1.EFFECT OF PHOSPHATE ON COLICIN V FORMATION BY STRAINS OF ESCHERICHIA COLI

ColV~+ strains of Escherichia coli produced larger inhibitory zones when these strains grown on nutrient agar containing phosphate after overlaid sensitive indicators. This appears that production of colicin V is increased by the addition of phosphate to nutrient agar. It was sure that stimulation of phosphate to colicin V formation results from its effect in reducing divalent cation levels in nutrient agar since adding EDTA to nutrient agar had the same effect as phosphate, but the addition of Mg~(2+) or Ca~(2+) had the oppsite effect. Therefore nutrient agar supplemented with phosphate can be used to isolate and identificate ColV~+ strains of E. coli.
2.Differences of spontaneous discharges of hippocampal pyramidal cells in rats of different age and memory groups

Objective To investigate the characteristics of spontaneous discharges of hippocampal pyramidal cells (PC) in rats in age and memory groups with the help of nonlinear theory. Methods Rats were randomly divided into age group and memory group. Age group included aged group (16-19 months) and young group (3-4 months) whereas memory group included good memory and poor memory groups in adult. Extracellular single cell recording was performed in vivo . Results No characteristics of the rhythm of spontaneous discharges of hippocampal pyramidal cells in distribution figure of interspike interval(ISI) were found, but the loss of complexity(C) and low percentage of favored patterns(PF) were found in age and poor memory groups. Conclusion The ISI complexity and the PF of the hippocampal pyramidal cells are correlated to age and memory, suggesting that the analysis of the complexity and favored patterns may be helpful for the discovery of the characteristics of the information coding in spontaneous discharges of hippocampal pyramidal cells.
3.Production and phenotype identification of specific expressed green fluorescent protein in male mice germ cells

Objective The aim of this study is production of organ specific animal model for studying reproductive toxicity in mice.Methods F1 generation was gotten by mating the Ddx4 -cre transgenic male mice with the Rosa26mT/mG transgenic female mice.F1 offspring and its parents phenotype was screened by molecular biological, histopathological and in vivo imaging technology.Results At molecular level, specific DNA fragment was only found in testis of F1 offspring; At the organ level, the expression of green fluorescent protein could only be observed in testis of F1 offspring; Testicular frozen sections and sperm fluorescence observation showed that green fluorescent protein were mainly expressed in the germ cell lineage such as secondary spermatocyte and spermatocyte and spermatozoon.Conclusions The production of the mice with specific germ cell expressed green fluorescent protein by Cre/loxP recombination system were built successfully.
4.Overexpression of HIV-1 P17 protein in Escherichia coli and its purification

Objective:To highly express HIV-1p17 in E.coli and purify the protein.Methods:①Recombinant plasmid was constructed by inserting HIV-1p17 gene amplified by PCR into plasmid vector,pET28c;②The recombinant plasmid was expressed in BL21,BL21(DE3),BL21(DE3) plysS and HMS174(DE3) of E.coli separately;③The target protein were purified with Ni-NTA resin;④The purified protein was detected by western blot and ELISA.Results:The expression of the P17 protein in BL21(DE3) represented up to 32% of total protein in E.coli,which was the most amounts compared with other kinds of E.coli.The purity of the purified protein reached 95%.The purified protein was recognized by HIV-1P17McAb as well as by HIV-1 positive serum.Conclusion:The recombinant plasmid is highly expressed in BL21(DE3) of E.coli that can be proceeded to the immunocompetence and the bioactivity research.The method of Ni-NTA resin is simple with low protein losing and high purity.And the purified p17 can be employed in early detection of HIV-1 infection and prediction of the clinical progression.
5.Analysis of 13 cases of low-pressure laparoscopic appendectomy during pregnancy

Objective To investigate the value of low-pressure laparoscopic technique for appendicitis surgery during early and mid-term pregnancy.Methods The clinical data of 13 pregnant women underwent low-pressure laparoscopic appendectomy from July 2007 to December 2010 were analyzed retrospectively.Results All the 13 cases were performed laparoscopic appendectomy successfully without laparotomy and cured completely before discharged from hospital No intestinal fistula,wound infection,abortion and other complications occurred.During follow-up,9 cases of full-term pregnancy patients gave birth to healthy babies,and there was no abortion and premature delivery on 4 patients still in their pregnancy.Conclusion For women at early or mid-term pregnancy with acute appendicitis,low-pressure laparoscopic appendectomy could reduce the stimulus of surgical operation on uterus and decrease the risk of abortion.
6.Characteristics and clinical significance of diffusing capacity in the patients with lung cancer.

BACKGROUNDTo explore the characteristics and clinical significance of diffusing capacity in the patients with lung cancer.
METHODSThe pulmonary diffusing capacity for carbon monoxide (DLCO) was measured with the rebreathing method in 138 patients with primary lung cancer, and 86 were performed pulmonary resection.
RESULTSThere was no significant difference in DLCOrb and DLCOrbc between the groups with different histological type and degree of ventilation impairment and general type (P > 0.05). DLCOrb/VA mildly decreased in the patients whose lung ventilation function was normal. DLCOc/VA decreased in the patients with restrictive ventilation dysfunction (P < 0.01), and DLCOrb/VA decreased in the patients with obstructive and mixed ventilation dysfuncion (P < 0.05). DLCOc/VA decreased in the patients with light and obvious lung dysfunction (P < 0.05). DLCOc/VA in the patients with central lung cancer was lower than that in the peripheral ones (P < 0.01). DLCOc/VA and D LCOrb in the group with postoperative respiratory failure were lower than that in the group without respiratory failure (P < 0.05). When DLCOc/VA of less than 80% and DLCOrb/VA of less than 70% were used to predict the postoperative respiratory failure, the correct ratios of dignosis and the diagnostic indexes were high.
CONCLUSIONSThe diffusing capacity decreases in the patients with lung cancer, and the main manifestation is the abnormal DLCOrb/VA and DLCOc/VA. The general type of lung cancer and the degree of pulmonary dysfunction may influence the diffusing function of the patients. DLCOc/VA of less than 80% and DLCOrb/VA of less than 70% should be selected for predicting postoperative respiratory failure.

BACKGROUNDTo explore the characteristics of exercise cardio-pulmonary function and its possible influencing factors in patients with lung cancer.
METHODSThe pulmonary function, ECG and exercise cardio-pulmonary function were measured in 198 patients with lung cancer and 20 healthy controls.
RESULTS1. Compared with healthy group, VO₂%P, VO₂/kg, AT, VO₂/HR%, VE and VT/VC significantly decreased in lung cancer patients with normal resting pulmonary ventilation, however, BR remarkably increased (P < 0.05 or P < 0.01). 2. In patients with normal resting pulmonary ventilation, there was no significant difference of exercise cardio-pulmonary function between the central and peripheral lung cancer groups. 3. The exercise cardio-pulmonary function was closely related to the TNM stages (P < 0.05 or P < 0.01). 4. W%, VO₂%P , AT and VO₂/HR% in patients with great vessel invasion were remarkably lower than those without great vessel invasion (P < 0.05 or P < 0.01).
CONCLUSIONSThe results suggest that exercise ventilation is impaired in lung cancer patients with normal resting ventilation. And the decrease of exercise cardio-pulmonary function may be related to TNM stage and to great vessel involvement.
9.Impacts of treating stratege for non-infarct-related artery on clinical prognosis in elderly patients with acute myocardial infarction after urgent percutaneous coronary intervention

Objective To investigate the impacts of treating stratege for non-infarct related artery on clinical prognosis in elderly patients with acute myocardial infarction (AMI) after urgent percutaneous coronary intervention (PCI). Methods From Augst 2007 to Augst 2010,a total of 75 elderly AMI patients (aged 75 years and over) were treated by urgent PCI and confirmed as multivessel coronary disease in our hospital. Among them,30 patients received medicine combined with PCI once again (PCI group) and 45 patients received only medicine treatemt (medicine group).The major adverse cardiovascular events (MACE) and results ot coronary angiography after treatment for average one year were compared between the two groups. Results There were no significant differences in the rates of in stent restenosis[1 case(2.2 %)vs.0 case],late loss of in-segment lumen [5 cases(11.1%)vs.3 cases(10.0%)],stent thrombosis[1 case(2.2%)vs.1 case(3.3%)] and larget vessel revascularization [2 cases (4.4 % ) vs.1 case ( 3.3 %) ] between medicine group and PCI group (x2=0.00,0.00.2.03 and 0.00,all P>0.05).The propertions of angina recurrence and second hospital admission for heart diseases were lower in PCI group than in medicine group [36 cases (80.0%)vs.14 cases(46.7%),18 cases(40.0%)vs.5 cases(16.7%),x2=9.00,4.61,P<0.01and P<0.05].However,no differences were found in the secondary heart failure,recurrent nonfatal myocardial infarction,severe arrhythmia,all- cause death and mortality rate of cardiovascular disease between the two groups (x2 =0.09,0.00,0.00,0.00 and 0.00,all P> 0.05). Conclusions Compared with single medicine therapy,the medicine combined with PCI for non- infarct-related artery may decrease the rates of angina recurrence and second hospital admission for heart diseases in elderly patients with AMI.
10.Evaluation of cardiopulmonary exercise test on predicting post-operative respiratory failure in patients with lung cancer.

BACKGROUNDTo evaluate the clinical significance of predicting post-operative respiratory failure in patients with lung cancer using cardiopulmonary exercise test (CPET).
METHODSBefore operation, 260 patients with lung cancer underwent CPET with incremental protocol. W%, VO₂%P, VO₂/kg, AT, MET, O₂ pulse, VTe, BF and VE were measured in the end of load exercise.
RESULTS(1) In patients after pneumonectomy, the values of the above indexes of CPET in the respiratory failure group were significantly lower than those in the non-respiratory failure group ( P < 0.05 or P < 0.01) except VTe. In patients after lobectomy, the values of 9 indexes of CPET in the respiratory failure group were similar to those in the non-respiratory failure group ( P > 0.05). However, when the patients after lobectomy were further divided into groups of upper and lower lobectomy, W% in the respiratory failure group was remarkably lower than that in the non-respiratory failure group after lower lobectomy ( P < 0.05). (2) Chi-Square test showed that abnormality of CPET indexes in different degrees was related to the morbidity of respiratory failure after pneumonectomy. Logistic regression showed that O₂ pulse < 80% and BF < 30/min correlated with the morbidity of post-operative respiratory failure. (3) For predicting post-operative respiratory failure, the sensitivity and specificity of VO₂%P < 60%, BF < 30/min, VE < 35 L/min were all more than 60% and their negative predictive values were all more than 90%.
CONCLUSIONSCPET is suitable to predict post-pneumonectomy respiratory failure. As a comprehensive index indicating cardiopulmonary function during exercise, VO₂%P < 60% should be selected to predict respiratory failure and evaluate indication of lung resection for patients with lung cancer.
