1.Construction and expression of Mtb8.4 gene vaccine and study on cellular immune response induced by the vaccine

Objective To prepare Mtb8.4 gene vaccine and to study the cellular immune response induced by the vaccine. Methods The gene encoding Mtb8.4 from Mycobacterium tuberculosis was amplified by PCR and cloned into the eukaryotic expression vector pcDNA3.1 (+). C57BL/6N mice were vaccinated three times with Mtb8.4 gene vaccine at 3 weeks interval. Four weeks after the final inoculation, mice were sacrificed to assess cytokine response and CTL induction. Results The IFN-? and IL-2 titers were 787.317?45.586pg/ml and 319.953?57.978pg/ml in Mtb8.4 gene vaccine group, 1 486.540?39.600pg/ml and 767.043?50.269pg/ml in BCG group, respectively. Objective: To investigate the effects of a Chinese herbal formula for calming liver and suppressing yang on the protein expressions of vascular tissues in spontaneously hypertensive rats (SHRs), and to explore the mechanism of efficacy. Methods: Twenty SHRs were randomly divided into model group and treatment group. Another 10 Wistar-Kyoto rats were selected as a normal control. SHRs in the treatment group were administered with the formula for calming liver and suppressing Yang for 4 weeks. During the course of treatment, blood pressure and heart rates were monitored every week and the ethology of rats, including irritability and rotation endurance was also evaluated. After treatment, thoracic aorta was obtained and its proteins were separated by 2-dimensional gel electrophoresis (2-DE). The differentially expressed proteins were identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) and database query. Results: The formula for calming liver and suppressing yang not only decreased the systolic blood pressure and heart rate, but also improved irritability degree and rotation endurance time of SHRs. This experiment had established the 2-DE graph of protein expressions of vascular tissues in SHRs. Compared with the normal group, the expressions of 15 proteins were down-regulated, and 12 proteins were up-regulated in vascular tissues of the model group. The formula for calming liver and suppressing yang treatment up-regulated expressions of 10 proteins in the 15 down-regulated proteins, and down-regulated 8 proteins in the 12 up-regulated proteins in vascular tissues of SHRs. After analysis, 16 obviously differentially expressed proteins were found, and 13 of them were identified. Conclusion: The formula for calming liver and suppressing yang can improve the ethology of SHRs. The mechanism is probably concerned with regulating the protein expressions of vascular tissues.

6.Correlation of serum uric acid level with carotid plaques and arterial stiffness in patients with essential hypertension

AIM:To study the correlation of serum uric acid ( UA) level with carotid plaques and arterial stiff-ness in the patients with essential hypertension ( EH) , and to explore the predictive value of serum UA for evaluating EH preclinically .METHODS:A total of 92 patients with EH and 30 healthy individuals were enrolled .The value of UA and other indicators were detected .B-mode ultrasound examination was performed to measure the common carotid artery intima -media thickness ( IMT) and the sites of plaque in the internal carotid-artery, external carotid artery and carotid bifurca-tions.Carotid-femoral arterial pulse wave velocity ( CFPWV) was assessed by Complior?atherosclerosis measurement in-strument.RESULTS:The serum level of UA in the patients with EH was higher than that in control group [(361.51 ± 83.81) μmol/L vs (317.03 ±62.22) μmol/L, P<0.05].The mean value and abnormal rate of IMT between hyperten-sion group and control group were significant difference [(0.69 ±0.14) mm vs (0.60 ±0.12) mm, 42.39%vs 10.00%, P<0.05].In 92 EH patients, 45 cases had carotid plaques .These 45 cases were divided into 3 groups according to the plaque severity, among which the serum UA level had statistically significant differences [(285.25 ±78.41) μmol/L, (341.19 ±63.99) μmol/L and (401.33 ±88.49) μmol/L, P<0.05].Compared with rigid plaque group ( n=34), the serum UA level in soft plaque group (n=11) was significantly higher [(389.00 ±69.45) μmol/L vs (323.03 ± 72.71) μmol/L, P<0.05].A stepwise multiple linear regression analysis demonstrated that age ( r=0.414), systolic blood pressure (r=0.224), pulse pressure (r=0.270) and uric acid (r=0.219) were predisposed factors for higher CFP-WV (P<0.05).CONCLUSION:UA is one of the risk factors causing hypertension .Serum UA level may reflect the sever-ity and stability of carotid plaques .The increased arterial stiffness is closely related to the increased serum UA level in EH .
8.Correlation between soluble receptor for advanced glycation endproducts and arterial stiffness in patients with metabolic syndrome

Objective investigate the correlation between plasma soluble receptor for advanced glycation endproducts (sRAGE) and arterial stiffness in patients with different types of metabolic syndrome (MS).Methods A total of 180 subjects were drawn from a epidemiologic follow-up study,including 60 cases non-metabolic syndrome (NMS),60 cases metabolic syndrome without diabetes mellitus (NDMMS),60 cases metabolic syndrome with diabetes mellitus (DMMS).Carotid femoral arterial pulse wave velocity (CFPWV) was assessed by the French KangPuLe atherosclerosis measurement instrument,and plasma sRAGE levels were measured by ELISA.Comparison of mean in multiple groups was conducted by analysis of variance.Multivariate analysis was done with multiple linear stepwise regression analysis.P < 0.05 was considered as statistically significant difference.Results Compared with NMS group,plasma sRAGE levels were significantly lower in DMMS and NDMMS groups [(635.07 ± 229.20) pg/mL vs.(671.17 ± 358.16) pg/mL vs.(992.99 ± 427.83) pg/mL,P =0.001].CFPWV of DMMS group was significantly higher than that of NMDMS and NMS groups (14.22 ±3.14) m/s vs.(12.15 ±2.79) m/s vs.(11.66 ± 2.52) m/s,P =0.002).Plasma sRAGE level was negatively correlated with CFPWV (r =-0.278,P =0.005).(3) Multiple linear regression analysis demonstrated that age (β =-0.091,95% CI-0.096 ~-0.095,P =0.031),HDL-C (β =1.295,95% CI 1.231 ~ 1.360,P =0.022) and sRAGE (β =0.119,95% CI 0.118 ~ 0.130,P =0.032) had a significant effect on CFPWV.Conclusions The increased arterial stiffness is closely related to the discreased plasma sRAGE levels in MS.Plasma sRAGE maybe a novel target for vascular disease prevention and treatment in patients with metabolic syndrome.
9.Mechanical and biodegradable properties of absorbable hydroxyapatite/poly-DL-lactide screws

BACKGROUND:Through compounding hydroxyapatite and poly-DL-lactide, the mechanical properties, physical and chemical properties of the implants can be enhanced.
OBJECTIVE:To detect the effect of hydroxyapatite/poly-DL-lactide composite internal fixation screws in canine femoral condyle cancel ous bone fracture repair.
METHODS:Forty-two beagle dogs were operated to bilateral femoral condyle fracture models. The left side was fixed using hydroxyapatite/poly-DL-lactide screws as experimental group;while the right side was fixed with pure poly-DL-lactide screws as control group. After 2, 4, 8, 12, 24, 36 and 48 weeks, conditions of fracture were observed using X-ray, femoral and screw specimens were observed histopathological y, and the bending strength and the average molecular weight were detected. The biological absorption rate, intensity decay rate and biodegradation rate were calculated.
RESULTS AND CONCLUSION:After 2-48 weeks, bilateral fractures were fixed wel and new bone grew wel , but the biological absorption rate of the screws in the experimental group was lower than that in the control group (P ≤0.01). In the first 2, 4 weeks, the bending strength of the experimental screws was higher than that of the control screw (P ≤0.05), but the biodegradable speed was slower in the former one (P ≤ 0.05 or P ≤ 0.01). The pathological changes were similar in the two groups. After 48 weeks, the fractures were healed and bone tissue reconstruction was completed. Compared with the pure poly-DL-lactide screws, the hydroxyapatite/poly-DL-lactide composite internal fixation screws have better fixation effects, biocompatibility, mechanical properties and biodegradability.
10.Air Circumstance in Laminar Flow Operating Room in Beijing:A Crosssection Investigation of 18 Hospitals

OBJECTIVE To investigate the management and sanitation status of the air circumstance in laminar flow operating rooms of grade two and above hospitals in Beijing.METHODS Air surveillance methods,cleaning methods of intakes,cleaning status of pipelines and frequency of renewing filter systems were investigated through interviewing charge nurse of 18 hospitals by questionnaires.RESULTS 50.0% of pipelines had never been cleaned,14.3% of intakes had never been cleaned.Air surveillance method of all hospitals was using sedimentation,with a low sensitivity.The monitoring systems were poor in 50.0% of hospitals.CONCLUSIONS Now there is only requirements of construction standard of clean surgery region,lacking routine regulations and technologies of contamination controlling standard,nor available dynamic contamination controlling system.Potential hazard exists in routine regulations of laminar flow operating room and it demands for making contamination controlling standard urgently in hospitals.
