1.LDRf classification for ectopic varices in gastrointestinal tract

Objective To study the feasibility of LDRf classification for gastrointestinal tract ectopic varices (EcV) outside the esophagus.Methods Data of 914 patients with gastrointestinal EcV were classified by LDRf and analyzed for EcV location (L),vascular diameter (D),and risk factor (Rf).The etiology of the portal hypertension (PH) was determined,and the patients were treated and followed up.Results The EcVs were located in duodenum of 198 cases,in jejunum and ileum of 93,in bile duct of 105,in colon of 65,and in rectum of 453.Diameters of blood vessels of EcV varied from 0.3 to 3.5 cm.PH causes were cirrhosis with portal hypertention in 630 patients (68.9%),in which 3 were autoimmune cirrhosis and 3 were portal spongiform liver disease (0.6%).Combined esophageal and gastric varices were found in 252 cases (27.6 %),including 4 cases (0.5%) of splenectomy.Various treatments were applied in 315 patients,including endoscopic tissue adhesive injection in 43,endoscopic sclerotherapy in 76,endoscopic ligation in 74,interventional treatment in 52,and surgical laparotomy in 70.A total of 19 patients died of variceal bleeding.The patients were followed up for 13 to 36 months,no varices relapsed,and 1-year survival rate was 100%.Conclusion LDRf classification,simple,applicable,standardized,is suitable for the whole gastrointestinal varicose veins.
2.Comprison among curative effects of different transplantion of BMSCs on rabbit model of cirrhosis

Objective To investigate the effect of bone marrow mesenchymal stem cells (bone marrow stem cells,BMSCs) through different approaches in the treatment of transplanted cirrhotic model in rabbit effect difference.Methods Establishment of rabbit model of carbon tetrachloride cirrhosis with liquid olive oil mixture of subcutaneous inj ection and ethanol fed,the liver cirrho-sis models were randomly divided into cirrhosis model group (n=10),hepatic artery transplantation group (n=10),ear vein trans-plantation group (n=10).Healthy New Zealand rabbits bone marrow,using the method of whole bone marrow adherent cultured bone marrow BMSCs,and then were transplanted and serum albumin,alanine aminotransferase in 3 ways;by HE staining,the de-gree of hepatic cirrhosis and detection of serum albumin by Western blot expression in liver.Results Through the vein group and ar-tery transplantation group BMSCs to cirrhotic model in rabbit,4 weeks after the discovery of liver transplantation group rabbits were significantly improved,serum albumin is liver cirrhosis without transplantation group increased significantly(P<0.05),transami-nase is liver cirrhosis without transplantation group decreased significantly (P<0.05).The therapeutic effect of 2 ways of BMSCs transplantation on rabbit liver cirrhosis had a significant difference (P<0.05).Conclusion Transplantation of BMSCs through the ear vein and the hepatic artery of rabbit after 4 weeks of treatment of liver cirrhosis,liver function can be improved.But the hepatic artery transplantation therapy is obviously better than that of intravenous transplantation of treatment effect.
3.Effects of different vascular diameter and pressure on complete ligation degree in vitro

Objective To explore the influence of different pressures and diameters in vitro on complete ligation degree of esophageal varices based on the principle of LDRf typing.Methods According to the different preset venous pressure,the experimental porcine veins were used to establish vascular pressure models and divided into three groups (group A 25-30 cmH2O,group B 35-40 cmH2O,group C 45-50 cmH2 O).The porcine esophagus was selected to establish models of esophageal varices,and divided into three groups according to the diameter of esophageal varices (group D1 0.4-1.0 cm,group D2 1.0-1.5 cm,group D3 1.6-2.0 cm).Influence of pressure and diameter on complete ligation degrees was evaluated by univariate and multivariate analysis.Results Group A completed 18 ligations (56.25%,18/32),group B 12 ligations (37.50%,12/32),and group C 11 ligations (33.33%,11/33),which were not significantly different by pressure (x2 =3.6126,P =0.0573).Group D1 completed 35 ligations (94.59%,35/37),group D2 6 ligations(16.67%,6/36),and D3 completed no ligation (0%,0/24),which were significantly different by diameter (x2 =38.0014,P =0.0000),indicating that the diameter of the varices exerted greater influence on the ligation degree.The logistic regression analysis showed that variceal pressure and diameter were the independent factors for complete ligation degree (P =0.000).Conclusion Complete ligation is effective and safe when the variceal diameter is 0.4-1.0 cm,or variceal pressure is 25-30 cm H2O.Endoscopic treatment for varices with the guide of LDRf typing is scientific and feasible.
4.The influence of venous pressure on porcine variceal ligation—an experiment in vitro

Objective To explore the influence of porcine esophageal variceal pressure on complete ligation for polycyclic ligator in vitro.Methods The experimental porcine venous vessels were selected to make the models of different venous pressure,which were divided into 3 groups according to the preset pressure range,P1 group 25-30 cmH2O(1 cmH2O =0.098 KPa),P2 group 35-40 cmH2O,P3 group 45-50 cmH2O.Ligation effects of each group were analysed.Results There were 75 complete ligations (47.77%,75/157) in group P1,41 in group P2 (32.28%,41/127),and 29 in group P3 (23.58%,29/123).There were significant differences among the three groups (x2 =19.558 5,P =0.000 6).Conclusion The higher the variceal pressure is,the worse the effect of ligation is.Variceal pressure could predict the effect of endoscopic ligation,and is helpful for the choice of endoscopic treatment.

Objective To investigate the changes of carotid, lipidemia and fibrinogen in type 2 diabetes mellitus patients complicated with cerebral infarction (DCI). Objective To explore the effect of non-directed musical therapy on the automatic thoughts and therapeutic effect of depression.Methods 60 people outpatient and inpatient who with depression disorder participated in this research.According to the medical order,we choose the people who with Cardinality order as the group of the musical therapy (study group) and the people with the even order as the group of drug therapy (control group).all the patients was test with the Hamilton's Depression Scale (HAMD-24) and Automatic Thoughts Questionnaire (ATQ) before and after 2,4,8 weeks treatment,and comparison of these questionnaires scores of the two groups.Results After the 2,4,8 weeks treatment,the HAMD scores of the study group was (15.70±2.63),(10.03±2.28),(6.00± 1.98) and the scores of control group was (17.43±2.62),(12.47±2.75),(7.50±2.03).The result of HAMD scores had significant difference(F=11.754,P=0.001).ATQ scores of the study group was (73.27± 17.60),(58.33± 13.66),(47.43± 11.94) and the scores of control group was(84.20±22.30),(72.53±20.86),(58.90± 18.61).The result of ATQ scores had significant difference(F=4.136,P=0.047).Conclusion Non-directive musical imagery could be an effective way to improve the automatic thoughts of people who with depression disorder and improve the effect of treatment.
7.Genetic detection of Plasmodium falciparum with Chelex-extracted DNA from thin blood smears

Objective To investigate the feasibility of Chelex DNA extraction from thin blood smears for genetic analysis, and to develop smear-based nested polymerase chain reaction (PCR) of the 18S RNA of Plasrnodium falciparum. Methods Chelex-100 which was chelating ion exchange resin was used to extract DNA from Giemsa-stained or unstained thin blood smears of different concentrations of Plasmodium falciparum. With the extracted DNA as the template, 18S RNA gene was amplified by nested PCR to test the susceptibility of Chelex method. Results Positive band of 205bp appeared in nested PCR with DNA extracted from Giemsa-stained or unstained thin blood smears of patient with falciparum malaria. Using the Chelex method, the detection limits of the smear-based nested PCR were 1.5 × 101 parasite/μL blood for Giemsa-stained and 1.5×10-1 parasite/μL blood for unstained thin blood smears. Conclusions Chelex DNA extraction is a simple and efficient method for extracting trace amount of DNA from thin blood smear. The smear-based nested PCR developed in this study is feasible to identify the gene from reserved thin blood smears and will provide a new approach for clinical diagnosis and study of molecular epidemiology.
8.Clinical characteristics of 745 cases of allergic conjunctivitis

Background Allergic conjunctivitis is common in clinical practice.But some patients,especially pediatric patients,are not diagnosed and treated correctly because of different classifications and other associated symptoms.A comprehensive understanding of the clinical characteristics of allergic conjunctivitis helps to correct diagnosis and management.Objective This study tried to analyze the clinical characteristics of allergic conjunctivitis.Methods A descriptive study was carried out.The clinical data of 745 cases of various classifications of allergic conjunctivitis who received treatment in Beijing Tongren Eye Center from April 2011 to March 2015 were retrospectively analyzed.The general status,disease history,family medical history,personal allergy history,medication history,disease course,attacked duration,affecting factors of symptoms,other non-ocular allergic diseases,other associated eye disease,the findings of the anterior ocular segment,treating outcomes were summarized and the correlation of age with symptoms was evaluated.Results In the 745 cases,75.44% cases (562/745 were seasonal allergic conjunctivitis (SAC) and perennial allergic conjunctivitis (PAC),21.88% cases (163/745 were vernal keratoconjunctivitis (VKC),2.42% cases (18/745) were giant papillary conjunctivitis (GPC),and 0.27% cases (2/745) were atopic keratoconjunctivitis (AKC).The number of patients was most in July (180,24.16%) and least in December (26,3.49%).The itching was found in 53.02% patients,redness in 36.64% patients,blinking in 21.88% patients.In the patients,58.26% patients were ≤ 14 years,34.09% patients were 15-39 years and 7.65% patients were ≥40 years,and a significant difference in gender was found in various age groups (x2 =89.431,P =0.000).The co-morbidities appeared to be allergic rhinitis,eczema,urticaria,allergic purpura and asthma,and the ocular coexisting conditions included dry eye,refractive error,blepharitis and meibomian gland dysfunction,corneal epithelial disorders,triehiasis and anterior uveitis.Logistic regression analysis revealed that blinking patients were more and dry eye,foreign body sensation and pain were less in ≤ 14 years patients than those in the >14 patiens (3=-2.039,P =0.000;3 =1.585,P=0.001;3=1.174,P=0.003;3=1.218,P =0.037).The symptoms of allergic conjunctivitis were relieved by the administration of dual-acting antiallergic agent/antihistamine drug,glucocorticoid or immunosuppressor agents.Conclusions SAC and PAC are more common classifications of allergic conjunctivitis,followed by VKC.July is a high-occurrence duration and clinical symptoms are diverse and often accompanied by other allergic diseases.The administration of dual-acting antiallergic agent/antihistamine drug,glucocorticoid or immunosuppressor agents is an effective approach to the management of allergic conjunctivitis.

10.Analysis of Cellulase Regulator ACEII Interaction with cbh1 Promoter Fragment in Trichoderma koningii

ACEI,ACEII and Xyr1 are transcriptional factors that regulate cellulase gene expression in Trichoderma koningii.In vitro experiments have shown that ACEI and Xyr1 bind to the cbh1 promoter fragment(-304 to-18) and regulate the gene transcription.However,whether ACEII binds to this 287bp fragment is still unclear.To further elucidate the regulatory mechanism of ACEII for cellulase,DNA-binding domains of ACEII from T.Koningii were expressed in E.coli.It could not show binding to the cbh1 promoter fragment(-304 to-18) by electrophoresis mobility shift assays,suggesting that it is Xyr1 but not ACEII binds playing an essential role during induction of cbh1 gene transcription.
