1.The Expression of TNF-? mRNA、IL-6mRNA in Liver of Rats with Alcohol Fatty Liver

Objective To measure the the expression of TNF-?、IL-6mRNA in liver of rats with alcohol fatty liver, and the amounts of TNF-?and IL-6, to observe the lesion of lipid superoxidation, to explore the pathogenesis of alcohol fatty liver. Methods 23 of SD rats were divided into the normal group(10) and the model group(13),all the rats in the model were given 50% alcohol by daily gavage, and drank 10% alcohol, after 14 weeks, all the rats were killed and the pathological change of liver were observed, the amounts of serum TG,TC,ALT,AST,FFA,MDA,SOD,GSH-PX,TNF-? and IL-6 were examined, the expression of TNF-?、IL-6mRNA was measured. Results In the model ,to different degree, there were some fatty steatosis , the amounts of TG、FFA、MDA、TNF-?and IL-6 were higher than that in the normal group, but the amounts of SOD and GSH-PX lower, in comparison with the normal, the expression of TNF-?mRNAand IL-6mRNA in liver of model were significantly increased. Conclusions long-term of ingestion of potation maybe cause the disorder of lipid metabolic system and alcohol fatty liver came into being. Simultaneously with the enhancement of oxidation stress/lipid superoxidation and abnormity of secretion of some cytokine such as TNF-?and IL-6.
2.Clinical study of the gestational diabetes mellitus in pregnant Women with HBsAg-positive

Objective To study the relationship between the pregnant women with HBsAg and the gestational diabetes mellkus.Methods Clinical data of 94 cases with pregnant women with HBsAg-positive in our hospital were analyzed retrospectively.119 cases with pregnant women with HbsAg-negative were selected at random according to their personal preference and were compared.The prevalence rate of the gestational diabetes mellitus and itS related factors were compared between the 2 groups.Results 32 cases were diagnosed the gestational diabetes mellitus in pregnant women with HBsAg-positive,according to 34.0%,13 cases were diagnosed the gestational diabetes mellitus in control group,according to 10.9%.It was significant difference in 2 groups(P<0.05).Compared wlth the control group,there were significant difference in the occurrence of thrombocytopenia,post partum hemorrhage(P<0.05).Compared with the control group,there were no significant changes in the ratio of cesarean section,BMI,occurrence of fetal macrosomia.Conclusion The prevalence rate of the gestational diabetes mellitus and post partum hemorrhage and thrombocytopenia in pregnant women with HBsAg-positive group was significantly higher than in the HBsAg-positive group.
3.The Changes of Serum Tumor Necrosis Factor and Epidermal Growth Factor in Rats with Experimental Liver Fibrosis.

Objective To study the the changes of serum tumor necrosis factor-?(TNF-?)and epidermal growth factor (EGF) in rats with experimental liver fibrosis and its significance. Methods All the SD rats were randomly divided into the normal group and the model group. The rats in the model group were subcutaneously injected with 40%CCl4 according to 0.3ml/100g body weight twice one week for the first four weeks, the initial dose was double. In the following five weeks, the rats were injected 40% CCl4 of 0.2ml/100g body weight twice one week.The rats in the normal group were subcutaneously injected with equal dosage of saline,mentioned aboveo.At the end of the ninth week,the serum was obtained,the levels of serum TNF-?, EGF were detected with radioimmunity, with HE stains, the severity of inflammation and fibrosis in all liver tissuses were measured by semiquantitative scoring. 5.Clinical Significance of Thrombo Molecular Markers in Blood-stasis Syndrome in Diabetes Mellitus

By applying the thrombo pre molecular markers, including plasmal GMP - 140, tPA, PAI - I TAT, PAP, ET, their correlation with blood stasis type diabetes were explored. Results showed that, the diabetic with blood -stasis type revealed the activation of platelets, injury of endothelial cells, changes of fibrinolysin. Their plasmal GMP-140, TAT, PAP, ET were all higher than that of the control group, indicating that the activation of platelets, injury of endothelial cells, and changes in the balance of blood coagulation/fibrinolysis are likely to benefit the incidence of blood stasis. The pathophysiological mechanism in blood stasis type diabetes is related to the changes in platelet, endothelial cells, blood coagulation and fibrinolysis.
6.Effect of misoprostol on gastrointestinal peristalsis after cesarean section

Objective To explore the effect of misoprustol on restoration of gastroinete stinal function after cesarean section.Methods 100 patients after cesarean section were randomly divided into two groups:50 patients were treated by dropping rectum with misoprostol 200μg.The other 50 patients were the control group.To compare the two groups of the time of wind breaking,the time of lactation and postpartum hemorrhage.Results The duration of there recovery time of the gastrointestinal functionin and lactation of misoprostol group was shorrer than that in the control group(P<0.05).Quicker involution of uterus and less quantity of red lochia were compared with the control group(P<0.05).Conclusion Misoprostol used after cesarean section can accelerate lactation and wind breaking and the involution of uterus.It would he used clinically for its safety,eonvenieney and without evident side effects.
7.Current status of standards for laboratory diagnosis of pertussis and clinical relevance of serological diagnosis

Despite high coverage of vaccinations, incidence of pertussis (whooping cough) has been increasing throughout the world and large outbreaks were reported in several countries.Adolescents and adults with atypical pertussis symptoms have become the main sources of infants′ pertussis.Since clinicians are lack of experience in making diagnosis based on atypical symptoms, laboratory methods are needed.The currently recommended laboratory methods include bacterial culture, PCR and serology ELISA.It is well known that sensitivity and specificity of the above-mentioned methods may vary depending on many factors such as status of vaccination, timing of specimen collection and onset of symptoms.Serology ELISA to measure specific anti-pertussis toxin IgG antibodies in serum has been proven to be one suitable method for the diagnosis of pertussis infection in adolescents and adults.In this review, we summarize the current status of methods used in China and other countries for the laboratory diagnosis of pertussis and discuss the clinical relevance of serology ELISA for pertussis diagnosis.
8.The Experimental Study on Liqihuatanquyu Formula(LF)Against NASH in Rats

Objective To investigate the effect of LF on blood biochemistry,cytokines in rats with NASH in order to approach possible mechanism of LF against NASH.Methods 50 male SD rats were randomly divided into the normal control group(10 rats),the model group(10 rats),the Essentinale group(10 rats)、the LF high dose group(10 rats)and the LF low dose group(10 rats).Except for the normal control group which was raised by standard animal feeds,others were all raised by high fat feeds to make NASH model and administered by Essentinale,high dose LF and low dose LF for 12 weeks.Hepatic lipid contents,plasma TG,CHO,HDL-C,LDL-C,ALT,AST and TNF-?,FFA,Leptin concentrations were evaluated,as were histopathological changes in the liver.Results In this NASH model,the liver FZ(liver weight /body weight),plasma CHO,LDL-C,ALT,AST and TNF-?,FFA,Leptin concentrations were higher than that in normal control group,but the levels of serum HDL-C and TG were reduced.In addition,the histopathological showed increased fatty droplets within hepatocytes and liver inflammation.The levels of serum ALT,AST,HDL-C,TNF-?,Leptin,FFA and hepatic CHO,TG contents in the LF high dose group and LF low dose group were reduced compared with that in the model group,while the levels of serum TG,HDL-C were enhanced remarkably.In addition,liver inflammation was improved evidently.Conclusions Treatment with LF might improve non-alcholic steatohepatitis,because LF inhibits the elevation in the level of plasma cytokine.
9.The Change and the Immpact of IFHL on Oxidative Stress in the Formation of NASH in Rats

Objective To study the change of oxidative stress in the formation of NASH in rats induced by high-fat diet,and observe the preventive and therapeutic effect of TFHL on NASH and explore its mechnism.Methods 110 rats were randomly divided into the normal group(40 rats),the model group(40 rats),the A.Nattermam group(10 rats) and high dosage and low dosage TFHL(10rats each).The normal rats were fed with the standard feedstuff the other rats fed with the high-fat diet.At the time of model-making,the rats in the A.Nattermam were fed the A.Nattermam suspension by daily gavage by 195.4mg/(kg?d),the rats in the high dosage and low dosage TFHL group were fed TFHL by daily gavage in 250、125mg/(kg?d),the rats in the normal and model groups were fed isometrical distilled water by daily gavage.At the ends of 2nd,4th,8th weeks of the experiment,10 rats in the nomal and model were killed respectively,at the end of experiment,all the rest were killed.pathological changes of liver tissues were observed,the levels of serum ALT,AST,the amount of TG,CHOL of livers tissue,the amount of MDA of serum and liver,the activity of OD、GSH-PX、T-AOC were measured.Results 1.the rats were fed with high-fat diet from 2 to 12w,the fatty degeneration of liver tissues were gradually serious,the amounts of TG,CHOL of livers were gradually increased.at 12th w the inflammatory cells in livers infiltrated obviously,the serum ALT,AST were obviously increased,it apeared a progressive process of NASH.2.from 4th w,after feeding with high-fat diet,the amounts of MDA of serum and liver were gradually decreased,the activity of SOD were gradually increased,the amount of T-AOC were gradually decreased as well and,increase or decrease with the development of inflammation.3.the inflammation degree of liver,the amount of liver CHOL and the serum ALT,AST in FMCL groups were lower than those in the corresponding model group;the amounts of MDA of serum and liver were decreased,the activity of SOD,GSH-PX and T-AOC were inceased than those in the corresponding model group,their oxidationresisting activity was better than those in the A.Nattermam group.Conclusions oxidation stress/Lipid Peroxidation plays an important role in the occurrence and development of NASH,TFHL can effectively treat and prevent the development of NASH,obviously ease oxidation stress,decrease Lipid Peroxidation of NASH,markedly enhance oxidationresisting abilily of body,which is the important mechanism of prevention and treatment for NASH.
10.Effects of the Therapy of Invigorating the Kidney and Qi and Dissipating Blood Stasis to Removing the Toxin on the Hepatocyte Apoptosis and Associated Genes in the Liver Fibrosis Rats

Objective To investigate the effects of the therapy of invigorating the kidney and qi and dissipating blood stasis to removing the toxin on the hepatocyte apoptosis and associated genes in the liver fibrosis rats,and to explore the treatment mechanism of liver fibrosis with TCM.Methods liver fibrosis model rats were induced by CCl4,and the Chinese herbs(Ganlike)with invigorating the kidney and qi and dissipating blood stasis to removing the toxin was given by daily gavage,after the experiment,the pathological changes of liver were observed with optical microscope.Hepatocyte apoptosis was detected with TUNEL assay,apoptosis-associated genes were detected with immunohistochemistry assay.Results The Chinese herbs Ganlike could obvously improve the pathological changes,alleviate the hepatocyte apoptosis and modulate the expression of apoptosis-associated genes.Conclusions The anti-fibrosis mechanism of Ganlike may be to regulate the hepatocyte apoptosis of liver fibrosis rats and to restrain the expressions of hepatocyte Fas/FasL,Bcl-2/Bax.
