Aim To investigate the effects of chloride on the injury of the ventricular myocytes from anoxia-reoxygenation. Methods Under the conditions of anoxia-reoxygenation injury, primary cultured rat ventricular myocytes were treated with 4-acetanide- 4′-isothiocya- natostilbene -2,2′-disulfonic acid (SITS),4,4′,-dii sothiocya-nostilbene-2,2′-disulfonicacid (DIDS) or replaced Cl~- with equimolar gluconate, respectively. Then the cell viability and the contents of Lactate dehydrogenase (LDH), malondialdehyde (MDA), superoxide dismutase (SOD)and glutathione peroxidase (GSH-Px) in the media were measured. Results The release of LDH and MDA was significantly increased in the anoxia-reoxygenation group, while the cell viability and the activity of SOD, GSH-Px decreased significantly compared with those in the control group. In both Cl~--free+ A-R group and SITS+A-R group, LDH and MDA release was noticeably lower than those of the A-R group, while the cell viability and the activity of SOD, GSH-Px significantly increased compared with those in the anoxia-reoxygenation group. But the cell viability and the contents of LDH, MDA, SOD and GSH-Px in the DIDS+A-R group had no significant change compared with those in the anoxia-reoxygenation group. Conclusion Cl~- plays an important role in anoxia reoxygenation injury. SITS provides effective protection to the cardiac myocyte subjected to anoxia reoxygenation injury, while DIDS cannot provide such protection.

Objective To investigate the value of laparoscopy in the treatment of liver cancer. Methods The clinical data of 128 liver cancer patients who received laparoscopic surgery at Southwest Hospital from March 2007 to October 2009 were retrospectively analyzed.Of all patients,116 were with primary liver cancer, and 12 with metastatic liver cancer. There were 107 patients who received laparoscopie bepatectomy,15 received laparoscopic radiofrequency ablation(RFA)and 6 received laparoscopic ligation of the right branch of portal vein. Results Of the 107 patients who received laparoscopic bepatectomy,7 were converted to open surgery, and 5 were converted to hand-assisted laparoscopic hepatectomy.Anatomical hepatectomy was performed on 88 patients, including left lateral lobectomy on 21,left hemihepatectomy on 15,extended left hemihepatectomy on 2,medial lobectomy on 1,right hemihepatectomy on 11,right posterior lobeetomy on 9 and hepatic segmentectomy on 29.Combined hepatic resection was performed on 4 patients, and nonanatomical hepatectomy on 15.The mean oporatire time and blood loss were(228±92)minutes and(393±213)ml,with no operative mortality. The mean postoperative hospital stay was(8±4)days,and the incidence of complications was 15%(16/107).A total of 126 patients were followed up for 1-42 months,12 patients with laparoscopic hepatectomy died within 16 months, with the mean survival time of(118±7)weeks and the mean tumor free survival time of(105±7)weeks;2 patients with laparoscopic RFA died within 11 months:2 patients with laparoseopie ligation of the right branch of portal vein received two-stage radical resection. Conclusion Laparoscopic surgery is safe and feasible with the advantages of minimal operative trauma and quick recovery of patients when it is applied to the treatment of liver cancer.

AIM: To study the distribution of CYP2C9∗3 and VKORC1-1639G>A gene polymorphism in Han population in Anhui province and their influence on the stable dose of warfarin. METHODS: The blood samples of 1 169 patients from 6 tertiary general hospitals in 5 areas of Anhui province from January 2020 to December 2021 were selected, the genotype of CYP2C9∗3 and VKORC1-1639G>A was detected by fluorescent staining in situ hybridization technique. RESULTS: The distribution of CYP2C9∗3 genotypes in 1 169 patients: the frequencies of AA, AC and CC genes were 90.16%, 9.24% and 0.60%, respectively; The distribution of VKORC1 genotype: the frequencies of AA, AG and GG genes were 84.26%, 14.71% and 1.03% respectively; There was no significant difference between the two genotypes in gender, age and regional distribution (P>0.05). The average daily warfarin dose of CYP2C9∗3 AA genotype in 755 patients with stable warfarin dose was (3.02±0.59) mg/d, which was significantly higher than patients with AC genotype and CC genotype; The average daily warfarin dose of patients with VKORC1-1639AA genotype was (2.72±0.40) mg/d, which was significantly lower than that of patients with AG genotype and GG genotype (P<0.05). And the difference was statistically significant (P<0.05); There are significant differences in gender, age and clinical diagnosis between patients with stable dose of warfarin and those without stable dose (P<0.05). CONCLUSION: CYP2C9 and VKORC1 genotypes are associated with the stable dose of warfarin. Clinical anticoagulation therapy guided by CYP2C9 and VKORC1 genotypes can provide guidance for individualized medication of warfarin.
