1.Application of SELDI-TOF-MS in searching for biomarkers for colorectal cancer

SELDI-TOF-MS is a novel proteomic technique.It reveals some new biomarkers which could be used for early diagnosis,preoperative staging and predicting response to radiochemotherapy of colorectal cancer.These new biomarkers were validated to be more sensitive and specific than traditional hiomarkers.SELDI-TOF-MS will be a useful tool for early diagnosis and tailor-made therapy of eolorectal cancer.
2.MTA1 in tumor progression and metastasis

MTA1 is a composition of Nurd,which may deacetylate histone and non-histone to affect gene transcription and protein expression. Since it is often overexpressed in many tumors and closely related to invasion,metastasis and prognosis of the malignancy, MTA1 might be exploited as a tumor marker for clinical application. This article reviews the structure and function of MTA1 and some new research progress on the tumor metastasis related to MTA1.
3.Effects of different degrees and duration of hypothermia on traumatic brain injury in dogs

Objective To investigate the effects of different degrees and duration of hypothermia on severe traumatic brain injury in dogs. Methods Thirty-six one-year-old healthy dogs weighing 13-15 kg were used in this study. Traumatic brain injury model was estabhshed according to the method described by Feeney. Orthogonal design was used in this experiment. Two empirical factors: temperature (factor A) and duration (factor B) were used. Body temperature was maintained at 38 ℃ (A1), 31 ℃ (A2) or35 ℃ (A3) for 6 h (B1) or 12 h (B2).The dogs were divided into 6 groups: group A1B1 , A1B2, A2B1, A2B2, A3B1 and A3B2. Blood gas analysis was performed after the target temperature was maintained for the target duration. Neurological deficit was assessed and scored (0 = no deficit, 500 = severe neurological deficit) and blood samples were obtained at 24, 48 and 72 h after brain injury for determination of serum concentrations of neuron-specific enolase (NSE) and myelin basic protein (MBP) and plasma S-100β protein concentration. Brains were removed at the 72 h after brain injury for determination of Bcl-2 and Bax expression and detection of apoptosis in the brain. Results Hypothermia significantly decreased serum NSE, MBP and plasma S-100β concentrations, neuronal apoptosis and NDS scores;up-regulated Bcl-2 expression and down-regulated Bax expression in brain tissue in the 4 hypothermia groups (group A2B1, A2B2, A3B1 and A3B2) as compared with the control groups (group A1 B1, A1 B2). Best neuroprotective effects were observed in group A3 B1 (35 ℃ for 6 h) in terms of serum NSE, NBP and plasma S-100β concentrations, neuronal apoptosis and cerebral Bax and Bcl-2 expression, but there was no significant difference in the NDS scores among the 4 hypothermia groups. Conclusion Hypothermia can provide neuroprotection in a dog model of traumatic brain injury but the neuroprotective effect is independent of the degree and duration of hypothermia.
4.Effects of Levocarnitine on Peripheral Blood T Cell Subsets and Inflammatory Factors in Diabetic Patients Underwent Peritoneal Dialysis

OBJECTIVE:To investigate the effects of levocarnitine on peripheral blood T cell subsets and inflammatory factors in diabetic patients underwent peritoneal dialysis. METHODS:A total of 100 diabetic patients underwent peritoneal dialysis in se-lected as research objects were divided into conventional therapy group and levocarnitine group according to random number table, with 50 cases in each group. Conventional therapy group received peritoneal dialysis and conventional therapy. Levocarnitine group was additionally given levocarnitine 1.0 g added into 0.9% Sodium chloride injection 20 mL intravenously,3 times a week,on the basis of conventional therapy group for 12 weeks. CD3+,CD4+,the proportion of Th1 and Th2,the contents of TNF-α,INF-γ, IL-10 and IL-4 in supernatant were all detected before and after treatment. The expression of T-bet and GATA-3 were compared be-tween 2 groups. RESULTS:After treatment,CD3+,CD4+,Th2 cells,GATA-3 expression and IL-10,IL-4 levels were significantly increased,the expression of Th1 cells,T-bet content and TNF-αand INF-γwere significantly reduced,and levcarnitine group was sig-nificantly better than conrentional therapy group,with statistically significant (P<0.05). CONCLUSIONS:Levocarnitine can im-prove inflammatory reaction and the expression of related transcription factors by promoting T cell subsets and Th1/Th2 cell subsets.
5.Effect of lipid emulsion on mitochondrial energy metabolism during bupivacaine-induced myocardiotoxicity in rats

Objective To investigate the effect of lipid emulsion on mitochondrial energy metabolism during bupivacaine-induced myocardiotoxicity in rats.Methods H9c2 cells were inoculated in 96-well plates at a density of 1 × 105cells/ml, and were randomly divided into 4 groups (n =18 each) using a random number table: control group (group C) , bupivacaine group (group B) , lipid emulsion group (group LE) , and bupivacaine + lipid emulsion group (group B + LE).The cells were incubated in the normal culture medium in group C.In group LE, the cells were incubated in the culture medium containing 1% lipid emulsion.In group B, the cells were incubated in the culture medium containing 1 mmol/L bupivacaine.In group B + LE, the cells were incubated in the culture medium containing 1 mmol/L bupivacaine and 1% lipid emulsion.After 24 h of incubation, the contents of ATP, ADP, and AMP were measured by high-performance liquid chromatography, and apoptotic rate was calculated by Hoechst33342/ PI staining.Results Compared with group C, the contents of ATP, ADP and AMP were significantly decreased, and apoptotic rate was increased in group B (P<0.05), and the contents of ATP and ADP in group LE and ATP content in group B + LE were increased, and no significant changes were found in apoptotic rate in LE and B+LE groups (P>0.05).Compared with group C, the contents of ATP, ADP and AMP were significantly increased, and apoptotic rate was decreased in LE and B+LE groups (P< 0.05).Compared with group LE, the contents of ATP, ADP and AMP were significantly decreased (P< 0.05), and no significant change was found in apoptotic rate in group B+LE (P>0.05).Conclusion The mechanism by which lipid emulsion reduces bupivacaine-induced myocardiotoxicity may be associated with improved mitochondrial energy metabolism in rats.
6.Preparation, Characterization and Application for Genomic DNA Extraction of a Magnetic Microsphere with Surface Charge Switching

SuperparamagneticSiO2@Fe3O4microspherewithcore-shellstructurewaspreparedbythe method of hydro-thermal and St?ber method, and modified by (3-aminopropyl) triethoxysilane ( APTES) . The obtained amino-Fe3 O4@SiO2 microsphere with controlled surface charge properties was characterized by SEM and TEM, and the surface electric property of amino-microsphere was investigated by Zeta potential measurement. The microsphere was used to extract DNA from human blood, and the solid phase extraction method by microspheres was developed. The mechanism of action between Aminated-Fe3 O4@SiO2 microsphere and DNA was explored, and the gel electrophoresis and PCR test were done on the extraction product. The result showed that genomic DNA with high purity was successfully extracted from whole blood by using amino-Fe3 O4@SiO2 microsphere, with the extraction rate about 70%, the saturated adsorption capacity for DNA was about 40 ng/μg, and the elution could be directly used for further bio-analysis.
7.Role of mitochondrial permeability transition pore in lipid emulsion-induced inversion of bupivacaine myocardiotoxicity in rats

Objective To evaluate the effect of mitochondrial permeability transition pore (mPTP) in lipid emulsion-induced inversion of bupivacaine myocardiotoxicity in rats.Methods H9c2 cells were inoculated in 6-well plates at a density of 105 cells/ml, and were randomly divided into 4 groups (6 wells in each group, 2 ml/well) using a random number table: control group (group C) , bupivacaine group (group B) , lipid emusion + bupivacaine group (group LB) , and lipid emusion + bupivacaine + atractyloside group (group LBA).Phosphate buffer solution 100 μl was added to the culture medium in group C.In group B, bupivacaine was added to the culture medium with the final concentration of 1 mmol/L.In group LB, lipid emusion and bupivacaine were added to the culture medium with the final concentrations of 1% and 1 mmol/L, respectively.In group LBA, lipid emusion, bupivacaine and atractyloside (an mPTP opener) were added to the culture medium with the final concentrations of 1%, 1 mmol/L and 30 μmol/L, respectively.All the cells were incubated for 24 h.After the end of incubation, the expression of Bcl-2, Bax, phosphorylated Bad (p-Bad) , caspase-3, activated caspase-3, caspase-9,activated caspase-9 and cytochrome c (Cyt c) was detected using Western blot.The expression of Bcl-2 mRNA, Bax mRNA, Bad mRNA, caspase-9 mRNA and Cyt c mRNA was detected using real-time reverse transcriptase polymerase chain reaction.The ratios of Bax/Bcl-2, activated caspase-3/caspase-3, activated caspase-9/caspase-9, and Bax mRNA/Bcl-2 mRNA were calculated.Results Compared with group C,the ratios of Bax/Bcl-2, activated caspase-3/caspase-3, activated caspase-9/caspase-9, and Bax mRNA/ Bcl-2 mRNA were significantly increased, the expression of p-Bad was down-regulated, and the expression of Cyt c, Bad mRNA, caspase-9 mRNA and Cyt c mRNA was up-regulated in group B (P<0.05) , and no significant change was found in the parameters mentioned above in group LB (P>0.05).Compared with group B, the ratios of Bax/Bcl-2, activated caspase-3/caspase-3, activated caspase-9/caspase-9, and Bax mRNA/Bcl-2 mRNA were significantly decreased, the expression of p-Bad was up-regulated, and the expression of Cyt c, Bad mRNA, caspase-9 mRNA and Cyt c mRNA was down-regulated in LB and LBA groups (P< 0.05).Compared with group LB, the ratios of Bax/Bcl-2, activated caspase-3/caspase-3, activated caspase-9/caspase-9, and Bax mRNA/Bcl-2 mRNA were significantly increased, the expression of p-Bad was down-regulated, and the expression of Cyt c, Bad mRNA, caspase-9 mRNA and Cyt c mRNA was up-regulated in group LBA (P < 0.05).Conclusion The mechanism underlying lipid emulsioninduced inversion of bupivacaine myocardiotoxicity is related to inhibited mPTP opening in rats.
8.Effects of endovascular cooling in different degrees on severe traumatic brain injury in dogs

Objective To investigate the effects of endovascular cooling in different degrees on severe traumatic brain injury in dogs.Methods Eighteen dogs of both sexes,weighing 12-15 kg,were randomly divided into 3 groups ( n =6 each):hypothermia at 31 ℃ group (group A),hypothermia at 35 ℃ group (group B) and normothermia group (group C).The model of severe traumatic brain injury was established according to the improved Feeney's free-fall method.Six hours of endovascular cooling was performed using an endovascular cooling system in A and B groups.The cerebrospinal fluid (CSF) was collected before traumatic brain injury (T0),and at 24,48 and 72 h after traumatic brain injury (T1-3) for measurement of the concentrations of neuron-specific enolase (NSE),S-100β,myelin basic protein (MBP),aspartate (Asp),glutamie acid (Glu),glycine ( G ly) and gammaaminobutyric acid (GABA).Results The concentrations of NSE,S-100β,MBP,Asp,Glu and Gly in CSF were significantly lower at T1-3,while the concentration of GABA in CSF was significantly higher at T1-3 in groups A and B than in group C (P <0.05 or 0.01).The concentrations of NSE,Asp,Glu and Gly in CSF were significantly lower at T1-3,while the concentration of GABA in CSF was significantly higher at T1-3 in group B than in group A ( P <0.05 or 0.01).Conclusion Endovascular cooling can reduce severe traumatic brain injury in dogs,and the efficacy is better when the temperature is reduced to 35 ℃ than that when reduced to 31 ℃.
9.Correlations between serum macrophage migration inhibitory factor and active rheumatoid arthritis

BACKGROUND: Macrophage migration inhibitory factor (MIF) is mainly concerned with macrophage mobilizing function, as the upper stream cytokine of tumor necrosis factor-α(TNF-α), interleukin-1β (IL-1β), which may have critical effect in the process of the onset of rheumatoid arthritis. OBJECTIVE: To explore the correlations between the change of serum MIF and the activity of rheumatoid arthritis (RA) disease.DESIGN, TIME AND SETTING: Non-randomized control and case study, which was carried out in the Bethune International Peace Hospital of Chinese PLA from September in 2005 to October in 2006.PARTICIPANTS: Sixty RA patients were included in this study, and other thirty healthy subjects were selected as the control group. There were significant differences in age and sex between the two groups. METHODS: Clinical data of sixty RA patients were selected by carrying out retrospective analysis, then on the basis of disease activity score (DAS) accumulated points, they were divided into active and inactive group respectively, who were contrasted with 30 health adults. MAIN OUTCOME MEASURES: ① Morning stiffness (in minutes), joint tenderness index, arthrocele index, semi-quantity rheumatoid factor (RF), C-reactive protein concentration (CRP), erythrocyte sedimentation rate (ESR), and platelet count (PLT) were recorded; ② To compare the level of serum MIF, IL-1β and TNF-α among active group, inactive group, and control group; ③ The correlation analysis was carried out among the level of serum MIF, inflammatory index and clinical observation index.RESULTS: There was significantly increased in serum MIF of patients in the active group compared to of inactive and normal groups (P < 0.05), but there were no significantly differences between inactive and control groups (P > 0.05). There were significant correlations between the serum MIF concentration and active inflammatory index of RA disease, blood sedimentationrate (ESR), C-reactive protein (CRP), platelet counting (PLT), interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), swell joint index (SJI) and tenderness joint, but no significant difference was observed between the serum MIF, age, disease course, morning stiffness and rheumatoid factor (RF).CONCLUSION: The serum MIF concentration is significantly increased in patients with RA, and it may be a useful parameter for monitoring disease activity of RA.
10.Hyphenation of C18 Gel Silica Column and Flow Injection withInductively Coupled Plasma-Mass Spectrometry andIts Application on the Seawater Analysis

An online enrichment-separation system of C18 and inductively coupled plasma-mass spectrometry (ICP/MS) with flow injection (C18-FI-ICP/MS) was established for the determination of trace heavy metals in seawater. The optimized experimental parameters including pH,C18 column,concentration of methol in elute and the velocity of elute were investigated. This system was applied to the online preconcentration , separation and determination of trace heave metals in seawater samples successfully. The detection limit (3σ) for Cd, Pb, Co, Ni and Zn was 0.03, 0.09, 0.1, 0.1, 0.3 μg/L respectively. The RSD of Cd, Pb, Co, Ni, and Zn for six measurements of seawater was 6.8%,3.4%,1.3%,2.6%,0.5%,respectively. The accuracy was verified with spiked recovery test. The recovery for five elements was 91.3%,95.1%,100.4%,100.3%,95.2%, respectively.
