1.The effects of human Th2 cytokine on expression of intercellular adhesive molecules on bovin aortic endothelial cells

Objective To investigate the protective effects of Th2 cytokine (hIL-4, hIL-10, hIL-13) on the bovine aortic endothelial cells(BAECs) stimulated by tumor necrosis factor alpher(TNF-?). Methods BAECs were activated by TNF-?(4ng/ml), and a varied dose of Th2 cytokine(2, 5, 10, 20 and 40ng/ml) was used to incubate BAECs for 2h before stimulation with TNF-?, and then co-incubate for 6h or 18h. The expression of cellular adhesive molecules on BAECs was examined by the cellular enzyme-linked immunoabsorbent assay (Cell-ELISA), and BAECs viability was determined by MTT method. Results BAECs pre-treated with Th2 cytokine could down-regulate the expression of E-selectin and ICAM-1 induced by TNF-? in a dose-dependent manner(P
2.LOCALIZATION OF NMDA RECEPTORS IN THE VENTRAL HORN =OF SPINAL CORD OF ADULT RAT AND DURING POSTNATAL DEVELOPMENT

Objective To investigate the cellular localization and developmental patterns of NMDA receptor subunits(NMDAR1 and NMDAR2A/B) in the ventral horn of rat spinal cord during postnatal stages. Method Immunohistochemical staining method was used in the present study. Results NMDAR1-or NMDAR2A/B-immunorectivity is abundantly observed in the lamina Ⅷ and Ⅸ of ventral horn.The immunopositive product is localized mainly in the neuronal somata,dendrites and axon terminals.Dynamic changes of NMDA receptor expression occur in early postnatal stages.NMDA receptor subunits express weakly at postnatal 7 days (P7),goes up at P14,reaches maximum level at P21,then remains this level until adult. Conclusion Neuronal functional NMDA receptors are composed of NMDAR1 and NMDAR2A in heteromeric configuration in the ventral horn of spinal cord;NMDA receptors may be involved in motor neuron maturation and plasticity during postnatal development.;
3.The study of short-term curative effect for recurring NPC with the treatment of intracavitary brachytherapy after interventional chemotherapy

Objective To evaluate the clinical curative effect and methods for recurring NPC with treatment of intracavitary radiotherapy after interventional chemotherapy. Methods 32 cases of recurring NPC were divided into two groups, each of which consisted of 16 cases. Group one was treated with intracavitary chemotherapy plus intracavitary radiotherapy. On the third day after interventional chemotherapy, the first intracavitary radiotherapy was performed (5 Gy) and the second treatment was done next day. Group two was only performed with intracavitary radiotherapy. The dose and interval were the same. The time of re examination was two weeks later. Results In the first group, clinical symptoms were improved, local swelling lymph nodes were shrunk, secretion on the nasopharyngeal surface was hardly left or disappeared. Tumours were shrunk and atrophy. On the intracavitary radiotherapy group, clinical symptoms changed a little, except the secretion on the mass surface was reduced. The white blood counts of the two groups showed no difference and there were no functional changes of liver, kidney and digestive system in the interventional group. Conclusions It is a new and effective therapy to treat recurring NPC with interventional chemotherapy plus intracavitary radiotherapy.
4.RESPONSE OF ASTROCYTES IN CENTRAL VISUAL BRAIN REGIONS OF MONOCULARLY AND BINOCULARLY DEPRIVED RATS

Objective Monocular deprivation(MD) and binocular deprivation(BD) were used to examine the experience\|dependent structural plasticity of astrocytes in the central visual brain regions of young rats. Methods Pups eyelids were monocularly or binocularly sutured on postnatal day 7(P7) and maintained until 80?d,while an unoperated light experienced group was used for comparisions(L).Immunohistochemical ABC method was employed to investigate the immunoreactive changes of astrocytic bodies and processes for glial fibrillary acidic protein(GFAP) in the central visual brain regions. Results The results showed that the amount of astrocytic bodies and processes in the brain was decreased both in MD and BD groups compared to L group.The amount of GFAP\|positive immunoreactivity in the optic chiasma,optic tract,and contralateral central visual brain regions (including suprachiasmatic nucleus,lateral geniculate nucleus,occipital visual cortex,optic nerve layer of the superior colliculus and pretectal area) was significantly decreased in MD group.GFAP\|positive structures presented complementary distribution in bilateral visual cortex.For bilateral brain regions mentioned above,the amount of GFAP\|positive structures basically disappeared in BD group.There was,however,an increase in the olfactory cortex of GFAP immunoreactive in MD and BD groups. Conclusion\ The data suggested that the structure of astrocytes might be influenced by visual experience during development.
5.Human Interleukin-13 Affects Expression of Adhesive Molecules on Bovine Aortic Endothelial Cells

Objective To investigate the effects of human interlukin-13 (hIL-13) on the expression of E- selectin and intercellular adhesion molecule-1(ICAM-1) on bovine aortic endothelial cells(BAECs) stimulated by tumor necrosis factor alpha(TNF-?), and to provide experimental basis for hIL-13 inducing immunity endure and relieving the repulsion reaction of xenograft. Methods BAECs were co-cultured with different concentrations of hIL-13 for 2 h and followed by co-cultured with 4 ng/ml TNF-? for 6 h or 18 h. The expressions of E-selectin and ICAM-1 on BAECs were detected by Cell-ELISA. The effect of hIL-13 on activity of BAECs was detected by MTT colorimetry.Results BAECs pretreateded with hIL-13 could inhibit the expression of E-selectin and ICAM-1 induced by TNF-?, and showed a does-dependent manner from 5 ng/ml to 20 ng/ml of hIL-13 (P
6.Up-regulation of FasL expression by TNF-? and IFN-? in human colon cancer cells

Objective: To investigate the role of TNF-? and IFN-? in modulation of Fas ligand(FasL) expression in human colon carcinoma cells. Methods: A total of 6 human colon cancer cell lines were examined for FasL protein expression and for TNF-? and IFN-? modulation of FasL expression by Western blot method. Results: The results showed that FasL protein was expressed in all of these 6 cancer cell lines.FasL expression was up-regulated by TNF-? and IFN-? for 10 h in DLD-1 colon cancer cells; FasL expression was also up-regulated by TNF-? and IFN-? as well as PMA plus ionomycin for 10 h in SW620 cells, and further up-regulation was got by TNF-? and PMA plus ionomycin for 48 h in SW620 cells,whereas slight up-regulation was obtained by IFN-?. Conclusion: These data suggest that TNF-? and IFN-? may enhance FasL expression in tumor cells and may facilitate to escape from the immune surveillance of the host.
7.Functional expression of Fas ligand in human colon cancer cells

Objective: To investigate the expression and function of Fas ligand (FasL) in human colon carcinoma cells. Methods: A total of 6 human colon cancer cell lines were examined for FasL mRNA and protein expression, Fas-dependent cytotoxicity and its modulation were detected by using RT-PCR, Western blot and 3H-TdR release assay. Results: FasL mRNA and protein was expressed in all of these 6 cancer cell lines RPMI 4788, HCT-116, DLD-1, LoVo, WiDr and COLO 320DM. Fas-dependent cytotoxicity to hFas transfectant was exerted in part of these cancer cells, such as DLD-1, LoVo and WiDr. The cytotoxicity was significantly enhanced by PMA and ionomycin. Conclusion: These data suggest that functional FasL is expressed, at least in part, in human colon cancer cell lines and may help to escape from immune surveillance of the host.
8.Late-course accelerated hyperfractionated radiotherapy combined with chemotherapy for advanced esophageal carcinoma

Objective To evaluated the treatment results of late-course accelerated hyperfractionated radiotherapy combined with chemotherapy for advanced esophageal carcinoma. Methods 72 patients with advanced esophageal cancer were randomized into two groups. The late-course accelerated hyperfractionated radiotherapy combined with chemotherapy(LCAF+CT)group received the induction chemotherapy for two cycles, followed by conventional fractionation radiotherapy to a dose of 36 Gy, then changed into accelerated hyperfractionated radiotherapy to a total dose of 60 ~ 66 Gy. The conventional fractionation radiotherapy combined with chemotherapy(CF+CT)group received the chemotherapy that it was similar to LCAF+CT group, and used the conventional fractionation radiotherapy to a total dose of 60 ~ 66 Gy. Results The 1-, 3- and 5-year survival rates were 82.3 %, 51.6 %, 38.2 % in the LCAF+CT group, and 73.8 %, 31.5 %, 18.6 % in the CF+CT group. The differences of 3- and 5-year survival rates were statistically significant (P 0.05). Conclusion Late-coures accelerated hyperfractionated radiotherapy combined with chemotherapy might improve the treatment efficacy of advanced esophageal cancer and lengthen the suvival time. The toxic and side effects of LCAF+CT group were more severe than those of CF+CT group, but they were well tolerable.
9.Clinical significance of classification and grading of ovarian serous tumors and its relationship with the expressions of p53 and Ki-67 proteins

Objective:To explore the clinical significance of classification and grading of ovarian serous tumors (OST) and the correlation of classification and grading with expressions of p53 and Ki-67 proteins, so as to provide a basis for accurate diagnosis and reasonable treatment of OST.Methods:A total of 100 paraffin-embedded ovarian tissues were collected from Haikou Affiliated Hospital of Central South University Xiangya School of Medicine from January 2012 to November 2017. The classification and grading of OST was based on the World Health Organization (WHO) ovarian tumor tissue classification, the work conference on borderline ovarian tumors which was held in August 2013 in Maryland, United States, and the two-level tissue classification system of the United States MD Anderson Cancer Center. The 100 cases of ovarian tissues included 10 cases of normal ovarian tissues (NOT), 12 cases of ovarian serous cystadenoma (OSA), 18 cases of ovarian serous borderline tumor (OSBT), 22 cases of low-grade serous carcinoma (LGSC), and 38 cases of high-grade serous carcinoma (HGSC). The expressions of p53 and Ki-67 proteins in paraffin-embedded tissues were detected by immunohistochemistry. The clinical characteristics of patients with different types of OST and their relationships with the expression of p53 protein and Ki-67 positive index were analyzed.Results:Among OSBT, LGSC and HGSC groups, the proportion of patients with onset age > 50 years old [38.9% (7/18), 45.5% (10/22), 73.7% (28/38)], poor differentiation [0 (0/18), 0 (0/22), 100.0% (38/38)], stage Ⅲ-Ⅳ [5.6% (1/18), 27.3% (6/22), 39.5% (15/38)] increased sequentially, and the differences were statistically significant (all P < 0.05). Among NOT, OSA, OSBT, LGSC and HGSC groups, there were significant differences in the positive rate of p53 protein [20.0% (2/10), 25.0% (3/12), 27.8% (5/18), 31.8% (7/22), 57.9% (22/38)] and Ki-67 positive index [(10.40±0.00)%, (31.49±6.53)%, (42.81±6.84)%, (74.29±6.54)%, (77.04±8.88)%] (all P < 0.05). In the patients with ovarian serous carcinoma (LGSC + HGSC), there was no significant difference in the positive rate of p53 protein and Ki-67 positive index between the onset age ≤ 50 years old and > 50 years old groups (both P > 0.05), but there were significant differences between the patients with different tissue differentiation grade, clinical stage and metastasis (all P < 0.05). There was significant difference in Ki-67 positive index among NOT, OSA, OSBT, LGSC and HGSC patients with positive expression of p53 protein ( P < 0.01). Conclusions:The classification and grading of OST is related to onset age, tissue differentiation and clinical stage, which can be used to guide treatment and judge prognosis. The positive rate of p53 protein and Ki-67 positive index increase with advancing grade of breast lesions, which may be related to the occurrence and development of OST.
10.Clinical study of external beam radiotherapy combined with californiam-252 neutron after loading intracavitary radiotherapy in the treatment of patients With T1 and T2 nasopharyngcal carcinoma

Objective The results and side effects of nasopharyngeal carcinoma treated by combined external radiotherapy and Californium -252 neutron after loading intracavitary radiotherapy. Methods From November 2005 to March 2007, 30 nasopharyngeal carcinoma patients with staged T1 and T2 by 1992 Fuzhou staging system, were treated by external beam radiotherapy combined with Californium-252 neutron after loading intraeavitary radiotherapy. Results All patients were followed up for 2 years. 2 cases recurred in the nasopharynx. 4 cases developed distant metastases. 2 cases developed trismus. No perforations in hand and soft palate occurred. Conclusion External beam radiotherapy combined with Californian-252 neutron after loading intracavitary radiotherapy for nasopharyngeal carcinoma is indicated for boosting the dose to the nasopharyngeal cavity; reducing the dose for external irradiation. It can improve local control rate of nasopharyngeal carcinoma and reduce occur rate for the trismus.
