2.The endothelin system and pulmonary arterial hypertension
Journal of Medical Postgraduates 2003;0(04):-
Endothelin-1 (ET-1) was characterized as a powerful vasoconstrictor and mitogen for smooth muscle. ET-1 binds to two types of receptors: ET_ A-R and ET_ B-R. Pulmonary arterial hypertension (PH) is a severe condition characterized by a progressive increase in pulmonary vascular resistance. The Endothelial dysfunction plays an important role in the development of pulmonary arterial hypertension. PH can be treated by antagonism of ET-1.

5.Cancer stem cells and tumor vascular heterogeneity
Chinese Journal of Clinical Oncology 2014;(2):134-137
Neovascularization is the fundamental process during tumorigenesis and tumor malignant progression. According the traditional neovascularization theory, tumor vasculatures are mainly developed through angiogenesis by sprouting from preexisting ves-sels and vasculogenesis via recruitment of endothelial progenitor cells from the bone marrow, and the endothelial-dependant vessels are the only way that provides tumor with blood. However, more and more studies indicate that tumor microcirculation network is heteroge-neous and cancer stem cells (CSCs) play an important role during tumor neovascularization. This review highlights the contribution of CSCs to tumor microcirculation modes and the potential anti-angiogenesis targets. Furthermore, this review presents insights for perti-nent studies in the future.
6.Efficacy of neoadjuvant chemotherapy on breast cancer and dynamic changes of circulating tumor cells
Chinese Journal of Clinical Oncology 2013;(23):1431-1435
Objective: This study aims to investigate the neoadjuvant chemotherapy effect of TC and TEC by detecting the change of circulating tumor cells (CTC) in peripheral blood before and after every neoadjuvant cycle of breast cancer patients. Meth-ods:A total of 96 locally advanced breast cancer patients were selected in our hospital from January 2010 to December 2012. These pa-tients were divided into TC (docetaxel and cyclophosphamide) group and TEC (docetaxel, epirubicin and cyclophosphamide) group. Ve-nous blood (5 mL) was extracted from 96 patients prior to neoadjuvant chemotherapy, 48 h after the first, second, third, and fourth cy-cles of neoadjuvant chemotherapy. The CTC values of these two groups of patients were detected by flow cytometry, and their chemo-therapy effect was compared. Results: No significant difference was observed for age, menopausal status, ER, PR, C-erbB-2, tumor size, and clinical stage between the two groups of patients before chemotherapy and for CTC between the two groups before chemother-apy. Before neoadjuvant chemotherapy, the CTC values were correlated with tumor size. The CTC values for patients with tumor size>5 cm were higher than those with tumor size<5 cm. After neoadjuvant chemotherapy, the CTC values of the two groups of patients de-creased (P<0.05), and the CTC values of the TEC group were lower than those of the TC group (P<0.05). Conclusion:After TC or TEC neoadjuvant chemotherapy, the tumors of patients were smaller, and the CTC values were lower than those prior to neoadjuvant chemotherapy. The efficacy of neoadjuvant chemotherapy can be evaluated by monitoring the values of circulating tumor cells in pe-ripheral blood changes.

8.EFFECTS OF INTERMITTENT OVERPRESSURE IN EAR CANAL ON ENDOLYMPHATIC HYDROPS IN GUINEA PIGS
Medical Journal of Chinese People's Liberation Army 2001;0(11):-
Intermittent overpressure of 20cmH 2 O was delivered to the external ear canal of guinea pigs. Histopathological changes in the middle and inner ear were examined under light microscope. Auditory evoked potential was recorded. The expression of atrial natriuretic peptide (ANP) was investigated by immunohistochemical SP method. We found that intermittent overpressure could minimize, but could not interrupt the development of endolymphatic hydrops, and could not improve hearing function. Expression of ANP decreased in the cochlear lateral wall after 2 week intermittent overpressure. The results suggested that intermittent overpressure of 20cmH 2 O was suitable in this experiment, and that ANP positive cells had an autocrine/paracrine function.

Objective The human IFN-inducible protein absent in melanoma 2 (AIM2) has been regarded as a tumour sup-pressor, however, the molecular mechanisms of its antitumor activity still remain unclear. This research is to study the molecular mech-anisms of AIM2 inducing breast cancer cell apoptosis. Methods Tet-Off induction model system was established to induce AIM2 ex-pression in great quantities, MCF-7 tTA-AIM2 cells as the experimental group and MCF-7 tTA-Luc cells as control group.Western blotting was used to detect AIM2 expression in breast cancer cell lines and subcellular localization.XTT assay was applied to analyze the effects of AIM2 on cell proliferation.Apoptosis were detected by Annexin V-FITC and propidium iodide staining, and the apoptosis mechanism were investigated by west blot. Results The established Tet-off guidance system could combine tTA to TRE, thereby promoting AIM2 gene transcription and inducing great expression of AIM2 protein.4 days after induction, the expression of AIM2 could be detected in cytoplasm and nucleus, and AIM2 expression increased with the increasing days.XTT assay detected the growth speed of MCF-7 tTA-AIM2 cell lines slowed down 6 days after induction. After abundant expression of induced AIM2, the percentage of FITC fluorescence apoptosis increased significantly (2.36%vs 14.45%, P<0.01) .Increased AIM2 expression inhibited the expression of the anti-apoptotic protein Bcl-xL, increased the expression of the apoptosis proteins Bad and Bax, and activated caspases, resulting in the cleavage of DNA repair protein PARP. Conclusion In breast cancer Tet-off induction system, AIM2 will express in cytoplasm and nucleus, influence cell proliferation, and stimulate the mitochondria to cell apoptosis.
