1.Autologous nucleus pulposus induces rat dorsal root ganglion damage without mechanical compression

BACKGROUND: Resent studies demonstrate that sciatica is caused by inflammatory reaction of nucleus pulposus, and dorsal root ganglion (DRG) is in involved in this process; however, its pathophysiological changes remain poorly understood. OBJECTIVE: To explore pathogenesis of sciatica related to nucleus pulposus protrusion without mechanical compression. METHODS: Totally 24 male SD rats were randomly divided into the experimental group and control group, with 12 rats in each group, The coccygeal intarvartebral discs of rats were opened to obtain 5 coccygeal nucleus pulposus, mixed with 50 μL physiological saline to prepare suspension. Rats in the experimental group were prepared non-compressive models by injecting suspension to the epidural cavum of rats. Meantime, the same volumes of physiological saline were injected into rats in the control group. The rat hind paw mechanical withdrawal thresholds were measured, pain-related behavior and dorsal root ganglion morphology were observed. RESULTS AND CONCLUSION: In the absence of mechanical pressure, disappearance of the nucleolus, degeneration of the nuclear membrane and significant hyperalgesia derived by auto-transplantation of nucleus pulposus to the epidural cavum of rats were observed. Dorsal root ganglion cells often had hyperemia, edema and intracytoplasmic vacuoles, with large and irregular nuclei. Prominent Nissl bodies were not seen. The Inflammatory reaction derived by autograft nucleus pulpcsus is an important factor in DRG injury and sciatica.
2.Medication and gene therapies for periprosthetic osteolysis

Aseptic loosening of the prosthesis due to periprosthetic osteolysis is one of the main causes that affect the service life following artificial joint replacement. The patient should undergo revision of artificial joints,but the surgery is of high difficulty,high risk and excessive cost,etc. Therefore,using nonsurgical technique to control artificial prosthesis osteolysis is an urgent and realistic task. Currently,the drug prevents osteolysis through inhibiting osteoclast bone resorption,responding to cytokines,and promoting bone formation. In addition,gene therapy and other treatments for osteolysis are under exploration.
3.Evolutionary Trace and Molecular Dynamics Simulation of the Beta2 Adrenergic Receptor Mutation-induced Activation

Beta2 adrenergic receptor (β2 AR) is one member of G protein coupled receptors (GPCRs), which is a key pharmaceutical target in the treatment of asthma. Evolutionary trace (ET) method was employed to analyze AR sequences and 44 conserved residues were identified. Then molecular dynamics (MD) simulation of the β2 wild-type receptor, D130N active mutant and D79N inactive mutant were carried out and tried to explore the structural/dynamic features characterizing functionally different states of the receptor,by means of investigating ET identified conserved basic residues in the wild-type receptor and its two mutants. Particularly, it was found that the departing of D130 from R131 of DRY motif and approaching to K149 are highly correlated with the receptor activation,and the movement of helix 2, 4 and 6 upon receptor activation is inferred from the observation that R151 and K270 interact with other residues in the receptor active state on the basis of little change of the side chain orientations. The results might provide further insights into the activating mechanism of β2 AR mutants, as well as the molecular bases of the diseases induced by the mutations of the receptor.
4.Effect of Fuming Tablet on ATP, MDA Content and SOD Activity in Retinal Tissue of Rabbit Retinal Detachment and Reattachment Model

Objective To observe the effect of Fuming Tablet on the content of ATP and MDA, and the activity of SOD in retinal tissue of rabbit retinal detachment (RD) and reattachment model. Methods 48 rabbits were randomly divided into 4 groups, with 12 in each. 12 rabbits were taken as healthy control and fed with normal saline, the others in model group, western medicine group and Fuming Tablet group were prepared as RD model by injecting Sodium Halyuronate (Helen) into subretinal spaces, and the detached retinas were reattached automatically in 10 ~ 14 days. They were administered with normal saline, mixed preparation of western medicines and Fuming Tablet suspension respectively, then the content of ATP, MDA and SOD activity were determined in postoperative 1 week (the retina remained detached) and 3 weeks (the detached retina reattached). Results Effect of fuming tablet of increasing the content of ATP and the activity of SOD, reducing the content of MDA was similar with that of the mixed preparation of ATP, VitB1 and Venoruton. Conclusion Fuming Tablet can significantly improve the oxidation resistance and depress the lipid peroxidation of retinal tissue, thus alleviate and inhibit the development of retinal pathological changes to some extent.

7.Experimerntal study of etanercept effects the wear debris-induced osteolysis

[Objective]To detect the effect of Etanercept on the Ti particles induced TNF-?IL-1 and IL-6 production by macrophage and to evaluate the validity of the etanercept on treatment of aseptic loosening of prosthesis.[Method]Separate and Cultivate mouse peritoneal macrophages were divide into 5 groups after 24 h.Group A was treated with M? alone,group B with M?+Ti particles,group C with M?+Ti particles + etanercept(10 ng/ml),group D with M? Ti particles + etanercept(100 ng/ml),group E with M? + Ti particles + etanercept(1000 ng/ml).After 18 h,the production of TNF-? IL-1 and IL-6 in culture supernatants was detected by ELISA.[Result]The levels of TNF-? IL-1 and IL-6 production were much higher in group B than those in group A,D,E(P0.05).[Conclusion]Is shows the Ti particles could stimulate M? to excrete profuse TNF-? IL-1 and IL-6 production,etanercept can significantly inhibited the production of TNF-? IL-1 and IL-6 secreted by Ti particles induced macrophages in a dose-dependent manner and hope to be a therapeutic candidate for the prevention of aseptic loosening.
8.Study on Legionella Contamination in Aerosols from Cooling Towers in Central Air Conditioning System

Objective To explore and describe the sampling methods for aerosol from cooling towers in the centralized air conditioning system, early-warning indicators, the effect factor and present status of contamination by Legionella.Methods Air-conditioning systems were sampled two rounds in 13 guesthouses, markest and bathing spots of Maanshan City in June-August, 2008.Aerosols were collected by fluid-striking microbial aerosol sample collector, while water samples from cooling tower and swelling box were obtained.Legionella were determined respectively for 67 samples by germiculture and polymerase chain reaction.Results The positive rate of Legionella in aerosols by germiculture was 0, meanwhile it was 50.0% by polymerase chain reaction for Legionella pneumophila.The positive rate in aerosols was 90.0% as the air-sampling volume exceeded 400 L.The positive rate of aerosols increased while the operating-time of cooling tower increased.The relative humidity in Legionella positive groups was higher than that in Legionella negative groups.Conclusion The positive result for Legionella pneumophila by polymerase chain reaction should be an important indicator for early-warning for the occurrence and spreading of Legionnaires' disease.The air-sampling volume of over 400 L is available by sampling collector for the detection of Legionella pneumophila by polymerase chain reaction.Risk of Legionella contamination in aerosol would be raised in high-humidity environment.
9.Self-provided well water solubility of total solid rapid determination method of research

Objective To establish a method for determination of total soluble solids (TDS) in self-provided well water .Methods The conductivity meter which has a function of determination of total soluble solid ,and was used to directly determinate the total soluble solids in self-provided well water .Results Compared with national standard method ,the results have no significant differ-ence .The relative standard deviation (RSD) was 0 .17% -0 .22% ,and the recovery rates of standard was 97 .5% -98 .6% .Conclu-sion The method is quick ,simple ,sensitive ,accurate and suitable for rapid determination of large quantities of waters .

Flavin-containing monooxygenase 3 (FMO3) is an important hepatic microsomal enzyme that oxidizes a number of drugs,xenobiotics and other chemicals.Many variants in the gene encoding FMO3 have been identified,some of which result in altered enzymatic activity,consenquently,altered substrate metabolism.Studies also implicate individual and ethnic differences in FMO3.Thus,it is anticipated that knowledge regarding functionally-relevant FMO3 genetic variability will become increasingly important for drug development and patient therapeutic choices.
