1.Thrombolysis for Treating Acute Deep Vein Thrombosis in the Lower Limbs

Objective:To observe the effect of thrombolysis in treating acute deep vein thrombosis (DVT) in the lower limbs.Methods:For the 14 patients diagnosed by colour Doppler ultrasonographic examination,intravenous drip of urokinase,dextran and mailuoning was ministered alternatively through the superficial venosus dorsalis pedis in the involved side.Meanwhile,oral intake of aspirin and dipyridamole was also prescribed.Results:Ten patients healed and four significantly improved.Conclusion:The efficacy of thrombolysis for treatment of DVT in the lower limbs is valid.This intervention is especially suitable for those patients who missed the optimum time window of operation.
2.Evaluation of Literature Quality of Acupuncture in Treatment of Chronic Fatigue Syndrome

Objective To assess the quality of literature of clinical studies on acupuncture in treatment of chronic fatigue syndrome.Methods The literatures between 1998-2008 were searched by means of electronic retrieval.Type and methodology,general condition,criteria for diagnosis and assessment of therapeutic effects,following-up and side effects etc.in clinical studies were evaluated according to principles and methods of clincial epidemiology and evidence-based medicine.Results Of the 78 literatures enrolled,76 were studied in China and published in Chinese journal of China,only 2 were English literature and published in English journal of China.Only 7 were authentic randomized controlled trials(RCTs),21 quasi-randomized controlled trials and RCTs can't contact with authors,3 non-randomized concurrent controlled trials and 47 narrative studies.50 had clear international recognized diagnostic ctriteria.22 mentioned enrolled or excluded criteria.26 adopted TCM criteria for therapeutic effects,27 evaluated therapeutic effects with grading.4 the sample content above 100,33 between 60 and 99,4 mentioned compliance evaluation,3 described retreat cases,2 reported follow-up.11 reported adverse reaction.Conclustion Clinical studies on acupuncture in treatment of chronic fatigue syndrome were mainly carried through in China,and the quality of study and literature is low comparatively.Therefore,high quality's RCTs with strict design are needed urgently in this area and the researchers should contribute to journals overseas.
3.Study of T-wave End Detection with Geometric Method

Purpose: To realize the detection of T-wave end with an efficient and accurate method. Methods: R-wave was lo-cated using wavelet transforming, and then the T-wave detection interval was identified accttrately by avoiding the influence of heart rate and P-wave. In addition, T-wave peaks were determined referring to the modulus maxima in the interval. Based on these, the T-wave end was located with a simple and practical geometrical method, which was applied for the evaluation of QT database. Results: The positioning capability of the above-mentioned method reached to (-0.35225±18.5869) ms which was comparable or even better than those manual annotations from QT database by cardiologists. Conclusions: The experimental results showed that our algorithm for T-wave end detection is robust to noise, baseline drift and waveform morphological varia-tions, and easy to calculate and to realize.
4.Coexistence of aldose reductase gene and endothelial nitric oxide synthase polymorphisms associates with diabetic nephropathy

BACKGROUND: It has been demonstrated that the C-106T polymorphism at promotor region of aldose reductase gene and GLU298ASP (894G→T) polymorphism at exon 7 of endothelial nitric oxide synthase (eNOS) gene are associated with diabetic nephropathy, it should be further studied wnether the risk for diabetic nephropathy will increase when the above polymorphic sites coexist.OBJECTIVE: To investigate the association between the coexistence of the polymorphisms of both the C-106T at promotor region of aldose reductase gene and GLU298ASP (894G→T) at axon 7 of eNOS gene in chromosome 7q35 region and the genesis of diabetic nephropathy in northern Chinese Hah people.DESIGN: A case-controlled, comparative observation.SETTING: Department of Endocrinology, the Affiliated Hospital of Medical College, Qingdao University.PARTTCIPANTS; Totally 139 inpatients with type 2 diabetes mellitus were selected from the Department of Endocrinology, the Affiliated Hospital of Medical College, Qingdao University from November 2002 to April 2005,including 54 males and 85 females, (64±8) years of age. Inclusive criteria: Accorded with the standards of diabetic diagnosis and classification by WHO in 1999. According to the 24-hour urinary albumin excretion rate (UAER), they were divided into diabetic nephropathy group (n =61) and non-diabetic nephropathy group (n =78). Meanwhile, 63 healthy controls were selected as the control group, including 24 males and 39 females, 50-78 years of age. All the enrolled subjects were Han people. Informed contents were obtained from all the subjects.METHODS: The genotypes and alleles of polymorphisms of GLU298ASP(894G→T) at exon 7 of eNOS gene and C-106T at promotor region of aldose reductase gene were detected by polymerase chain reaction restriction-fragment length polymorphism technique (PCR-RFLP), DNA sequencing technique and agarose gel electrophoresis separation technique.Then the frequency of genotypes and alleles were compared.MAIN OUTCOME MEASURES: ① Polymorphisms of eNOS gene and aldose reductase gene; ② Results of the multivariant stepwise regression analysis of risk factors for diabetic nephropathy; ③ Polymorphisms of aldose reductase C-106T and eNOS 894G→T and the relative risk of diabetic nephropathy.RESULTS: All the 139 patients with type 2 diabetes mellitus and 63 healthy adults were involved in the analysis of results. ① The frequencies of the T allele and TG genotype at exon 7 894G→T polymorphic site of eNOS gene in the diabetic nephropathy group were significantly higher than those in the non-diabetic nephropathy group and control group (χ2=8.261, 19.629, P < 0.05). ② The frequencies of the T allele and CT genotype at promotor region of aldose reductase gene in the diabetic nephropathy group were significantly higher than those in the non-diabetic nephropathy group and control group (χ2=6.343, 8.940, P < 0.05, 0.01). ③ After associated analysis on the above gene polymorphisms, it was found that the frequency of TG/CT genotype in the diabetic nephropathy group were significantly higher than that in the non-diabetic nephropathy group and control group (χ2=6.972, P < 0.01); whereas the frequency of GG/CC genotype was significantly lower than that in the non-diabetic nephropathy group and control group (χ2=13.304, P < 0.05). ④Glycosylated hemoglobin (GHbA1c), systolic blood pressure, total cholesterol, polymorphisms of 894G→T at exon 7 of eNOS gene and C-106T at promotor region of aldose reductase gene were all the independent risk factors for diabetic nephropathy (Wald =5.627, 4.92, P < 0.05). ⑤ GG/GC genotype might be the protective genotype for diabetic nephropathy (OR=0.25, P < 0.01); The risk for diabetic nephropathy in the carrier of GG/CT or TG/CC genotype increased by 2.3 times and risk for diabetic nephropathy in the carrier of TG/CT genotype increased by 4.8 times.CONCLUSION: The coexistence of polymorphisms of 894G→T at exon 7 of eNOS gene and C-106T at promotor region of aldose reductase gene can increase the risk of diabetic nephropathy in patients with type 2 diabetes mellitus. And the TG/CT haplotype formed by the coexistence of polymorphism of these two genes is probably the predisposing genotype for diabetic nephropathy.

6.3M-Z350 Nano-resin and 3M-Z100 composite resin for cosmetic restoration of anterior teeth with vital pulp: Evaluation on postoperative sensitivity and pain

BACKGROUND: Resin filling, full crown and veneer are used in cosmetic restoration of anterior teeth. Doctors prefer resin because of its perfect clinical effect and small hurt. Some patients complain of sensitivity after rsein filling and we are interest in choosing a better resin with less sensitivity.OBJECTIVE: To compare the incidence of postoperative sensitivity and the difference of the patients' self sensitivity alter cosmetic restoration of anterior teeth with vital pulp by 3M-Z350 Nano-resin and 3M-Z100 composite resin. METHODS: A total of 112 teeth with vital pulp because of caries or traumatic tooth fractures of 68 patients demanding cosmetic restoration were selected from the Department of Endodontics, Stomatological Hospital of Luzhou Medical College from August to December 2006. The teeth were random divided into 3M-Z350 group and 3M-Z100 group, with 56 teeth in each group. 3M-Z350 nano-rasin and 3M-Z100 composite resin were used for cosmetic restoration of anterior teeth with vital pulp. Ryge criteria were used to evaluate the sensitivity to the cold stimulation following cosmetic restoration, and the level of pain was detected by the pain ruler.RESULTS AND CONCLUSION: Compared with 3M-Z100 group, the incidence of sensitivity and self-pain level were decreased in the 3M-Z350 group at 1 day, 1 week, and 1 month after restoration (P < 0.01 or P < 0.05). There was no significant difference in the sensitivity and self-pain level between the two groups (P > 0.05). Anterior teeth with vital pulp were well restored by 3M-Z350 nano-resin which might reduce the incidence of postoperative sensitivity.
7.Identification of telomerase activity of human adipose-derived mesenchymal stem cells from various donors and at varied passages

BACKGROUND:It is not certain that whether human adipose-derived mesanchymal stem cells express telomerase reverse transcriptase (TERT).OBJECTIVE:To investigate whether human adipose-derived mesenchymal stem cells express telomerase reverse transcriptase or not.DESIGN,TIME AND SETTING:The cytology in vitro experiments were performed at TEDA Life and Technology Research Center,Institute of Hematology and Blood Diseases Hospital,Chinese Academy of Medical Sciences and Peking Union Medical College in August 2009.MATERIALS:Adult adipose tissue was obtained from 5 healthy donors undergoing liposuction at the Tianjin Yili Medical Cosmetology Plastics Outpatient.Hela cells were supplied by the Cell Center of Basic Medical Sciences,Institute of Basic Medical Sciences,Chinese Academy of Medical Sciences.METHODS:Human adipose-derived mesenchymal stem cells were isolated from 20 mL human edipose tissue of each healthy donor by collagenase digestion.Cells were digested by trypsin when 80% confluency.Hela cells served as controls,and treated with 1640 medium containing 10% fetal bovine serum,and then digested using trypsin when 80% confluency.MAIN OUTCOME MEASURES:Morphology,phenotype and differentiation of human adipose-derived mesenchymal stem cells.TERT of human adipose-derived mesenchymal stem calls were detected by reverse transcription-polymerase chain reaction (RT-PCR).Telomerase activity was measured by telomeric repeat amplification protocol assay ELISA (TRAP-ELISA).RESULTS:Human adipose-derived mesenchymal stem cells adhered to the flask,presented spindle shape,and whirlpool-shape when cell density was large.Human adipose-derived mesenchymal stem cells were labeled positively for CD73,CD90 and CD105,but negatively for CD19,CD34,CD11b,CD45,and HLA-DR.Following adipogenic induction,oil red O staining showed significant red oil drop.Following ostesgenic induction,Von Kossa staining showed black particles,which indicated calcium deposition.Human adipose-derived mesenchymal stem cells at P1,P4 and P7 did not express telomerase reverse transcriptase gane.Human adipose-derived mesenchymal stem cells from 5 different donors did not expression telomerase reverse transcriptase gane.Hela cells had telomerase activation,but human adipose-derived mesanchymal stem cells did not express telomerase activation.CONCLUSION:Human adipose-derived mesanchymal stem cells do not express mRNA of hTERT,nor the activity of telomerase reverse transcriptase.
8.Questionnaire investigation on cost-effectiveness analysis of acupuncture for migraine.

OBJECTIVEBy using questionnaire method to understand the evaluation regarding cost-effectiveness of acupuncture for migraine in both doctors and patients.
METHODSBy using questionnaire method, questionnaires for doctors and patients were made respectively; by using network platform, two sets of questionnaires were publicly released, which were filled in online. The results were real-time background collected and then analyzed.
RESULTSTotally 696 effective questionnaires for patients and 114 questionnaires for doctors were collected. The results indicated that (1)the direct cost (including treatment fee and material fee) for preventive treatment of migraine was 109 yuan per time in regular acupuncture, 152 yuan per time in regular acupuncture + electroacupuncture (EA) and 238 yuan per time in acupoint catgut embedding. The travelling expense was 42 yuan per time within the city and 193 yuan per time between cities. The indirect cost included cost for loss of working time (208 yuan per time), cost for treatment time (acupoint catgut embedding: 10 to 30 min per time, once one to three weeks; the remaining two treatments: 20 to 50 min per time, once to 5 times every week), time cost for arriving and departing hospital (1 to 3 hours per time within the city and 4 hours per time between cities). From the angle of treatment course, acupoint catgut embedding reduced the treatment frequency, leading to a lower total cost. (2)A mong the reasons to select different acupuncture methods to treat migraine, the results of patient questionnaire indicated that the focus in the patients who selected acupoint catgut embedding was different from that in the patients who selected two other treatments. The main advantage of acupoint catgut embedding was superior and lasting efficacy with low cost. The results of doctor questionnaire indicated the main reason to select acupoint catgut embedding was "lasting efficacy after single treatment" (87. 5%) and "lower frequency of treatment" (75. 0%). (3) The main reason to obstruct the popularization of acupoint catgut embedding was this method was not widely known.
CONCLUSIONThe acupoint catgut embedding is one ideal preventive treatment for migraine, which has higher cost-effectiveness, however, currently it is not widely applied. In future advertisement and training program should be strengthened to perform targeted popularization of acupoint catgut embedding for migraine.

9.Soluble expression, purification and bioactivity of hemangiopoietin protein

AIM: To prepare a soluble hemangiopoietin(HAPO) protein and to construct pET22b(+) expression vector, to obtain pure recombinant HAPO protein and to measure its bioactivity. METHODS: HAPO cDNA was amplified using RT-PCR method from a commercial human fetal liver cDNA library. The resulting product was cloned into pET22b(+) vector and transformed into E.coli BL21(DE3). The recombinant protein was isolated and purified by Ni~(2+)-NTA chelating resin and the chromatographies of SP Sepharose FF. The adhesion of human umbilical vein endothelial cells (HUVEC) were measured by adhesion assay. RESULTS: HAPO gene with a reading frame of 897 bp was successfully cloned from human fetal liver cDNA library, the expressed pET22b(+)-HAPO fused protein existed in a soluble form, with the yield above 10% total bacterial protein and its purity achieved above 80%. The activity assay showed that the treatment of HAPO enhanced total adherence of HUVEC in a concentration-dependent manner. CONCLUSION: HAPO protein can be expressed in a soluble form. HAPO may facilitate the homing of hematopoietic stem/progenitor cells in vitro.
10.Nano-cellulose proteinversusacellular matrixinoral mucosarepair

BACKGROUND:The sensitivity andmucus secretion of theoral mucosamake oral soft tissues difficult to repair, so patients cannot achieve satisfactory outcomes after treatment. Nano-celulose protein mainly composed of glycine, alanine and serine has good histocompati bility. However, there is a lack of comparative study about the effect of nano-celulose protein and acelular matrix in oral mucosa repair, and the clinical effects of the two materials are stil under discussion.
OBJECTIVE:To investigate the effects of nano-celulose proteinver susacelular matrix in oral mucosa repair.
METHODS:Oral mucosadefect models were preparedinrats, and these rat models were randomly divided into four groups:oral mucosa defectswere repaired by vaseline (control group), nano-celulose protein, bovine skin acelular matrix and human skin acelular matrix, respectively. Repair effects were compared among different materials within 2 months after surgery.
RESULTS AND CONCLUSION:The diameter of oral mucosa defect measured using a vernier caliper, had no significant differences among groups at 1 day after surgery (P> 0.05); the diameter of oral mucosa defect in the nano-celulose protein group was significantly lower than that in the other groups at 3, 5 and 7 days after surgery (P<0.05); the diameter of oral mucosa defect in the bovine and human skin acelular matrix groups was significantly lower than that in the control group at 5 and 7 days after surgery (P< 0.05). Morphological observationoftheoral mucosa under light microscope showed: the number of newborn capilary endothelialcels in the defect region had no significant differences among nano-celulose protein, bovine acelular matrix and human acelular matrixgroupsat 1, 3, 5 and 7 weeks after surgery (P> 0.05);but there were significant differences in the number of newborn capilary endothelialcels between the control group and the other three groups (P< 0.05). Furthermore, at 21 days after surgery, closely aligned and thicker new epithelialtissuecould be found in the nano-celulose protein group; in the bovine acelular matrix group,thedefect regionwasrepaired wel, new epithelialtissueappeared andthe number of inflammatory cels decreased; in the human acelular matrix group, inflammatory cels appearedobviously, and new epithelialtissueformed with the normal thickness. In contrast,abundant inflammatory cels and thinner epithelial tissues appeared in the control group. To conclude,bothnano-celulose protein and acelular matrix can accelerate wound healing by promoting oral mucosal epithelial hyperplasia.
