1.The relationship between the anxiety and depression of parents of children with congenital heart disease after surgery and family cohesion and adaptation

Objective To investigate the relationship between family cohesion and adaptation and the anxiety and depression level of parents of children with congenital heart disease after surgery.Methods 302 parents of children with congenital heart disease and 330 parents in child health care clinic were conveniently selected and investigated with Hospital Anxiety and Depression Scale (HADS).And Family Cohesion and Adaptation Questionnaire (FACES Ⅱ-CV) was applied to investigate the former.Results The mean score of the anxiety and depression level of parents of children with congenital heart disease was higher than that of the parents in child health care clinic.The difference was significant.The score of anxiety and depression of parents of children with congenital heart disease was significantly correlated with dissatisfaction of family coherence and adaptation; while negatively correlated with real family cohesion and adaptation.Multiple linear regression showed that real family adaptation,dissatisfaction of family adaptation were influential important factors of parents' anxiety.Dissatisfaction of family adaptation was also the influential important factors of the parents' depression.Conclusions The anxiety and depression of parents was above the average level,we must pay more attention to improve it,make full use of the function of families,and to improve their anxiety and depression condition.
2.The relationship between positive experience and perceived social support of parents with a cerebral palsy ;child

Objective To investigate the relationship between positive experience and perceived social support of the parents with a cerebral palsy child. Methods 105 parents with a cerebral palsy child who were hospitalized in the Third Affiliated Hospital of Zhengzhou University between November , 2014 and January , 2015 were conveniently selected and investigated about their positive experience and perceived social support with general information questionnaire, positive aspects of caregiving and perceived social support scale. The correlation between the two was analyzed. Results The mean score of the parent′s positive experience was (30.82 ± 5.26); the dimension of self affirmation was (16.71 ± 3.06) and the dimension of life expectation was (14.10 ± 2.85). The scores of the parents′ positive experience were significantly different among gender, education of parents and the age,gender of children. The score of the parents′ positive experience was positively correlated with the total score of perceived social support (r = 0.214, P = 0.029) and the score of other support (r = 0.227, P = 0.020). Conclusions The parents′ positive experience was medium of the high level, which is worth of more attention in order to improve it. The higher perceived social support score is , the more positive experience parents will have.
4.Effect of Exercise on the Interleukin-1 beta(IL-1β) Expression in Muscle Tissue of Insulin-Resistant Rats

Objective To investigate the effect of swimming exercise on insulin sensitivity and interleukin-1 beta(IL-1β)mRNA expression in muscle tissue of insulin-resistant (IR) rats induced by high-fat diet.Methods The IR rat model was induced by high-fat diet feeding for 10 weeks and assessed by hyperinsulinemic-euglycemic clamp technique (HEC). The rats were divided into the fllowing three groups: normal group without exercise, IR groups with and without swimming exercise. After 4 weeks swimming excersie, insulin sensitivity and serum IL-1β levels were measured.Results Glucose infusion rate (GIR) of the IR rats was significantly elevated than that of non-exercise IR rats (P<0.01). Moreover, serum IL-1β levels were reduced markedly in IR rats (P<0.01). Conclusion High-fat diet feeding can partly reverse IR of rats to normal value. Swimming exercise improves insulin sensitivity and reduces the levels of IL-1β in IR rats induced by high-fat diet.
5.Three kinds of cell factors on the expression of integrinαvβ6 in the vaginal epithelial cell line VK2/E6E7

Objective Few researches are reported on the association of integrinαvβ6 with vaginal mucous infection and de-fense.This study aimed to investigate the effects of IL-6, TGF-β, and IFN-γon the expression of integrinαvβ6 in the vaginal epithelial cell line VK2/E6E7. Methods Immortalized human vaginal epi-thelial cells (VK2/E6E7) were cultured in vitro and treated with gra-dient concentrations of IL-6 (1, 10, 50, and 100 ng/mL), TGF-β(0.1, 1, 10, and 100 ng/mL), and IFN-γ(50, 500, 2500, and 5000 U/L) , respectively.After 48 hours, the cells were collected and total RNA extracted by the Trizol method to be reversely tran-scribed to cDNA. The expressions of integrin αand β6 subunit mRNA were detected by real-time quantitative PCR. Results In the IL-6-treated VK2/E6E7 cells, the integrin αand β6 subunit mRNA expressions were significantly lower in the 1 ng/mL and 10 ng/mL groups but remarkably higher in the 100 ng/mL group (1.14 ±0.12 and 1.37 ±0.25) than in the blank control (1.00 ±0.09) (P<0.05), and only the expression ofβ6 subunit mRNA was elevated in the 50 ng/mL group (P<0.05), with the expressions increased in a concentration-dependent manner.In the TGF-βtreated VK2/E6E7 cells, the expressions of integrinαandβ6 subunit mRNA were significantly lower in the 0.1 ng/mL and 1 ng/mL groups but remarkably higher in the 100 ng/mL group than in the blank control (1.00 ±0.09) (P<0.01), and only the expression ofβ6 subunit mRNA was elevated in the 10 ng/mL group (4.31 ±0.78, P<0.01), with the expressions increased in a concentration-dependent manner.In the IFN-γtreated VK2/E6E7 cells, the expressions of both integrinαandβ6 subunit mRNA were significantly lower in the 50 U/L, 500 U/L, 2500 U/L, and 5000 U/L groups than in the blank control (all P<0.01). Conclusion IL-6 and TGF-βhave an inhibitory effect the expression ofαvβ6 in VK2/E6E7 cells at low concentrations, which gradually diminishes with the increased concentration of inflammatory factors.In the early stage of inflammation, IFN-γcan effectively suppress the expression ofαvβ6 at a high concentration.However, with the progression of inflammation and decrease of its concentration, IFN-γloses its inhibi-tory effect and therefore does not help inflammation control.
6.Study of lymphocyte subsets and its activated-molecules in patients with paroxysmal nocturnal hemoglobinuria.

OBJECTIVETo investigate the relationship between subsets of lymphocytes and between its activated status and the clinical manifestations in patients with PNH, and to unfold immunological mechanism in the pathogenesis of PNH.
METHODSThe peripheral blood mononuclear cells (PBMNC) from 18 PNH patients and 20 controls were separated into two subpopulations using anti-CD(59) monoclonal antibody combined with goat-anti-mouse IgG immunomagnetic beads. CD(3)(+), CD(4)(+) and CD(8)(+) lymphocyte subsets were detected by flow cytometry. In 6 newly diagnosed patients, phenotypes associated with T cell activation such as CD(28)(+)/CD(4)(+) or CD(8)(+) cells, CD(8)(+) CD(38)(+) cells, and HLA-DR(+)/CD(4)(+) or CD(8)(+), and NK (CD(3)(-) CD(16)(+)) cells were detected in the peripheral blood.
RESULTPatients with PNH showed significantly increased CD(3)(+) CD(8)(+)/CD(3)(+) CD(4)(+) ratio as compared with controls (1.22 +/- 0.51 vs 0.86 +/- 0.27, P < 0.05), and the CD(3)(+) CD(8)(+)/CD(3)(+) CD(4)(+) ratio in CD(59)(-) PBMC was higher than that in CD(59)(+) PBMC (2.31 +/- 1.56 vs 0.62 +/- 0.27, P < 0.05). The ratios of CD(4)(+) CD(28)(+)/CD(4)(+) markedly decreased and CD(8)(+)HLA-DR(+)/CD(8)(+) increased.
CONCLUSIONPatients with PNH appear to have abnormalities in their lymphocytes. Increased ratios of CD(3)(+) CD(8)(+)/CD(3)(+) CD(4)(+) and HLA-DR(+) CD(8)(+)/CD(8)(+) lymphocytes as well as declined ratio of CD(4)(+) CD(28)(+)/CD(4)(+) lymphocytes might be involved in the pathogenesis of PNH.

7.Application of improved PCNN algorithm in retinal macular edema segmentation.

In order to extract the outlines of macular edema from OCT images of macular, and estimate the volume of edema, we have to accurately segment the macular edema region. In this paper, an improved PCNN algorithm was proposed to conduct the above process. Combined with the adaptive base threshold, and the simplified neural network parameters, a binary image of macular edema was produced. According to the principle of maximum image entropy, the optimal number of iterations was determined as 8, which was evaluated by its misclassification rate. Simulation showed that the proposed algorithm could extract the macular edema region rapidly and accurately, providing the basis for further OCT image analysis.

8.Ex vivo expansion of CD34(+) CD59(+) cells from bone marrow of paroxysmal nocturnal hemoglobinuria patients.

OBJECTIVETo study the separation, purification and ex vivo expansion of CD(34)(+) CD(59)(+) cells from patients with paroxysmal nocturnal hemoglobinuria (PNH), and explore the new treatment for the PNH patients.
METHODSCD(34)(+) CD(59)(+) cells were selected from the bone marrow mononuclear cells of PNH patients by means of immunomagnetic microbead-flow cytometry two step sorting method, followed by ex vivo expansion of the cells with combination of hematopoietic factors for two weeks.
RESULTSThe best combination for the ex vivo expansion was SCF + IL-3 + IL-6 + FL + Tpo + Epo, and the seventh day was the most suitable time for the best harvest when the CD(34)(+) CD(59)(+) cells were 22.42 +/- 3.73 fold expanded. After ex vivo expansion, the cells remained CD(59) positive and potent capacity of colony formation, but their potentialities to multilineage differentiation were decreased.
CONCLUSIONThe present study shows that ex vivo expansion of CD(34)(+) CD(59)(+) cells from PNH patients might promise the possibility of performing ABMT or APBSCT clinincally for the patients.

9.Proliferative capacity of the isolated single CD(34)(+) glycosylphosphatidylinesitol-anchored (GPI) protein negative and positive hematopoietic cells in paroxysmal nocturnal hemoglobinuria.

OBJECTIVESTo investigate the stroma-independent growth ability, multilineage differentiation and expansion of the single hematopoietic stem/progenitor cell from patients with paroxysmal nocturnal hematoglobinuria (PNH).
METHODThe CD(34)(+) CD(59)(+) and CD(34)(+) CD(59)(-) cells from PNH patients and CD(34)(+) CD(59)(+) cells from normal volunteers were sorted as each single cell into a well of 96 well culture plates containing culture medium supplemented with SCF, IL-3, Epo, GM-CSF, G-CSF, IL-6, Tpo and Flt-3 ligand.
RESULTS(1) Single PNH CD(34)(+) CD(59)(-) cell had a higher capacities for plating efficiency, colony (>/= 50 cells) formation and cell expansion than that of the PNH CD(34)(+) CD(59)(+) cells (P < 0.05); (2) Both the single CD(34)(+) CD(59)(-) cells from PNH patients and the single CD(34)(+) CD(59)(+) cells from normal controls had similar capacities for cell plating efficiency and colony and large colony formation. The PNH CD(34)(+) CD(59)(-) cells had a lower average cell production and cell expansion capacity. (3) The single CD(34)(+) CD(59)(+) cells from both PNH patients and normal controls showed the same capacities for cell plating efficiency and colony formation. The PNH CD(34)(+) CD(59)(+) cells exhibited much lower capacity for large colony formation, average cell production and total cell expansion. (4) A diminished secondary colony formation ability was also observed in the PNH CD(34)(+) CD(59)(+) and CD(34)(-) CD(59)(-) clones.
CONCLUSIONThe single PNH CD(34)(+) CD(59)(-) cells had growth advantage over the single PNH CD(34)(+) CD(59)(+) cells to some extent, but they both had impaired growth abilities as compared with CD(34)(+) cells from normal volunteers.

10.Protective role of γδ T cells in concanavalin A-induced liver injury.

OBJECTIVETo investigate the role played by γδ T cells in acute liver injury using the concanavalin A (ConA)-induced liver injury mouse model.
METHODSAcute liver injury was induced by intravenous injection of 10 mug/g of ConA into male C57BL/6J mice with wild-type or T cell receptor-γ knockout (TCR δ-/-) genetic backgrounds. Mice injected with PBS alone served as negative controls. The degree of liver damage was assessed by measuring serum levels of transaminase and cytokines at post-injection hours 3, 6, 12, 24, 48, and 72. The percentage of γδ T cells and proportions of different subsets in liver lymphocytes were measured by flow cytometry.
RESULTSThe TCR δ-/- mice showed significantly higher levels of the inflammatory cytokines IFN-γ, TNFα and IL-4 than the wild-type mice at post-injection hour 3. The percentage of liver γδ T cells increased with increased injury degree, and the extent of increase was significantly higher in the TCR δ-/- mice than the wild-type mice (post-injection hour 6: 6302.61+/-592.06 vs. 1319.26+/-355.48, 12: 6569.44+/-1060.98 vs. 3415.53+/-343.90, 24: 6514.29+/-757.26 vs. 2062.73+/-365.67, 48: 1262.61+/-558.07 vs. 113.66+/-113.26, and 72: 226.54+/-98.20 vs. 42.35+/-21.51 U/L; all P less than 0.05). In addition, compared to the negative control mice, the ConA-induced mice showed a higher proportions of Vγ4 γδ T cells to total γδ T cells (17.78+/-2.95 vs. 25.26+/-2.43) and to total liver lymphocytes (0.47+/-0.07 vs. 0.66+/-0.05). Similarly, compared to the negative control mice, the ConA-induced mice showed a higher proportion of Vγ1 γδ T cells to total γδ T cells (38.37+/-6.10 vs. 50.19+/-5.52) but the proportion to total liver lymphocytes was not significantly different among the groups (0.76+/-0.18 vs. 0.78+/-0.25). Reinfusion of Vγ4 γδ T lymphocytes into TCR δ-/- mice led to lower serum ALT levels than reinfusion of Vγ1 γδ T lymphocytes (5054.10+/-1748.51 vs. 12333.56+/-663.535 U/L).
CONCLUSIONγδ T cells play a protective role in ConA-induced liver injury and this effect maybe mediated by the Vγ4 γδ T cell subset.
