Objective:To study correlation analysis between Ki-67 and p53 expression in breast cancer tissues and preoperative neoadjuvant chemotherapy (NAC).Methods:From January 2014 to September 2015 in our hospital,73 cases of breast cancer patients treated with NAC were selected as the research object.NAC was administered to all patients for 3 weeks, and then the surgery was performed, and the biopsy specimens were performed before and after chemotherapy.Analysis of the effect of the patients and the expression of p53 and Ki-67.Results:In the 73 patients,CR had 24 cases, accounting for 32.88%,PR had 39 cases, accounting for 53.42%,OR had 63 cases, accounting for 86.30%.The results of pathological grading showed that grade 5 were 13cases,grade 4 in 26 cases, grade 3 in 22 cases, grade 2 in 9 cases, grade 1 in 3 cases. There was no significant difference in the expression of p53 in all patients before and after chemotherapy(P>0.05).However,the expression of Ki67 was significantly lower than that before chemotherapy, and the difference was statistically significant (P<0.05).The percentage of p53 positive expression in OR patients after chemotherapy was 34.92%,significantly lower than that of the negative expression of 65.08%,and the positive expression of Ki-67 was 71.43%,significantly higher than that of negative expression of 28.57%,the difference was statistically significant (P<0.05).According to Pearson analysis, the negative expression of p53 in breast cancer tissues was positively correlated with the efficacy of chemotherapy(r=0.689,P<0.06);Ki-67 positive expression was positively correlated with the efficacy of chemotherapy (r=0.714,P<0.05).Conclusion:The expression of p53 and Ki-67 in breast cancer tissues and its effect on NAC were closely related to the preoperative, and it can be used to determine the effect of NAC by monitoring the expression of p53 and Ki-67.

OBJECTIVE To establish and apply a method for simultaneous determination of the contents of dicentrine, protopine and coptisine in Tibetan Corydalis pallida of different origins, and to provide reference for origin determination and quality control of the kind of medicinal materials. METHODS Ultra performance liquid chromatography-tandem quadrupole mass spectrometry method was used. The determination was performed on Agilent EC-C18 column (100 mm×2.1 mm, 2.7 μm) with mobile phase consisted of acetonitrile-0.1% formic acid by gradient elution. The flow rate was 0.2 mL/min, and the column temperature was set at 35 ℃ . MS detection was carried out by electrospray ionization in positive modes, multiple reaction monitoring mode was used for quantitative analysis. RESULTS The injection mass concentrations of dicentrine, protopine, coptisine ranged from 5.88 to 117.60, 53.70 to 1 074.00, and 4.85 to 97.00 ng/mL, respectively, showing a good linear relationship with their respective peak areas (r=0.998 2, 0.991 9, and 0.999 6, respectively). The limits of quantitation were 2.35, 1.07 and 1.46 ng/mL; the limits of detection were 1.17, 0.54, 0.49 ng/mL, respectively. RSDs of precision, stability (24 h) and repeatability tests were all lower than 2.0%. The average recovery rates were 97.41%, 98.89% and 105.44%( all RSDs<5.0%, n=6). CONCLUSIONS The established method has good selectivity and high accuracy, and is suitable for the rapid analysis of dicentrine, protopine and coptisine in Corydalis. The total contents of three alkaloids in different original medicinal materials are from high to low in order of C. chrysosphaera, C. mucronifera, C. pygmaea, C. hendersonii and C. conspersa. The alkaloid contents in C. chrysosphaera and C. mucronifera are relatively similar, but no dicentrine has been detected in C. conspersa and C. hendersonii.

Aortic stenosis (AS) is the most common primary valve lesion requiring surgery or transcatheter intervention in modern era. Its prevalence is rising rapidly as a consequence of the aging population. Transcatheter aortic valve replacement (TAVR) as a therapy option for older high-risk symptomatic severe AS patients has emerged and is currently extending its indications towards surgery intermediate- and low-risk subjects. Considering the common characteristics of frailty and high comorbidity among AS patients, cardiac rehabilitation (CR) has been proven to improve not only survival but also quality of life in previous reports. CR as a classⅠ recommendation in guidelines for the prevention and treatment of cardiovascular disease has been widely used in clinical practice. The purpose of this article is to sort out the current CR programs for TAVR patients in global medical management, and explore the CR optimization program fit for China medical model in post COVID-19 pandemic era.
