1.Expression of Schwann cell derived neurotrophic factor in developing lumbar spinal cord of rats

BACKGROUND: Schwann cell can excrete many neurotrophic factors,protect neuron, and be nutritious to neuron. It also can promote the recover of injury of nerve.OBJECTIVE: To observe the expression of Schwann cell derived neurotrophic factor(SDNF) in developing lumber spinal cord of normal adult and different embryo rats as well as immature rats, and the effect on spinal cord in various developing period.DESIGN: Randomized and Controlled animal study.MATERIALS: The experiment was completed in the Animal Laboratory of Nanhua University in September 2003. Totally 30 Wister rats, in each gender,weighing 150-300 g, were selected from Animal Laboratory of Nanhua University.INTERVENTIONS: ① Selection and section: The normal lumbar spinal cord and dorsal root ganglia of normal adults rats with body mass in 180-220 g and 4-6 mature stages were fixed. Frozen sections were selected from 8 pregnant female rats including various embryol rats and immature rats. ② Immunohistochemical staining: Regarding protein antibody of SDNF as the first antibody,avidin-biotin peroxidase method was used to stain, the ash value was observed and the demi-quantitative value was worked out by scanning staining depth in order to analyze the expression of SDNF in lumbar spinal cord.MAIN OUTCOME MEASURES: ① Immunohistochemical staining of normal lumbar spinal cord; ② Immunohistochemical staining of developing lumbar spinal cord.RESULTS: Eight female rats were pregnant, and all samples were selected from the fetal rats and the immature rats. ① There appeared well-distributed SDNF, staining the whole white matter and Ⅲ, Ⅳ, Ⅴ stratum of the poster horn of gray matter of adult rats, and the dorsal root ganglia is much more intensively stained than lumber spinal cord. ② There was a inclination of early increase and late declination of SDNF immunoreactivity and the staining reaches a maximum at 21-day embryo, and then decreased at 7-day embryo and 12-day embryo after birth.CONCLUSION: There is lower expression of SDNF in lumber spinal cord of adult rats. But the expression of SDNF decreases gradually in the developing lumber spinal cord of rats. This means that SDNF may promote the development of spinal cord of embryo rats.
2.Fracture strength of custom-fabricated celay all-ceramic post and core.

OBJECTIVEThe purpose of this study is to compare the fracture strengths of custom-fabricated Celay all-ceramic post-core, custom cast metal post-core, and prefabricated stainless steel post (Parapost) plus composite resin core with or without a 2.0 mm dentine ferrule.
METHODSA total of 60 recently extracted human maxillary central incisors were endodontically treated and divided into five groups of 12. They were given the following treatments: Group A: Celay ceramic post-core with 2.0 mm dentine ferrule, Group B: Celay ceramic post-core with no dentine ferrule, Group C: cast metal post-core with 2.0 mm dentine ferrule, Group D: cast metal post-core with no dentine ferrule, and Group E: prefabricated post and composite core with 2.0 mm dentine ferrule. All specimens were stored at 100% humidity at room temperature for 30 days before testing. Each specimen was in a special jig at a 45 degrees angle to the long axis and subjected to a load on MTS 810 universal material testing machine until failure, with crosshead speed of 0.02 cm/min. Analysis of variance followed by the Newman-Keuls pairwise multiple comparison test was used to compare the results of the groups tested.
RESULTSThere was a statistically significant difference between five groups (P < 0.01). Celay ceramic post-core with 2.0 mm dentine ferrule (758.35 N +/- 119.26 N) and cast metal post-core with 2.0 mm dentine ferrule (756.63 N +/- 166.22 N) had a significantly larger mean failure threshold for fracture than the other three groups which had no significant difference between each other. There was a statistically significant difference between the fracture resistance of Celay post-core restored teeth with and without 2.0 mm dentine ferrule.
CONCLUSIONThe custom-fabricated Celay post-core could be a choice for clinical use in endodontically treated tooth when the final restoration is an all-ceramic crown and the preparation has a 2.0 mm dentine ferrule.

3.Clinical performance of fiber post restorations in elderly patients with residual root and crown

Objective To investigate the clinical performance of fiber post restorations in elderly patients with residual root and crown and to analyze the factors affecting the clinical success rate.Methods A total of 66 elderly patients requiring post restoration were selected.They were randomly divided into two groups:the observation group (n=33,restored with fiber post-resin cores) and the control group (n=33,restored with cast metal post-cores).All patients were rechecked 1 year and 2years after the restoration.The clinical efficacy and the failure types were recorded.The differences in clinical performance between the 2 groups were compared by using Fisher's exact test.Results The differences in the gingival index and periodontal index in the 2 groups between the experimental teeth and the control teeth were zero.There were no significant differences in dental morphology,root canal treatment failure,tooth color matching and marginal coloration between the 2 groups (all P>0.05).1 case with post debonding and 1 case with fiber post fracture were found in the observation group and 1 case with marginal coloration was found in control group.There was no significant difference in clinical success rate between the 2 groups (93.94% vs.96.97%,P>0.05).Conclusions The clinical success rate in fiber post-resin core restoration is the same as in cast metal post-cores restoration in elderly patients.
4.THE PREVENTIVE AND THERAPEUTIC EFFECT OF SHEEP BONE COLLAGEN PEPTIDE ON OSTEOPOROSIS OF OVARIECTOMIZED RATS

Objective To study the preventive and therapeufic effect of sheep bone collagen peptide (SBCP) on osteoporosis of ovariectomized rats. Method Thirty-two 3.5 mon unmated SD female rats were randomly divided into 4 groups by weight:blank group (sham operation), control group (ovariectomized), sample 1 group (ovariectomized and fed freeze drying SBCP) and sample 2 group (ovariectomized and fed spray drying SBCP). Seven days after ovariectomy,the blank group and ovariectomized group were given distilled water (1 ml/100 g?d), the sample 1 and sample 2 group were given sample 1 and sample 2 (1000mg/kg.d). Body weight, feed efficiency, serum bone metabolizing index and bone density (BD), length and diameter of femur were measured 10 w later. Results The alkaline phosphatase (ALP) of ovariectomized or control group was significantly increased, but the increase was restrained in sample 1 and sample 2 group. The BGP of ovariectomized group was significantly higher than that of the blank group, and the BGP of sample 1 and sample 2 group was lower than that of the ovariectomized group, nearly equal to the blank group. The BD of ovariectomized group was declined significantly, but that of sample 1 and sample 2 group was declined slowly. The BD of sample 2 group washigher than that of ovariedomized group, even similar to the blank group. The length and diameter of femur in the sample groups were larger than those of ovariectomized group. The effect of sample 1 and sample 2 on serum bone metabolism indices and femur size was not significantly different. Conclusion Sheep bone collagen polypeptide can reduce the resorption of bone, promote bone growth, and also reduce postmenopausal osteoporosis effectively. The different drying methods have no difference in the anti-osteoporosis effect.
5.Research progress of traditional Chinese medicine intervention in pre-diabetes

With the continuous improvement of people's living standard, more and more people are in pre-diabetes state. Pre-diabetes is the key to the development of diabetes, and early intervention can reduce the incidence of diabetes, and prevent transforming pre-diabetes to diabetes in order to maintain the health status of the patient. By retrieving the Chinese Biomedical Literature Database of nearly five years on pre-diabetes intervention literature, it was found that traditional Chinese medicine interventions in pre-diabetes have a relatively new understanding. Through the traditional Chinese medicine, acupuncture, acupoint massage, and combine traditional Chinese and western medicine, medicinal food, eight brocade etc intervention therapy in patients with pre-diabetes, the occurrence and development of diabetes and its complications can be effectively prevented.
6.Influence of post-core on the strength of endodontically treated and crowned teeth.

OBJECTIVETo investigate the influence of post-core structure on the strength of endodontically treated and crowned teeth with or without a 2.0 mm dentine ferrule.
METHODSA total of 60 recently extracted human maxillary central incisors were endodontically treated and randomly divided into five groups of 12. They were given following treatments: Group A, endodontically treated; Group B, endodontically treated and crowned (PFM); Group C, cast metal post-core with 2.0 mm dentine ferrule and crowned (PFM); Group D, cast metal post-core with no dentine ferrule and crowned (PFM); Group E, prefabricated post and composite core with 2.0 mm dentine ferrule and crowned (PFM). All specimens were stored at 100% humidity at room temperature for 30 days before testing. Each specimen was in a special jig on the MTS 810 universal material testing machine and subjected to a load at a 135-degree angle to the long axis until failure, with crosshead speed of 0.02 cm/minute. Analysis of variance followed by the Newman-Keuls pairwise multiple comparison tests was used to compare the results.
RESULTSThere was a statistically significant difference between different restorative methods. The cast metal post-core with 2.0 mm dentine ferrule and crowned teeth had the highest fracture strength (1793.59 +/- 387.93N), followed by endodontically treated intact teeth (1466.68 +/- 240.11N). No significant difference in the fracture strength was found among the other three groups (958.49 +/- 286.02N; 992.98 +/- 291.00N; 994.94 +/- 285.04 N). There was a statistically significant difference in the fracture resistance between crowned teeth with and without 2.0 mm dentine ferrule (P < 0.01).
CONCLUSIONSNot all post-core structure could improve the strength of endodontically treated teeth. The dentine ferrule can effectively improve the fracture resistance of endodontically treated and crowned teeth.

7.A comparison of efficacy of sedation with dexmedetomidine versus midazolam in mechanically ventilated patients

Objective To compare the efficacy of sedation with dexmedetomidine versus midazolam in mechanically ventilated patients.Methods Sixty patients aged 20-64 yr,with body mass index 21-25 kg/m2,APECHE Ⅱ score 10-25,requiring 24 h of nechanical ventilation in intensive care unit (ICU),were randomly divided into 2 groups (n=30 each): midazolam group (group M) and dexmedetomidine group (group D).A loading dose of midazolam 0.05 mg/kg was injected intravenously,followed by infusion at 0.03-0.20 mg· kg-1 · h-1 in group M.A loading dose of dexmedetomidine 1 μg/kg was injected intravenously,followed by infusion at 0.2-0.7 μg· kg-1 · h-1 in group D.Ramsay sedation score was maintained at 2-4.The satisfaction of the doctors in ICU with the sedative efficacy,hypotension and bradycardia were recorded.Delirium was recorded starting from the begging of sedation to 2 h after the end of sedation.The emergence time and occurrence of falling asleep again within 2 h after waking were also recorded.Results Compared with M group,the satisfactory level of the doctors in ICU with the sedative efficacy was significantly increased,the emergence time was significantly shortened and the incidences of falling asleep again within 2 h after waking and delirium were significantly decreased (P < 0.05 or 0.01),and no significant change was found in the incidence of hypotension and bradycardia in group D (P > 0.05).Conclusion The efficacy of sedation with dexmedetomidine is better than that of midazolam in mechanically ventilated patients.
8.The role of Wnt/β-Catenin signaling pathway in the endotoxin-induced acute lung injury during the treatment with mesenchymal stem cells

Objective To investigate the role of Wnt/β-Catenin signaling pathway in the endotoxin induced acute lung injury (ALI) during the treatment by mesenchymal stem cells (MSCs).Methods Six SPF male SD rats were isolated and bone marrow mesenchymal stem cells were cultured.A total of 72 SPF male SD rats with 6-week-old were randomly (random number) divided into 4 groups:control group (n =18) in which phosphate buffered solution (PBS) used instead of lipopolysaccharide (LPS);LPS group (n =18) in which LPS used to induce acute lung injury;LPS + MSCs group (n =18) in which MSCs directly transplanted after injection of LPS;Control + MSCs group (n =18) in which MSCs transplanted after injection of PBS.And then 6 rats of each group were sacrificed at 6 h,24 h,and 48 h separately after injection of LPS.At 24 h after the modeling,lung tissue was taken and the levels of Wrnt signaling pathway components were detected by using immunohistochemistry and Western blot.In addition,quantitative realtime PCR was used to detect the expression of Wnt signaling pathway target genes.Results Compare with the PBS control group,significant decrease in lung dry-to-wet ratio and increase in arterial oxygen partial pressure (PaO2) were found in MSCS transplantation groups.According the immunohistological results,Wnt 5a was significantly increased in the LPS-induced ALI rats and decreased after MSCs transplantation.Moreover,decrease in levels of GSK-3β phosphorylation and β-catenin was found in the lung tissue after MSCs transplantation.In addition,the expressions of Wnt signaling target genes Vegf,Axin2 and Klf4 were decreased significantly after MSCs transplantation.Conclusions In the setting of ALI,the therapeutic effect of MSCs was exerted by decreasing the expressions of Wnt 5a,GSK-3β phosphorylation,β-catenin,and Wnt signaling target genes Vegf,Axin2 and Klf4.Wnt signaling implicated in the therapeutic effect of MSC in the setting of ALI.
9.The clinical value of amino-terminal pro-brain natriuretic peptide in septic children

Objective To investigate the prognostic value of amino-terminal pro-brain natriuretic peptide (NT-proBNP) in patients with sepsis.Methods A total of 162 patients was enrolled with consecutive pediatric intensive care unit (PICU) admissions during the study period of Jan 1st,2013 to June 30th,2013 at Hunan Children's Hospital.The 162 septic patients were divided into sepsis group and severe sepsis group,sepsis group and septic shock group,and survival group and death group.NT-proBNP was tested in the first and third day after 162 septic patients in hospital.Pediatric critical illness score (PCIS) score was assessed in all patients.NT-proBNP was compared between groups.The change of NT-proBNP was summarized between groups.The relationship between NT-proBNP and PCIS was analyzed.Results (1) The plasma NT-proBNP level of the first day after admission in the severe sepsis group and septic shock group was higher than sepsis group,the PCIS in the sepsis group was higher than severe sepsis group and septic shock group,with significant difference (P < 0.05).(2) The plasma NT-proBNP level of the first day after admission in the death group was higher than the survival group,the PCIS in the death was lower than the survivor group,with significant difference (P < 0.05).In the death group,the plasma NT-proBNP level of the third day after admission was higher than the NT-proBNP of the first day after admission (P =0.037) ; contrarily,the plasma NT-proBNP level of the third day after admission was lower than the NT-proBNP of the first day after admission in the survival group (P =0.023).Conclusions NT-proBNP could be used to assess the condition of septic patients,and dynamic test NT-proBNP can help to predict septic patient’s prognosis.
10.Association of serum chemokine levels with cognitive dysfunction in newly diagnosed T2DM patients

Objective Decline in cognitive function caused by diabetes has become a research hotpots.The article aims to explore the relationship between serums regulated upon activation normal T cell expressed and secreted(RANTES) and cognitive dysfunction of newly diognosed T2DM patients, and provide a new way of prevention and treatment for newly diagnosed T2DM patients with cognitive dysfunction.Methods We retrospectively analyzed the general information and clinical biochemical indexes of the 123 patients who were first diagnosed of T2DM from March 2015 to September 2016 in Tangshan Worker''s Hospital.The levels of serum RANTES were measured by enzyme-linked immunosorbent assay (ELISA), and the cognitive function of all patients was assessed by Mini-mental State Examinatlon(MMSE)and Repeatable Battery for the Assessment of Neuropsyehologic Status(RBANS).Finally, newly diognosed T2DM patients were divided into T2DM non-cognitive disorder group and T2DM cognitive disorder group according to the MMSE score.We analyzed whether there are differences among general information,serum RANTES level and RBANS cognitive function score of two group patients.The correlations of RANTES with general information and cognitive scores were analyzed by single factor correlation and multiple stepwise regression analysis.Results ①The level of serum RANTESin T2DM cognitive disorder group[(2.62±0.37)mmol/L] was significantly higher compared to that in T2DM noncognitive disorder group[(2.29±0.36)mmol/L], and there was significant difference(P<0.001).②The instant memory,visual span,attention,delayed memory score and RBANS score of T2DM cognitive disorder group were(70.90±14.71)、(92.90±15.50)、(87.80±16.45)、(88.02±14.28)、(82.92±11.07), which were significant declined compared to those of T2DM non-cognitive disorder group [(85.28±13.97),(104.18±12.69),(101.51±12.94),(96.42±10.30),(95.84±9.94)], and there was significant difference (P≤0.05).There was no statistically significant difference between the two groups′ verbal function score [(96.08±7.87),(99.31±9.83)] (P=0.056).③The RANTES was negatively correlated with the total score of instant memory, visual span, verbal function, delayed memory score and RBANS score in T2DM patients(the valuue of r were-3.48、-2.35、-2.01、-3.02、-4.17).Conclusion There was a significant correlation between serum RANTES level and cognitive dysfunction, and elevated serum RANTES level could be used as an important indicator for monitoring newly diognosed T2DM patients with cognitive dysfunction.
