1.The research development of LncRNA in colerectal cancer

With the progress of genetic studies,it has been found that long non-coding RNA( LncRNA) in many regulations which play a key role in development of tumor.Its own individual pieces can also have differ-ent adjustive effects;some LncRNA can even have different regulation and controlling effect in a variety of tumor. How the newly discovered LncRNA expression in colorectal cancer?This review mainly outlines the research de-velopment of LncRNA in colerectal cancer in brief.
2.Inhalation fluticasone propionate in treatment of children with cough variant asthma

Objective To observe and compare the clinical efficacy,recurrence rate and safety of differ-ent courses of inhalation fluticasone propionate in treatment of children with cough variant asthma. Methods A total of 101 children with cough variant asthma in our hospital were randomly divided into three treatment groups,all with the same therapy drug -fluticasone propionate and with the treatment time of two months, six months and nine months respectively. Clinical efficacy and treatment compliance were followed up during the treatment. After discontinuation,observation for the relapse of cough was followed up for 6 months. In inhalation therapy before and after,bone mineral density( BMD) and blood cortisol,electrocardiogram were monitored re-spectively to observe its safety. Results Three groups of children with cough were all get effective control,with no significant difference(H=5. 2894,P=0. 0710). The two months treatment group(A group)was more easy to relapse than the six months treatment group( B group) and the nine months treatment group( C group) after drug withdrawal,statistically significant(χ2 =4. 858 1,P=0. 027 5;χ2 =4. 482 6,P=0. 034 2). But after drug with-drawal,there was no significant difference in the rate of relapse in B and C group(χ2 =0. 022 2,P=0. 881 4). There were few changes in laboratory values about morning serum cortosol concentrations. There was no signifi-cant decrease in BMD after treatment. No other adverse events were observed in this study. Conclusion Flutica-sone propionate inhalation treating children cough variant asthma has good effection. A long period of treatment scheme(6 months)has better curative effect and lower recurrence rate after drug withdrawal.
3.Analysis on related factors of post-stroke depression

Objective To investigate the morbidity rate and risk factors of post-stroke depression in middle-aged and elderly per-sons with the stroke .Methods From March 2011 to March 2012 ,68 cases of stroke patients were followed up for 6 months after the first stroke .Hamilton Depression Scale-17 ,Activity of Daily Living Scale ,Eysenck Personality Questionnaire ,Scores of Neuro-logical Deficit and Family APGAR Index were used to investigate the morbidity rate and risk factors of PSD in 6 months after the attack of stroke .Results The morbidity rate of PSD was 36 .76% (25/68) in 6 months after the first stroke .Logistic regression a-nalysis indicated that the morbidity rate of PSD was without a clear relationship with age ,whether or not combined with basic disea-ses ,personality ,daily life ability and neurologic impairment .It was closely related with stroke position ,gender ,economy and family care .Conclusion The morbidity rate of PSD is considerable and the major risk factors of PSD are female ,left brain stroke ,poor e-conomic condition and lack of family care .
4.The relationship between EphB receptor and the development of tumor

Currently ,EphB receptor has been demonstrated as an important factor on the biological be -havior of normal cells .Some studies found that it plays an important role in the regulation of normal cell morpholo -gy,cell adhesion ,migration and angiogenesis .There is a certain relationship between abnormal expression of EphB receptor with a variety of tumor development .Visiblely,EphB receptor is important research direction in cancer studies,understanding the relationship between EphB receptor and tumor can better realize the progress of the tumor,and further to provide more scientific basis for cancer treatment and prevention .
5.Clinical observation of inhalation combined salmeterol plus fluticasone propionate aerosol in treatment of asthma in preschool children

Objective To study the effect of inhalation combined salmeterol plus fluticasone propionate in treatment of asthma in children between 4 ~ 5 years old.Methods Seventy-four children (age range,4 ~ 5 years) were enrolled for our study,they were randomly divided into 2 groups (one group inhaled combined salmeterol plus fluticasone,37 children;the other group inhaled fluticasone,37 children).Comparative studies of clinical efficacy,quantified symptom score,and regular follow-up observations were conducted at the interval of one week,one month,three months.Results In the first week and first month during the treatment,there were no difference between salmeterol plus fluticasone group and fluticasone group (P > 0.05).In the third month during the treatment,the daytime ratings(0.16 ± 0.09 vs 0.32 ± 0.11,P < 0.05) and night ratings(0.04 ± 0.01 vs 0.14 ±0.00,P <0.05) of salmeterol plus fluticasone group were significantly better than those of fluticasone group.After three months,more children in salmeterol plus fluticasone group were responsive to the decreased dose of the drug than those of fluticasone group (x2 =4.57,P < 0.05).Conclusion In the follow-up of exacerbatious,treatment with combined salmeterol and fluticasone propionate was as efficient as that of fluticasone in alleviating symptoms.During remission of asthma,combined salmeterol and fluticasone propionate was probably better than fluticasone for control of symptoms.
6.Clinical Application of Tidal Flow-volume Measurement and Its Impact Factors

Objective To study the clinical application and its impact factors of tidal flow - volume measurement in infants with bronchiolitis and pneumonia. Methods Thirty infants with bronchiolitis and thirty infants with pneumonia were studied. Tidal breathing flow- volume loops were obtained after the patients were sedated. The tidal volume per kilogram(VT/kg) ,the fraction of tidal volume ex-heled at peak tidal expiratory flow (PTEF) to total tidal volume (VPTEF/VE), and the fraction of exhaled time at PTEF to total expirato-ry time (TPTEF/TE) were measured. Results VT/kg was not significant different between the bronehiolitis group and the pneumonia group. VPTEF/VE and TPTEF/TE were lower in the bronehiolitis group than those in the pneumonia group. Conclusion The measure-ment of tidal breathing flow - volume loops is a good technology, which would be helpful to evaluate airway obstruction in wheezing in-fants. The impact factors should be controlled during the measurement. Otherwise, the results would be affected.
7.Diagnosis and surgical treatment of carotid body tumor

Objective:To investigate the diagnosis and therapy approaches of carotid body tumor (CBT).Method:Retrospective study of diagnosis and treatment of 10 patients with CBT was conducted.Results:Five patients were examined by B ultrasonic technique and all were not diagnosed;eight were examined by color Doppler flow imaging, six of whom were diagnosed as with CBT;six were examined by CT, three of whom were diagnosed;three was examined by digital subtraction angiography(DSA) and all were diagnosed;two were examined by magnetic resonance imaging(MRI) and one was diagnosed.There was no patient died during operation.One patient died 4 days after operation because of anoxemia in brain. Pathological examination after operation confirmed the diagnosis.Conclusion:Synthetical examinations should be taken to confirm the diagnosis of CBT. Color Doppler flow imaging and DSA are significant. Selection of operation methods should be based on the practice.
8.The Inhibitory Effect and Mechanism of Ester Catechin Monomers-EGCG, GCG on the Growth of Human Colon Carcinoma Cell Line SW480

Objective To investigate the inhibitory effect of ester catechin monomers-EGCG, GCG on the growth of human colon carcinoma cell line SW480 and its potential mechanism. Methods MTT assay, soft-agar colony formation test, Hoechst 33258 stain and flow cytometry analysis were used to determine the inhibitory effect of theasinesin(TS), and its monomers-EGCG, GCG on SW480 growth. Results EGCG, GCG and TS significantly inhibited the growth of human colon carcinoma cell line SW480 in dose-dependent manner, and their half inhibitory concentration (IC 50 ) was 108.88, 183.21, and 83.36?g?ml -1 , respectively. 24 hours after treatment with IC 50 of EGCG, GCG and TS, the colony formation rate of SW480 cells in the experimental group was obviously lower than that in the control group. Hoechst 33258 staining showed typical apoptotic features such as cell shrinkage, nuclear condensation and cell splinter in the experimental group. A subdiploid peak before G 0/G 1 phase in the experimental group was observed by flow cytometry. Conclusion In accord with TS, EGCG and GCG could inhibit the growth of colon carcinoma cell line SW480 cells, the mechanism of which may be related to inducing cell apoptosis.
10.High-level Expression of Foreign Genes In vivo and In vitro by Improved DNA-Based Replicon Vector Derived From Semliki Forest Virus

The design of DNA-based alphavirus vectors significantly improves the utility of these replicon vectors. The DNA-based replicon vectors can be used in expressing foreign genes and preparing RVP in virto efficiently, also in developing replicon vaccines and gene therapy vectors in vivo. The approach involved the conversion a RNA-based replicon vector into a layered DNA-based replicon vector by the RNA polymerase Ⅱ promoter and transcription termination/polyadenylation signal transcribed replicon RNA from DNA. When DNA-based alphavirus vector tranfected into cells, the first layer includes a eukaryotic RNA polymerase Ⅱ expression cassette that initiates transcription of RNA in nucleus. Following transport of this RNA from the nucleus to the cytoplasm, the second layer, autocatalytic amplification of the RNA vector corresponds to virus RNA replication cycle and results in high level expression of foreign gene. DNA and RNA-based bifunctional replicon expression vector pSCTA and helper vector pSHCTA were successfully constructed by replacing the SP6 promoter used in the original system pSFV1 and pSFV-helper2 derived from Semliki Forest virus (SFV) with CMV promoter and T7 promoter, and inserting BGH transcription termination and polyadenylation signal downstream 3′-untranslated region (UTR). In order to obtain DNA-based highly efficient replicon vectors, they were further modified to construct additional three DNA-based SFV replicon expression vectors and corresponding helper vectors. To investigate the efficiency of foreign gene expression level by the four different DNA-based SFV expression vectors and recombinant virus particle (RVP) prepared by cotranfecting with corresponding helper vectors, improved DNA-based replicon vectors pSCAR and pSHCAR derived from SFV were developed. high level protein could be generated using the new vector system by transfecting DNA into BHK21 cells and High titer of RVP produced by cotranfecting with helper vector. Antigen genes were also expressed in cells by the replicon expression vector. Additionally, reporter gene expression was observed in mice muscle following injection with SFV DNA vector. Anti-?-Gal antibody response and cell-mediated immune response were induced after intramuscular inoculation of the ?-Gal-encoding SFV replicon DNA. The results suggested that highly efficient DNA-based replicon vectors pSCAR and pSHCAR were constructed by modifying the SFV vectors. The improved DNA-based replicon vectors enhance the utility of them, and can be developed as potentially replicon vaccines and gene therapy vectors.
