Objective Stem cell therapy has been extensively used in clinical practice. However ,the methods available are unable to non‐invasively detect the distribution of cells in vivo. This study was aimed to establish a novel molecular imaging probe for stem cell labeling ,which would monitor cells via fluorescence imaging. Methods 1‐iodododecane was used to modify polyethyleneimine (PEI) to form amphiphilic macromolecules. These macromolecules encapsulated the hydrophobic quantum dots by self‐assembly to form imaging probes. Results Strong fluorescence signals of the probe occurred at 630 nm ,and the probe could be effectively endocyted by mesenchyme stem cells. In vivo imaging showed that significant optic signals occurred in probe‐labeled mesenchyme stem cells compared with the control cells (blank group:1.47e8 [p/s/cm2/sr]/[μW/cm2 ];test group:2.78e8 [p/s/cm2/sr]/[μW/cm2 ]). Conclusion The nanoprobes have good optic imaging effects ,and can be used to la‐bel cells.

The aquaporins (AQPs) are small, integral-membrane proteins which selectively transport water across cell plasma membranes. AQPs are over-expressed in tumor cells of different origins, particularly aggressive tumors. AQPs-expressing cancer cells show enhanced migration in vitro and greater local tumor invasion, tumor cell extrava-sations, and metastases in vivo. AQPs inhibition will provide a help for tumor therapy.

Objective To research the influences of gambogic acid on rats'cytochrome P450 isoforms by using the Cocktail probe drugs. Methods Rats were randomly divided into several groups. One group of rats received gambogic acid solution containing 0.5 %CMC-Na (2 mg?mL-1,once per day) orally for six days. At the same time, the rats in the other group received 0.5 %CMC-Na solution serving as blank control group. Blood samples were collect at different time point, and the plasma concentration of cocktail probe drugs was determined by HPLC. Results There was insignificant difference of caffeine and dapsone metabolism between the gambogic acid group and the blank group, but the chlorzoxazone was eliminated faster in the gambogic acid group, and the half life of chlozoxazone was shorter. Conclusion Gambogic acid has no significant influence on the cytochrome P450 isoforms of CYP1A2 and CYP3A4,but has an induction on the CYP2E1.

AIM: To investigate the effect and the mechanism of apolipoprotein (a) [apo (a) ] on proliferation of vascular smooth muscle cells ( VSMCs). METHODS: All VSMCs used in experiments were serial subcultured from primary cells and were identified by immunohistochemistry staining of a - actin. Cell growth assay was observed as cell counting and MTT assay. Western blotting was also employed to detect the related mechanism. RESULTS: All cells used in experiments were confirmed as VSMCs. Although apo (a) enhanced VSMCs proliferation, this effect was attenuated by anti -integrin α_vβ_3, LM609.Use these reagents alone had no effect on VSMCs growth. The results of Western blotting demonstrated that focal adhesion kinase (FAK) was activated by apo (a) and the expression of total or phosphorylated transforming growth factor β_1 (TGF -β_1) was also decreased. However, these effects described above were all blocked by LM609.CONCLUSION: Apolipoprotein (a) enhances VSMCs proliferation and this effect is mediated by integrin α_vβ_3, which activates FAK and attenuates TGF - β_1 and phospho -TGF - β_1 expression.
