1.Mechanical and biodegradable properties of regenerated fibroin fibers

BACKGROUND: The molecular weight of fibroin is inevitably reduced in the course of the dissolution of regenerated fibroin fiber, and this is why we have not entered the stage of the practical application of the regenerated fibroin fiber till now.OBJECTIVE: To obtain biodegradable regenerated fibroin fiber.DESIGN: A single sample experiment.SETTING: Jiangsu Provincial Key Laboratory of Silk Project of Material Engineering College of Soochow University.MATERIALS: Leftover silk pieces were provided by Suzhou Silk Import & Export Corp.METHODS: This study was performed at Jiangsu Provincial Key Laboratory of Silk Project of Material Engineering College of Suzhou University between January 2003 and December 2004. By controlling the molecular of abandoned natural fibroin fiber during neutral salt dissolution process, wet spinning technique and draft method were applied to spin regenerated fibroin fibers that have certain mechanical and biodegradable properties.The specific procedure is as follows: ①fibroin preparations. ② Spinning solution preparations. ③ Regenerated fibroin fibers were spun with wet spinning method. ④Sodium lauryl sulphate-polyacrylamide gel electrophoresis method was used to detect the molecular weight of LiBr dissolved fibroin solution. ⑤To detect the crystallinity and orientation of fibroin fibers. ⑥ To detect the mechanic properties of regenerated fibroin fibers. ⑦ To detect the in vitro enzyme degradation rate of fibroin fibers.MAIN OUTCOME MEASURES: ①The relative molecular weight of LiBr dissolved silk fibroin. ② Result of X-Ray diffraction. ③The in vitro enzyme degradation rate of regenerated fibroin fibers.RESULTS: ①After LiBr dissolution, the molecular weight of fibroin spinning solution was mainly below 100 000. ②During the coagulation and pulling process of regenerated fibroin fiber wet spinning, fibroin fiber conformation changed from random convolution to the co-existence of β-folding and random coi1/α-spiral conformation in molecular chain. ③ Actinozyme was used to assess the in vitro enzyme degradation rate of hexafluoroisopropanol spun regenerated silk fibroin, and the 30 day-degradation rate of regenerated fibroin fibers was 37.16%, compared with 10.70% of natural fibroin fiber.CONCLUSION: Wet spinning technique and draft method can be used to spin regenerated fibroin fibers possessing certain mechanical and biodegradable properties by controlling the molecular weight of abandoned natural fibroin fibers during neutral salt dissolution process.
2.Effects of pegylated interferon combined thymopeptideenteric -coated tablets on CXCR3 , CCR6 and serum IL-2 and soluble ligand in chronic hepatitis

Objective To investigate the effect of pegylated interferon combined thymopeptideenteric-coated tablets on CXCR3, CCR6 and serum IL-2 and soluble ligand in patients with chronic hepatitis.Methods 125 patients with chronic hepatitis B patients from June 2015 to June 2016 in our hospital were selected and divided into observation group 62 cases and control group 63 cases.The observation group was treated with pegylated interferon combined thymopeptideenteric-coated tablets, and the control group was treated with thymopeptideenteric-coated tablets, the expression of T cells in patients with CXCR3 and CCR6 was detected by flow cytometry, the serum levels of interleukin-2 and soluble interleukin-2 receptorwere measured by enzyme-linked immunosorbent assay, and compared the treatment of two groups of patients.Results After treatment, the levels of IL-2 and sIL-2R in the observation groupwere (16.4 ±5.6), (9.4 ±2.1), significantly lower than that in the control group, while CXCR3 and CCR6 were (5.8 ±0.9), (5.54 ±1.2), significantly higher than the control group, the difference was statistically significant (P<0.05), the levels of HBsAg, HBeAg and HBV-DNA in the observation group were (1.5 ±0.4), (1.6 ±0.9), (1.6 ±0.3), significantly lower than those in the control group, the difference was statistically significant (P<0.05), the levels of ALT, AST, TBIL, IBIL in the observation groupwere (34.5 ±8.7), (31.4 ±6.9), (14.6 ± 6.9), (9.4 ±2.9), significantly lower than those in the control group, the difference was statistically significant (P<0.05).Conclusion Interferon combined thymopeptideenteric-coated tablets therapy for chronic hepatitis B is helpful to improve the level of chemokine receptor and inhibit the degree of inflammatory reaction.It can help to improve liver function and inhibit viral replication in patients with hepatitis B.It is better than the treatment of chronic hepatitis with thymosin alone.
3.Exploration of Biological Mechanism of Lingering Toxins Theory in Ulcerative Colitis

Objective To explore the biological mechanism of the Lingering Toxins Theory in ulcerative colitis (UC) by clinical trials. Methods Forty patients with UC at mild, moderate active stage and at remission stage were enrolled into the experiment. Twenty cases of UC patients at active stage were divided into Chinese medicine intervention group and western medicine treatment group, 10 in each group. Twenty cases of UC patients at remission stage were also evenly divided into Chinese medicine intervention group and western medicine treatment group. All patients were given conventional western medicine treatment, and additionally, Chinese medicine intervention group were given oral use of Chinese medicine decoction, the treatment course covering 8 weeks. Before and after treatment, plasma matrix metalloproteinase-1 (MMP -1), tissue inhibitor ofmetalloproteinase-1 (TIMP-1) and prostaglandin E2 (PGE2) expression levels were observed by enzyme-linked immunosorbent assay (ELISA) . Results Plasma MMP-1,TIMP-1, and PGE2 expression levels in UC patients were positively correlated with disease severity (P<0.001) . After treatment, the expression levels of MMP-1, TIMP-1, and PGE2 in active-stage patients of both groups were decreased compared with those before treatment (P<0.001) , but the difference between the two groups was insignificant ( P>0.05) . After treatment, the expression levels of MMP-1 and TIMP-1 in remission-stage patients of Chinese Medicine intervention group were decreased (P<0.001) , and the decrease was superior to that of Western medicine treatment group (P<0.05) . Difference of PGE2 expression level in remission-stage patients of both groups was insignificant ( P>0.05) . Conclusion There is a relation between plasma MMP-1, TIMP-1 expression levels and the “lingering toxins” in the field of traditional Chinese medicine. The biological mechanism of toxins-removing therapy in treating UC may be associated with the decrease of the plasma MMP-1 and TIMP-1 expression levels.
4.Analysis of infection status and patterns of hepatitis B among population of health examination in Chongqing

[Abstract ] Objective To understand the infection status of hepatitis B among population of health examination in Chongqing ,and to analyze the modes of hepatitis B outcomes .Methods The 30170 cases of serological results of hepatitis B (HB‐sAg① ,HBsAb② ,HBeAg③ ,HBeAb④ and HBcAb⑤ ) among the individuals of health examination in our hospital from June to December 2013 were performed the retrospective analysis .Results The positive rate of HBsAg among the population of health ex‐amination aged 0-96 years old in Chongqing was 8 .02% (2 419/30 170) ,and the differences in the positive rates among different age groups and between different genders had statistical significance (χ2 =188 .045 ,67 .813 ,P<0 .01) .The infection rate of HBV was 28 .64% (8 641/30 170) ,and the difference in the infection rates among different age groups and between different genders also had statistical significance(χ2 =202 .347 ,36 .171 ,P<0 .01) .30 170 cases of hepatitis B detection results of 5 indexes were conclu‐ded into 14 kinds of modes .As the two common modes of healthy population ,all negative and single HBsAb positive ,their percenta‐ges were 25 .76% (7 772/30 170)and 45 .60% (13 757/30 170)respectively .As the common modes of HBV infection ,“① ④ ⑤”posi‐tive ,“① ③ ⑤” positive and“① ⑤” positive ,their percentages were 6 .18% (1 863/30 170) ,0 .59% (177/30 170) and 1 .21% (366/30 170) respectively .Conclusion The positive rate of HBsAg and the infection rate of HBV among population of health examina‐tion in Chongqing are close to or lower than the national average levels .Analyzing the different combination modes can reflect the different situation of HBV infection and has the guidance significance for the diagnosis and prognosis of the disease .

7.Effects of N-linked glycosylation on specific cell fusion in rubella virus

Objective To explore the effects of glycosylation in E2 and E1 protein on specific cell fusion in rubella virus(RV)strain JR23.Methods Site-directed mutagenesis was used tO obtain mutants containing new enzyme sites on the E2 and E1 gene of RV JR23.All the mutants and wild type proteins were expressed in BHK21 cells and treated with acid medium to induce specific cell fusion.The fusion functions were assayed with Giemsa staining and reporter gene method for qualitative and quantitative analysis,respectively.Expression efficieneies of mutant proteins on cell surface were quantified with fluorescence-activated cell sorter(FACS).Hemadsorption assays were performed to detect binding activity of mutant proteins qualitatively and quantitatively.Results Mutant proteins E2 N53G,S73I and S131V had 62.73%,66.66%and 55.12% of fusion activities,and E1 T78A,T179A and T211A had 66.93%,87.33%and 90.18%of fusion activities,respectively,as compared with the wild type protein.The FACS indicated that the expression efficiencies of all the mutant proteins except E2 S131V were lower than that of the wild type protein.Hemadsorption assays demonstrated that binding abilities of E2 S73I and E1 T78A decreased slightly,but that of the other four mutant proteins Wns almost same as the wild type protein. Conclusion Glycosylation on E2 N53,N71,N129 and E1 N76 were important for the specific cell fusion,but E1 N177 and N209 were almost not.
8.Clinical effects of different surgical methods on papillary carcinoma of the thyroid with clinically negative lymph nodes

Objective:To explore the efficacy and safety of different surgical methods in papillary thyroid carcinoma (PTC) patients with negative cervical lymph nodes (cN0) and to provide theoretical basis for the selection of surgical procedures. Methods:A total of 225 PTC cN0 patients were selected and divided into two groups according to the operation method. Only 110 patients underwent total thy-roidectomy, whereas the 115 cases in the lobectomy group underwent ipsilateral lobectomy and isthmus resection. Al patients were treated with ipsilateral central lymph node dissection. We analyzed both groups of patients in terms of surgery, hospitalization, pathological characteristics, complications, follow-up, and recurrence. Results:(1) The length of incision, blood loss, operation time, and length of stay were not significantly different between the two groups (P>0.05). (2) In the total resection group, the number of multiple foci of the cancer cases was significantly more than that of the lobectomy patients (P<0.05). The average tumor diameter in both groups were not significantly different (P>0.05). Between the two groups, the number of central lymph node dissection and total lymph node metastasis, and the number of cases with central lymph node metastasis were not significantly different (P>0.05). Likewise, the incidence rates of mistakenly cut parathyroid, extremely low blood calcium levels, temporary larynx return nerve paralysis, and superior laryngeal nerve injury were not significantly different between the two groups (P>0.05). No permanent hypocalcemia or permanent recurrent laryngeal nerve injury occurred. (3) All patients were followed up for 2-4.5 years. No recurrent cases were reported in the total resection group. For the lobectomy group, 8 patients with adenocarcinoma had contralateral tumor metastasis and underwent contralateral lobe resection, whereas 4 patients suffered from ipsilateral neck lymph node metastasis at the side area and underwent functional lymph node clearance. Significant differences were noted in the metastasis and recurrence rates between the two groups of patients (P<0.05). No deaths were reported during the follow-up period. Conclusion:For PTC cN0 patients, total thyroidectomy with joint ipsilateral central lymph node clearance can reduce the residual tumor, metastasis, and risk of recurrence,compared with surgery limited to the affected lobe and isthmus. Postoperative complications were not significantly increased.
9.Influence of flurbiprofen axetil for preemptive analgesia on stress response during recovery period in neurosurgical patients

Objective To investigate the effects of flurbiprofen axetil for preemptive analgesia on hemodynamics and stress levels in patients undergoing neurosurgery.Methods 60 patients undergoing neturosurgery were enrolled in the study.The patients were randomly divided into the flurbiprofen group (Group F,n =30) or the control group (Group C,n =30).HR,MAP and serum norepinephrine (NE),epinephrine (E) levels were measured at 30min before the end of operation (T0),immediately after the end of operation (T1),immediately after extubation (T2).Results The changes in MAP,HR and serum NE,E levels in Group F at T1 and T2 were less than those of Group C (all P < 0.0 1).Conclusion Flurbiprofen axetil for preemptive analgesia may suppress the stress response during recovery period in neurosurgical patients,and keep the hemodynamic index stable.
10.Study of left ventricular transmural mechanics at peri - infarct myocardium in vivo using tissue Doppler strain imaging

Objective To observe left ventricular transmural peak radial strain and strain time-to-peak of peri-infarct different layers myocardium using tissue Doppler strain imaging, to assess its mechanical pattern during acute myocardial ischemia.Methods Left anterior descending coronary artery (LAD) were ligated in experimental open-chest Beagle dog models (n = 9),the two-dimensional apical short-axis views of left ventricle in three complete cardiac cycles were acquired and stored in TDI-Q workstation at baseline(the control group of peri-infarc myocardium) and during acute myocardial ischemia respectively.Sampling volume was uesd to measure the peak radial strain and the strain time-to-peak consesquently on the derived M -mode tissue Doppler velocity images at peri-infarct myocardium before and after ischemic segments and different layers(subendocardium, medium, subepicardium).Statistical analies was performed using student's t- test or Pearson's correlations.Results Peak radial strain decreased at peri-infarct subendoeardium (P<0.05) with no significant difference between those at baseline and at peri-infarct medium (P >0.05), the peak radial strain increasd at peri-infarct subepieardium (P < 0.05) ,and the strain time-to-peak at different layers of peri-infarct myoeardium was significantly postponed (P< 0.05).There was a good correlationship of peak radial strain between subendocardium and segment as well as between the medium and segment (r = 0.617, P<0.01 ; r = 0.556, P<0.01).This correlationship disappeared at peri-infarct myocardial segment (r = 0.287, P > 0.05, r = 0.243, P > 0.05).Conclusions The left ventricular transmural mechanical remodeling at peri-infarct myocardium is the integrant of result of mechanical interactions between ischemic and nonischemic myocardium,which might be one of the trigger the structural and fundational remodeling processes involving in the pathophysiological foundation of ischemie cardiomyopathy.
