1.Quality standard for Ganmao Qingre Granules

AIM: To study the quality standard for Ganmao Qingre Granules(Herba Schizonepetae,Radix Saposhnikoviae,Radix Puerariae,etc). METHODS: Radix Saposhnikoviae,Spica Schizonepetae,Radix Bupleuri,Herba Platycodonis in Ganmao Qingre Granules were identified by TLC,and the content of puerarin was determined by HPLC. RESULTS: Radix Saposhnikoviae,Herba Schizonepetae,Radix Bupleuri,Herba Platycodonis could be identified by TLC.The concentration curve of hesperdin showed linearity over the range of 0.086 8-8.680 ?g(r=0.999 9).The average recovery was 99.09%,RSD was 1.12%(n=9). CONCLUSION: The method is simple,specific and accurate and can be used effectively for the quality control of Ganmao Qingre Granules.
2.Efficacy of propofol compared with sevoflurane on inflammatory factor and liver function after hepatectomy

Objective To evaluate efficacy of propofol compared with sevoflurane on inflammatory factor and liver function after hepatectomy. Methods 40 cases with hepatectomy were divided into propofol group ( P group) and sevoflurane group ( S group) , 20 cases in each group.The P group and S group respectively were given propofol or sevoflurane maintained anesthesia.The alanine aminotransferase( ALT) , total bilirubin( TBil) , aspartate amino transferase(AST), albumin(ALB), plasma tumor necrosis factor(TNF-α), interleukin-6(IL-6), interleukin-10(IL-10)and complication rate were compared between two groups.Results Postoperation 1d, 3d and 7d, the AST, ALT, and TBiL levels of S group were significantly lower than those of P group (P<0.05).There was no significant difference of ALB level between two groups.The plasma TNF-α, IL-6 and IL-10 levels of S group were significantly lower than those of P group (P<0.05).There was no significant differece of the rate of postoperative complications between S group and P group [6(30.0%) vs.10 (20.0%); χ2 =1.67, P >0.05].Conclusion Sevoflurane inhalation could effectively inhibit the plasma inflammatory cytokines secretion after hepatectomy, and promote liver function recovery, its effect is better than that of propofol.
3.Effect of intrathecal injection of dexmedetomidine on the behavioral activity, pain degree and expression of protein kinase C in the spinal dorsal horn in rat models of chronic neuropathic pain

Objective To analyze the effect of intrathecal injection of dexmedetomidine on the behavioral activity, pain degree and expression of protein kinase C in spinal dorsal horn of rat models of chronic neuropathic pain, and to investigate the analgesic mechanism of dexmedetomidine.Methods 75 healthy male rats were randomly divided into observation group, model group and control group, 25 rats in each group.Chronic sciatic nerve injury model was established in the observation group and model group.After modeling, intrathecal dexmedetomidine intervention was used in the observation group.The model group was treated with saline injection and there was no intervention in the control group.Before the modeling (BM)and at 3(D3), 5(D5), 7(D7), and 14 (D14)days after medicine administration, the behavioral capacity was evaluated by cumulative evaluation method and movement function evaluation, and the assessment of pain degree (mechanical withdrawal method and thermal withdrawal latency pain threshold detection method), PKC staining score (immunohistochemical SABC method), PKC mRNA assay (RT-PCR method) and PKC protein expression (Western blot) were conducted and the data were statistically analyzed.Results ① Before modeling, the behavior, the cumulative scores of motor function, MWT, and TWL showed no significant differences between the different groups (P>0.05).After modeling, the model group and observation group showed that the cumulative scores and motor function scores were increased significantly, MWT and TWL decreased significantly, and the changes in the observation group were significantly lower than those in the model group (P<0.05).After modeling, the cumulative scores, motor function scores, MWT, and TWL were significantly different between the groups (P<0.05).② The expression of PKC was negative in the control group and positive in the model group.In the observation group, after the initial establishment of model, the PKC was strongly positive, and along with the prolonged treatment, the PKC expression intensity was decreased, and only weakly positively expressed at 14 d.③ After modeling, the observation group and model group showed that the PKC mRNA and PKC protein expression levels were significantly higher than that of the control group (P<0.05).With the continuous drug administration, the PKC mRNA and PKC in the observation group were decreasing, and reached a level close to that of the control group at 14 d of drug administration.From the third day after modeling, at the same time points, the amount of PKC expression in the observation group was significantly lower than that in the model group (P<0.05).Conclusions Intrathecal injection of dexmedetomidine can improve the behavior of rat models with chronic neuropathic pain, and reduce the degree of pain.It may be related to the inhibition of protein kinase C expression in the spinal dorsal horn.
4.Relationship between Hypertension Prevalence and Body Weight in Students in Beijing Changping Area

Objective To explore the relationship of hypertension prevalence and body weight in children and teenagers in Changping district of Beijing. Methods With stratified cluster sampling, 4 247 students (2 090 female and 2 157 male) aged 7~18 yrs from primary and middle schools in Changping district were investigated in May to October, 2007, and examined on their body height, weight, systolic pressure, diastolic pressure, fasting blood glucose, etc. Data was analyzed with SPSS11.5. 5.Clinical significance of the serum fibrosis parameters in Chronic hepatitis B patients

Objective To study the relationship between serum fibrosis parameters levels and chronic viral hepatitis B patients.Methods Radioimmunoassay and chemiluminescence were used to test the levels of hyaluronic acid(HA),collagen type Ⅳ(Ⅳ-C),laminin(LN) and procollagen type Ⅲ(PC Ⅲ) in 146 cases of chronic hepatitis B and 40 of normal controls,and the relationship with clinical parameters of liver function were analyzed.Results The levels of HA,Ⅳ-C,LN,PC Ⅲ in chronic hepatitis B patients were higher than those of normal control group (P <0.05 or P < 0.01),there were statistical differen in different course of disease (P < 0.05 or P < 0.01) ; The levels of serum Ⅳ-C and PCⅢ had positive correlation with plasma PT,the same to HA with TBil and PCⅢ with ALT;The levels of HA,Ⅳ-C and LN had negative correlation with Alb,the same to LN with CHE.Conclusion The levels of HA,Ⅳ-C,LN and PC Ⅲ in chronic hepatitis B patients may reflect the situations of hepatic fibrosis and the degrees of liver function damage.
6.Determination of glycated hemoglobin standardization situation

Glycated hemoglobin is internationally recognized as the gold standard for assessment of long-term glycemic status in diabetic patients,but our determination of glycosylated hemoglobin standardization work is still at primary stage at present.Consistency and accuracy of glycated hemoglobin results are still needed to be solved.
7.Comparison of dual-energy X-ray absorptiometry T-score concordance between the measurement sites of hip and lumbar spine in the elderly

Objective To compare dual-energy X-ray absorptiometry T-score concordance between hip and lumbar spine, and to estimate the value of T-score of the hip for diagnosing osteoporosis in the elderly. Methods From September 2000 to May 2002, 123 males aged 60-89 years (average 72.0?7.4) and 137 females aged 60-88 years (average 67.3?6.1) were selected in the study based on the exclusion of subjects with apparent abnormalities affecting bone mass or taking medications which might affect bone metabolism. Detailed questionnaire included lifestyle factors such as smoking, drinking, exercise and dietary habits, etc. Radiographs of the thoracic and lumbar spine were taken. Patients' BMD were measured by DEXA (DMS, Challenger) at lumbar spine (L 2-4) and left hip in posterior-anterior projection. The right hip was scanned if patients had a history of fractures or any bone disease in the left hip. All data were statistically analysed using SPSS 8.0 system. 8.Retrospective study on the dorsal metacarpal flap and compound flap

Objective To report the long-term outcomes of repairing finger soft tissue defects using the dorsal metacarpal flap and compund flap. Methods Through a review of 122 cases,who were treated using the dorsal metacarpal flap and compound flap,the indication and long-term outcomes of the flap and characteristics of blood supply and compound tissues transferred and venous return were analyzed. Results All cases survived unevently.The fingers which were repaired using the flap were normal,and the contours of the donor sites were normal. Conclusions The dorsal metacarpal flap and compund flap are an optimal method to repair finger soft tissue defects.
9.Effect of matrilysin and its antisense oligonucleotide on proliferation of lung adenocarcinoma cell line A549

Objective To study the effect of the recombinant matrilysin (rMMP-7) and its antisense oligonucleotide transfected by liposome (LIPO-MAON) on the proliferation of lung adenocarcinoma cell line A549 and human umbilical vein endothelial cells (HUVEC). Methods Antisense oligonucleotide of matrilysin was constructed by liposome transfection. The proliferation of HUVEC and A549 was detected by MTT assay in case of rMMP-7 or LIPO-MAON existence. The expression of MMP-7 mRNA in both HUVEC and A549 was examined by semi-quantitative RT-PCR assay. Results The proliferation of both HUVEC and A549 cell line was accelerated by high level of rMMP-7 (0.75, 1.0 ?g/ml), and the inhibited proliferation was only found in A549 cell line treated with high concentration of LIPO-MAON (7.5, 10 nmol/ml), but not in HUVEC. 10.Recognition of Specific Antigens by Specific IgG and IgE During Anaphylactic Shock Induced by Echinococcus granulosus in Sheep

Objective To determine the specific recognition of Echinococcus granulosus (E.g.) cyst fluid crude antigen (EgCF) and antigen B (EgB) by serum specific IgG and IgE using Western blotting during anaphylactic shock induced by E. g. in sheep, and to investigate the importance of defined characteristics and molecular weight of the specific antigens. Methods EgCF was obtained through local slaughterhouses in Urumqi from the cysts of infected sheep liver. Western blotting was used to analyze total specific IgG and IgE antibodies in serum from sheep infected with E.g. using either crude antigen of E. g. and EgB, and to understand specific recognition of hydatid cyst antigens by serum total IgG and specific IgE. Results SDS-PAGE and Western blotting showed that there were differences between EgB and EgCF in electrophoresis pattern. EgB was recognized by IgG from sera of infected sheep in a series of bands with molecular weight ranging from 31, 43 to 66.2 kDa. No binding of IgG against EgCF was observed in any serum from the infected sheep during anaphylactic shock. In contrast, specific IgE antibodies in E. g. infected sheep obviously recognized the single 43 kDa subunit of EgCF, but no binding of specific IgE against EgB was observed in sera of the infected sheep. Conclusion EgCF is consisted of antigenic components in which there is a specific antigen against IgE with a molecular weight of over 43 kDa. This component may lead to an anaphylactic shock induced by E. g. . EgB is a specific antigen against the total IgG but not to the specific IgE.
