1.Inhibitory effect of MMP-7 antisense oligodeoxynucleotide on proliferation of human pneumonic adenocarcinoma cell line A549

Objective To investigate the possibility of inhibiting the malignant proliferation of tumor cells in vitro through down-regulating the expression of MMP-7 by antisense gene technology. Methods A Phosphorothioate antisense oligodeoxynucleotide (ASODN) and scrambled oligodeoxynucleotide (SCODN) against MMP-7 were respectively transfected into A549 cells mediated by liposome. The expression of MMP-7 and cell proliferation in the cells were respectively examined by immunohistochemical assay and MTT. Flow cytometry was used to detect the cell cycles. Results After A549 cells were transfected with MMP-7 ASODN, the cell number was decreased. MTT method indicated that the proliferation of A549 cells was inhibited by MMP-7 ASODN at different concentrations, with the inhibition reaching the peak at 48 h. ASODN transfection downregulated the expression levels of MMP-7 protein, and inhibited the transition period of G_ 0 /G_ 1 phase to S phase. Conclusion The artificial MMP-7 ASODN can specifically inhibit the expression of MMP-7 protein and regulate cell cycle and cell proliferation in A549 cells.
2.Syphilis in 100 Married Couples

Objective To uncover the possible factors of rapid increasing incidence of syphilis. Methods The clinical data of 100 patients firstly diagnosed as syphilis and their spouses were analyzed. Results There were 65 couples (130 cases) both were infected with syphilis, accounting for 65%. Among these cases, 25 males firstly diagnosed, 15 of them were primary syphilis; and 40 females firstly diagnosed as syphilis, 32 of them were secondary syphilis. Among 65 syphilis couples, 48 spouses were quickly diagnosed as syphilis through contact tracing, but the other 17 spouses were not diagnosed as syphilis until 1~3 months later, and most of them were latent syphilis, accounting for 83.2%(54/65). Chancre, which may be noticed by the patients themselves, was not so common in this group of patients, accounted for only 28.5%(37/130), and mainly in males. Conclusions The infections rate of syphilis between spoused is high, especially latent syphilis, and chancre is low, which might be one of the factors for the rapid spreading of the disease.
3.Efficacy of propofol compared with sevoflurane on inflammatory factor and liver function after hepatectomy

Objective To evaluate efficacy of propofol compared with sevoflurane on inflammatory factor and liver function after hepatectomy. Methods 40 cases with hepatectomy were divided into propofol group ( P group) and sevoflurane group ( S group) , 20 cases in each group.The P group and S group respectively were given propofol or sevoflurane maintained anesthesia.The alanine aminotransferase( ALT) , total bilirubin( TBil) , aspartate amino transferase(AST), albumin(ALB), plasma tumor necrosis factor(TNF-α), interleukin-6(IL-6), interleukin-10(IL-10)and complication rate were compared between two groups.Results Postoperation 1d, 3d and 7d, the AST, ALT, and TBiL levels of S group were significantly lower than those of P group (P<0.05).There was no significant difference of ALB level between two groups.The plasma TNF-α, IL-6 and IL-10 levels of S group were significantly lower than those of P group (P<0.05).There was no significant differece of the rate of postoperative complications between S group and P group [6(30.0%) vs.10 (20.0%); χ2 =1.67, P >0.05].Conclusion Sevoflurane inhalation could effectively inhibit the plasma inflammatory cytokines secretion after hepatectomy, and promote liver function recovery, its effect is better than that of propofol.
4.Effect of Xinjihuoliyin on Myocardium Injury Induced by Adriamycin in Rats

Objective To observe the protective and antioxidative effect of Xinjihuoliyin on the myocardium injury induced by adriamycin (ADM) in rats. Methods 80 SD rats were randomly divided into four groups:normal group, control group, Xinjihuoliyin group and positive control group (Huangqishengmaiyin group). In control group, Xinjihuoliyin group and Huangqishengmaiyin group, ADM was intraperitonial injected at a dose of 2.5 mg/kg, three times every week, for 4 weeks, while normal saline was intraperitonial injected at a dose of 3 mL/kg in normal group. In Xinjihuoliyin group, Xinjihuoliyin was intragastric administrated once a day at a dose of 10 g/kg. In Huangqishengmaiyin group, Huangqishengmaiyin was intragastric administrated once a day at a dose of 7 mL/kg and distilled water was given to other two groups. The activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) of serum and myocardium were assayed. The myocardial pathomorphism was detected at the same time. Results Compared with the control group, MDA of serum and myocardium was lower, and SOD of serum and myocardium was higher in Xinjihuoliyin and Huangqishengmaiyin group. The scale of myocardial pathological damage in Xinjihuoliyin and Huangqishengmaiyin group was improved. Conclusion Xinjihuoliyin has protective effect and antioxidative effect on the myocardium injury induced by adriamycin in rats.
5.Construction and identification of recombinant baculovirus of the novel CEA vaccine

Objective To design a novel efficient CEA vaccine of fission epitopes with the vehicle of HPV16L1,and construct the recombinant baculovirus in Bac-to-Bac baculovirus expression system.Methods The recombinant baculovirus expression vector pFastBac1-V was constructed.HPV16L1 and CEA-PADRE synthesis primers were inserted into bacmid in E.coli DH10Bac with the help of Tn7 transposition system.Then the recombinant baculovirus was got by infecting insect cell sf9.Results HPV16L1 and CEA-PADRE synthesis primers that were cloned to the baculovirus transfer vector pFastBac1 were assessed by restriction analysis and direct sequencing.The recombinant baculovirus of the novel vaccine had been constructed.After infected sf9,the correct fragment was amplified by PCR.Conclusion The baculovirus of CEA vaccine of fussion epitopes with the vehicle of HPV16L1 was constructed.
6.Therapeutic effect and mechanism of intrapleural administration of cisplatin bound to activated carbon particles for treating malignant pleural effusion

Objective To evaluate the therapeutic effect and possible mechanism of intrapleural administra- tion of cisplatin bound to activated carbon particles for treating malignant pleura] effusions(MPE). Methods ①60 patients with MPE confirmed in the Third Hospital of Changzhou and the Second Hospital of Changzhou from 2004 to 2007 were randomly divided into treatment group (n=30)and control group(n=30). Chest catheters were inserted percutaneously into the pleural space to drain the effusions. Cisplatin mixed with activated carbon particles for the treatment group and only cisplatin for the control group were injected into pleural cavity. Whether above-mentioned treatment should be repeated was determined by ultrasonic B every week until up to four times. Curative effect and side-effect were compared between two groups a month later. ②20 cases, randomly selected from both groups respectively, underwent whole body and chest SPECT scan to image lymphatic system by means of 99Tcm-DX after effusions drainage but before intrapleural injection of drug. Cases whose imaging graphics were abnormal would undergo the above SPECT again 2 weeks after intrapleural injection of drug so as to find changes in imaging graphics. Results ① The overall response rate was significantly higher in treatment group than in control group(100% (30/30) vs 66.7% (20/30), χ2=12.00, P<0.01)and that intrapleural injection was needed only once in most cases. Gastrointestinal upset and leucoponia were less and milder in treatment group than in control group (16.7% (5/30) vs 30.0% (9/30) and 6.7% (2/30) vs 20.0% (6/30) respectively),but there was no statistical difference between the two groups(χ2=1.49 and 1.30,P>0.05). ②The rate of improvement in lymphatic imaging was significantly higher in treatment group than in control group (78.6% (11/14) vs 37.5% (6/16),P<0.05). Conclusion The treatment of malignant pleural effusions with intrapleural administration of cisplatin bound to activated carbon particles is extremely effective and safe, and can improve lymphatic drainage as well.
7.Comparative study of two proteases for mass spectrometric method for the determination HbA1c

Objective To explore the difference of two kinds of proteolytic enzymes (GLU-C and Lys-C trypsin),hemoglobin A1c were measured in mass spectrometry.Methods Based on the IFCC recommended hemoglobin A1c reference measurement method by mass spectrometry,the blood samples were preparied (the number of these blood samples:201201,201202,201203).Hemoglobin in the samples were enzymed respectively by two kinds of proteolytic enzymes (Glu-C and Lys-C),before the mass spectra which be used solid phase extraction method,under the optimal experimental conditions,hemoglobin A1c in the sample were measured by the liquid chromatography tandem mass spectrometry and screened of variant hemoglobin with sequest software.All results by mass spectrometry are required to use the Sequest software to search RAW file in the human IPI Database,retrieve four common hemoglobin variant published results and compare hemoglobin variant sites,using statistical software SPSS13.0 and Excel 2003 on the results in the correlation analysis,compare between groups by the independent sample t test.Results In the detection of hemoglobin A1c,respectively Glu-C and Lys-C blood which the samples were enzymed,results of hemoglobin A1c by Lys-C were (34.70 ± 2.80),(51.76 ± 1.60),(73.39 ± 1.11) mmol/mol,results of hemoglobin A1c by Glu-C were(33.12 ± 1.48),(54.54 ± 2.50),(75.40 ± 3.60) mmol/mol,the difference between groups was not statistically significant,the results of two methods of drawing curve,all R2 > 0.995.Searching the human database with sequest software,four common hemoglobin variant were not be finded.Conclusions In the detection of hemoglobin A1c by the mass spectrometry,the application of the above two kinds of proteolytic enzyme in the blood samples,that can be more consistent results,test results of two methods have a good correlation.
8.The study of E-cadherin gene abnormality in non-small cell lung cancer

Objective To observe the E cadherin gene abnormality in non small cell lung cancer(NSCLC). Methods E cadherin gene mutations in exons 6, 7, 8 and 9 in 24 cases of non small cell lung cancer were studied by PCR SSCP and DNA sequence analysis. Results Abnormality of the E cadherin gene was found in exon 9 by means of insertion, deletion and mutation in 2 cases. Conclusion Abnormality of E cadherin gene is present in NSCLC by means of insertion and deletion in extracelluar matrix. The main cause of the decrease in epithelial adhesion may be resulted from the abnormality gene change of the E cadherin.
9.The evaluate of Paraelectronic effusion by medical thoracoscopy

Objective To determine chest ultrasound locating treatment of paraelectronic effusion by medical thoracoscopy.Methods We chose 40 Xinxiang center hospital patients in respiratory wards from June 2012 to June 2014,who was randomly divided into two teams,one was electron-assisted Thoracoscopic surgery,the other was thoracic cavity closed drainage combination of intrapleural injection of urokinase.It was 3rd reviewed by chest ultrasound,we evaluated curative effection ac cording to the drainage effect,we statisticsed two draining days and total inpatient days,and total hospital costs.The results two teams were compared between gender,age,nutritional status,effusion volume (length x width x depth),pleural effusion conventional,biochemical,lactate dehydrogenase,adenosine deaminase,carcinoembryonic antigen,the difference was not statistically significant (P > 0.05).Pleural fluid culture results were negative.Medical thoracoscopy treatment group hospitalization days,drainage time was significantly shorter than the thoracic drainage combined with pleural urokinase group,The same was the cost of hospitalization.Medical thoracoscopy poor drainage treatment group was significantly lower than the thoracic drainage combined with pleural urokinase group (P < 0.05).Conclusion To the middle-large number encapsulated paraelectronic effusion,chest ultrasound locating treatment by medical thoracoscopy early can reduce the formation of empyema,reduce the formation of pleural fiberboard.It is a safe,effective and worthy of promotion treatment.
10.Application of diffusion weighted imaging in severity assessment and prognosis prediction of diffuse axonal injury

Objective To discuss the value of diffusion weighted imaging (DWI) in severity assessment and prognosis prediction of diffuse axonal injury (DAI). Methods A retrospective study was performed on the clinical imaging data and the follow-up results at six months after injury in 29 patients with DAI. The detection rate of DAI lesion by DWI and conventional MRI was compared by means of one-way ANOVA. The correlation between the number of lesion in different areas with GCS and GOS was analyzed with Spearman rank correlation test. Results (1)The average detected DAI lesions were 19.24±5.72 on DWI, 14.41 ±4.50 on FLAIR, 10.58±3.79 on T2WI and 4.83 ±2. 11 on TIWI, with the highest detection number of DAI lesion on DWI (P < 0, 05). (2) There was a significant correlation of the number of central lesions (in corpus callosum, basal ganglia and brain stem) with GCS and COS (P < 0. 05), but there was no correlation of total lesion number or periphery lesion with GCS and COS (P > 0.05). Conclusions DWI is a potentially useful imaging modality in detecting DAI lesion, when the number of central lesion on DWI can be served as an objective marker in severity assessment and prognosis prediction of DAI.
