ankle-Foot Retractor on Ankle Joint Motion and Equilibrium Function of Stroke Hemiplegic Patients

Objective To study the effects of ankle-foot retractor on ankle joint motion and equilibrium function of stroke hemiplegic patients.Methods 74 patients were divided into two groups: observation group (n=36) and control group (n=38). The control group was trainedby routine rehabilitation training program and electro-uprise bed, the observation group was trained by ankle-foot retractor based on routinerehabilitation training program. The effect was evaluated after 8 weeks. Results After training, either the observation group or the controlgroup showed significant improvement at motion of ankle joint, activity of daily living (ADL), and equilibrium function, but the observationgroup was better than the control group (P<0.001). Conclusion Ankle-foot retractor plays a positive role on motion of ankle joint, ADL andequilibrium function in stroke patients.
7.Interference from Filter Film Adsorption for Dissolution of Clarithromycin

Objective:To find the reasons for the low dissolution of clarithromycin tablets and study the interference from filter film adsorption at various time points to explore the appropriate processing approach for dissolution solution of clarithromycin tablets. Meth-ods:Clarithromycin tablets from two different manufacturers were used. The dissolution solution was prepared according to Japanese Orange Book. The dissolution was determined after different processing and the adsorption rate of the filter film was calculated. Re-sults:Totally 14 kinds of filter films were tested with different adsorption for clarithromycin, and the adsorption rate of some kinds of filter films exceeded the prescribed limit. Conclusion:The absorption rate of filter films for clarithromycin can be decreased by boiling the films and using American membranes. The interference from filter film adsorption can be reduced and inhibited by rejecting the first filtrate above 5ml or centrifuging dissolution solution.
8.Multislice spiral CT and digital subtraction angiography in interventional therapy of hepatocellular carcinoma

Objective To investigate the value of multislice spiral CT(MSCT,)in interventional therapy of the hepatocellular carcinoma(HCC)emphasising on transcatheter hepanc arterial chemoembolization(TACE).Methods MSCT were performed in 60 cases of HCC before interventional procedure,CT findings of hepatic artery phase,portal venous phase and hepatic venous phase were observed respectively,among which CTA were done in 15 cases,and the anatomy of celiacartery and its branches were observed in 45 cases.The schemes of interventional therapy were worked out according to the findings of MSCT.Results MSCT showed 250 lesions,10 cases of tumor thrombosis in portal vein and 19 cases of hepatic arterioportal shunt.There was no significant difference between MSCT and digital subtraction angiography(DSA)in positive rate of in showing number of tumor or tumor thrombosis in portal vein(P>0.05),but the 3D construction of celiac artery branches in CTA was better than that in DSA,while angles between celiac artery and abdominal aorta in MSCT were more convenient than that in DSA.MSCT showed 5 eases of hepatic artery original abnormality,according to that in DSA.Conclusion MSCT is of importance for guidance of interventional therapy of hepatocellular carcinoma.
9.Controllable synthesis of different size hydroxyapatite nanoparticles and their biological effects on osteoblast-like cells

ObjectiveTo investigate the size effect of hydroxyapatite nanoparticles on proliferation and apoptosis of osteoblast-like cells.MethodsCetyltrimethylammnonium bromide (CTAB) was used to regulate the size of nano hydroxyapatite (nHAP) particles.All obtained particles were characterized by transmission electron microscopy (TEM),X-ray diffraction,dynamic light scattering and chemical analysis.HAP films were obtained by slowly coating cover glasses with 1% HAP particle suspension.MG-63 cells on three different films(20HAP,40HAP and 80HAP) were cocultured for up to 5 days.Cell proliferation assay was obtained by methyl thiazolyl tetrazolium (MTT).Cell apoptosis was detected by flow cytometric detection.Cell ultrastructure morphology was observed by TEM observation.ResultsnHAP with diameter of 20 nm,40 nm and 80 nm were synthesized and and analyzed.The MG-63 cells were cultured on three different fihns.The optical density value of cells on 20HAP was 1.22±0.13 after 5 days incubation,and there was no different compared to the control group(F=6.843,P=0.124).Cell number and viability were significantly higher on 20HAP compared to large nHAP after 5 days incubation.The percentage of apoptotic cells increased with increasiug nHAP particle size.TEM images showed 20HAP was found in cytoplasm and cell morphology had no changes.ConclusionBoth cell proliferation and cell apoptosis are related to the size of the nHAP particles.20HAP was the most effective on promoting cell growth and inhibiting cell apoptosis.
10.The effective study of Miller pyramid demonstration applied in the teaching of shoulder physical examination for visiting orthopaedic surgeons

Objective To explore the effect of Miller pyramid demonstration in the teaching of shoulder physical examination for visiting orthopaedic surgeons and aim at finding a more effective teach-ing method in this area. Method 96 visiting surgeons in our department were chosen and divided even into two groups randomly from 2011 to 2013. Traditional teaching method was applied in group A, while Miller pyramid demonstration which combined theory with practice was used in group B. Basic knowledge, manipulative skill and clinical diagnosis were assessed in group A and group B respectively. Finally, the results were statistical analyzed using t-test with SPSS 19.0 software. Results In the basic professional knowledge test, the average score of Group B was better than Group A [(92.8 ±5.0) vs. (83.0 ±8.5)]; in terms of operational skills assessment, the average score of Group B was better than Group A [(94.0 ±4.1) vs. (79.3± 3.9)]; in the clinical diagnosis of the application, the average score of Group B was better than Group A [(90.6±2.3) vs. (85.1±1.9)]. There were significant differences between the two groups (P<0.05). Conclusion Miller pyramid demonstration was more effective than traditional teaching method in the teaching of shoulder physical examination for visiting orthopaedic surgeons.
