Objective To investigate the effect of skull acupuncture on cerebral infarct volume and plasma SOD activity and MDA and NO contents. Method Forty-eight male SPF-grade SD rats were randomized into sham operation, model and skull acupuncture groups. A rat model of middle cerebral artery occlusion (MCAO) was made by intraluminal thread occlusion. Rat nerve function was assessed by Zea Longa neurological function scoring. Rat cerebral infarct volume was measured by TTC staining. Plasma superoxide dismutase (SOD) activity and malondialdehyde (MDA) and nitric oxide (NO) contents were measured by colorimetry. Result Rat neurobehavioral score was significantly lower on day 1, 3 and 7 in the model group than in the sham operation group (P<0.01). TTC measurement showed that cerebral infarct volume was significantly smaller in the skull acupuncture group than in the model group (P<0.01). Plasma SOD activity was lower and MDA and NO contents were higher in the model group than in the sham operation group (all P<0.01). Plasma SOD activity increased and MDA and NO contents decreased after skull acupuncture treatment compared with the model group (all P<0.01). Conclusion Skull acupuncture has a protective effect against cerebral ischemia/reperfusion injury and can increase plasma SOD activity and decrease plasma MDA and NO contents in rats with cerebral ischemia/reperfusion injury.

[Objective]To evaluate the effects of sutureless technique in comparison to conventional techniques for repair of total anomalous pulmonary venous connection(TAPVC)with the method of propensity score analysis.[Methods]From October 2007 to December 2013,179 consecutive patients were enrolled in this study. Patients were operated with sutureless technique(n = 81)or Conventional technique(n=98),and followed up at an interval of 1 month,3 months,6 months and then once a year post-operation. During analysis, three type of propensity-score matching methods, including nearest neighbor caliper matching,Mahalanobis metric matching with propensity score ,optimal full matching were used to create balanced groups of patients receiving each treatment. Surgeons’performance difference was assessed with random frailty proportional hazards models with gamma. Composite endpoints was defined by postoperative death or late death or postoperative pulmonary venous obstruction(PVO),which was evaluated with Kaplan-Meier curve and multivariable Cox proportional hazard model, adjusted by Preoperative-PVO,age,gender,weight and TAPVC type.[Results]Nearest neighbor caliper matching method was the best choice during propensity score analysis. After matching ,sutureless group included 73 patients and Conventional group73 patients. In sutureless group, cardiopulmonary bypass(CPB)time(Z=2.18, P=0.030),cross-clamp time(Z=3.63,P<0.001),rate of composite endpoints(HR 95%CI=0.20(0.06~0.61),P=0.005),late death(HR 95%CI=0.03(0.01~0.55),P=0.017)were significantly better than that in Conventional group. In subgroup analysis ,for patients with pre-PVO,decreased composite endpoints was seen in sutureless group.[Conclusion]Comparison using thepropensity score analysis demonstrated that sutureless strategy for primary repair of TAPVC may associate with decreased mortality rate of post-PVO and CPB time and cross-clamp time.

ObjectiveForskolin (FSK) analogs, isoforskolin (isoF),deacetylforskolin(deaF),and 1-acetylforskolin(1-aF),extracted from Coleus forskohlii native to Yunnan, were assayed for their adenylate cyclase stimulating activities in vitro and for effects of two analogs on ocular hypertension (OHT) in water-loaded rabbits.MethodsAdenylate cyclase stimulation was determined by protein-binding method of radioimmunoassay, and intraocular pressure was monitored by pneumatonometer.ResultsIt showed that isoforskolin and forskolin stimulated adenylate cyclase in vitro with almost equal activity, deacetylforskolin with milder activity, and 1-acetylforskolin with little activity in vitro.1% deaF and 1-aF suppressed rabbit OHT induced by water-loading for at least 3h,with the maximal inhibitory rates of 6.0,10.9% respectively.ConclusionThis study suggests that two foskolin analogs (isoforskolin, deacetylforskolin) possess adenylate cyclase stimulation activities in vitro;deacetylforskolin and 1-acetylforskolin suppress OHT induced by water-loading in rabbits.

Objective To establish a rabbit model of radiation-induced skeletal muscle injury in order to study the ultrastructural pathological changes and underlying mechanism. Methods 28 New Zealand rabbits were randomly divided into 2 groups with 16 rabbits in experimental group and 12 rabbits in control group. The experimental rabbits were irradiated on hip with a single dose of 80 Gy of 9 MeV electrons from a linear accelerator.1 month and 6 months after irradiation the pathological changes were respectively observed under light microscope and electron microscope. Results One month after irradiation, the morphologic changes including degeneration, necrosis of muscle cells, and hemorrhage between the muscle cells were observed under light microscope and the swelling of myofibrillae, blurring of light and shade band, vacuolar degeneration of mitochondria and amorphous areas of necrosis were observed under electron microscope.Six months after irradiation, the morphologic changes of nucleolus chips, fibrous connective tissue, thickening of vascular wall and vascular congestion between the muscle cells and the amorphous areas of necrosis in the experimental group were much more serious than those of 1 month after irradiation. In addition, the myofilaments were lost in degeneration areas and the sarcomere became shorten.Observation with electron microscope showed that the mitochondrial size and its morphological changes were varied and the amounts of collagen between myofibrillaes were increased 6 months after irradiation. Conclusions A rabbit model of high-dose irradiated skeleton muscle injury was successfully established with a single dose of 80 Gy of 9 MeV electrons from a linear accelerator. The degeneration and necrosis of muscle cells may be promoted by mitochondrial and vascular injury, degeneration of vessel and nerve fiber.

Objective:To analyze the impact of previous exposure to macrolide, quinolones and nitroimidazole antibiotics on eradication rate of bismuth quadruple therapy (BQT) in newly diagnosed patients with Helicobacter pylori( H. pylori). Methods:A total of 469 patients with H. pylori initially treated at the Third Hospital of Peking University from September 2017 to August 2020 were retrospectively recruited. The therapeutic regimens were BQT containing clarithromycin/levofloxacin/metronidazole recommended by Chinese guidelines. Clinical data were collected, including general demographic data, exposure history of antibiotics, CYP2C16 metabolic pattern, endoscopic diagnosis, bacterial density, H.pylori resistance, eradication results, etc. Univariate analysis, Chi-square test, Fisher exact probability test, Kruskal-Wallis H test and Logistic regression model were used as statistical methods. Results:Among different eradication therapies, univariate and multivariate analyses suggested that previous exposure to macrolides ( OR=3.37,95 %CI 1.04-10.98, P<0.05) was relevant to the decreased eradication rate of BQT containing clarithromycin. This may be due to increased resistance to clarithromycin ( OR=6.12,95 %CI 3.99-9.40, P<0.01).The previous exposure to quinolones ( OR=3.65, 95 %CI 1.27-10.49, P<0.05) was relevant to the decreased eradication rate of BQT containing levofloxacin, which was probably explained by the increased resistance to levofloxacin ( OR=2.50, 95 %CI 1.69-3.71, P<0.01). But the previous history of nitroimidazole did not impact the efficacy of BQT containing metronidazole. Conclusions:In patients newly diagnosed with H.pylori infection, the previous exposure to macrolide or quinolones antibiotics is related to lower eradiation rates of H. pylori. Although the exposure to nitroimidazole also indicates drug resistance to metronidazole, the clinical efficacy of BQT with metronidazole 400 mg four times a day is not affected.
